Yellowfin tuna (Thunnus albacares) is a valuable recreational and commercial species in Grenada and the Caribbean region. Myxozoans, specifically the monotypic Kudoidae family within the Order Multivalvulida, are marine parasitic organisms that can negatively impact fish health and marketability. On 8 September 2023, muscle fillets from a yellowfin tuna caught off the coast of Grenada and containing numerous white nodules were submitted for diagnostic evaluation. Gross examination of the muscle showed firm, spherical to ovoid, white polysporous cysts approx. 0.8-1.3 mm in diameter and approx.

BACKGROUND: Despite population-level control efforts, human echinococcosis continues to pose a public health challenge in China. Targeting modifiable risk factors at the individual level could be another recipe, but the priority targets for intervention remain unclear. Our aim was to quantify the potential health gains of hypothetical interventions on modifiable risk factors for human echinococcosis, informing effective control strategies.
METHODS: We collected and aggregated data on the association between modifiable risk factors and human echinococcosis and national total echinococcosis (cy

Inhibition of agonist-induced M2 muscarinic receptor (M2R) activation by functional anti-M2R autoantibodies has been associated with cardiac parasympathetic dysfunction in patients with chronic Chagas disease (CD). This study explored the allosteric nature of that inhibitory effect by assessing the ability of serum IgG from patients with CD and dysautonomia (DCD IgG) to modulate the interaction between M2R and β-arrestins in HEK 293T cells using bioluminescence resonance energy transfer. DCD IgG alone did not stimulate arrestin-2 or arrestin-3 recruitment. When cells were preincubated with DCD

Chagas disease, caused by Trypanosoma cruzi, is a neglected parasitic infection. The very limited arsenal of anti-T. cruzi treatments calls for the development of new drugs. Recently, a library of 3-benzylmenadione derivatives was synthesized, with cruzidione being the most efficient and specific compound against the parasite. To decipher its mode of action, we used the yeast Saccharomyces cerevisiae as a model. Evidence pinpointed at the heme A synthase Cox15 as a primary target of cruzidione: (i) a mutation in Cox15 (i.e., S429F) renders the yeast cells highly sensitive to the drug, (ii) tre

The apicoplast organelle of the malaria parasite, Plasmodium falciparum, is essential for parasite replication, though its cell cycle regulation remains poorly understood. We developed a dynamic live-imaging platform with analytical capabilities to track subcellular structures throughout the parasite's 48-h intraerythrocytic life cycle. Our analysis revealed four distinct morphological stages in apicoplast development that correlate with nuclear replication. We identified a critical "Crown" morphology stage required for nucleus-apicoplast attachment, where the apicoplast stretches across multi

This review paper outlines the history of giardiasis caused by Giardia duodenalis. Discovered in 1681, G. duodenalis is probably the first protozoan parasite ever observed by a human. Despite the early discovery, the taxonomic status of the protozoan remained uncertain for a long time. The reason for this is that G. duodenalis is a species complex comprising several phylogenetically distinct genotypes termed assemblages. Initially, it was thought that G. duodenalis is a primitive eukaryote because it lacks many subcellular organelles. However, recent research has shown that the protozoan has d

Plasmodium falciparum malaria is fatal if left untreated. Treatment is hampered by drug-resistant variants of the malaria parasite, highlighting the need to explore unique pathways for the development of new drugs with different mechanisms of action. Kinases in the inositol phosphate signaling pathway (IPP), and its products play many important roles in energy metabolism and signal transduction, making them attractive drug targets. In this exploratory study we investigated the potential of P. falciparum IPP as a novel and attractive pathway for antimalarial drug discovery, employing a combined

Leishmaniasis is a zoonotic disease where parasites of the genus Leishmania are transmitted among hosts by sandfly vectors of the subfamily Phlebotominae. In Paraguay, studies on sandfly biodiversity are limited, even though the disease is present in its tegumentary and visceral forms. This pioneering ecoepidemiological study analyzes the diversity, abundance, and detection of Leishmania spp. DNA in sandflies from primary, secondary, and degraded forest habitats within the Mbaracayú Forest Biosphere Reserve. Over a year of sampling, 954 sandflies were captured, comprising 599 females and 355 m

Competition between insects and their endosymbiotic bacteria for environmentally limited nutrients can compromise the fitness of both organisms. Tsetse flies, the vectors of pathogenic African trypanosomes, harbor a species and population-specific consortium of vertically transmitted endosymbiotic bacteria that range on the functional spectrum from mutualistic to parasitic. Tsetse's indigenous microbiota can include a member of the genus Spiroplasma, and infection with this bacterium causes fecundity-reducing phenotypes in the fly that include a prolonged gonotrophic cycle and a reduction in t

Schistosomes, parasitic flatworms responsible for the neglected tropical disease schistosomiasis, are protected by a skin-like tegument, and tegument maintenance is controlled by a schistosome ortholog (p53-1) of the tumor suppressor TP53. To understand p53-1 function, we characterized a schistosome cyclin-dependent kinase inhibitor homolog (cki). Knockdown of cki resulted in hyperproliferation that, combined with p53-1 knockdown, yielded tumor-like growths, indicating that cki and p53-1 are tumor suppressors in Schistosoma mansoni. cki homologs are ubiquitous in parasitic flatworms but are ab

Fasciolosis is a foodborne anthropozoonotic disease caused by Fasciola spp. that affects multiple hosts worldwide. Genetic characterization studies have revealed considerable diversity within F. hepatica populations owing to human and animal migration and intermediate snail hosts. Molecular markers such as microsatellites and mitochondrial DNA sequences are useful tools for assessing parasite population dynamics and evolutionary history. In this study, 16 F. hepatica samples were analyzed using cytochrome c oxidase 1 (CO1) and nicotinamide dehydrogenase (NADH) genes. Parasites were collected f

BACKGROUND: Malaria remains a major public health concern, and sub-Saharan Africa accounts for a significant proportion of global cases. A key challenge in malaria control is the increasing resistance of malaria vectors to commonly used insecticides, particularly pyrethroids. This study assessed the susceptibility of Anopheles gambiae s.l. to permethrin and deltamethrin in Zuba, located in the Gwagwalada Area Council of Abuja, Nigeria.
METHODS: Mosquito larvae were collected in June 2024 and reared under controlled insectary conditions until adulthood. Adult mosquitoes were then tested using

The marker compounds sesquiterpene lactones, Dehydrocostus lactone, and Costunolide have been isolated from Aucklandia costus Falc. The α-methylene-γ-Lactone (αMγL) moiety is a highly reactive functional group in sesquiterpene lactones and is responsible for biological effects. However, the αMγL group is alkylated by -SH groups of enzymes as well as amino acids like cysteine, and glutathione that leads to alterations in biological systems. In the present work a series of derivatives have been synthesised by Michael addition reaction between substituted piperazines and isolated sesqueterpene la

Malaria is an infectious disease that imposes a significant global health burden. Increasing drug resistance creates an urgent demand for novel treatment options. We have previously synthesised a new class of heterospirocyclic compounds with novel chemical linkages, which have shown preliminary antimalarial activity. Compounds 25 and 26 display antimalarial activity within 24 h against a panel of drug-resistant strains of Plasmodium falciparum, the most virulent of human malaria parasites. Untargeted metabolomics analysis of P. falciparum-infected red blood cells revealed that the mechanism of
