Transcriptomic Profiling of mRNAs, circRNAs, and lncRNAs in Mouse Liver at the Early Stage of Echinococcus multilocularis Infection.

Alveolar echinococcosis (AE) is a significant zoonotic disease caused by the larval form of Echinococcus multilocularis (E. multilocularis), posing substantial health risks. However, the roles of circular RNAs (circRNAs), long non-coding RNAs (lncRNAs), and messenger RNAs (mRNAs) in the early stages of E. multilocularis infection, particularly regarding liver involvement, remain unclear. This study aimed to investigate the molecular mechanisms of AE infection in mice during the early stage of secondary infection. The expression profiles of mRNAs, circRNAs and lncRNAs in mouse livers were analy

Intestinal parasitic infections associated with nutritional status and inflammatory markers among young children in Huye district, Rwanda.

BACKGROUND: Intestinal parasitic infections (IPIs) are a public health issue affecting young children in low and middle income countries (LMICs). These factors may induce malnutrition, as well as systemic and/or intestinal inflammation, depending on the species, intensity of infection, and host response. This study aimed at determining the effect of intestinal parasites on nutritional status and inflammatory responses in pre- and school-aged children in rural areas of the southern province of Rwanda. METHODS: A cross-sectional study involving 127 children under 12 years of age was conducted a

Structural basis of double-stranded RNA recognition by the J2 monoclonal antibody.

Double-stranded (ds) RNAs are major structural components of the transcriptome, hallmarks of viral infection, and primary triggers of innate immune responses. The J2 monoclonal antibody is the gold-standard method to discover and map endogenous dsRNAs across subcellular locations and cell surfaces, detect exogenous RNAs in viral infection, and surveil mRNA prophylactics and therapeutics for inflammatory dsRNAs. To define its epitope, specificity, and mechanism, we determine a 2.85 Å co-crystal structure of J2 antigen-binding fragment (Fab) bound to dsRNA. J2 uses its heavy and light chains in

Asymptomatic Plasmodium infection and predictors among schoolchildren in Bahir Dar Zuria District, Northwest Ethiopia.

BACKGROUND: Asymptomatic Plasmodium infection is a protozoan disease transmitted by vectors, in which the parasite remains inside the host without causing signs and/or symptoms. It poses a significant challenge to malaria control and elimination efforts due to its role in sustaining silent transmission within communities. Despite ongoing progress toward the 2030 malaria elimination targets, the disease remains a significant public health issue in Ethiopia. Schoolchildren in endemic areas are common carriers of the parasite and deserve special attention, as they represent a highly vulnerable po

Seasonal dynamics and species diversity of Anopheles mosquitoes in malaria endemic districts of Southern Odisha India.

Mosquitoes of the Anopheles, Aedes, and Culex genera are responsible for transmitting major vector-borne diseases. Malaria remains a significant public health concern in Odisha, primarily due to the state's conducive environment for Anopheles mosquito breeding. This study, conducted between March 2021 and February 2023 across 11 traditionally hyper-endemic districts in southern Odisha, aimed to assess seasonal variations in Anopheles diversity, composition, and abundance. A total of 10,807 Anopheles mosquito's species were collected manually indoors (house dwellings and cattle sheds) and outdo

Effect of inoculation dose on infection kinetics and immune responses to Giardia.

Giardia duodenalis is a globally prevalent enteric parasite capable of establishing both acute and chronic infections. While infection dose is known to influence early infection dynamics, its effect on immune activation and chronicity remains poorly defined. In this study, we investigated how varying inoculum sizes affect infection kinetics and immune development in both immunocompetent (C57BL/6J) and T cell-deficient (TKO) mice. High infection doses induced earlier cyst shedding and accelerated interleukin-17A (IL-17A) responses, while lower infection doses resulted in delayed immune activati

Practicability of a malaria rapid diagnostic self-test among artisanal gold miners in French Guiana: a cross-sectional study.

BACKGROUND: In French Guiana (FG), artisanal and small-scale gold mining (ASGM) workers represent mobile, hard-to-reach population with high malaria burden. Limited access to rapid diagnostic test (RDT) delays treatment, favours inappropriate use of artemisinin-based combination therapy (ACT), and increases transmission and resistance risk. The Malakit intervention enables ASGM workers to self-diagnose and self-treat using RDTs and ACT in a free kit distributed by community health workers (CHWs) after a dedicated training session. This cross-sectional study assessed the praticability of the Bi

Computational evaluation of indole alkaloids as inhibitors of 3-hydroxykynurenine transaminase in malaria vector control.

The enzyme 3-hydroxykynurenine transaminase (3-HKT) is critical for the production of xanthurenic acid, a key metabolite required for Plasmodium falciparum gametogenesis in Anopheles mosquitoes. Recognizing the importance of disrupting this pathway, the study explores the potential of indole alkaloids as inhibitors of 3-HKT, leveraging their natural insecticidal properties. Computational methods, including virtual screening, molecular dynamics, and MMPBSA analyses, were applied to evaluate the binding affinity and stability of indole alkaloids. Virtual screening results revealed spiroindolone

Bayesian assessment of three serological tests for the screening of Trypanosoma cruzi in hunting dogs of Virginia and West Virginia, USA.

Trypanosoma cruzi, the causative agent of Chagas disease (CD), is a vector-borne disease that affects several mammalian species, including humans. An endemic cycle in wild animals has been described, where dogs with an outdoor lifestyle are at higher risk. CD diagnosis is challenging, requiring multiple tests. Although there are several serologic tests available, they were originally developed for human diagnosis. Currently, there is limited knowledge about their performance in other species. The objective of this research was to estimate the performance of three serologic assays for CD screen

Inhibiting catalytic activity of Plasmodium falciparum aspartate protease plasmepsin V: A biochemical approach to malaria intervention.

Plasmepsin V, an aspartate protease, is crucial for the survival of the malarial parasite Plasmodium falciparum, making it a promising target for antimalarial drug development. In this study, the quinoline-based drugs amodiaquine (AQ) and clioquinol (CQ) are repurposed to inhibit the catalytic activity of PfPlmV. Fluorescence quenching assays suggest that both AQ and CQ exhibit moderate binding affinity for PfPlmV. Temperature-dependent thermodynamic analyses indicate the involvement of both H-bonding/electrostatic interactions and hydrophobic interactions in the binding of AQ/CQ with PfPlmV.

Acceptability and preferences for dual-active ingredient long-lasting insecticidal nets in rural Tanzania: a mixed-methods study.

BACKGROUND: The World Health Organization (WHO) recommends dual-active ingredient long-lasting insecticidal nets (dual-AI LLINs) for protection against malaria in areas with insecticide resistance. The effectiveness of LLINs, however, depends on user compliance, influenced by community perceptions of malaria, prevention methods, and the acceptability of LLINs. Understanding these factors is essential for the success of large-scale implementation. METHODS: This study was part of the two cluster-randomized controlled trials (RCTs) evaluating the efficacy of dual-AI LLINs on malaria indicators i

Harmless parasites? Infections with Hemolivia mauritanica (Apicomplexa: Adeleorina: Karyolysidae) and Haemocystidium spp. (Apicomplexa: Haemosporida: Haemoproteidae) have a negligible impact on white cell counts in tortoise hosts.

The pathogenicity of haemogregarines and their effects on the health status of ectothermic hosts remain largely unexplored. In this study, we examined the impact of Hemolivia mauritanica (Sergent et Sergent, 1904) infection on the differential leukocyte count (DLC) as a measurable indicator of health in tortoise hosts. A total of 206 blood smears were analysed, including 181 from spur-thighed tortoises (Testudo graeca Linnaeus) and 25 from marginated tortoises (Testudo marginata Schoepff). Light microscopy was used to identify infected individuals, determine DLC, and quantify parasitaemia leve
