Differential antioxidant enzyme profiles reveal early molecular signatures of virulence in Trypanosoma cruzi DTU-TcI and DTU-TcVI strains.

BACKGROUND: Trypanosoma cruzi, the causative agent of Chagas disease, exhibits significant genetic and phenotypic diversity that influences clinical outcomes. Antioxidant enzymes are crucial for parasite survival under host-induced oxidative stress, yet their role as determinants of virulence remains underexplored. OBJECTIVE: This study aimed to characterize and compare early antioxidant responses between the virulent T. cruzi CL Brener clone and a non-virulent strain isolated from Arequipa, Peru, to test the hypothesis that elevated antioxidant enzyme expression is associated with parasite v

Macroscopic and Microscopic Aspects of Echinococcus multilocularis and Echinococcus granulosus in Humans: A Guideline for Pathologists.

BACKGROUND: Echinococcosis is a parasitic disease, caused by the larval stage of the genus Echinococcus. Echinococcus multilocularis and Echinococcus granulosus cause alveolar echinococcosis (AE) and cystic echinococcosis (CE) in humans. Pathologists may be confronted with the larval stage of both species. Diagnosis may be difficult due to the low incidence in daily routine. However, right diagnosis of the pathologist is still the gold standard and crucial for further treatment of the patient. SUMMARY: Macroscopically, specimens of AE show multiple small cysts and a bread-like appearance. Ins

Functional and phenotypic analysis of CD4 T cell dynamics in peripheral blood of human visceral leishmaniasis patients confers increased frequencies of CD25 expressing regulatory T cells that contribute to disease pathogenesis.

INTRODUCTION: Regulatory T cells (Tregs) have been reported to control immune responses in microbial infections. However, their possible role in visceral leishmaniasis (VL) has not been well defined. To address this, we carried out extensive studies to investigate the frequency, phenotype and functions of kala-azar patients' peripheral blood Tregs pre and post treatment. METHODS: Fresh blood and peripheral blood mononuclear cells (PBMCs) were used to delineate the frequency and phenotype in VL through flow cytometry. Further for functional characterization, PBMCs of VL patients were depleted

Pentatrichomonas hominis Infection Induces Chronic Intestinal Inflammation in Immunocompetent Mice.

Pentatrichomonas hominis (P. hominis) has traditionally been regarded as a commensal or opportunistic inhabitant of the intestine. However, a growing number of studies have identified this protozoan as the sole pathogen in cases of intestinal disorders, including diarrhea, in both humans and animals, suggesting it may be a neglected zoonotic pathogen. To investigate the pathogenicity of P. hominis, we here systematically evaluated its effects on healthy immunocompetent BALB/c mice. Our findings revealed that infection with P. hominis induced injury in the large intestine, marked by inflammator

Emerging Trichomonad Infections in Companion Animals: Rapid Visual Detection of Pentatrichomonas hominis and Tritrichomonas foetus Using an RPA-CRISPR/Cas12a Assay.

Pentatrichomonas hominis (P. hominis) and Tritrichomonas foetus (T. foetus) are prevalent intestinal protozoa. P. hominis is associated with chronic diarrhea in humans and animals, whereas T. foetus causes gastrointestinal disease in companion animals and reproductive-tract infection in cattle. Rapid and accurate identification of these infections at the point-of-care (POC) is crucial for the diagnosis and effective management of zoonotic diseases. In this study, we developed two novel recombinase polymerase amplification (RPA) assays coupled with CRISPR/Cas12a detection. The dual-species assa

Sacubitril-Valsartan vs Enalapril in Heart Failure Due to Chagas Disease: Primary Results of ANSWER-HF Randomized Trial.

BACKGROUND: Chronic Chagas cardiomyopathy (CCC) represents a common cause of nonischemic heart failure with reduced ejection fraction (HFrEF) in Latin America, yet evidence from randomized trials remains scarce. Sacubitril-valsartan has shown benefits in HFrEF of other etiologies, but less is known about its effects on CCC. OBJECTIVES: The authors sought to compare the effects of sacubitril-valsartan vs enalapril on cardiac remodeling, functional outcomes, biomarkers, and safety in patients with CCC and HFrEF. METHODS: ANSWER-HF was a randomized, double-blind, controlled trial in Brazil. A t

IL-27 limits HSPC differentiation during infection and protects from stem cell exhaustion.

Many inflammatory stimuli can induce progenitor cells in the bone marrow to produce increased numbers of myeloid cells as part of the process of emergency myelopoiesis. These events are associated with trained immunity and have long-term impacts on hematopoietic stem and progenitor cell (HSPC) development but can also compromise their function. While many cytokines support emergency myelopoiesis, less is known about the mechanisms that temper these events. When mice that lack the cytokine IL-27 were infected with Toxoplasma gondii, there was enhanced generation of monocyte progenitors and incr

From treatment to prevention: reimagining schistosomiasis control through WASH and environmental management.

Schistosomiasis remains a persistent global health challenge, primarily controlled through Mass Drug Administration (MDA) of praziquantel. While MDA has reduced prevalence and morbidity in endemic regions like China and Sierra Leone, its limitations, including rapid reinfection, logistical barriers to coverage, emerging drug resistance, and failure to interrupt transmission, highlight the inadequacy of a purely pharmacological approach. This review advocates for a paradigm shift from treatment-focused to prevention-oriented strategies, integrating Water, Sanitation, and Hygiene (WASH) interven

Intestinal immunopathological evaluation of mice reinfected with Trypanosoma cruzi Y strain.

BACKGROUND: Chagas disease, caused by the protozoan Trypanosoma cruzi, is influenced by both host-related factors and parasite characteristics, including reinfection and strain variability. While the heart is the primary focus of reinfection studies, the effects on the intestinal form of the disease remain poorly understood. METHODOLOGY/PRINCIPAL FINDINGS: In this study, C57BL/6 male mice were infected and subsequently reinfected with the T. cruzi Y strain to assess the immunopathological consequences in the intestinal tract. We analyzed parasitemia levels, systemic cytokines, anti-T. cruzi i

Detection of Echinococcus granulosus sensu lato microRNAs in cystic echinococcosis patients: An exploratory study using quantitative PCR and digital PCR.

BACKGROUND: Cystic echinococcosis (CE) is a zoonosis caused by the larval stage of Echinococcus granulosus sensu lato (s.l.). The adult parasite typically resides in the intestine of canids, while the larval stage, a fluid-filled cyst, primarily resides in the liver and lungs of ungulates and humans. The diagnosis of abdominal CE in humans is mainly based on ultrasound (US) complemented by serology, but both techniques present limitations. Therefore, new diagnostic methods are needed. MicroRNAs (miRNAs) are potential suitable biomarkers for parasitic diseases, as well as for the diagnosis and

Detection and response to Haemaphysalis longicornis and Theileria orientalis Ikeda on a cow-calf farm in Tennessee (USA).

Bovine theileriosis is a vector-borne disease poised to become one of the most economically significant diseases of cattle in the United States. The hemoparasite Theileria orientalis Ikeda genotype causes bovine theileriosis (Piroplasmida: Theileriidae), which is primarily transmitted by Haemaphysalis longicornis Neumann (Acari: Ixodidae) (longhorned tick, Asian longhorned tick). Since its initial detection in New Jersey in 2017, H. longicornis has rapidly spread to 24 states and Washington, DC, as of July 2025. Given the exotic and invasive nature of this vector and its unique biological char

Hydatid disease of the distal humerus treated with wide resection and reconstruction with endoprosthesis.

A man in his 40s from a rural area presented with progressive pain and functional limitation of the right arm without trauma. Imaging revealed an aggressive lytic lesion with pathological fracture of the distal humerus. Staging studies excluded other lesions. Percutaneous biopsy confirmed osseous hydatid disease. Preoperative albendazole was administered, followed by wide en bloc resection of a 15 cm humeral segment with preservation of neurovascular structures. Reconstruction was achieved using a modular distal humerus endoprosthesis. Postoperative albendazole continued for 6 weeks. At 12 mon

A congress report of the new Emerging Pathogens and Parasitology Subgroup within the ISBT Working Party on Transfusion-Transmitted Infectious Diseases.

In 2025, the Parasitology Subgroup of the International Society of Blood Transfusion (ISBT) Transfusion-Transmitted Infectious Diseases (TTID) Working Party (WP) transitioned into the Emerging Pathogens and Parasitology (EPP) Subgroup (referred to here as the EPP). This followed recognition that the parasitology subgroup's relevance was limited in scope given the small number of transfusion-transmissible parasites that still lacked effective mitigation. The EPP was proposed to address themes that are not adequately covered by existent subgroups of the TTID WP. In addition to maintaining a focu

Prevalence of porcine toxoplasmosis using ERP-iELISA in Hubei Province.

Toxoplasma gondii (T. gondii) is an intracellular parasite that can infect almost all mammalian species, including humans and important economic animals such as pigs and sheep, thereby causing serious public health security issues. In pigs, such infections are common and economically significant, yet distinguishing between infection routes remains challenging. The current commonly used detection methods mainly aim to quickly detect positive cases of toxoplasmosis but cannot distinguish between different modes of infection. To distinguish whether cysts or oocysts are the source of T. gondii inf

Investigating genetic profiles of cases of Schistosoma spp. imported into Europe: a cohort from the European Society of Clinical Microbiology and Infectious Diseases Study Group for Clinical Parasitology.

BACKGROUND: The potential of schistosomiasis to spread across borders, coupled with the considerable delay by which infected travellers and migrants are diagnosed in Europe, calls for better surveillance of the distribution of this disease. This study explored the geographical origin and genetic profiles of Schistosoma infections imported into Europe and diagnosed in a network of 11 European centres specialized in traveller and migrant health. METHODS: Genetic profiles were obtained from DNA extracted from concentrated Schistosoma eggs or Schistosoma-positive samples (faeces, urine, biopsy) c
