The dynamic of Trypanosoma cruzi transmissibility in field-caught Mepraia spinolai (Hemiptera: Reduviidae) of contrasting seasons: A developmental stage-dependent study.

A competent vector can harbor a pathogen and infect a high number of hosts. Scarce information is available on how triatomine vectors accomplish this task in nature, particularly without knowing the number of parasites they excrete. We study the dynamics of Trypanosoma cruzi transmissibility in the triatomine Mepraia spinolai collected in summer (December 2018) and winter (July 2019) of an austral endemic area. The insects received two feedings in the laboratory, the day after arrival and 45 days after the first feeding. Excreta samples were collected, DNA was extracted, and parasite loads wer

Cascades of effectiveness of next-generation insecticide-treated nets against malaria, from entomological trials to real-life conditions.

As insecticide resistance spreads in Africa, next-generation insecticide-treated nets (ITNs) are increasingly being deployed to protect vulnerable populations against malaria. While these nets provide greater entomological efficacy against resistant mosquitoes, their effectiveness against malaria transmission also depends on other factors, such as durability, access, usage, and activity patterns of hosts and vectors. Here, we quantify the impact of two next-generation ITNs, namely Interceptor®G2 (chlorfenapyr-pyrethroid) and Olyset® Plus (piperonyl butoxide-pyrethroid), in a cascade from entom

Serological and molecular detection of Toxoplasma gondii in naturally infected red foxes (Vulpes vulpes) from Victoria, Australia.

Toxoplasma gondii is a zoonotic parasite with a global distribution that can infect a wide range of warm-blooded hosts. This study investigated, for the first time, the seroprevalence and genetic variability of T. gondii in red foxes (Vulpes vulpes) from Victoria, Australia. Animals from both regional Victoria and the metropolitan Melbourne area were sourced from trappers and shooters involved in pest control. Sera were screened for anti-T. gondii IgG antibodies using a modified agglutination test, and tissue samples were tested using a qPCR assay targeting the 529 bp repeated element. qPCR po

Surgical Management of Pincer Nail Deformity: A Simplified Technique.

INTRODUCTION: Pincer nail deformity (PND) involves excessive transverse nail plate over-curvature which pinches up distal nail bed and causes pain. Conventional surgical options are invasive with high complication risks. We treated pincer nails with a simplified, reproducible technique. METHODS: Records of PND patients treated over 7 years with a simplified technique were retrospectively analyzed. After confirming the absence of any distal bony abnormality, lateral 1/5th-1/6th of the nail plate was avulsed on both sides, with chemical matricectomy of the lateral matrix horn. Thereafter, the d

Giardiasis in pediatric populations: A case series highlighting nutritional and growth impacts.

Giardiasis, a common cause of parasitic diarrhea, is caused by the protozoan parasite Giardia lamblia (Giardia intestinalis or Giardia duodenalis). This intestinal flagellate protozoan is prevalent among children worldwide and is a significant source of traveller's diarrhea. It is estimated that there are about 2.5 million cases of giardiasis-related diarrhea and nutritional deficits in children every year, with the majority occurring in developing countries. Since 2004, the World Health Organization has classified it as one of the neglected tropical diseases. Those most at risk for complicati

Assessing allergenic potential, exposure and risk of Anisakis spp.

Parasites in seafood are a recognised food safety concern worldwide. Anisakiasis is a human disease caused by ingestion of larvae of parasitic nematodes of the genus Anisakis , typically present in raw or undercooked seafood. It is considered a significant emerging foodborne disease and was recently ranked among the top 10 of 24 parasitic foodborne infections in Europe, with a rapidly increasing potential to cause illness. Many clinical cases are often misdiagnosed as other gastrointestinal disorders, leading to misinterpretation of data and likely a substantial underestimation of their true p

Nematocidal activity of wasabi paste and allyl Isothiocyanate against Anisakis third-stage larvae and their stress response characterized by heat shock protein expression.

Anisakis pegreffii (Nematoda: Anisakidae) is a marine parasitic nematode responsible for anisakiasis, a zoonotic disease acquired through the consumption of raw or undercooked fish. With the rising popularity of dishes such as sushi and sashimi, the risk of infection has grown, prompting the need for effective food-safety measures. This study investigates the nematocidal effects of commercial wasabi substitute paste (WSP) and its bioactive compound, allyl isothiocyanate (AITC), on third-stage larvae (L3) of A. pegreffii, and explores their associated stress-response mechanisms. WSP at 1 g/mL a

In silico detection and characterization of novel Picornavirales mined from publicly available insect RNA-seq data sets.

Picornavirales is a hyperdiverse order of viruses that infect assorted hosts, including arthropods. Members of this group contain a positive-sense, single-stranded RNA genome with a polyadenylated tail. As such, they are often sequenced as "bycatch" in RNA-seq experiments targeting other organisms. We developed a simple-to-follow bioinformatic pipeline to assemble and annotate genomes of the order Picornavirales using publicly available insect metatranscriptome data sets. Using our pipeline, 40 novel viruses were discovered, including putative new members of the families Dicistroviridae, Iflav

Harnessing controlled human infection models to accelerate vaccine development for neglected tropical diseases: Lessons from leishmaniasis.

BACKGROUND: Controlled Human Infection Models (CHIMs) offer a powerful approach to expedite vaccine development by enabling early evaluation of vaccine candidate efficacy and immune responses. Their role is increasingly relevant for neglected tropical diseases (NTDs), where traditional trial approaches may be slow, costly, or unfeasible. This review explores the scientific, ethical and translational dimensions of CHIMs, with a focus on the recently developed Leishmania major CHIM for cutaneous leishmaniasis (CL). METHODS: A narrative synthesis of peer-reviewed literature and regulatory guidan

First record of chigger mites (Acari: Trombiculidae) parasitizing birds in the Ogasawara Islands.

Larvae of trombiculid mites (Acari: Trombiculidae), known as chiggers, are ectoparasites that parasitize various animals and transmit pathogens such as Orientia tsutsugamushi. We report the first record of chigger mites in the Ogasawara Islands, collected from Bonin Greenfinch Chloris kittlitzi and Warbling White-eyes Zosterops japonicus. By morphological and molecular analysis, the chiggers were identified to the genus Toritrombicula which predominantly parasitize avian hosts. Although the mites could not be identified to the species level, this finding provides new insight into the distribut

Global Protein Interaction Network for Trypanosoma cruzi.

Trypanosoma cruzi, the causative agent of Chagas disease, poses a significant health challenge due to limited therapeutic options and an incomplete understanding of its biology. Approximately half of the genome encodes hypothetical proteins with unknown functions, underscoring the need for systematic functional annotation. Protein-protein interactions (PPIs) underpin essential cellular processes, yet no large-scale PPI map has been developed for T. cruzi ─a critical gap that impedes both functional annotation of its proteome and drug discovery. This study presents the first comprehensive PPI n
