Experimental models of liver-stage malaria: Progress, gaps, and challenges.

For decades, achieving malaria eradication has proven difficult. Plasmodium parasites first multiply asymptomatically in the liver before causing cyclic erythrocytic infections and clinical symptoms. Unlike other species, Plasmodium vivax can remain dormant for months or years as hypnozoites within hepatocytes. When these latent parasites reactivate, they cause clinical episodes in most vivax malaria cases. Thus, targeting the liver stage is essential to prevent disease progression and relapse. Current therapies are effective but pose risks for individuals with glucose-6-phosphate dehydrogenas

Pilot study to assess the impact of feed-through insecticide on the sand fly density in an endemic area of zoonotic cutaneous leishmaniasis in Morocco.

BACKGROUND: Cutaneous leishmaniasis (CL) remains a significant public health concern in Morocco. Leishmania major, the principal cause of zoonotic CL (ZCL) in Morocco, is transmitted by the female sand fly Phlebotomus papatasi, with the great gerbil (Meriones shawi) serving as the primary reservoir host. Current control strategies predominantly rely on strychnine-treated bait targeting gerbils. Recently, insecticide-treated rodent bait has emerged as a promising alternative for ZCL control. This study evaluated the village-level impact of feed-through insecticide, involving two intervention vi

Transcriptomics analysis of in vivo Opisthorchis viverrini: Stage-specific gene expression and novel genes with stable expression in mammalian stages for opisthorchiasis intervention.

Opisthorchis viverrini, Southeast Asian liver fluke, is a parasitic flatworm that has been widely spread in Asia and is a major risk factor for bile duct cancer - cholangiocarcinoma (CCA). Over 12 million people are at risk of opisthorchiasis, and the associated CCA is known to have claimed around 20,000 deaths per year in Thailand alone, with the number projected to be underestimated elsewhere. There is an opportunity to develop more efficient drug targets and diagnostic biomarkers, and these are urgently needed. Despite previous studies on gene expression analysis and reference genome, the k

Global burden of lower respiratory infections and aetiologies, 1990-2023: a systematic analysis for the Global Burden of Disease Study 2023.

BACKGROUND: Lower respiratory infections (LRIs) remain the world's leading infectious cause of death. This analysis from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2023 provides global, regional, and national estimates of LRI incidence, mortality, and disability-adjusted life-years (DALYs), with attribution to 26 pathogens, including 11 newly modelled pathogens, across 204 countries and territories from 1990 to 2023. With new data and revised modelling techniques, these estimates serve as an update and expansion to GBD 2021. Through these estimates, we also aimed to

Histopathological changes, dynamics of macrophage polarization and deposition of type I and III collagen along the course of experimental hepatic toxocariasis.

Toxocariasis is a helminthic infection that predominantly affects the liver and induces significant pathological changes mediated by host immune response. Both M1 and M2 macrophage phenotypes exert opposing yet complementary functions during infection. This study aimed to characterize the sequential immunopathological and fibrogenic events in hepatic toxocariasis over 16 weeks post-infection (wpi) in a murine model. Liver samples were collected from infected mice at different time points, and immunohistochemical (IHC) examination revealed the presence of both macrophage phenotypes up to 8 wpi

In vitro expression and seroreactivity of Toxoplasma gondii GRA1 protein encoded by unmodified and nucleoside-modified mRNA constructs.

Toxoplasmosis is an infectious parasitic disease caused by Toxoplasma gondii, with a global seroprevalence estimated to affect more than one-third of the human population. T. gondii can be a serious health concern for pregnant women and immunocompromised patients. Currently, there is no effective and safe vaccine for humans. However, mRNA technology is a promising platform to develop efficient and safe vaccines against toxoplasmosis. Therefore, we designed nucleoside-modified and unmodified mRNA vaccine transcripts expressing T. gondii recombinant GRA1 protein and analyzed their in vitro prote

Randomized field trial of a therapeutic vaccine against Trypanosoma cruzi natural infection in dogs and correlates for efficacy.

Chagas disease, caused by Trypanosoma cruzi, is a vector-borne parasitic disease, and dogs are a major domestic host of the parasite. We performed the first randomized field trial of an immunotherapeutic vaccine among client-owned dogs with natural infections and investigated immune correlates of efficacy. Thirty one dogs with T. cruzi infection received three doses of Tc24-C4 and TSA1-C4 recombinant proteins with MPLA (N = 16) or saline control (N = 15) and were followed for six months. Therapeutic vaccination decreased parasite burden, prevented/stopped cardiac alterations measured by electr

Dosing and safety of amphotericin B deoxycholate in paediatric American cutaneous leishmaniasis in Peru: a case series.

BACKGROUND: Cutaneous leishmaniasis (CL) is a parasitic disease endemic to the Americas, with a high prevalence in Peru. In resource-limited settings where liposomal amphotericin B is often unavailable, amphotericin B deoxycholate (AmB-d) remains a second-line option. However, data on its safety and effectiveness in children are scarce. This study describes the clinical experience of 20 paediatric patients with CL treated with AmB-d in a referral centre in Peru. METHODS: We conducted a retrospective descriptive study of patients ≤14 years with confirmed localised CL who had failed parental so

A "Familiar Foe Revisited": examining the relationship between endemic Burkitt's lymphoma and changing malaria admissions in the coastal region of Kenya.

BACKGROUND: Burkitt’s lymphoma (BL) is a type of non-Hodgkin lymphoma that may account for more than 40% of childhood malignancies in tropical Africa. The endemic version is common in equatorial Africa, where a BL belt has been mapped. The role of P. falciparum malaria infection in BL has been postulated but not substantiated. The decrease in P. falciparum malaria infection offers an opportunity to examine this association. METHODS: In this study, we utilized data collected over three decades (1990–2020) and examined the trends in annual admission incidence rates of Burkitt’s lymphoma among pe

Interactions between Plasmodium falciparum-infected red blood cells and their extracellular vesicles with megakaryocytes: implications for platelet-like particle formation.

BACKGROUND: Thrombocytopenia commonly accompanies Plasmodium falciparum (P. falciparum) malaria, yet the role of impaired megakaryopoiesis and platelet production remains unclear. This study examined how P. falciparum-infected red blood cells (pRBCs) and their extracellular vesicles (EVs) modulate megakaryocytic differentiation and platelet-like particle (PLP) formation. METHODS: MEG-01 cell line, a human megakaryoblastic leukaemia model, was differentiated with phorbol 12-myristate 13-acetate (PMA) in the presence or absence of fetal bovine serum (FBS) and co-cultured with pRBCs, normal RBCs

A retail audit of mosquito control products in Bidibidi refugee settlement, northwestern Uganda.

BACKGROUND: Uganda remains one of the highest malaria burden countries globally, with refugee populations disproportionately affected. While government-led initiatives, such as free distribution of insecticide-treatesd nets (ITNs) and indoor residual spraying, are key to the vector control response, limited data exist on the role of retail markets in providing complementary mosquito control products (MCPs) in refugee settings. METHODS: As part of the Advancing Evidence for the Global Implementation of Spatial Repellents project, we conducted a retail audit across 61 refugee and host community

Asymptomatic parasitaemia in a malaria pre-elimination setting in the Putumayo area of the Ecuadorian Amazon basin.

BACKGROUND: Ecuador is approaching malaria elimination, but its Amazonian Putumayo canton borders regions in Colombia and Peru with high transmission intensity. This 'frontier' setting represents a critical vulnerability where asymptomatic infections may sustain transmission, undetected by routine surveillance. METHODS: In a repeated cross-sectional study during the 2023 rainy season, we surveyed 293 asymptomatic individuals in four riverine communities in Putumayo. We used a tiered diagnostic approach: rapid diagnostic tests (RDTs), expert microscopy, and real-time qPCR targeting the 18S rRN

Fine-scale spatial mapping of urban malaria prevalence for microstratification in an urban area of Ghana.

BACKGROUND: Malaria is a focal disease and more localized in low endemic areas. The disease is increasingly becoming a concern in urban areas in most sub-Saharan African countries. The growing threats of Anopheles stephensi and insecticide resistance magnify this concern and hamper elimination efforts. It is, therefore, imperative to identify areas, within urban settings, of high-risk of malaria to help better target interventions. METHODS: In this study, a set of environmental, climatic, and urban covariates were combined with observed data from a malaria prevalence study in Ghana and geospa

African leaders and regional institutions need to take the bull by the horns: a perspective on the impact of the 2025 funding cuts to malaria programmes.

Despite global malaria programmes already operating within resource constraints, 2025 saw a significant decrease in funding following the US government's termination of most of its global health programmes, as well as the decline in development aid spending by the UK, France, Germany, Canada, Switzerland, and other countries. The disruption of funding was sudden, with most African countries lacking adequate contingency plans. This, despite most of the malaria burden being in Africa (94% of 263 million cases in 2023), accounting for a reduction in gross domestic product of up to 1.3% annually,

Ecology and molecular validation of species of vector snails of fascioliosis and schistosomiosis in Bangladesh.

Freshwater snails are important intermediate hosts for several parasitic diseases, including fascioliosis and schistosomiosis, with significant impacts on human and animal health. In Bangladesh, vector snails have been identified only by morphology. Here, we validate the species of freshwater snails acting as intermediate hosts for Fasciola and Schistosoma spp. using molecular and bioinformatics tools. Following morphology and morphometrics, we identified 9 species of snails: Lymnaea auricularia, Lymnaea luteola, Indoplanorbis exustus, Physa acuta, Viviparus bengalensis, Brotia spp., Thiara sp

Efficacy of oral afoxolaner and a combination of afoxolaner, moxidectin, and pyrantel against Amblyomma maculatum in dogs.

BACKGROUND: Amblyomma maculatum is a tick with a broad host range that is undergoing an expansion of its range within the USA. When feeding the predilection sites on the host are the head and ears and due to the long mouthparts, it can cause significant lesions that can lead to infection. It has also been implicated as a vector of Hepatozoon americanum, the causative agent of canine hepatozoonosis and a spotted fever group Rickettsia, Rickettsia parkeri. METHODS: Two randomized, blinded, negative controlled studies were conducted to determine whether treatment with afoxolaner (NexGard®, Boehr

Genetic variation of cutaneous leishmaniasis causative species in Ethiopia.

In Ethiopia, three Leishmania species are linked to Cutaneous Leishmaniasis (CL): Leishmania aethiopica, L. tropica, and L. donovani. Among these, L. aethiopica is the most commonly reported cause. The clinical presentations of CL are broadly classified into localized (LCL), mucocutaneous (MCL), and diffuse cutaneous CL (DCL) forms. There is limited data on the genetics of these pathogens, despite the different clinical forms and estimated 50 thousand new cases of CL per year. In this research, we sequenced the ITS1 region from 72 clinical specimens. In the in-silico PCR-RFLP, 67 were identifi

Verification and comparison of pig, mouse, and human genome similarities: use of manual assembly and analyses.

BACKGROUND: Recently there have been numerous attempts to improve the genome of the pig. Despite these efforts, there is a substantial amount of work remaining to obtain a "finished version" of the genome; analysis of incomplete versions can lead to incorrect biological interpretations. To that end, we manually assembled and annotated a non-redundant, 16,146 RNA and 15,613 pig protein sequence libraries. We used it to assess the assembly and annotation status of the 3 latest builds of the genome and to the mouse and human genomes. RESULTS: Our analysis of 6,135 protein-coding genes reveals th

Pathogenic piscine intestinal coccidia infection alters gut microbiome in juvenile Asian seabass (Lates calcarifer).

BACKGROUND: Piscine intestinal coccidiosis has been associated with increased susceptibility to secondary bacterial infections, potentially through disruption of the gut microbiota. While many gut bacteria are commensal, infection-related alterations in microbial composition may favor the proliferation of opportunistic taxa. However, the specific effects of intestinal coccidial infections on the gut microbiome of fish remain insufficiently described. RESULTS: This study examined the influence of intestinal coccidial infection on gut microbiota composition and the relative abundance of potentia
