Molecular surveillance of antimalarial drug resistance genes in Nigeria: a systematic review and roadmap to malaria elimination.

BACKGROUND: Malaria remains a significant public health challenge in Nigeria, accounting for a substantial proportion of global malaria cases and deaths. Although artemisinin-based combination therapies (ACTs) are the recommended first-line treatments, resistance to antimalarial drugs continues to threaten malaria control efforts. This systematic review provides an overview of the molecular surveillance of antimalarial drug resistance in Nigeria, aiming to identify prevalent resistance markers and inform future research and policy directions. METHODS: A systematic search was conducted using d

Hemozoin induces neuronal injury primarily characterized by axon rupture and mitochondrial damage in experimental cerebral malaria.

BACKGROUND: Cerebral malaria (CM) is the most serious and fatal neurological complication of Plasmodium falciparum infection, which can cause death or long-term neurological sequelae. Neuronal injury is a primary cause of these sequelae in patients with CM; however, the underlying mechanisms remain incompletely elucidated. Hemozoin (Hz), the metabolic byproduct of hemoglobin digested by Plasmodium parasites, is closely associated with the severity of CM. However, it is not clear whether Hz is a direct contributor to neuronal injury. METHODS: C57BL/6 J mice were infected with the Plasmodium be

Mechanisms of parasite-mediated disruption of brain vessels.

The brain vasculature is a critical barrier to maintain central nervous system (CNS) homeostasis. Parasitic infections can profoundly disrupt the brain vasculature, with consequences ranging from subtle neurological alterations to severe, life-threatening pathologies. In this review, we explore the diverse mechanisms by which endoparasites interact with, modulate and breach CNS blood and lymphatic vessels. We highlight how these pathogens manipulate endothelial function, alter barrier permeability and exploit vascular surface molecules to access or influence the brain. These interactions often

Epidemiology and spatiotemporal patterns of human echinococcosis in China, 2006-2020.

China accounts for over 90% of global alveolar echinococcosis (AE) cases caused primarily by Echinococcus multilocularis. If left untreated, AE can have a 10-year mortality rate of 94%. Understanding its epidemiological patterns is essential for targeted control strategies. Surveillance data from 2006 to 2020 were obtained from the Public Health Scientific Data Center, and spatial and temporal trends were analyzed using spatial autocorrelation, hot spot analysis, and centroid migration techniques. A total of 51,403 echinococcosis cases were reported from 2006 to 2020, with an average annual in

Cerebral Malaria and Multi-Organ Dysfunction Caused by Plasmodium vivax in an Adult: A case from Oman with a literature review.

Cerebral malaria is a life-threatening complication most commonly associated with Plasmodium falciparum. To the best of the authors' knowledge, this is the first documented case in Oman of a previously healthy 38-year-old Pakistani male who presented to the Emergency Department of a tertiary care hospital in 2025 with cerebral malaria and multi-organ dysfunction syndrome (MODS) caused by Plasmodium vivax, confirmed by peripheral blood smear and polymerase chain reaction (PCR). The patient was treated with intravenous (IV) artesunate and supportive intensive care, resulting in full neurological

[Taeniasis and appendicitis: a rare parasitic connection].

Taeniasis, primarily caused by Taenia solium or Taenia saginata, is the most common zoonotic infection affecting the gastrointestinal tract. Although appendicitis related to this parasitic infection is rare, it represents an important differential diagnosis in patients presenting with abdominal pain. This case report highlights a unique case of appendicitis associated with Taenia infestation, emphasizing the need for clinical awareness of potential parasitic origins in appendicitis. A 35-year-old Ethiopian female, non-diabetic and non-hypertensive, presented with a one-day history of moderate

Long-term trends in parasite diversity and infection levels: approaches and patterns.

Parasites exist in every ecosystem, affecting nearly all organisms and playing a complex role in human societies. On the one hand, they contribute substantially to biodiversity and support ecosystem stability by performing essential ecological functions. On the other, they can impose health burdens on their hosts, causing diseases in both animals and humans. Despite their significance, our understanding of how parasitic organisms are affected by human-driven environmental change remains poor. In other well-studied groups such as free-living birds, mammals and insects, long-term ecological data

Highlights of Medicinal Chemistry Optimization Strategies in Peptidomimetic Antimalarial Drug Discovery.

Progress in the fight against malaria appears to have stalled, as reflected by the nearly unchanged mortality figures reported in 2022 and 2023-600,000 and 597,000 deaths, respectively. In addition to reports of resistance to artemisinin-combination therapies (ACTs) in South-East Asia, resistance to ACTs has now been confirmed in Africa. Therefore, new drugs are urgently needed. In this perspective, we summarize different medicinal chemistry strategies employed to address various liabilities in hit antimalarial peptidomimetics. These include an increase in Log P; macrocyclization; chemically r

Comparative analysis of radiological and serological methods for diagnosis of cystic echinococcosis.

Cystic echinococcosis (CE) diagnosis relies on clinical signs, serological tests, radiological findings and histopathology. Lack of pathognomonic features or clinician inexperience can make CE diagnosis challenging. In this study, we aimed to investigate the demographic, serological, and radiological characteristics of individuals with a preliminary diagnosis of CE. A total of 1,410 serum samples from 1,265 patients were analysed for the presence of IgG antibodies Anti-Echinococcus granulosus IgG using chemiluminescence immunoassay (CLIA) and indirect haemagglutination assay (IHA). Among these

The Deanes' malarias and the antimalarial drugs in 1939 and the 1940s.

Malaria was one of the main research themes of Leônidas de Mello Deane (1914-1993) and Maria José von Paumgartten Deane (1916-1995), a couple of parasitologists - known among their peers as "the Deanes" - who in the late 1930s and 1940s ventured into areas of the Amazon, where the disease was endemic, and of the northeastern Brazil, where an important epidemic outbreak ecloded. Despite their knowledge of the disease and the adoption of preventive measures, both contracted malaria, he the malignant tertian fever (Plasmodium falciparum) and she the benign form of the disease (Plasmodium vivax).

TPEDCC-PEG-based photodynamic therapy exhibits potent therapeutic efficacy against echinococcosis by inducing oxidative stress and mitochondria-mediated apoptosis.

Alveolar echinococcosis (AE) is a fatal zoonosis caused by the larval stage of Echinococcus multilocularis, but there are no fully effective treatments for this disease to date. The aim of this study was to investigate the potential therapeutic implication of photodynamic therapy (PDT) for E. multilocularis. We synthesized a novel nanophotosensitizer, TPEDCC-PEG, with aggregation-induced emission (AIE) properties and investigated its in vitro and in vivo PDT effects against the larval stage of E. multilocularis, as well as the anti-Echinococcus mechanism. TPEDCC-PEG exhibited excellent physico
