A national atlas of tsetse flies and African animal trypanosomosis in Ghana.

BACKGROUND: Tsetse-transmitted African animal trypanosomosis (AAT) remains a widespread and major constraint to sustainable development and food security in vast areas of sub-Saharan Africa. In Ghana, like in most endemic countries, data on the occurrence of the tsetse vector and the disease are available at the country level, but for a long time information has been scattered and poorly harmonized, thus hampering proper analysis and evidence-based decision-making. METHODS: The Tsetse and Trypanosomosis Control Unit (TTCU) in Ghana collated, harmonized, georeferenced and centralized in a sing

Isoalantolactone exhibits activity against Echinococcus multilocularis associated with mitochondrial perturbation and changes in the lesion microenvironment.

BACKGROUND: Alveolar echinococcosis (AE) is a severe parasitic disease caused by Echinococcus multilocularis. Current therapy relies mainly on benzimidazole drugs and is largely parasitostatic, highlighting the urgent need for new candidate compounds with improved efficacy. We hypothesized that isoalantolactone (ISO) exerts direct anti-Echinococcus multilocularis activity and evaluated whether ISO treatment was associated with changes in the lesion microenvironment and host immune responses during AE progression in vivo. METHODS: In vitro experiments were conducted using isolated Echinococcus

Paroxetine activity against Trypanosoma cruzi: in vitro and in vivo studies.

BACKGROUND: Chagas disease (CD) lacks effective therapy. However, the long time and high financial investments for the development of new drugs are limiting obstacles. The repositioning of drugs is an interesting approach for novel antiparasitic medicines. Previous results suggest that paroxetine (PXT) may represent a promising anti-Trypanosoma cruzi agent. OBJECTIVES: Our aim was to evaluate the activity and cellular targets of PXT on T. cruzi infection in vitro and in vivo. METHODS: PXT and benznidazole (BZ) were assayed against intracellular and bloodstream forms for different periods of

Control of M. tuberculosis infection requires CD28 to generate lung-homing T cells and CTLA-4 to limit immunopathology.

Mycobacterium tuberculosis (Mtb)-specific CD4 T cells differentiate into protective lung parenchyma-homing and non-protective intravascular subsets, but the signals that instruct these fates are poorly understood. Here we show that CD28 co-stimulation regulates host protection during Mtb infection. Cd28-/- mice are susceptible to Mtb infection, displaying higher bacterial loads and reduced survival compared to wild type mice. Cd28-/- Mtb-specific T cells expand normally but are skewed towards the lung intravascular phenotype. CD4 T cells primed in the absence of CD28 co-stimulation in B7 defic

Seroprevalence of Toxoplasma gondii infection, associated risk factors, and circulating IL-6 and VEGF-A levels in colorectal cancer patients: a case-control study from western Iran.

BACKGROUND: Colorectal cancer (CRC) development is influenced by inflammatory, angiogenic, and lifestyle-related factors. Latent Toxoplasma gondii infection has been implicated in modulating host immune and inflammatory responses, potentially affecting tumour biology. This study investigated the relationship between T. gondii seropositivity, risk factors, and circulating Vascular endothelial growth factor (VEGF-A) and Interleukin-6 (IL-6) levels with CRC risk in a population from Lorestan Province, western Iran. METHODS: This case-control investigation was conducted on 215 newly diagnosed pat

Association of Blastocystis subtype 3 with elevated IL-8-mediated inflammation in patients with irritable bowel syndrome.

BACKGROUND: The role of Blastocystis in the pathophysiology of irritable bowel syndrome (IBS) remains controversial, with emerging evidence suggesting subtype-specific differences in pathogenicity. Interleukin-8 (IL-8), a key pro-inflammatory chemokine, has been implicated in mucosal immune activation in both IBS and parasitic infections. OBJECTIVE: This study aimed to investigate the association between Blastocystis subtypes and clinical manifestations of IBS, with a particular focus IL-8 as part of the systemic pro-inflammatory immune response associated with functional GIT disorders and in

Repurposing Ionophores to Tackle Antimicrobial Resistance with a Focus on PBT2.

Antimicrobial resistance (AMR) remains a growing global health challenge. While resistant bacterial strains continue to emerge and spread, the development of new antibiotics has remained slow. This gap has increased interest in drug repurposing as a promising strategy to combat multidrug-resistant (MDR) pathogens. Ionophores are a structurally diverse group of natural and synthetic compounds that transport ions across biological membranes. Many ionophores were originally developed and widely used for the prevention and treatment of coccidiosis, a common protozoan parasitic intestinal disease i

Water, sanitation, and hygiene in soil-transmitted helminth control: A scoping review of evidence gaps across transmission pathways.

Soil-transmitted helminths (STH) are parasitic intestinal worms infecting humans through contact with, or ingestion of, soil contaminated with eggs shed by infected individuals. STHs include roundworms (Ascaris lumbricoides, Strongyloides stercoralis), whipworm (Trichuris trichiura) and hookworms (Ancylostoma duodenale, Necator americanus, Ancylostoma ceylanicum). Their transmission is influenced by water, sanitation and hygiene (WASH) services and practices. Unlike past reviews, this scoping review did not assess effect sizes or strength of associations. Instead, it aimed to: map WASH-related

Parasitic diseases of marine fishes and their impact on human health: focus on the Egyptian biosphere: A review.

Recently, food deficiency has become the most critical problem and a major global challenge. Rapid population growth requires increased food production to meet the resulting demand. High meat and chicken prices prompted the world to focus on fish production to meet ongoing food needs. Seafood is rich in vitamins, minerals, and omega-3s. Fish-borne parasitic diseases are primarily found in people in most countries, causing severe illness and death. This review article will focus on the most important parasites affecting marine fishes and the most popular parasitic diseases affecting fish global

Imported mixed malaria caused by Plasmodium falciparum and Plasmodium ovalecurtisi: An Italian case report.

Malaria remains a major global health threat, particularly in endemic regions of sub-Saharan Africa. In 2024 alone, an estimated 282 million cases were reported worldwide, with approximately 610,000 associated deaths. Beyond the high disease burden, mixed-species Plasmodium infections represent an additional clinical challenge, as they are frequently underdiagnosed and may significantly complicate both diagnostic accuracy and therapeutic management even in non-endemic countries. We report the case of a 67-year-old man from Guinea who presented to the Emergency Department with high-grade fever

Placental immunopathology and congenital transmission of Zika virus and Trypanosoma cruzi: comparative mechanisms, immune disturbances, and potential co-infection scenarios.

Congenital transmission of Zika virus (ZIKV) and Trypanosoma cruzi (T. cruzi) remains a major public health concern in the Americas. Both pathogens cross the placental barrier and cause severe neonatal outcomes, yet no confirmed congenital co-infections have been reported, and their combined effects on maternal-fetal health are unknown. Critical gaps persist regarding placental immunopathology, fetal immune programming, and long-term consequences of prenatal exposure. This review synthesizes current evidence on the routes and mechanisms of placental transmission of ZIKV and T. cruzi, their tro

Utility of recombinant Schistosoma bovis 22.6 kDa antigen for diagnosis of human urogenital schistosomiasis by ELISA: a cross-sectional study in Yewa North LGA, Ogun State, Nigeria.

BACKGROUND: In many endemic regions, accurately diagnosing urogenital schistosomiasis remains challenging, particularly in cases of low-intensity infection and limited laboratory capacity. Exploring antigens that cross-react between related schistosome species provides a promising alternative to conventional diagnostic approaches. Schistosoma bovis-an animal parasite closely related to S. haematobium and sharing the same Bulinus snail hosts in overlapping habitats- is a strong candidate for such studies. This work therefore assessed whether the recombinant S. bovis 22.6 kDa antigen (rSb22.6 kD

Gallopamil exhibits antiplasmodial activity against Plasmodium falciparum and Plasmodium vivax and shows predicted interactions with Plasmodium hexose transporters.

Malaria remains a major parasitic disease caused mainly by Plasmodium falciparum and Plasmodium vivax, and drug-resistant parasites continue to challenge disease control. Drug repurposing may help identify compounds with antimalarial potential. Here, we evaluated gallopamil, an L-type calcium channel blocker, using in vitro, ex vivo, in vivo and in silico approaches. Gallopamil inhibited the chloroquine-resistant P. falciparum W2 strain in vitro (IC₅₀ = 3.65 µM) and showed ex vivo activity against clinical isolates of P. falciparum and P. vivax from CEPEM/RO, with median IC₅₀ values of 6.4 and
