Next-generation CRISPR-based molecular diagnostics for veterinary parasitology: From laboratory innovation to one health surveillance.

Veterinary parasitic diseases remain a major threat to livestock production, companion animal health, wildlife conservation, and public health worldwide. Conventional diagnostic methods, including microscopy, serology, and culture-based techniques, often suffer from limited sensitivity, poor specificity, prolonged turnaround time, and dependence on specialized laboratory facilities. Recent advances in Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-based diagnostics have introduced highly sensitive, rapid, and field-deployable platforms for parasite detection. This review cr

Mechanosensation of rice root border cells activates serotonin-mediated pre-invasion behaviors in Meloidogyne graminicola via DEG/ENaC channels.

BACKGROUND: Plant-parasitic nematodes (PPNs) locate host roots primarily through chemosensation, yet the sensory events that convert root contact into the stylet thrusts required for host penetration remain obscure. Root border cells (RBCs), terminally differentiated cells shed from the root cap, constitute the first point of physical contact between the root and invading nematodes, but whether and how RBCs trigger pre-invasion behaviors is unknown. RESULTS: Here we showed that rice (Oryza sativa) RBCs induce preparasitic second-stage juveniles (pre-J2) of the rice root-knot nematode Meloidog

The multifaceted role of mesenchymal stem cells in infectious diseases: antimicrobial properties, immunomodulation and tissue repair.

BACKGROUND: Mesenchymal stem cells (MSCs) have emerged as promising therapeutic agents for infectious diseases owing to their antimicrobial, immunomodulatory, and tissue regenerative properties. Beyond their well-established role in tissue repair, MSCs and their secretome directly influence pathogen clearance through the secretion of antimicrobial peptides, extracellular vesicles, iron-sequestration mechanisms, and modulation of both innate and adaptive immune responses. KNOWLEDGE GAP: Despite extensive preclinical investigations, MSC responses are highly pathogen-dependent, with outcomes tha

Cytomics-driven insights for investigating host-pathogen interactions.

Cytomics, the high-dimensional profiling of immune cells, provides a systems-level view of host responses to infection. Unlike traditional diagnostics focused on pathogen detection or early immune activation, cytomics captures rare and functionally specialized immune subsets linked to disease severity and clinical outcome. In this review, we explore how advances in spectral cytometry, mass cytometry, and multimodal single-cell platforms enable unprecedented resolution of immune complexity during infection by summarizing cytomics evolution, high-dimensional single-cell and multimodal technologi

Transcriptomic, untargeted metabolomic, and gut microbiota studies on the anticoccidial activity of the combination formula in broilers.

UNLABELLED: Coccidiosis, a devastating intracellular parasitic disease, poses a significant threat to the poultry industry. Traditional control methods, which involve the inclusion of anticoccidial drugs in feed, have raised concerns over veterinary drug residues. Consequently, there is a pressing need to identify suitable alternatives or complementary strategies to anticoccidial drugs, with plant-derived natural products emerging as a promising candidate. Our laboratory has previously identified a combination formula that demonstrates good anticoccidial activity. However, the underlying mecha

Implementation of Targeted Selective Treatment (TST) in a Commercial Smallholder Sheep Flock: Reduction in Anthelmintic Expenditure.

The adoption of targeted selective treatment (TST) is an alternative for the sustainable management of gastrointestinal nematodes in sheep. TST mitigates the development of anthelmintic resistance by reducing the number of animals drenched while preserving the parasite refugia population. This study analysed the reduction of anthelmintic (AH) usage and assessed the AH cost reduction derived from a TST program compared to the previous whole-flock treatment regimen on a commercial smallholder sheep farm. Data acquisition spanned a 4.75-year period on a smallholder meat sheep farm in Colombo, Par

Synthesis of 1,2,3-Triazole Derivatives of N,N-Dimethyltryptamine Isolated From Mimosa tenuiflora Root Bark: Antiplasmodial Activity In Silico and In Vitro.

Malaria is a severe parasitic disease, with Plasmodium falciparum as one of the main etiological agents. The growing resistance to antimalarial drugs underscores the urgent need for the development of new therapeutic agents. This study describes the semisynthesis of eight new 1,4-disubstituted 1,2,3-triazole derivatives (3-10) from N,N-dimethyltryptamine (1), isolated from Mimosa tenuiflora. The derivatives were obtained through copper(I)-catalyzed alkyne-azide cycloaddition (CuAAC) reactions and characterized by NMR and mass spectrometry. Compound 1 showed low in vitro activity against the W2

Antifibrotic activity of Schistosoma japonicum-derived sja-miR-2a-3p in carbon tetrachloride-induced liver fibrosis.

BACKGROUND: Liver fibrosis is a common consequence of chronic liver injury and is driven largely by persistent activation of hepatic stellate cells and excessive extracellular matrix deposition. We previously identified Schistosoma japonicum-derived sja-miR-2a-3p as a parasite-derived microRNA with antifibrotic activity in schistosomiasis-associated liver fibrosis. Whether this cross-species regulatory molecule also modulates fibrogenesis beyond the context of schistosome infection remains unclear. METHODS AND RESULTS: We investigated the effects of sja-miR-2a-3p in a carbon tetrachloride-ind

Comparative palatability and efficacy of two praziquantel isomers in rainbow trout infected by eyefluke.

Praziquantel is a widely used anthelmintic in both human and veterinary medicine, but low palatability of the commonly used compound, which is a mixture (M) of two isomers (D and L forms), may result in rejection of oral treatments. Purification of the D-isomer of praziquantel allows a comparative evaluation of both palatability and efficacy of the isomers. We showed that rainbow trout (Oncorhynchus mykiss) preferred feed containing D-PZQ feed (60% acceptance) compared to M-PZQ feed (20% acceptance) (inclusion of 400 g PZQ/kg feed). Subsequent efficacy studies using eyefluke Diplostomum pseudo

A cohort-based population model for the Poultry Red Mite Dermanyssus gallinae (Acari: Dermanyssidae): Application to explore potential pathways for control strategies using Fluralaner as a case study.

This project addresses the challenge of controlling Dermanyssus gallinae infestations in poultry farms. A cohort-based model is developed that couples the pharmacokinetics of Fluralaner (Exzolt ®) in the plasma of laying hens with the dynamics of the mites, enabling the simulation of various treatment administration strategies while balancing costs. The study reveals that administering a reduced dose of Fluralaner (Exzolt ®) at high frequencies, in conjunction with a customized infestation tolerance threshold for each farm, provides a cost-effective solution for mite control. Specifically, thi

Wolbachia outcompetes mosquito-borne viruses by reprogramming host ATPSβ1-driven ATP metabolism toward symbiotic proliferation.

Wolbachia endosymbionts confer broad-spectrum resistance to mosquito-borne viruses. While the antiviral metabolites in Wolbachia-Aedes have been identified during viral replication, the core metabolic gene underlying this defense across the entire viral infection cycle remains undefined. Here, we demonstrated for the first time that Wolbachia strongly suppressed the emerging alphavirus Venezuelan equine encephalitis virus, extending its known antiviral spectrum. Transcriptomic and functional analyses revealed ATP synthase beta subunit 1 (ATPSβ1) as a key metabolic hub hijacked by both MBV and
