Effectiveness of benznidazole in the treatment of Chagas disease in children and adults: a long-term retrospective cohort study.

Chagas disease, caused by Trypanosoma cruzi, is endemic to the Americas but has spread worldwide due to migration. Benznidazole is effective, but long-term follow-up efficacy studies are scarce and include only small numbers of patients. We analyzed data from a retrospective cohort of Chagas disease patients treated with benznidazole at a large tertiary center in Argentina (1980-2019). Treatment response was assessed through clinical, parasitological, and serological follow-up. Parasitemia was evaluated by direct visualization or PCR. Among 567 treated patients, 464 met selection criteria, inc

Spatial and genetic dynamics of Plasmodium falciparum histidine-rich protein2 in sub-Saharan Africa.

BACKGROUND: Malaria rapid diagnosis tests (mRDTs) detecting Plasmodium falciparum histidine-rich protein 2 (Pfhrp2) are the most widely adopted diagnostic tool in malaria-endemic areas. However, extensive polymorphisms in Pfhrp2 could modify Pfhrp2 epitopes, rendering it undetectable by Pfhrp2-based mRDTs. This study investigates the spatial distribution and population dynamics of Pfhrp2 in sub-Saharan Africa (sSA). METHOD: Pfhrp2 sequences (215) were retrieved from public repositories and analysed with MAFFT v7, DnaSP, and various packages in R. We computed genetic diversity metrics, neutral

From islands to landscapes: Integrating genomic and advanced eco-geographic tools in area-wide tsetse control.

Tsetse (Diptera: Glossinidae) are the exclusive cyclical vector of human and animal African trypanosomoses and continue to threaten health and agricultural productivity across sub-Saharan Africa. Area-wide integrated pest management (AW-IPM) strategies provide a coordinated framework for combining tools such as the sterile insect technique (SIT), insecticide-treated targets and cattle, and others. This review examines the success and challenges of AW-IPM in tsetse control and advances in population genetics, species distribution modeling, and landscape genetics that improve identification of c

Dynamics and functional heterogeneity of FOXP3+ regulatory T cells in Trypanosoma cruzi infection: a systematic review.

BACKGROUND: The role of regulatory T cells (Tregs) in Trypanosoma cruzi infection remains unresolved, with conflicting evidence suggesting either protection from immunopathology or impairment of parasite control. METHODS: We performed the first systematic review integrating data on Treg phenotype and function in human and experimental Chagas disease. A comprehensive search of PubMed, Embase, Scopus, and LILACS (2003-2025) identified studies assessing Tregs during T. cruzi infection. Study selection, data extraction, and risk-of-bias assessment followed PRISMA 2020 and Cochrane guidelines. RE

Modified wet mount staining improves the microscopic diagnosis of Trichomonas vaginalis in vaginal discharge.

Conventional microscopic diagnosis of Trichomonas spp. in vaginal discharge specimens relies entirely on organism motility. Herein, we established a novel modified methyl green-acid fuchsin staining method to differentiate Trichomonas vaginalis from leukocytes and epithelial cell nuclei based on staining characteristics, rather than motility. To evaluate the performance of this novel method, 2133 vaginal discharge specimens were subjected to clinical testing using safranin staining microscopy, Trichomonas culture, polymerase chain reaction (PCR), and the proposed method, with an expanded gold

Genetic diversity and morphology of South Korean Aedes koreicus (Diptera: Culicidae) provide insights into the origin of European populations.

BACKGROUND: Aedes koreicus (Edwards, 1917) is native to Korea, northern China, and the Russian Far East, and has gained attention following its first detection in Europe in 2008 and subsequent spread across Europe. The Korean Peninsula, which includes the type locality of Ae. koreicus, has been regarded as a potential source region for invasive populations of this species, along with other parts of its native range. However, little is known about the genetic and morphological attributes of Korean populations. METHODS: We obtained DNA from 172 specimens of Ae. koreicus from 19 populations in S

Disease of South American camelids in England and Wales: Analysis of diagnoses from Animal and Plant Health Agency laboratory submissions between 2012 and 2021.

BACKGROUND: South American camelids (SACs) have become increasingly popular in the UK. Consequently, knowledge among veterinary practitioners about diseases in these non-native species has improved, although gaps remain. The remit of this study was to identify common diseases of SACs based on Animal and Plant Health Agency (APHA) surveillance data from testing of carcass and sample submissions. METHODS: A total of 3541 data sets of SAC submissions to APHA between 2012 and 2021 were analysed. RESULTS: The overall diagnostic success rate was 39.5% but was significantly higher in carcass submis

Estimation of Adult Census Size From Close-Kin Dyads in the Malaria Mosquito Anopheles gambiae.

Accurate estimates of adult mosquito abundance are central to the design and evaluation of vector control strategies, yet they are difficult to obtain from natural populations. Conventional mark-recapture methods for estimating adult mosquito census size pose logistical and other challenges. A recently developed close-kin mark-recapture (CKMR) approach is a promising alternative. However, application of CKMR has been largely confined to long-lived vertebrates. Validation on empirical data from short-lived, highly fecund insects is lacking. Here, we apply CKMR to a natural population of Anophel

Rotational grazing management as a strategy for the integrated epidemiological control of parasites in cattle in Brazil.

The bioeconomy of livestock is influenced by several factors, including parasitic diseases. The occurrence of these diseases is related to factors involving the parasite, the host, the environment and management of the property. This study seeks to assess parasite control in cattle using the Lone Tick rotational grazing system. Cattle were kept in a paddock and were subjected to a rotational management system to test the Lone Tick system for tick and helminth control. A total of 37 male Angus cattle were used in this experiment. The animals were kept in a pasture (paddock) for 28 days and retu

Development of a novel organoid-based two-dimensional system to model the intestinal host-parasite interface in fasciolosis.

BACKGROUND: The earliest interactions between Fasciola hepatica newly excysted juveniles (FhNEJ) and the intestinal epithelium remain poorly understood, partly due to the limited availability of physiologically relevant in vitro models. Here, we establish a human duodenum organoid-derived epithelial monolayer (ODM) co-culture system to investigate the early host-parasite interface, providing a controlled experimental platform for analysing interactions between FhNEJ and the intestinal epithelium. METHODS: ODMs representing the human duodenum were co-cultured with either live FhNEJ or a tegume

Abdominal hydatid disease and its mimics: a comprehensive pictorial review of imaging clues to the differential diagnosis.

BACKGROUND: Hydatid disease (HD), also known as echinococcosis, is a parasitic disease caused by tapeworms. HD is endemic in many parts of the world and can affect virtually any organ system. Due to the rapid growth of worldwide migration, especially in recent decades, the disease can be diagnosed anywhere, particularly among immigrants in non-endemic areas. OBJECTIVE: To illustrate the spectrum of abdominal imaging manifestations of HD and to review the principal benign and malignant abdominal cystic lesions that can mimic HD, using an organ-based approach with emphasis on imaging features t

Molecular and morphological characterization of hard ticks (Acari: Ixodidae) infesting rodents in Southern Iran: evidence of a cryptic Haemaphysalis species.

BACKGROUND: Hard ticks are important vectors of various zoonotic diseases. Identifying closely related tick species using morphological methods alone can be challenging. This study aimed to assess the taxonomic status of hard ticks infesting rodents in southern Iran using morphological and molecular approaches. METHODS: Ticks were collected from the ear bases and rears of rodents and identified morphologically. Genomic DNA was extracted, and the cytochrome oxidase subunit 1 (COX1) and internal transcribed spacer 2 (ITS2) genes were amplified via polymerase chain reaction (PCR). Sequencing and

Use of pour-on formulations of fluralaner in prophylactic treatments against Cochliomyia hominivorax in newborn calves under extensive grazing systems.

The aim of this study was to evaluate the efficacy of pour-on fluralaner formulations for the prevention of cutaneous myiasis caused by Cochliomyia hominivorax in newborn calves under field conditions. Three field trials were conducted in Argentina, including subtropical and temperate regions. In Trial 1, calves with unhealed navels were assigned to two fluralaner-treated groups with different commercial pour-on formulations and an untreated control group. In Trial 2, two groups of newborn calves were treated either with fluralaner pour-on or injectable doramectin. Trial 3, where 84 calves wer

Preliminary Ultrasonographic Optic Nerve Sheath Diameter Values in Clinically Healthy Adult Rahmani Sheep.

BACKGROUND AND AIMS: Sheep reared in tropical and subtropical areas may develop neurological disorders associated with infectious, parasitic, metabolic, or nutritional diseases. Some of these disorders may be accompanied by increased intracranial pressure (ICP). Direct ICP measurement methods are invasive, and technically unavailable in clinical field settings. Ultrasonographic measurement of optic nerve sheath diameter (ONSD) has been reported as a noninvasive method of ICP indicator in humans and animal species. However, baseline ONSD values for clinically healthy sheep are not yet reported.

Haemophagocytic Lymphohistiocytosis Triggered by Visceral Leishmaniasis Masquerading as Epstein-Barr Virus (EBV)-Associated Haemophagocytic Lymphohistiocytosis in Rheumatoid Arthritis: A Case Report and Literature Review.

Haemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory syndrome caused by uncontrolled immune activation. In adults, it is usually secondary to an underlying condition, such as infection, malignancy, or autoimmune disease. Identifying this trigger is essential, as treatment depends on the cause. Visceral leishmaniasis (VL) is a systemic parasitic infection that may mimic or trigger HLH, particularly in immunosuppressed patients. We report a 59-year-old man with seropositive rheumatoid arthritis, for which he received immunosuppressive treatment with methotrexate and

Autoamputation of pelvic adnexa: two case reports.

BACKGROUND: Parasitic ovarian teratoma is a rare consequence of chronic torsion or auto-amputation of the adnexa. Delayed diagnosis may result in a free-floating pelvic mass without identifiable adnexal tissue. CASE DESCRIPTION: Respectively, two young women (23 and 29 years) presented with ignored pelvic pain a few months ago. Both had previously self-medicated with oral analgesics without further treatment. The ultrasound revealed an isolated solid-cystic pelvic mass with absent colour Doppler signals. No normal ipsilateral ovarian or tubal tissue was visualised. Both patients had no positi

Five ways to find a parasite: comparison of five molecular assays for the detection of Strongyloides spp.

UNLABELLED: Strongyloidiasis, caused by nematodes of the genus Strongyloides, is a neglected tropical disease with significant public health implications. Accurate diagnostics are essential for disease surveillance and control, yet traditional stool-based and serological methods have limitations in sensitivity and specificity. Molecular assays offer an alternative diagnostic approach. In this study, we compared four probe-based real-time PCR (qPCR) assays and one loop-mediated isothermal amplification (LAMP) assay using human fecal DNA samples from Gabon and Papua New Guinea with both Strongyl
