Duffy-negatives were previously thought to be immune to Plasmodium vivax infections due to Duffy binding protein's (PvDBP1) inability to invade erythrocytes lacking Duffy antigen receptor for chemokines (DARC) expression. Nevertheless, reports of P. vivax cases are growing throughout Africa and among Duffy-negative people. Although there are alternative invasion mechanisms by P. vivax, the exact mechanisms in Duffy-negative individuals are unclear. Sudan, with a mixed Duffy-negative and Duffy-positive population, is ideal to study differences between these infections on epidemiological and gen

Effective control of parasitic diseases in veterinary medicine relies heavily on anthelmintic drugs, particularly combinations targeting both nematodes and cestodes. However, simultaneous quantification of multi-component anthelmintic formulations presents significant analytical challenges, especially with cost-effective UV-Vis spectrophotometry due to inherent spectral overlap. This study introduces a novel, rapid, and environmentally conscious UV-Vis spectrophotometric method, leveraging advanced chemometrics, for the simultaneous determination of Fenbendazole (FN), Pyrantel embonate (PN), a

The advent and global spread of plasmid-carried resistance genes, including the tigecycline resistance gene tet(X3/X4) and the colistin resistance gene mcr-1, present a substantial threat to public health that requires immediate resolution. This study successfully screened and identified commercial anti-parasitic drugs that efficiently inhibit Tet(X4) enzyme activity (IC50 = 79.42 µg/mL), revealing that mefloquine exhibited a synergistic antibacterial effect with tetracyclines against tet(X3/X4)-positive bacteria (FIC < 0.5) and an additive effect with tetracyclines against tet(X3/X4)-negative

BACKGROUND: Giardiasis is a common intestinal disease caused by the protozoan parasite Giardia duodenalis. The transmission of giardiasis is influenced by a complex interplay of environmental hazards such as contaminated water sources, markets, and/or roads or pathways including anthropozoonotic domestic animal-related factors and accessibility to healthcare. However, the risk factors of acquiring G. duodenalis including its assemblages and sub-assemblages have not been previously mapped in high burden populations in rural Africa. To mitigate transmission in rural African settings with special

Coccomyxa parasitica-like algae pose a growing threat to bivalve aquaculture. In this work, for the first time under controlled conditions, the effect of the green parasitic microalgae of genus Coccomyxa sp. in the Sea of Japan on the immune and antioxidant protection of Mytilus galloprovincialis was studied in two ways of infection-through filtration (with feed) and injection (into an adductor). By day 7, mortality in both experimental groups reached 68%. The phagocytic activity of hemocytes significantly decreased in the feed group, which may be due to the masking of the parasite as a food p

Background: Chronic Fatigue Syndrome (CFS), also known as Myalgic Encephalomyelitis (ME), is a debilitating condition characterized by persistent fatigue and multisystemic symptoms, such as cognitive impairment, musculoskeletal pain, and post-exertional malaise. Recently, parallels have been drawn between ME/CFS and Long COVID, a post-viral syndrome following infection with SARS-CoV-2, which shares many clinical features with CFS. Both conditions involve chronic immune activation, raising questions about their immunopathological overlap. Objectives: This study aimed to compare immune biomarker

UNLABELLED: Alpha-lipoic acid (ALA) is well-known for its antioxidant and anti-inflammatory functions in various disease models. Here, we tested whether pre-exposure to ALA can protect the testes from cellular damage caused by scrotal heat shock (HS) in mice.
METHODS: Thirty-six Swiss albino mice were divided into control (CTRL, n = 6), HS (n = 10), and two ALA dose (HS + ALA 200 mg/kg, n = 10; and HS + ALA 400 mg/kg, n = 10) groups. ALA supplementation was administered orally for 30 days. Subsequently, the animals, except the controls, were subjected to an HS water bath at 43 °C for 20 min.

Background: Neurocysticercosis is a parasitic infection of the central nervous system caused by the larval stage of Taenia solium. This disease is endemic in some countries in Central and South America, South and South-East Asia, and sub-Saharan Africa. In North America, Europe, Japan, and Australia, only sporadic cases are documented. Moreover, reports of bacterial superinfection arising within neurocysticercotic lesions remain exceptionally scarce. Methods: We report a clinically severe and diagnostically challenging case of suspected neurocysticercosis with cerebral streptococcal superinfec

Coccidiosis is one of the most serious parasitic diseases in poultry, with Eimeria-induced apoptosis of IECs recognized as a key pathogenic mechanism. This review systematically delineates the molecular mechanisms governing this apoptotic process. The invasion process of Eimeria app. is mediated by the AMA1-RON2 moving junction complex and secreted effector proteins. An integrated model of apoptotic regulation is proposed. This model comprises the mitochondrial, death receptor, and endoplasmic reticulum stress pathways, which are coordinated by signaling hubs, such as PI3K/Akt, NF-κB, and JNK/

Trichinella spp. are zoonotic nematodes with a global distribution, primarily maintained through wildlife reservoirs which complicates eradication efforts. In Europe, four species-T. spiralis, T. britovi, T. nativa, and T. pseudospiralis-circulate in wild carnivores and omnivores, with T. britovi and T. spiralis being the most prevalent in wild and domestic hosts. This study aimed to assess the presence and species distribution of Trichinella spp. in wild mammals. This study presents the results of five years of wildlife surveillance carried out within the long-standing monitoring program impl

The increasing threat of zoonotic malaria parasites of humans, such as Plasmodium knowlesi, make the search for improved pharmacotherapy imperative. Using protein sequence and structural analyses, phylogenetics, protein network mapping, protein-ligand interaction, and small molecule docking studies, we have identified for the first time the predicted structure, function, and druggability of the P. knowlesi heat shock protein 90s (PkHsp90s). Four isoforms were identified (in the cytosol, endoplasmic reticulum, mitochondrion, and apicoplast), and key structural differences were elucidated compar

Haemonchus contortus is a blood-feeding gastrointestinal nematode that significantly impacts the health and productivity of small ruminants. The burden of parasitism and the escalating incidence of anthelmintic resistance necessitate alternative control methods. Here, we characterize the enzymatic activities of five mammalian cell-expressed recombinant H. contortus cysteine proteases (HcCPs), which include two cathepsin B-like proteins (HcCBP1 and HcCBP2) and three cysteine protease 1 proteins (HcCP1a, HcCP1b, and HcCP1c). We hypothesize that these enzymes degrade host blood proteins, thereby

Trypanosoma cruzi, the causative agent of Chagas disease, can cross the placental barrier and be transmitted congenitally, yet the mechanisms underlying this process remain incompletely understood. Recent evidence suggests that T. cruzi-derived extracellular vesicles (TcEVs) may facilitate placental invasion by modulating host-pathogen interactions. In this study, we examined the effects of TcEVs on human placental explants (HPEs), focusing on their capacity to disrupt tissue architecture and modulate matrix metalloproteinases MMP-2 and MMP-9, enzymes critical for extracellular matrix remodeli

The development of new alternative models is essential to overcome the limitations of traditional two-dimensional (2D) cell cultures and animal models. Three-dimensional (3D) models, such as organoids, better mimic the structural and functional complexity of mammalian organs, thereby reducing the ethical and economic issues related to animal experimentation. These systems provide more physiologically relevant environments, improving the accuracy of disease modeling and drug response prediction. In this context, we have developed mouse hepatic organoids from livers of adult BALB/c mice and char

Microsporidia MB is a natural symbiont of Anopheles arabiensis Patton, 1905 that blocks Plasmodium transmission without compromising mosquito fitness. Developing this symbiont for malaria control requires a thorough understanding of its transmission dynamics across generations under environmentally relevant conditions. This study aimed to evaluate the transgenerational spread of Microsporidia MB in semi-field settings. From field-collected infected females (G0), several independent replicates of Microsporidia MB-infected An. arabiensis were established and maintained under semi-field condition

Rumen development is critical for optimal growth, feed efficiency and health in pre-weaned dairy calves. Butyric acid, a short-chain fatty acid produced in the rumen, has been shown to stimulate epithelial development and modulate immune responses. This study evaluated the effects of butyric salt supplementations, as metabiotics, on rumen morphometry and investigated the potential associations with feed intake, growth performance and health indicators in pre-weaned dairy calves. Thirty calves were randomized into three groups: controls (G1), 25 g (G2) or 50 g (G3) sodium butyrate from birth un
