A poly(vinyl alcohol)-based dissolvable bilayer film for buccal and sublingual delivery of FITC-dextran 4000 as a model macromolecule.

Oromucosal administration of peptides remains challenging due to the limited permeability of the buccal and sublingual mucosa to hydrophilic macromolecules. This study evaluated the permeation-enhancing performance of the bile salt, sodium glycodeoxycholate (GDC), by investigating its incorporation into a semi-dissolvable bilayer film platform using fluorescein isothiocyanate-labelled dextran 4000 (FD4) as a model macromolecule. In human TR-146 buccal epithelial monolayers, GDC increased FD4 permeability in a concentration-dependent manner, with 1 mM identified as the highest non-cytotoxic con

The molecular landscape of nodular fasciitis: novel USP6 fusions, genomic stability, and an exploratory clinicopathological model - a 20-year retrospective study.

Nodular fasciitis (NF) is a benign myofibroblastic neoplasm characterized by rapid growth and recurrent USP6 rearrangements. Despite its benign nature, NF may exhibit worrisome histologic or clinical features that mimic sarcoma, posing diagnostic challenges. The relationship between molecular diversity and morphologic heterogeneity remains incompletely understood. We retrospectively analyzed 283 NF cases diagnosed between 2000 and 2024 at a tertiary referral center. Molecular testing for USP6 rearrangements was performed in 44 cases using targeted RNA sequencing. Histologic features, clinical

Preparation and evaluation of degradable imaging drug loaded embolization microspheres for hepatocellular carcinoma.

Transarterial chemoembolization (TACE) necessitates the use of embolic agents with intrinsic radiopacity and controlled biodegradability to enhance therapeutic precision. In addressing the limitations inherent in current drug-eluting microspheres (DEMs), this study introduces the development of inherently radiopaque and biodegradable (DOX+Lipiodol) @PLGA @CaCO3 @Alg core-shell microspheres. These microspheres were synthesized using rotating membrane emulsification and surface-induced ion crosslinking, with calcium carbonate serving multiple roles: a solid surfactant for Pickering stabilization

Analytical and clinical validation of the EdenDx methylation assay as a noninvasive supportive triage tool for early detection of endometrial cancer in cervical scraping specimens.

OBJECTIVE: Endometrial cancer (EC) is the most common gynecologic malignancy; however, early detection remains challenging due to overlapping symptoms and the absence of validated noninvasive triage testing. Diagnosis relies on invasive procedures such as endometrial biopsy or dilation and curettage. The EdenDx assay detects hypermethylation of CDO1 and CELF4 genes, which are specifically associated with EC and have been shown to be epigenetically altered prior to symptom onset. Here, we evaluate the clinical and analytical performance of the EdenDx assay as a noninvasive triage tool for the e

Enhanced reproductive toxicity following combined exposure to ethinylestradiol and levonorgestrel: evidence of non-additive endocrine disruption in male rats.

Ethinylestradiol (EE2) and levonorgestrel (LNG) are synthetic hormones widely used in contraceptives and increasingly recognized as endocrine-disrupting contaminants. Although their individual reproductive effects are documented, their combined toxicity remains poorly understood. This study evaluated the sub-chronic reproductive toxicity of EE2 and LNG, administered alone or in combination, in male Wistar rats, with emphasis on potential non-additive endocrine-disrupting interactions. Animals were orally treated with EE2 (1 µg/kg bw), LNG (5 µg/kg bw), or both for four weeks. Individual exposu

Association of admission C-reactive protein levels and histological chorioamnionitis in patients with preterm premature rupture of membranes: a systematic review and meta-analysis.

BACKGROUND: Preterm premature rupture of membranes (PPROM) is a high-risk obstetric complication. Ascending infection and histological chorioamnionitis (HCA) frequently occur after membrane rupture, contributing to severe adverse maternal and fetal outcomes. Early prediction of HCA using non-invasive biomarkers allows timely clinical management, while the predictive efficacy of C-reactive protein (CRP) for subsequent HCA in PPROM patients remains inconsistent across existing studies. This meta-analysis aimed to systematically evaluate the predictive association. METHODS: We systematically sea

Leveraging deep learning for prognostic stratification of hepatocellular carcinoma using histopathological images.

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common and lethal malignancies, whose prognostic prediction is particularly challenging. Histopathological biomarkers are critical for cancer prognosis assessment. Advancements in artificial intelligence (AI) have made it possible to automate the classification and detection of information from whole-slide pathology images using deep learning. This study aims to utilize AI deep learning to analyze histopathological slides of HCC patients for prognostic stratification, thereby assisting in the development of clinical treatment plans.

3D printing of a microfluidic chip-generated liposome-loaded hydrogel scaffold for accelerated wound closure and reduced fibrotic remodeling in burn wounds: an in vivo study.

Growth factor-based therapies for burn wound healing are limited by rapid clearance, poor bioavailability, and inadequate efficacy of single agents. Here, we report a 3D printed chitosan hydrogel scaffold loaded with PEGylated liposomes encapsulating TGF-β3 and cationic lipoplexes carrying miR-132 mimic for localized co-delivery. The optimized formulation exhibited favorable printability and rheological properties, and liposome incorporation modulated swelling, accelerated degradation, and altered the tensile profile while maintaining scaffold structural integrity, as confirmed by SEM, DSC, an

Carboxylesterase 1 is a core metabolic vulnerability of platinum-resistant high-grade serous ovarian carcinoma.

BACKGROUND: Platinum-resistant high-grade serous ovarian carcinoma (HGSOC) is a major clinical challenge, with high mortality rates and no effective therapeutic options. Metabolic adaptations-including enhanced lipid catabolism, oxidative phosphorylation (OXPHOS), and autophagy-are major drivers of chemoresistance; however, the molecular mechanisms underpinning this metabolic reprogramming remain poorly understood. Carboxylesterase 1 (CES1), an NF-κB-regulated neutral lipase, supports malignant cell survival under nutrient-depleted conditions in other cancers, but its role in HGSOC has not bee

Low-Cost Methotrexate Chemoselection to Predict Radiotherapy Response in Advanced Oral Squamous Cell Carcinoma: A Prospective Feasibility Study From South India.

IntroductionOral squamous cell carcinoma (OSCC) represents a disproportionate cancer burden in South Asia, where advanced-stage presentation, limited radiotherapy infrastructure, and constrained chemotherapy resources necessitate pragmatic approaches to treatment selection. Platinum-based combinations remain the standard for induction chemotherapy; however, a substantial proportion of OSCC patients present with malnutrition, poor performance status, and limited financial resources that preclude these regimens. This is a hypothesis generating feasibility study which explores whether early respo

Nuclear protein in testis midline carcinoma of nasal cavity: a case report and review of the literature.

Nuclear protein in testis (NUT) midline carcinoma (NMC) is an extremely rare, highly aggressive, and poorly differentiated malignant neoplasm that predominantly arises from the midline epithelial structures of the head and neck. It is driven by genetic rearrangements of the NUTM1 gene, with the most common aberration being a translocation that forms a fusion gene with BRD4 on chromosome 19p13.1. The diagnosis of NMC is challenging due to its non-specific clinical and histological features, and it typically requires a combination of immunohistochemical detection of NUT protein and molecular ass

Excision of a giant cervical-thoracic aggressive fibromatosis: a case report and review of literature.

BACKGROUND: Aggressive fibromatosis is a rare, locally invasive tumor with a marked tendency to recur, and it is often diagnosed only after the lesion has already reached a considerable size. Although histologically benign, these tumors can behave aggressively at the local level. CASE DESCRIPTION: We report the case of a male patient with a large mass occupying the entire left hemithorax and extending into the left side of the neck. The tumor compressed the surrounding organs and produced corresponding symptoms. It encircled the left subclavian artery and vein. The patient underwent a complet

Immune checkpoint inhibitor-associated interstitial nephritis: let's move forward addressing the unknowns.

Immune checkpoint inhibitors (ICIs) are now well recognized as a cause of acute interstitial nephritis (ICI-AIN), and their increasingly widespread use explains the growing incidence of reported cases. This clinicopathological entity is characterized by acute kidney injury with a tubulointerstitial pattern occurring in a patient exposed to an ICI, and histological confirmation is required to establish the diagnosis. Histopathological examination typically reveals a lymphocytic infiltrate, supporting the use of corticosteroid therapy for ∼12 weeks, which enables recovery of kidney function in >

Negative Appendicectomy Rate and Surgery Despite Negative Triple-Screen Inflammatory Markers in Children: A Single-Centre Retrospective Cohort Study.

Introduction  The negative appendicectomy rate (NAR) is an important measure of diagnostic accuracy and quality of care. Diagnosing appendicitis in children remains challenging due to frequently atypical and non-specific clinical presentations. Despite advances in clinical scoring and imaging, NAR in the UK remains high. The primary aims of this audit were to determine the NAR at Queen's Hospital Burton against the Getting It Right First Time (GIRFT)-reported national figure, and to establish how often surgery proceeded despite negative Academy of Medical Royal Colleges (AoMRC) triple-screen i

Post-radiation Uterine Carcinosarcoma: A Case Report.

Uterine carcinosarcoma is a rare, malignant tumor of the endometrium composed of both high-grade epithelial (carcinomatous) and mesenchymal (sarcomatous) components, representing one of the most aggressive histological subtypes of endometrial carcinoma. Prior pelvic radiotherapy has been associated with an increased risk of subsequent aggressive endometrial malignancies, although the latency between irradiation and tumor development may vary considerably. Diagnosis may be delayed because early symptoms can mimic benign post-radiation sequelae. We report a woman in her 60s who, seven years afte
