A Defined Strategy for Multi-Lineage Differentiation of Gastric Organoids.

In the gastric glands, adult stem cells differentiate into multiple epithelial lineages, including pit mucous cells, acid-secreting parietal cells, and enzyme-producing chief cells. Efficient and controllable lineage specification from adult gastric stem cells is essential for modeling gastric homeostasis and disease in vitro. Organoids are a good model for investigating cell differentiation and organ formation. However, methods to achieve appropriate differentiation of multiple gastric lineages are inefficient. Here, we provide a detailed stepwise protocol for the differentiation of functiona

Automated Quantification and Morphometric Analysis of Pluripotent Stem Cell Colonies Using ColonyQuant.

Alkaline phosphatase staining is routinely used to evaluate the undifferentiated state of embryonic stem cells, yet quantitative interpretation of colony assays frequently depends on manual inspection and categorical scoring that introduce subjectivity and limit scalability. ColonyQuant provides a standardized, automated framework for objective analysis of alkaline phosphatase-stained colonies from conventional bright-field images. The software performs adaptive colony detection, per-colony intensity measurement, and extraction of eight geometric descriptors that collectively characterize colo

Alkaline Phosphatase Staining of Embryonic Stem Cells.

Alkaline phosphatase (AP) is a widely utilized histochemical marker for assessing the pluripotent state of embryonic stem (ES) cells. Mouse ES cells maintained in an undifferentiated state exhibit high levels of tissue-nonspecific alkaline phosphatase (TNAP) activity, which sharply declines upon differentiation. AP staining provides a rapid, cost-effective, and reliable method to evaluate ES cell colony morphology, confirm maintenance of pluripotency during routine culture, and monitor the efficiency of differentiation protocols. Here, we describe a detailed protocol for AP staining of mouse E

Animal-Free Intestinal Epithelial 2D "Organoid-Sheet" Cultures Using Invasin.

The intestinal epithelium consists of a single layer of polarized cells that serves as a crucial barrier against pathogens while mediating food digestion and nutrient absorption. The differentiated villus cells, important for the intestine's function, originate from rapidly dividing Lgr5+ stem cells located in the intestinal crypts. These stem cells can be expanded in vitro and form three-dimensional (3D) "mini-organs" known as intestinal organoids, which closely phenocopy the intestinal epithelium. The development and maintenance of these 3D organoids depend on a defined growth factor cocktai

Intestinal Organoids Culture in Synthetic Hydrogels.

Derived from adult intestinal stem cells, organoids form intricate three-dimensional structures that closely mimic the cellular diversity and spatial organization of the human gut epithelium. Patient-derived organoids (PDOs) can recapitulate patient-specific tumor heterogeneity, making them highly valuable for colorectal cancer research and personalized medicine. However, their long-term in vitro culture still depends mainly on animal-derived materials (e.g., Matrigel) with undefined composition, batch variability, and limited tunability. To overcome these limitations, we introduced the use of

Chlamydia trachomatis Infection in a Microphysiologic Cell Culture Model.

Chlamydia trachomatis is an obligate intracellular bacterium that infects the columnar epithelium of the human endocervix. While conventional two-dimensional cell cultures and animal models have been instrumental in advancing our understanding of C. trachomatis biology, they are limited in capturing the multicellularity, architecture, and physiological microenvironment of the human cervix. This chapter describes the use of a three-dimensional (3D) microphysiologic model to study Chlamydia trachomatis infection. The model is inexpensively made without specialized equipment and is designed to re

Diagnostic performance and testing costs of pooled testing for molecular detection of tuberculosis in seven countries: a prospective, observational, diagnostic accuracy study.

BACKGROUND: Globally in 2024, only 54% of people diagnosed with tuberculosis received an initial WHO-recommended rapid molecular diagnostic test. Pooled testing could expand access to molecular detection using existing laboratory capacity. We aimed to evaluate the performance of pooled tuberculosis testing in a multicentre diagnostic accuracy study. METHODS: In this prospective diagnostic accuracy study, we enrolled individuals with presumptive tuberculosis through facility-based case finding (district hospitals and primary health centres) and community-based case finding in Bangladesh, Brazi

Comprehensive Isolation Strategy for Intestinal Epithelial and Fibroblast Cells from Healthy and Tumor Human Tissues.

Organoids and fibroblasts derived from patient tissue serve as physiologically meaningful in vitro models to investigate tissue biology, diseases, and treatment responses. In this study, we present a robust protocol for the simultaneous isolation and long-term culture of patient-derived organoids (PDOs) and fibroblasts from both healthy and colorectal cancer samples. The workflow uses enzymatic dissociation followed by efficient separation into epithelial and stromal cell fractions. Organoids are embedded in Matrigel or Matrigel-Collagen I mixtures to support three-dimensional growth, whereas

Oleic acid supports mitochondrial function during lipolytic and pro-oxidant challenges in bovine adipocytes.

Increased energy demand during the transition period in dairy cows leads to excessive lipolysis, oxidative stress, and mitochondrial dysfunction in adipose tissue (AT). Mitochondria play a central role in fatty acid (FA) oxidation and redox regulation during negative energy balance. In humans, palmitic acid (PA) is linked to mitochondrial stress, whereas oleic acid (OA) supports normal function. However, the impact of acute lipolytic or pro-oxidant challenges on mitochondrial function in bovine adipocytes, and whether that impact can be mitigated by FA supplementation, remains unclear. This st

Mitochondria-mediated anticancer effects of two previously characterized COX-2 inhibitors in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality and often shows limited response to current therapies like sorafenib. This study investigates the selective anticancer activity and mitochondria-mediated apoptotic effects of two previously characterized COX-2 inhibitors, ZMA-M3 (imidazo[1,2-a]pyrazine) and ZMA-M4 (imidazo[1,2-a]pyridine), in isolated HCC and normal rat hepatocytes. Isolated HCC and normal rat hepatocytes were treated with various concentrations of ZMA-M3 and ZMA-M4 to determine their half-maximal inhibitory concentrations (IC50). Apoptosis was evalu

Rational design and synthesis of a novel cereblon ligand class enabling selective molecular glue and potent PROTAC.

The expansion of E3 ligands provides elemental value for the research and drug design of both molecular glues and proteolysis-targeting chimeras (PROTAC). This study first reports the rational design of a novel class of sulfur-incorporated CRBN ligands (SCLs) with enhanced binding affinity over classical immunomodulatory drugs (IMiDs). Based on this platform, we developed: (1) GT-02624, a GSPT1 molecular glue degrader with small size, degradation selectivity among the tested canonical CRBN neosubstrates and corresponding anti-proliferative activity; and (2) GT-01956, a BET-targeting PROTAC exh

Centering Patient Health Over Legal Status: A Student-Driven Cannabis Risk Stratification Framework for Undergraduate Medical Education.

BACKGROUND: Cannabis use is increasing worldwide, both for evidence-supported indications and conditions with limited evidence such as anxiety, depression, and chronic pain. Although incongruence between state and federal cannabis laws has delayed expanding cannabis education within medical schools, patients will continue to use cannabis and physicians must be prepared to counsel them. Therefore, evidence-informed cannabis education is needed to prepare medical students to discuss cannabis use using a patient-centered and harm reduction-oriented framework rather than relying on legal status al

Discovery and Modulation of an Unusual Terminal Mismatch Bypass Activity Involving 3'-Phosphorothioated Primers and Commercial Strand Displacement DNA Polymerases.

Phosphorothioated (PST) oligonucleotides are extensively used in RNA therapeutics and biosensing applications. PST-modified linkages remain part of 13 approved oligonucleotide drug backbones (out of 23 total approved drugs). Despite the widespread utilization of PST linkages in oligonucleotide therapeutics, the full spectrum of their interactions with nontarget nucleic acids and DNA polymerases remains inadequately characterized. In this study, we report a novel discovery and the modalities (sequence dependence and length) of an unusual 3'-mismatch bypass activity involving 3'-PST primers, cir

Maillard-like Reactions Generate Secondary Organic Products with Oxidative Potential: Mechanisms and Photoaging Enhancement.

The formation mechanisms and atmospheric evolution of secondary organic aerosols contributing to the oxidative potential (OP) remain poorly understood, limiting the assessment of their potential health impacts. Here, we demonstrate for the first time that atmospheric aqueous Maillard-like reactions constitute an important pathway for generating secondary organic products with OP. Six representative dicarbonyl-nitrogen reaction systems were designed to systematically investigate the formation kinetics, molecular characteristics, and photoaging evolution of the products generated from the Mailla

Characterization of F-30 as an FGFR2/4-selective covalent inhibitor with activity against clinically relevant mutations.

Fibroblast growth factor receptors FGFR2 and FGFR4 are oncogenic drivers in multiple cancers, yet most approved FGFR inhibitors lack subtype selectivity and are prone to acquired resistance. Here we identify F-30 as, to our knowledge, the first covalent inhibitor with preferential activity toward both FGFR2 and FGFR4 through covalent targeting of a conserved P-loop cysteine. Structural analyses reveal that differential engagement of the P-loop and surrounding binding environment contributes to paralog discrimination. Consistent with these structural observations, F-30 showed an approximately 1

Protective cucurbitane-type triterpenoids from the stems of Cucumis melo against cisplatin nephrotoxicity.

Six new cucurbitane-type triterpenoids, named melonoids A-F (1-6), were isolated from the stems of Cucumis melo L., along with eight known compounds (7-14). Their structures were elucidated through comprehensive analysis of 1D and 2D NMR, HRESIMS, UV, and single-crystal X-ray diffraction. Melonoid F (6) is the first example of cucurbitacin bearing an additional furan moiety at C-24. In vitro evaluation showed that compounds 1-7 protected against cisplatin-induced nephrotoxicity in a concentration-dependent manner, with cell viability increasing by 24%-35% relative to the model group at a compo
