Dual-functional fluorescent composite based on carbon dots‑europium ions: intelligent ratiometric detection and adsorption of tetracyclines.

Residual tetracycline antibiotics (TCs), especially tetracycline (TC) and doxycycline (DOX), pose critical threats to ecosystems and human health. There is an urgent demand for approaches capable of rapid, on-site monitoring and removal of TCs. Herein, a ratiometric fluorescent hybrid probe (HA-CDs/Eu3+) was engineered by integrating HA-CDs with Eu3+, featuring specific fluorescence response toward TCs (superior sensitivity to TC and DOX) and adsorption capacity. Upon the introduction of TC, quenching of the blue emission of HA-CDs occurred via the inner filter effect (IFE), while the red emis

Pharmaceutical residues in food samples: occurrence, toxicity, fluorescence quenching mechanisms, and analysis using carbon dot-based fluorescent probes.

Carbon dots (CDs) are an important class of carbon-based nanomaterials and represent a relatively new type of fluorescent nanomaterial that has attracted extensive research interest. Their popularity has increased rapidly due to several advantageous characteristics, including low toxicity, chemical stability, tunable fluorescence behavior, good water solubility, and favorable physicochemical properties. Owing to these features, CDs have been widely explored as sensing materials for the detection of various targets. In this review, pharmaceutical residues in food samples and their associated to

Does macrolide resistance play a role in the clinical outcome of patients with Mycoplasma pneumoniae infection?

BACKGROUND: A marked increase in Mycoplasma pneumoniae infections was observed across Europe in 2023, while data on resistance to macrolides, the recommended first-line therapy, remained limited. This study assessed the prevalence of macrolide-resistant M. pneumoniae (MRMP) and its impact on clinical outcomes. METHODS: Patients diagnosed with M. pneumoniae infection by multiplex PCR between January 2024 and January 2025 were included. Macrolide-resistance mutations were retrospectively identified by PCR and Sanger sequencing of domain V of the 23S rRNA gene. Clinical characteristics, treatmen

Clitocine for Translational Readthrough in Junctional Epidermolysis Bullosa.

Junctional epidermolysis bullosa (JEB) caused by laminin-332 deficiency (JEB-LAM332) is a severe inherited skin disorder characterized by impaired dermal-epidermal adhesion, chronic wounds, and increased cancer susceptibility. The management of JEB-LAM332 is primarily symptomatic, focusing on wound care and general supportive measures; however, many therapeutic needs remain unmet. While gene and stem cell therapy has been effective in JEB-LAM332, it is not yet broadly available owing to its complexity and cost. Translational readthrough-inducing drugs (TRIDs) represent an alternative strategy

Cerebrospinal fluid penetration of new antibiotics in the treatment of central nervous system infections.

BACKGROUND: Multidrug-resistant (MDR) bacterial infections present significant challenges to the management of central nervous system (CNS) infections, particularly in neurosurgical patients and those with cerebrospinal fluid (CSF) diversion devices. Achieving adequate CNS exposure is complicated by variable blood-brain barrier permeability and physiological changes, and CSF concentrations are not reliably predicted by plasma concentrations. Several new antibiotics have been introduced in response to rising MDR rates. To date, none are licensed for CNS infections, and data on CSF penetration a

Pneumococcal carriage and selected respiratory virus in children with clinical pneumonia in Salvador, Brazil during PCV10 era.

BACKGROUND: In 2010, Brazil introduced the 10-valent pneumococcal conjugate vaccine (PCV10GSK). Assessing pneumococcal carriage among children with clinical pneumonia in the context of a mature PCV10GSK program can inform decisions about use of next-generation PCVs. METHODS: We prospectively enrolled children aged 2-59 months meeting a standardized case definition for clinical pneumonia at two emergency care units in Salvador, Brazil. Nasopharyngeal specimens were tested for pneumococcus by culture and qPCR with serotyping by Quellung and/or PCR or qPCR; isolates underwent antimicrobial susce

Potential clinical actionability of metagenomic next-generation sequencing for antimicrobial stewardship for urinary tract infections: a retrospective, single-centre, BI-Biotia UTI cohort study.

OBJECTIVES: Culture and antimicrobial susceptibility testing (AST) take 48-72 hours, leaving empirical therapy for urinary tract infections (UTIs) without resistance data. Metagenomic next-generation sequencing (mNGS) reports resistance within hours, but its potential clinical actionability for prescribing is underexplored. METHODS: In a retrospective single-centre cohort (Bangkok tertiary hospital, 2018-2020), BIOTIA-DX results were compared with culture and AST in 251 culture-paired samples. Potential clinical actionability was assessed for 54 same-day empirical prescriptions and a post hoc

Campylobacter jejuni Across the Food Chain: Integrating Pathogenesis, Persistence, Antimicrobial Resistance, and Emerging One Health Control Strategies.

Campylobacter jejuni continues to be a globally important foodborne pathogen and is one of the most common bacterial causes of gastroenteritis in humans. This fastidious microaerophilic organism is seldom cultured outside of animal hosts, yet it repeatedly populates poultry, livestock, water, processing environments, retail foods, domestic kitchens, and humans. Here, we critically integrate the evidence in a transmission-connected, farm-to-fork pathway spanning colonization of animal reservoirs and carcass contamination to environmental persistence, human exposure, and disease (including antim

Gut microbial dynamics and antibiotic resistance during rifaximin therapy in severe cirrhosis: a ProPILA-Rifax ancillary study.

INTRODUCTION: Increasing evidence implicates gut dysbiosis in the pathophysiology of advanced cirrhosis. This is an ancillary study of the ProPILA-Rifax trial (NCT03069131), which evaluated rifaximin (RFX) as primary prophylaxis of spontaneous bacterial peritonitis (SBP) in patients with severe cirrhosis. Although the combined use of RFX and lactulose is effective in preventing recurrent hepatic encephalopathy (HE), its longitudinal impact on microbial communities and antibiotic resistance remains poorly characterized. METHODS: We characterized longitudinal changes in the gut microbiome of 42

Trials, tribulations, and formulations in access to new TB drugs for children: learning from rifapentine.

Historically, there has been considerable delay between stringent regulatory authority approval for new TB drugs in adults, and subsequent trials, registration, and access for children. This has been due to the exclusion of older children from phase 3 trials, the failure to study novel drugs in younger children earlier on, and delays in developing and testing paediatric formulations. Specific barriers include the failure to develop paediatric investigational plans early on, regulatory barriers, inefficient trial design and implementation, limited site capacity, and lack of advocacy and funding

Occult gram-negative bacterial colonization of explanted alloplastic implants incidentally detected during revision rhinoplasty: a microbiologically confirmed case report.

BACKGROUND: Postoperative infection following rhinoplasty is uncommon but can have serious consequences, particularly after revision procedures. Latent bacterial colonization and biofilm formation on implanted materials may contribute to delayed or subclinical infection, yet microbiologically confirmed colonization of implants in fully asymptomatic patients is rarely documented, and Gram-negative organisms are seldom reported in this setting. CASE PRESENTATION: A 35-year-old woman underwent revision rhinoplasty for a persistent nasal deformity after ten prior procedures involving alloplastic

A process-resolved risk prioritization framework reveals hidden antibiotic control deficiencies in wastewater treatment trains.

Antibiotic prioritization in wastewater treatment plants (WWTPs) commonly relies on influent-effluent changes. Such assessments may mask differences among configurations, treatment contexts, and endpoint media. Here, we established a process-resolved antibiotic Environmental Health Prioritization index (PA-EHPi). At the configuration × antibiotic scale, PA-EHPi integrates effluent occurrence, apparent removal, ecological risk, and antimicrobial resistance selection pressure. Across anaerobic-anoxic-oxic (AAO), sequencing batch reactor (SBR), and membrane bioreactor (MBR) treatment configuratio

Comparative study of antibiotic resistance and virulence profiles of exoU and exoS Pseudomonas aeruginosa isolates from urinary tract infections and microbial keratitis in Bangladesh and India.

Pseudomonas aeruginosa is an important opportunistic pathogen in urinary tract infections (UTIs) and microbial keratitis (MK), but differences in resistance and virulence between exoU and exoS P. aeruginosa isolates across regions and infection sites remain poorly understood. This study compared antibiotic resistance and virulence profiles of exoU and exoS P. aeruginosa isolates from Bangladeshi and Indian UTIs, and from Indian UTIs and MK collections. A total of 23 Bangladeshi UTI isolates were analyzed by broth microdilution for susceptibility to ciprofloxacin, levofloxacin, gentamicin, and

Coming Into Resistance: Clinically Important Antibiotic Resistance Genes in NICU vs. Healthy Infants.

IMPORTANCE: Antibiotic-resistant infections in neonates often arise from the infant's own colonizing bacteria harboring antibiotic resistance genes (ARG). This study evaluates ARG that impact antibiotic selection and drive resistance-related mortality, termed clinically important antibiotic resistance genes (CI-ARG). OBJECTIVE: To determine differences in gut CI-ARG abundance in children from birth through 3 years old, stratified by neonatal intensive care unit (NICU) admission and antibiotic (abx) exposure in the first two months. DESIGN: Cohort study (sequencing, analysis completed 2025-20

Exploring the evidence for use of cefiderocol as part of combination therapies: a systematic review of in vitro, in vivo, and clinical studies.

BACKGROUND: Cefiderocol is a siderophore cephalosporin with potent activity against carbapenem-resistant Gram-negative bacteria (CR-GNB). While monotherapy is recommended for susceptible Enterobacterales and Pseudomonas aeruginosa, the role of cefiderocol-based combination therapy remains uncertain. We conducted a systematic review of preclinical and clinical evidence supporting cefiderocol combination regimens. METHODS: MEDLINE, ESCMID global and IDWeek sources (January 2015-January 2025) were systematically searched for in vitro, in vivo and clinical studies evaluating cefiderocol-based com

Genetic characterization of gut-associated Clostridium perfringens isolates from Austrian animals using a combination of established and novel methods.

INTRODUCTION: Clostridium perfringens (C. perfringens) is a pathobiont of the intestinal microbiota that can cause severe systemic and enteric diseases. C. perfringens colonizes a variety of hosts and environmental niches, which is the theorized reason for its large genome, low guanine-cytosine content, and small stable core genome. Over 20 putative virulence factors have been described, but only six are included in the used typing scheme. Epidemiological analysis is limited, and to the authors' knowledge, this is the first study for Austria. Furthermore, C. perfringens frequently harbors diff
