Enantioselective Decarboxylative Synthesis of Chiral α-Silylamines and α-Germylamines.

Metalloids bridge the properties of metals and nonmetals, creating unique opportunities in drug discovery. However, the potential of silicon- and germanium-containing scaffolds in organic synthesis and medicinal chemistry remain underexplored, largely owing to the lack of general stereocontrolled synthetic methods, particularly for value-added chiral α-silylamines and α-germylamines. Here, we report a robust nickel-catalyzed enantioselective decarboxylative cross-coupling of amino acid or peptide-derived redox-active esters with silylzinc and germylzinc reagents. The method exhibits a broad su

HIV-1 cell-to-cell infection of macrophages escapes restriction by the BST-2 host cell factor independently of the Vpu counteracting viral protein.

As HIV-1 target cells, macrophages can be infected by cell-to-cell virus transfer from infected T cells through cell-cell fusion leading to the formation of virus-productive multinucleated giant cells (MGCs). We recently demonstrated that this route of infection escaped host cell restriction acting on the early steps of the viral life cycle. Here, we investigated whether restriction factors acting on late steps of HIV-1 replication were also bypassed in virus cell-to-cell infection of macrophages. We focused on the impact of the interferon(IFN)-induced factor BST-2/tetherin acting on virus rel

Chiral Boron Complex-Catalyzed Asymmetric [2π+2σ] Cycloaddition of β-Fluoroalkyl-α,β-Unsaturated Ketones with Bicyclo[1.1.0]butanes.

Bicyclo[2.1.1]hexanes (BCHs), three-dimensional benzene bioisosteres characterized by high sp3 carbon content, hold great promise for diverse applications in medicinal chemistry. While racemic BCH synthesis has advanced significantly, highly enantioselective approaches are still comparatively scarce. Here, we report an enantioselective [2π+2σ] cycloaddition of β-fluoroalkyl-α,β-unsaturated ketones with bicyclo[1.1.0]butanes (BCBs) catalyzed by a chiral (R)-3,3'-(3,5-(CF3)2-C6H3)2-BINOL-boron complex. This approach overcomes major limitations of existing metal-catalyzed and photochemical method

Electrochemical Synthesis of Macrocyclic Iminoisocoumarins by Rh-Catalyzed Intramolecular Vinylic C-H Activation/Heterocyclization.

Herein, we report a new macrocyclization strategy via electrochemical Rh(III)-catalyzed intramolecular vinylic C-H activation and subsequent heterocyclization. This approach affords diverse iminoisocoumarin-braced macrocycles with 13- to 28-membered rings bearing various functional groups in good yields with excellent regioselectivity under mild electrooxidation conditions. Mechanistic studies, including isolation and characterization of a key Rh(I) sandwich intermediate, along with cyclic voltammetry analysis, provide evidence for a rhodium(III/I) catalytic regime.

Palladium-Catalyzed Radical Annulation of α-Iodo-gem-Diborylalkanes with Alkenes: A Facile Approach to Semisaturated Cyclic Scaffolds.

Semisaturated cyclic scaffolds are increasingly valued in medicinal chemistry due to their balanced sp2/sp3 character and unique physicochemical properties, yet their synthesis often relies on lengthy de novo sequences. Herein, we report a palladium-catalyzed radical annulation of α-iodo-gem-diborylalkanes with alkenes, enabling a two-phase approach to these architectures from readily accessible precursors. The key transformation proceeds via a tandem Giese addition/homolytic aromatic substitution sequence and accommodates a broad range of (hetero)aromatic systems with diverse electronic prope

Effects of fenpropathrin on adult zebrafish reproduction and offspring development.

Fenpropathrin (FEN), a widely used insecticide, is frequently detected in aquatic environments, raising significant concerns about its ecological risks. In this study, a subacute exposure experiment was conducted on adult zebrafish exposed to FEN for 28 d to evaluate reproductive toxicity and developmental effects on the F1 and F2 generations, as well as underlying mechanisms. FEN exposure significantly increased the proportion of immature oocytes in females and decreased the percentages of spermatids and spermatozoa in males. These gonadal alterations were accompanied by dysregulation of game

Modelling herpes simplex virus type 2 and Zika virus replication in mouse vaginal organoids and human vaginal spheroids.

Sexually transmitted viral infections disproportionately affect women and have adverse outcomes on maternal and fetal health. Since they are acquired through the vaginal tract, the vaginal epithelium is an important site to study host-viral interactions. An in vitro model of mouse vaginal epithelial organoids that can be infected by herpes simplex virus was recently established. Here, we used multiple vaginal organoid systems and studied their response to infection by herpes simplex virus type 2 (HSV-2) and Zika virus (ZIKV), both of which can be sexually transmitted and result in adverse effe

A Sulfur-Modified Cryptolepine-Based Type I/II AIE Photosensitizer for Efficient Eradication of Planktonic Bacteria and Biofilms.

In recent years, the application of aggregation-induced emission photosensitizers (AIE PSs) in photodynamic therapy has gained significant traction as an effective approach for eradicating bacteria and biofilms. We developed a series of AIE PSs based on structural modifications of the natural product cryptolepine. Among them, TPA-CRYS exhibited exceptional type I/II reactive oxygen species (ROS) generation capability. At a low concentration of 20 nM, TPA-CRYS exerted potent antibacterial activity, achieving over 99.7% eradication of both Staphylococcus aureus and MRSA. Notably, TPA-CRYS effect

Random Hybrid Polypeptides Regulate Immune Signaling through Promiscuous and Dynamic Interaction Landscapes.

We developed a high-throughput synthesis and screening workflow combined with machine-learning-assisted optimization to identify functional compositions for immunomodulation in random hybrid polypeptides (RHPs). Using a selenium-containing polypeptide platform, we constructed a nucleoside-functionalized RHP library incorporating nucleoside, cationic, and anionic interaction motifs. Machine-learning-assisted optimization of this library identified a lead RHP, termed IDP-IMP (intrinsically disordered protein-inspired immunomodulatory polypeptide), that suppresses CpG DNA-Toll-like receptor 9 (TL

Pharmacokinetic diversity of baloxavir marboxil across rare avian species.

High pathogenicity avian influenza virus (HPAIV) poses a serious threat to captive rare birds. Because effective countermeasures are limited, the use of human antiviral drugs to treat HPAIV infections in captive rare birds has been considered. Although the human antiviral drug, baloxavir marboxil (BXM), is not approved for the treatment of influenza virus infection in birds, previous studies demonstrated the antiviral efficacy of BXM in experimental avian models. Pharmacokinetic studies have also revealed substantial interspecies differences following BXM administration, suggesting the need to

Crystal structures of SARS-CoV-2 main protease and its complex with PBI-0451.

The SARS-CoV-2 main protease (Mpro) has a pivotal role in mediating viral genome replication and transcription of the coronavirus, making it a prime target for antiviral drug development. This study presents four high-resolution crystal structures of SARS-CoV-2 Mpro in the apo state. Among these structures, we identify a previously unobserved, stable inactive conformation, characterized by a major displacement of the oxyanion loop (especially Phe140, Leu141 and Asn142), the rotation of some important residues (including His163 and His172) and the loss of many key interactions around the active

In vitro antiviral activity of the RdRp inhibitor 4'-fluorouridine against a porcine teschovirus clinical isolate.

Porcine Teschovirus (PTV) is an economically significant swine pathogen with a global distribution that can cause severe neurological, respiratory, and reproductive diseases. However, no specific vaccines or antiviral drugs are currently available. To address the absence of specific antiviral therapies for PTV, this study aimed to identify a clinically promising antiviral candidate through in vitro compound screening. We began by isolating and purified a contemporary PTV strain (PTV/Shandong/2024) from a clinical swab of a dead piglet. We also established a strain-specific RT-qPCR assay to acc

Mitochondrial Dysfunction in Diabetic Neuropathy: An Evidence Map of Molecular Mechanisms, Translational Biomarkers, and Emerging Implications for Oral Neuropathy-A Scoping Review.

Diabetic peripheral neuropathy (DPN) is a common and disabling complication of diabetes, but treatment still focuses largely on symptom control rather than modification of the underlying nerve injury. Mitochondrial dysfunction is increasingly implicated in the response of peripheral nerves to chronic hyperglycemia, although the literature is spread across different mitochondrial pathways, experimental models, biomarkers, and clinical settings. We therefore conducted a scoping review, following the Joanna Briggs Institute framework, to map this evidence and examine how far it has extended to or

Akkermansia muciniphila enhances chicken resistance to infectious bronchitis virus by boosting GABA synthesis and suppressing the NF-κB inflammatory pathway.

Infectious bronchitis virus (IBV) is a major viral pathogen causing substantial economic losses in the global poultry industry. However, the limited efficacy of commercial vaccines and the absence of approved antiviral drugs underscore the urgent need for novel strategies to combat IBV, particularly in the context of antibiotic-free poultry production. Emerging evidence suggests that gut microbiota plays a crucial role in shaping host immunity and antiviral resilience. In this study, we demonstrated that gut microbiota composition is a key determinant of IBV resistance in chickens, as revealed

Antiviral Activity of an Ethanolic Extract of Cinnamomum cassia and Cinnamaldehyde Against the Herpesviruses HSV-1 and VZV.

Herpes simplex virus type 1 (HSV-1) and varicella-zoster virus (VZV) are clinically important alphaherpesviruses that establish lifelong latent infections and remain significant causes of morbidity despite the availability of antiviral drugs. The emergence of drug resistance and the inability of current therapies to eliminate latent virus reservoirs underscore the need for novel antiviral agents with alternative mechanisms of action. In the present study, we evaluated the antiviral activity of an ethanolic extract prepared from commercial cinnamon bark marketed as Cinnamomum cassia and cinnama

DNAzymes and Oligodeoxyribonucleotides Immobilized on Aminopropylsilanol and Polylysine-Anatase Nanoparticles as Active Agents Against Influenza A/H5N1 Virus.

The nanocomposites based on aminopropylsilanol (Si~NH2) and polylysine-anatase (Ans/PL) nanoparticles bearing DNAzymes (Dz) and oligodeoxyribonucleotides (ON) can effectively inhibit the replication of the influenza A/H5N1 virus in cell culture. Nanocomposites based on both types of nanoparticles exhibit high activity. The Dz-containing nanocomposites exhibited higher antiviral activity than those containing ON, reducing influenza A/H5N1 virus production by 3.5 log versus 2.4-3.0 log, respectively. Notably, the effect of Dz incorporated into the nanoparticles was an order of magnitude higher t

Computational Drug Repurposing Reveals Lopinavir and Ritonavir as Potential Inhibitors of APOL Proteins for Combating Kidney and Neuropsychiatric Disorders.

INTRODUCTION: The Apolipoprotein L (APOL) family, particularly APOL1, APOL2, and APOL4, has been implicated in the pathogenesis of kidney injury, neuroinflammation, and several psychiatric disorders. However, there are currently no approved drugs available that can directly target these proteins, except for some APOL1-directed therapeutics. Identifying compounds that can modulate multiple APOL paralogs may therefore offer a promising therapeutic approach. METHODS: In this study, a structure-based drug repurposing approach integrating molecular docking and MD simulations was employed to identi
