[Surveillance of hypothyroidism treatment].

The efficacy of substitution treatment in primary or secondary hypothyroidism is based on the diappearance of the clinical signs of the disease and the return of blood levels of thyroxine and TSH within normal limits. The dosage should be adapted to the age of the patient the duration of the course of the hypothyroidism and the state of the heart. The cardio-vascular consequences of hypothroidism are so serious that one should know how to detect them straight away, and thus prevent complications with would prevent proper substitutive treatment.

The influence of beta-adrenoceptor blocking agents on plasma thyroxine and triiodothyronine.

The effect of the beta-adrenoceptor blocking agent propranolol on plasma levels of thyroxine and triiodothyronine was studied in eleven hyperthyroid patients and in six hypothyroid patients on L-thyroxine substitution therapy. In all patients a decrease in plasma-triiodothyronine was found. The per cent decrease of plasma-triiodothyronine was the same in both groups. Plasma levels of thyroxine and thyrotropin increased in the hypothyroid patients, but remained constant in the hyperthyroid subjects. The decrease in plasma-triiodothyronine during propranolol medication is most likely caused by a

[Characteristics of the structure and functional potentials of the metaepiphysis of tubular bones].

During early postnatal ontogenesis in the metaepiphyseal plate of the developing rat tibial bone, the borderline zone, the columnar cartilage zone and the zone of macrovesiculous cells differ in their cell composition, structure of basic substance and in functional and reactive potencies, which manifest themselves most distinctly after injection of growth-promoting hormones to the animals. The organic specificity of the metaepiphyseal borderline zone depends, first of all, on its intermediate position between the cartilage, fulfilling further bone growth lengthwise as an organ, and a developin

Thyroid hormones stimulate erythropoiesis in vitro.

Thyroid hormones have important effects on erythropoiesis in man and animals. We performed in vitro culture studies with murine and human bone marrow in order to define the interaction of these hormones with erythroid and granulocyte-monocyte progenitor cells. The methylcellulose clonogenic assay was used with the appropriate addition of erythropoietin or colony-stimulating activity. L-thyroxine, D-thyroxine and L-triiodothyronine potentiated erythropoientin-stimulated erythroid colony formation in concentrations of 50-100 ng/ml. These hormones had no effect on granulocyte-monocyte colony form

Effects of hypo- and hyperthyroidism on the activity of cystathionase in mammalian parenchymatous organs during early development.

In rats after neonatal thyroid destriction, cystathionae in liver and pancreas increased, but the enzyme activity in kidneys decreased. Substitution with thyroxine corrected these changes. Excess of thyroxine, too, had an effect on tissue cystathionase.

Urinary excretion of carnitine and serum concentrations of carnitine and lipids in patients with hypofunctional endocrine diseases: involvement of adrenocorticoid and thyroid hormones in ACTH-induced augmentation of carnitine and lipids metabolism.

The promoting effect of ACTH on carnitine and lipid metabolism was studied in patients with various endocrine hypofunctions. The results were compared with those of normal subjects. In adrenocortical insufficiency, hypothyroidism and hypopituitarism urinary excretion of carnitine was significantly lower than in normal subjects. On intramuscular injection of synthetic beta1-24 ACTH-Z urninary excretion of carnitine in normal subjects increased sixfold on the day of the injection and returned to the pretreatment level on the third day. Serum concentrations of carnitine and FFA increase in parall

[Neonatal hypothyroidism induced by the ingestion of potassium iodide during pregnancy (author's transl)].

This concerns a case of an infant born with goiter and hypothyroidism whose mother had taken potassium iodide during pregnancy in the form of a respiratory eupneic syrup. Laboratory tests revealed the etiology of the goiter by demonstrating that it concerned a transitory, acquired hormone production disturbance. The tests gave an indication of the course of the condition, leading to substitute hormone therapy being stopped after six weeks. The euthyroidism was then checked on two occastions.

Interaction between thyroid hormones and erythrocyte membranes: competitive inhibition of binding 131 I-L-triiodothyronine and 131 I-L-thyroxine by their analogs.

Molecular structural characteristics of thyroid hormones which influence binding to the erythrocyte membranes were investigated by competitive binding experiments. The ability of thyroid hormone analogs to displace 131 I-L-thyroxine and 131 I-L-triiodothyronine from the membranes was considered evidence of their competitive binding. The diphenyl ether linkage (thyronine) was essential as compounds with a single aromatic ring were weakly competitive. The presence of three iodine atoms at 3, 5 and 3' positions on thyronine was optimal for maximal competitive binding. There was weak competitive b

Analogues of human calcitonin. IV. Influence of leucine substitutions in positions 12, 16 and 19 on hypocalcaemic activity in the rat.

The replacement of the three aromatic amino acids in positions 12, 16 and 19 of human calcitonin (HCT) by leucine residues, which occupy the corresponding positions, in ultimobranchial, e.g. salmon and eel, calcitonins, increased the hypocalcaemic potency of the peptide, as determined by bioassay on the rat, about 10-fold. The individual substitutions were not all equally augmentative: leucine (12) enhanced the activity of HCT about 4--5 times, but leucine (16) and (19) afforded no increase at all. Combination of leucine (12) with a deamino cysteine at the N-terminus yielded an analogue 10 tim

Thyroparathyroid conditioning of hydrocortisone and glucagon-induced hypocalcemic effect in rats.

A single dose of 2.5 mg hydrocortisone induced hypocalcemia in both intact and parathyroidectomized rats. This effect disappeared in the thyroparathyroidectomized group and did not reappear after the substitution with parathormone. A single dose of 0.2 mg glucagon produced a significant hypocalcemic effect in intact, but not in parathyroidectomized rats. In the latter group, however, the hypocalcemic effect of glucagon reappeared after the administration of parathormone. It was concluded that the hypocalcemic effect of hydrocortisone may be conditioned by the presence of calcitonin secreting t

[Behavior of the TSH level in thyroidectomized patients after thyrotropin releasing hormone (TRH)].

Following operation or/and radioiodine therapy in 69 patients with malignant tumors of the thyroid gland the plasma level of TSH was increased independently of the level of thyroid hormone and of a clinical finding of hypothyroidism. In all cases it was possible to stimulate the secretion of TSH with TRH (p less than 0,0005). Therefore, application of TRH may be helpful to increase the uptake of iodine by metastases. On the other hand substitution of thyroid hormones--thyroxine or triiodothyronine or both combined--resulted in normal levels of TSH and normal response to TRH.

[Transitional symptoms, adverse effects and incidents in hormone-substitution therapy].

Patients underlying the permanent endocrine substitution need a particular control and a competent conduction on account of their endangering by intercurrent events. Highly specialised knowledge of the physician and intensive collaboration of the patient from this reciprocity lead to essential aspects of the prophylaxis of the crisis-like exacerbations exhibited in detail. The optimum substitution is supplemented by issuing information and emergency cards. When the patient possesses such cards they will become of decisive importance in an urgent therapy necessary outside the controlling facili

The importance of growth hormone and liothyronine in induced hypocalcaemia in hypophysectomized animals.

The resistance to hypocalcaemia induced by synthetic salmon calcitonin (SCT) was studied in intact and hypophysectomized rats. The recovery from hypocalcaemia was greatly impaired in the latter group, but this was normalised by small doses of purified human growth hormone (GH). The initial hypocalcaemic response to SCT was delayed in the hypophysectomized rats. This was not affected by treatment with GH, but normalised by substitution with liothyronine. We conclude that GH accelerates the recovery from hypocalcaemia in hypophysectomized animals, perhaps by a stimulation of parathyroid activity

Synthesis of new cyclic nucleotides and their differential stimulatory effects on thyroid function in mice.

Various 8-substituted cAMP analogues were converted into the corresponding N6,O2'-dibutyryl, N6-monobutyryl, or O2'-monobutyryl derivatives and the parent compounds as well as the new derivatives were tested for TSH-like stimulation of the thyroid function in mice in vivo by means of the McKenzie bioassay. It was found that a pronounced stimulatory effect on the thyroid secretion can be produced by certain cyclic nucleotides. The most potent compounds, 8H2N-cAMP, 8MeS-cAMP, and 8N3-cAMP at 15-30 mg/kg iv, showed an activity approximately one order of magnitude higher than that of cAMP and comp

Sheehan's syndrome with complete bone marrow aplasia: long-term results of substitution therapy with hormones.

A case of Sheehan's syndrome with pancytopenia and bone marrow aplasia is presented. The diagnosis was made 6 years after an ante-partum haemorrhage. The presenting feature was an anaemia unresponsive to usual therapy. Hormonal therapy alone (cortisone and thyroid extract with triiodothyronin, oestrogen and progestogen given sequentially) produced full haematological recovery, which has continued for 2 years since the treatment was begun.

[Therapy of acute pancreatitis].

The unpredictable course of acute pancreatitis needs a careful surveyance of the patient in the first days of the acute attack in order to apply therapeutic measures adequate to the severity of the symptoms. The avoidance of food or drink and gastric suction appears to be sufficient to prevent endogenous stimulation of the gland while there is probably no benefit of anticholinergic drugs or carboanhydrase inhibitors. Early adequate substitution of fluids using watery solutions, plasmaexpanders or blood is of decisive importance. For treatment of pains spasmoanalgetics, synthetic opium derivati

Radioimmunoassay of thyrotrophin releasing hormone in human serum and its clinical application.

A radioimmunoassay for thyrotrophin releasing hormone (TRH) in human serum is described. Inactivation of TRH immunoreactivity by serum is prevented by a mixture of 8-hydroxyquinoline sulphate and Tween 20. This persisted for a long period and at the concentration used in this study did not affect the TRH value. The TRH was extracted from serum with methanol. The results of the recovery experiment, dilution curve of high TRH serum, intra-assay reproducibility and inter-assay variation were quite satisfactory. The lowest detectable amount in this system was 5.0 pg/ml. Immunoreactive TRH levels i
