Combined oral contraceptives and pharmacokinetic drug interactions: impact on efficacy and safety.

INTRODUCTION: Combined oral contraceptives (COCs), commonly referred to as the 'pill,' are a reliable form of contraception used by a vast majority of women of reproductive potential. COCs contain two primary components: estrogens and progestins. In addition to protection against pregnancy, COCs also confer several significant non-contraceptive benefits, such as the treatment of polycystic ovary syndrome. Given that polypharmacy is a common practice, drug interactions between COCs and concomitant therapies can adversely impact efficacy and safety. AREAS COVERED: This article systematically re

Effect of extended hormonal suppression in patients with adenomyosis undergoing embryo transfer.

Adenomyosis is an estrogen-dependent uterine disorder increasingly recognized as a major cause of infertility and adverse obstetric outcomes, yet optimal medical preparation before embryo transfer (ET) remains uncertain. Adenomyotic lesions create a hyperestrogenic, inflammatory, and architecturally distorted uterine environment that may impair endometrial receptivity and increase miscarriage risk, compromising assisted reproductive technology (ART) success. This review summarizes current evidence on extended hormonal suppression in patients with adenomyosis undergoing in vitro fertilization a

Hormonal contraceptives and EEG biomarkers for antidepressant treatment response in women.

BACKGROUND: Women are disproportionately affected by major depressive disorder (MDD). Widely used hormonal contraceptives are linked to depression and altered brain function, yet their unstudied impact on EEG-biomarkers of antidepressant response may confound efforts to develop biomarkers for women. METHODS: In 60 unmedicated premenopausal women with MDD: non-users (n = 25), combined oral contraceptive users (COC, n = 19), and progestin-only contraceptive users (POC, n = 16), we assess five EEG-biomarkers before eight weeks of 10-20 mg escitalopram: Alpha peak frequency (APF), Vigilance level

Knowledge of High School Students About Cancers of the Genital System.

Cancers of the genital tract are one of the most common causes of morbidity and mortality in women in the fourth-fifth decade of life. The incidence of cancers of this system in men is also increasing, testicular cancer in the younger age groups, and prostate cancer in the fifth and subsequent decades of life. The purpose of this study was to analyse knowledge of college students about genital system cancer risk factors. The study included 3108 adolescents aged 15-19 years, students of the first and final grades of 20 high schools located in Mazovia (Poland). The participants were asked about

[Male contraception-scientific foundations and contemporary progress].

Despite decades of assiduous inquiry and burgeoning societal demand, contraceptive options for men remain circumscribed. Women have benefitted from highly efficacious hormonal modalities, but the burden of contraceptive responsibility continues to rest almost exclusively upon them. In light of the potential sequelae of female hormonal agents and the inviolable right to bodily autonomy, the quest for safe, reversible male methods is assuming urgency.Hormonal contraception for men relies on suppression of spermatogenesis through exogenous testosterone combined with progestins. Gene Expression Profiling of Peripheral Blood and Endometrial Cancer Risk Factors: Systems Epidemiology Approach in the NOWAC Postgenome Cohort Study.

INTRODUCTION: The increasing incidence of endometrial cancer (EC) requires an extensive search for novel preventive tools and early intervention approaches. However, the development of reliable predictive models is impossible without knowledge of genetic alterations prior to diagnosis. In this work, we aimed to establish whether known EC risk factors are associated with peripheral blood gene expression changes in a prospective design and whether such associations differ between women who later developed EC and matched controls. METHODS: First, we selected variables (parity status, lifetime nu

Sex hormones and arrhythmia risk in women with long QT syndrome.

Sex-related differences in the QT interval become evident from puberty to menopause. Sex steroid hormones have been suggested to play a role in cardiac repolarization, though their impact remains a matter of debate. These effects are especially pronounced in women with inherited long QT syndrome (LQTS) and may face a higher risk of QT variability associated with endogenous or exogenous hormonal fluctuations. This narrative review aimed to summarize the impact of physiological hormonal fluctuations on QT interval in women with LQTS across the female lifespan, including menstrual cycle, pregnanc

Breast cancer risk in peri- and postmenopausal women with a history of hormonal contraceptive use: A narrative review.

Breast cancer is the most common cancer among women worldwide, and its incidence is projected to increase substantially by 2050. As hormonal exposure is a key modifiable risk factor, understanding the impact of exogenous hormones-including hormonal contraception-is essential. Although hormonal contraceptives are primarily used during the reproductive years, they are increasingly prescribed to perimenopausal women for both contraception and symptom management. We conducted a structured literature review using Ovid MEDLINE, identifying studies on hormonal contraception and breast cancer risk, in

A concept for a new approach to combined oral contraception from adolescence to perimenopause: Continuous use of the oral GnRH antagonist Relugolix and the fetal estrogen Estetrol.

The introduction of oral GnRH antagonists has created the possibility to replace progestins in combined oral contraceptives as the suppressor of ovarian function. A concept for this new approach to oral hormonal contraception is the combination of 40 mg relugolix, known to inhibit ovulation, and a low dose of the fetal estrogen estetrol. Major noncontraceptive benefits of this regimen will be the establishment of continuous amenorrhea and a smooth and symptomless perimenopausal transition to the postmenopausal status, creating a life cycle endocrine management approach for women (the RE4Women

Negative selection assisted isolation of a highly selective DNA aptamer for the detection of 17α-ethinylestradiol.

Estrogenic compounds, including both natural estrogens such as estrone (E1) and 17β-estradiol (E2) and synthetic 17α-ethinylestradiol (EE2), are widely recognized for their endocrine-disrupting effects in the environment. EE2 is a key ingredient in oral contraceptives, and it poses environmental challenges due to its resistance to biodegradation and bioaccumulation in aquatic systems. In this work, EE2 was used as a target to select DNA aptamers, and extensive negative selections were employed using E2 as a counter target to obtain highly selective aptamers. After 18 rounds of selections, two

Effects of Initial Periodontal Therapy on Intraoral Bacterial Flora in Patients With Oral Contraceptive-Related Severe Gingivitis.

Gingivitis is a common inflammatory condition of the gingival tissues, influenced by microbial plaque and various systemic factors, including hormonal changes. Oral contraceptives containing estrogen and progestin are known to exacerbate gingival inflammation. We report the case of a 20-year-old Japanese woman who presented with gingival swelling and bleeding despite self-care, with a history of orthodontic treatment and oral contraceptive use. Clinical examination revealed probing pocket depths of ≥ 4 mm and bleeding on probing in more than half of the sites, a periodontal inflamed surface ar
