New cannabidiol structure-related terpene N-acyl-hydrazones with potent antinociceptive and anti-inflammatory activity.

Inflammation is the organism's protective mechanism to restore cellular and tissue homeostasis. Cannabidiol has been reported for its ability to bind to diverse receptors related to or not related to the endocannabinoid system, with good safety being one of the most promising phytocannabinoids for therapeutical purposes. CBD has shown in vitro and in vivo ability to significantly reduce the production of cytokines and other inflammatory mediators, with an unclear mechanism of action. Herein, we report the design and synthesis of a novel series of eight terpene N-acylaryl hydrazone analogues an

Enhanced Analysis of Curcuminoids in Turmeric via Selective Homodecoupled 1D 1H NMR.

Curcuminoids, including curcumin, demethoxycurcumin, and bisdemethoxycurcumin, are vital for quality control in food, nutraceuticals, and pharmaceuticals. Conventional 1D 1H NMR can face challenges in spectral interpretation when dealing with overlapping signals and complex coupling patterns, especially in structurally similar compounds like curcuminoids. This study explores the use of selective homodecoupled 1D 1H NMR spectroscopy as a complementary technique to enhance spectral resolution and facilitate peak assignment in curcuminoid analysis. By collapsing multiplet structures such as doubl

Epilepsy and autophagy modulators: a therapeutic split.

Epilepsy is a neurological disease characterized by repeated unprovoked seizure. Epilepsy is controlled by anti-epileptic drugs (AEDs); however, one third of epileptic patients have symptoms that are not controlled by AEDs in a condition called refractory epilepsy. Dysregulation of macroautophagy/autophagy is involved in the pathogenesis of epilepsy. Autophagy prevents the development and progression of epilepsy through regulating the balance between inhibitory and excitatory neurotransmitters. Induction of autophagy and autophagy-related proteins could be a novel therapeutic strategy in the m

Evaluation of dose linearity in the systemic availability and pharmacokinetics of topically administered diclofenac: A 14C-microdosing study in healthy volunteers.

An important safety consideration for topically administered drugs is the extent of systemic exposure they achieve. The aim of this study was to evaluate whether a topically administered microdose of the model drug diclofenac can predict the systemic availability, plasma, and tissue pharmacokinetics of a topical therapeutic dose. Eight healthy participants (6 men and 2 women) participated in a 4-period, crossover study. In period 1, a topical microdose (62 ± 6 μg) was administered; in period 2, a single intravenous microdose (0.95 ± 0.03 μg) was administered; in period 3, a topical therapeutic

Assessment of the molecular mechanisms of drug-induced hidden cardiotoxicity by a multi-omics approach: The example of rofecoxib.

BACKGROUND AND PURPOSE: Hidden cardiotoxicity is defined as drug-induced cardiotoxicity that becomes obvious only in the presence of comorbidities. However, the molecular mechanisms of hidden cardiotoxicity are not always known. Therefore, unbiased multi-omics approaches could assist in revealing regulatory pathways. The most notable representative of hidden cardiotoxic drugs is the cyclooxygenase-2-inhibitor, rofecoxib. We previously reported increased mortality in rats because of proarrhythmic effects of rofecoxib in ischaemic hearts. Here, we aimed to identify molecular mechanisms of hidden

Investigation of renal COX-2 distribution of prenatally administered diclofenac sodium in postnatal rats using stereological and immunohistochemical methods.

While cyclooxygenase-2 (COX-2) plays a critical role in embryonic kidney development. Inhibition of this enzyme by nonsteroidal anti-inflammatory drugs (NSAIDs), commonly used during pregnancy, can adversely affect fetal kidney development. In this study, we examined the postnatal expression and distribution of COX-2 in the kidneys of murine offspring born to mothers administered diclofenac sodium, a COX-2 inhibitor, during gestation, using stereological and immunohistochemical techniques. Twelve female Wistar albino rats were divided into diclofenac sodium (DSG) and control groups (CG). From

A Cross-sectional In Vitro Study on the Synergistic Neuroprotective Effects of Phytochemicals Ferulic Acid and Ginkgolide B against Amyloid Beta-induced Oxidative Stress and Modulation of Multifunctional Enzyme APE1/Ref-1 in Human Neuroblastoma SH-SY5Y Cells.

The Apurinic/Apyrimidinic endonuclease 1 (APE1/Ref-1) is essential for controlling the oxidative DNA base damage via the Base Excision Repair (BER) pathway. While APE1's N-terminal region is important in redox function (named redox effector factor 1, Ref-1) and protein-protein interactions, its C-terminal region is involved in DNA repair activities. In this work, we employed the Aβ25-35 peptide to produce oxidative stress in order to assess the protective effects of two phytochemicals, ferulic acid (FA) and Ginkgolide B (GB), individually and in combination, with a focus on APE1 in different s

Deciphering the formation rule and anti-retrogradation potential of starch/ferulic acid/ovalbumin composite gels: A ratio modulation and interaction perspective.

To improve the quality of starch gels and enhance their anti-retrogradation behavior, this work developed a wheat starch composite gel system incorporating ovalbumin (OVA) at various ferulic acid (FA) ratios. RVA revealed that the incorporation of OVA and FA effectively facilitated the starch pasting process. FT-IR analysis indicated that OVA and FA disrupted the intermolecular hydrogen bonding network of starch during the modification process, consequently leading to a reduction in the energy storage modulus of the gel. This reduction in the energy storage modulus of the gel was observed. Con

CD-MOF Based pH/ROS Sensitive 5-ASA Delivery System for Ulcerative Colitis Therapy.

5-Aminosalicylic acid (5-ASA) is the first-line drug for ulcerative colitis (UC), which encounters therapeutic constraints due to the poor water solubility and absorption by the upper gastrointestinal. Here, a cyclodextrin-based metal-organic framework (CD-MOF) is used as a carrier for 5-ASA (ASA@CM) to enhance its solubility. To improve the targeted release 5-ASA, ASA@CM is incorporated into sodium alginate/L-arginine complex (SA-LA) to obtain the desired microspheres (ASA@CM/SL) by electrospray. ASA@CM/SL safeguards 5-ASA against the harsh gastrointestinal environment and provides favorable
