Gut Microbiota Mediate the Metabolism of Colonic Prostaglandins.

Prostaglandins (PGs) are endogenous lipid signaling molecules that regulate diverse physiological and pathological processes, and their biosynthetic pathways are targets of widely used drugs such as aspirin and other nonsteroidal anti-inflammatory drugs (NSAIDs)1-7. However, previous research of PG biosynthesis has focused on host metabolic pathways; as such, the contribution of gut microbiota remains unknown1. Here, we demonstrate a colonic pathway in which gut microbes directly participate in PG metabolism, thereby regulating intestinal PG levels and their biological effects. Comparison of g

Curcumin for the Treatment of Inflammatory Bowel Disease: From Mechanism to Clinic.

Inflammatory bowel disease (IBD) is characterized by chronic symptoms, frequent relapses, and an increased risk of complications such as colorectal cancer. These qualities significantly impair patients' quality of life, and while various treatment options are available, they are often limited by inconsistent responses, unclear long-term safety, considerable individual variability, restricted drug accessibility, high costs, and the complexity of combination therapies. Such challenges thus highlight the urgent need for novel, safer, and more effective treatments. Curcumin, a natural compound wit

Withanolides from Datura stramonium: two unprecedented B-ring-seco derivatives and anti-inflammatory active analogues.

UV-guided separation of the Datura stramonium L. extract led to the elucidation of thirteen previously unreported withanolides (1-13), together with nine known analogues (14-22). Compounds 1 and 2 are unprecedented withanolides featuring B-ring cleavage and bearing two formyl (-CHO) groups. The structures of the new withanolides were elucidated on the basis of UV, HRESIMS, and NMR data. The inhibitory effects of the isolated compounds on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in BV2 microglial cells were evaluated. Compounds 16 and 22 exhibited potent in vitro anti-infla

Network modeling of sporadic colorectal cancer reveals the importance of off-target effects of Cyclooxygenase inhibitors.

Cyclooxygenase (COX) inhibitors (COXIBs) have shown preventive and therapeutic potential for colorectal cancer (CRC). In addition to inhibiting COX, approved COXIBs also target COX-independent pathways, including NF-κB and AKT. We evaluated how inhibition of various COXIB targets affects cell proliferation and apoptosis using a Boolean model incorporating the effect of several genetic mutations linked to CRC. The mutations activate two positive feedback loops and cause increased cell proliferation and survival. When simulating loss of function of APC, proliferation and apoptosis can be restore

Modelling of Rheological Properties of Pharmaceutical Powder Mixtures for Direct Compression: A Statistical Approach.

The rheological properties of pharmaceutical mixtures are exceedingly difficult to understand and control. Despite their impact on final product quality, no generally valid approach to define and predict mixture rheology has been discovered so far. The cause is a complexity of factors affecting the particulate mixture behaviour. This study aimed at addressing part of this problem by suggesting an approach to evaluate and subsequently predict flow properties of binary particulate mixtures from parameters of individual components to better guide the formulation development. This study investigat

Integrative ensemble learning framework for forecasting controlled drug release based on Raman spectral signatures.

Modeling drug-release kinetics from polysaccharide-coated oral controlled-release formulations remains challenging due to nonlinear diffusion–dissolution behavior, complex polymer–drug interactions, and the limited interpretability of conventional machine-learning approaches. In this study, we develop and validate a predictive framework for targeted colonic delivery of 5-aminosalicylic acid (5-ASA) from polysaccharide-coated solid oral dosage forms using Raman spectroscopy–derived molecular fingerprints and time-resolved dissolution data. The dataset comprises 155 formulation samples, each cha

Formulation and Characterization of Multiparticulate System for Proton Pump Inhibitor using Combination Therapy for Peptic Ulcer.

INTRODUCTION: The purpose of this study was to develop sustained-release enteric-coated granules of Amla extract and esomeprazole magnesium for the treatment of peptic ulcer. It is well known that Amla possesses anti-ulcer activity due to the phenolic compound gallic acid, which inhibits a gastric H+/K+ ATPase pump. Therefore, enteric-coated Amla extract granules were combined with esomeprazole magnesium granules to enhance the synergistic effect, mitigate adverse effects associated with esomeprazole magnesium, and improve overall anti-ulcer activity, while the enteric-coated multiparticulate

Adhesive Capsulitis in a Postmenopausal Woman With Thyroid Dysfunction.

Adhesive capsulitis, also known as frozen shoulder, is characterized by progressive pain and loss of shoulder mobility due to capsular inflammation and fibrosis. This is a case of a 50‑year‑old postmenopausal woman with a history of Graves' disease treated by radio‑iodine ablation who presented with a year‑long history of gradually worsening left‑shoulder pain and stiffness. Imaging revealed mild acromioclavicular arthrosis and supraspinatus tendinosis without rotator cuff tear, findings consistent with adhesive capsulitis. Conservative management with physiotherapy, home exercise, and non‑ste

Interplay between solid state properties and tissue-associated release of acetylsalicylic acid from PLGA microneedles across in vitro, ex vivo and in silico models.

Despite the fact that acetylsalicylic acid (ASA) remains clinically important for inflammatory conditions, conventional oral administration is hampered by gastrointestinal adverse effects and variable systemic exposure. Whilst polymeric microneedles (MNs) enable transdermal delivery as an alternative to oral delivery through bypassing the stratum corneum; how drug solid-state behaviour affects the release kinetics of sustained-release MN systems has yet to be thoroughly elucidated. Herein, ASA-loaded poly (lactic-co-glycolic acid) (PLGA) MNs were developed to examine how retained but altered A

Design and Optimization of Indomethacin Nanocrystals using Machine Learning and Molecular Dynamics Simulations.

In this study, a stable nanocrystalline drug delivery system for indomethacin (IND) was rapidly developed by integrating machine learning methods with Hummer Acoustic Resonance (HAR) technology. This system effectively enhanced the solubility of IND and demonstrated excellent scalability. High-throughput screening using HAR technology identified P188-PVA as the optimal composite stabilizer for IND nanocrystal suspensions. Molecular dynamics simulations were employed to thoroughly investigate the interaction mechanisms between the drug and stabilizers. Systematic design and optimization of IND

Celecoxib enhances explosive exercise capacity in hypoxic mice and attenuates glutathione-associated oxidative stress in hypoxic C2C12 cells.

Under hypoxic conditions, exercise capacity declines, accompanied by the significant activation of the arachidonic acid (AA) metabolism. The AA metabolism pathway has been confirmed to participated in the regulation of hypoxic inflammation. However, the relationship between hypoxia and AA metabolism remains largely unclear and whether targeting AA metabolism represents an effective strategy to improve exercise capacity is not clear. Celecoxib, a cyclooxygenase-2 (COX-2) selective inhibitor, has been showed to effectively inhabit AA metabolism with minimal side-effects. In this study, celecoxib

Predicting in vivo performance of piroxicam and apalutamide drug nanocrystals from in vitro flux measurements combined with in silico modelling.

The development and optimization of pharmaceutical nanocrystal formulations is often complex and resource-intensive, as particle properties depend on composition, manufacturing parameters, and batch variability, all of which influence in vivo performance. Predictive in vitro-in silico approaches can streamline formulation development by reducing the need for extensive animal and clinical testing. This study reports on a methodological evaluation of an integrated workflow combining dissolution-permeation data with a mechanistic absorption model based on the gastrointestinal unified theoretical

Ischemic Stroke in a 28-Year-Old Young Adult Associated With Chronic Triptan and Non-Steroidal Anti-Inflammatory Drug (NSAID) Use.

Ischemic stroke in young adults is an uncommon but clinically significant condition that frequently occurs in the absence of traditional cardiovascular risk factors. In this population, identification of nontraditional and potentially iatrogenic contributors is essential for accurate diagnosis and effective secondary prevention. We report the case of a 28-year-old woman with normal body mass index who presented with acute-onset expressive aphasia accompanied by right-sided motor and sensory deficits. Her medical history was notable for chronic migraine treated with daily sumatriptan use over t

Characterizing the frequency of clinical events and assessing biomarkers in propionic acidemia: a natural history study.

BACKGROUND: Propionic acidemia (PA) is a rare disease resulting in toxic accumulation of metabolites that cause metabolic decompensation events (MDEs). Natural history characterization of PA is limited partly due to its rarity and complexity. This study examined clinical events in participants with PA and their correlation with biomarkers to better understand the disease. METHODS: Participants diagnosed with PA (early- versus late-onset) were evaluated every 6 months from baseline to month 36. Outcome measures included annualized MDEs and changes in 3-hydroxypropionic acid (3-HP), 2-methylcitr

NMR Crystallography at 1 GHz: Insight into a Rich World of Binary Forms of Meloxicam.

NMR crystallography, which is based on both diffraction- and solid-state NMR-based observables, can deliver a precise description of the crystal structure of powdered samples of pharmaceuticals. On the other hand, it is by no means a straightforward technique, not least because of the overcrowding of solid-state NMR spectra and difficulties in reliably assigning all resonances, even when advanced 2D techniques are applied. In this work, we showcase the advantages of using very high magnetic field spectrometers in NMR crystallography to solve the structural puzzles posed by four new binary form
