A ShbHLH34-ShPUR Module Activates ShTPS1-mediated β-caryophyllene Biosynthesis for Insect Resistance in Sugarcane.

Terpenoids represent one of the largest and most structurally diverse classes of plant secondary metabolites, playing essential roles in defense against herbivorous insects. Despite their importance, the mechanisms regulating terpenoid biosynthesis remain largely unexplored. Here, in sugarcane, we identified and characterized a terpene synthase gene from Saccharum spp. Hybrids, ShTPS1, which is strongly induced by feeding from fall armyworm Spodoptera frugiperda and sugarcane stem borer Chilo infuscatellus. Enzymatic assays in vitro revealed that ShTPS1 catalyzes the biosynthesis of volatile t

ZmTPS18-mediated β-caryophyllene biosynthesis contributes to resistance against southern corn rust in maize.

Southern corn rust (SCR), caused by the obligate biotrophic fungus Puccinia polysora Underwood, is a major foliar disease of maize associated with substantial yield losses in many production regions. Terpenoids are important components of plant defense, but the terpene synthases and upstream regulators involved in pathogen-induced terpenoid biosynthesis in maize are still poorly defined. In this study, volatile profiling showed that β-caryophyllene emission was induced in the SCR-resistant maize inbred line Yanzi F7016K after pathogen inoculation, reaching levels 3.18-fold higher than those in

Successful treatment of hairy cell leukemia with TP53 abnormality using cladribine combined with low-dose rituximab: a case report and literature review.

Hairy cell leukemia (HCL) is a rare mature B-cell malignancy characterized by the BRAF V600E mutation. TP53 abnormalities in HCL are associated with poor response to standard therapies and a higher risk of relapse. We report an elderly male patient diagnosed with classical HCL (BRAF V600E-positive) who also presented with a TP53 deletion detected by fluorescence in situ hybridization (FISH) and bone marrow fibrosis. The patient was treated with one cycle of cladribine combined with low-dose rituximab (100 mg). The treatment was well tolerated. At the 7-month follow up, the patient achieved com

Long-term comparative outcomes of rituximab and belimumab in lupus patients.

OBJECTIVES: Although rituximab and belimumab are recommended for systemic lupus erythematosus (SLE) and lupus nephritis, direct long-term comparative evidence is limited. We aimed to compare the effectiveness of rituximab versus belimumab in a real-world setting for treating SLE. METHODS: This prospective, single-centre, observational cohort study analysed SLE patients who initiated biologics in the BILAG-BR registry at Addenbrooke's Hospital (2011-2025). We compared treatment periods with rituximab and belimumab among patients stable on therapy, without switching or entering clinical trials.

Ultrasound-modulated whey protein isolate fibrils for curcumin delivery: Structural changes, functional properties, and bioaccessibility.

Whey protein isolate fibrils (WPIF) are promising biomacromolecular carriers, but their entanglement-prone structure and limited interfacial activity restrict hydrophobic bioactive substance delivery. This study evaluated the effects of ultrasound power (0-600 W) and treatment time (0-15 min) on WPIF structural characteristics, functional properties, and curcumin (Cur) delivery efficiency. Circular dichroism, FTIR, intrinsic fluorescence, and surface hydrophobicity analyses revealed that moderate ultrasound disrupted intermolecular interactions, reduced β-sheet content, and exposed hydrophobic

Emulsion gels stabilized by millet chaff prolamin-gallic acid or -sucralose conjugates: characterization, application in curcumin, and mechanism insights.

To develop a safe, low-cost, and efficient curcumin delivery system, emulsion gels were prepared using millet chaff prolamin (MCP) and the conjugates of MCP with gallic acid (MCP-GA) and MCP with sucralose (MCP-S). Gallic acid-binding and sucralose-glycosylation improved the emulsifying properties of MCP, increased hydrophobic interactions (from 0.43 to 0.67), crystallinity (from 27.34% to 67.58%), bound water content, and viscosity of the MCP-emulsion gel, and reduced the loss factor, thereby improving its gel structure, thermal and oxidative stability, hardness, elastically, and cohesiveness

Pullulan acetate with tailored degree of substitution as a promising polymer for oral drug delivery: Effects on enzymatic degradability and drug release.

This study investigates how the degree of substitution (DS) governs the behaviour of polysaccharide-based materials relevant for oral drug delivery, using pullulan acetate (PA) as a model system. A PA library with controlled DS (0.1-2.8) was synthesised via acetylation with acetyl chloride to enable systematic evaluation of structure-property relationships, including physicochemical properties, enzymatic accessibility, and drug-release performance. Structural and functional characterisation was performed using spectroscopic and thermal techniques, while solubility and compressibility were asse

Early preeclampsia screening and prevention: An update for general practice.

BACKGROUND: Preeclampsia is a disorder of pregnancy that causes significant adverse perinatal outcomes. Risk factor-based screening performs poorly when compared with algorithm-based approaches. The role of general practitioners in early screening for preeclampsia is crucial in predicting and preventing the condition. OBJECTIVE: This article discusses the different screening approaches and interventions available for prevention of preeclampsia, making the case for a screen-and-treat model based on individual risk calculation for all pregnant women. DISCUSSION: The Fetal Medicine Foundation (

Joint-brain communication in rheumatoid arthritis: glial mechanisms, pain phenotypes, and translational evidence.

Rheumatoid arthritis (RA) is a systemic autoimmune disease in which persistent pain, fatigue, cognitive impairment, depression, and anxiety may involve altered central nervous system (CNS) processing. This narrative review synthesizes preclinical and clinical evidence concerning proposed glial cell-mediated joint-brain communication in RA. Joint-derived neural inputs and systemic inflammatory signals may communicate with the CNS through humoral, cellular, neural, neuroendocrine, and neurovascular pathways. Preclinical studies implicate microglia and astrocytes in neuroinflammatory responses, s

Dihydroquercetin, a naturally occurring flavonoid, has limited applications in food systems because of its poor aqueous solubility and low bioaccessibility. A food-grade delivery system was developed to improve the stability and bioaccessibility of dihydroquercetin. Alkali heat treatment was applied to modify the conformational structure and enhance the functional properties of quinoa protein. Covalent quinoa protein-tannic acid nanoparticles were prepared under alkaline conditions and used to stabilize Pickering emulsions for dihydroquercetin encapsulation. The nanoparticles were comprehensiv

Low-dose colchicine: a potential augmenting agent in the treatment of refractory mood disorders.

Treatment-resistant mood disorders, including major depressive disorder (MDD) and bipolar disorder (BD), remain a significant clinical challenge. A confluence of emerging evidence implicates chronic low-grade inflammation in the pathophysiology of at least a subset of mood disorders, potentially mediating their poor response to traditional therapies. Colchicine, a well-established anti-inflammatory agent used in gout and more recently (at low dose) cardiovascular disease, inhibits microtubule polymerisation and suppresses NLRP3 inflammasome activation. Recent advances in immunopsychiatry sugge

Targeting the NLRP3 inflammasome in rheumatoid arthritis fibroblast-like synoviocytes: pathogenic mechanisms, therapeutic strategies, and translational perspectives.

Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease characterized by persistent synovitis, pannus formation, cartilage degradation, and bone erosion. Although conventional synthetic, biologic, and targeted synthetic disease-modifying antirheumatic drugs have improved disease control, many patients fail to achieve sustained remission, lose responsiveness over time, or develop treatment-limiting safety concerns. Fibroblast-like synoviocytes (FLSs) are increasingly recognized as long-lived pathogenic stromal cells that sustain synovial inflammation, promote matrix degradation, supp

Low-Cost Methotrexate Chemoselection to Predict Radiotherapy Response in Advanced Oral Squamous Cell Carcinoma: A Prospective Feasibility Study From South India.

IntroductionOral squamous cell carcinoma (OSCC) represents a disproportionate cancer burden in South Asia, where advanced-stage presentation, limited radiotherapy infrastructure, and constrained chemotherapy resources necessitate pragmatic approaches to treatment selection. Platinum-based combinations remain the standard for induction chemotherapy; however, a substantial proportion of OSCC patients present with malnutrition, poor performance status, and limited financial resources that preclude these regimens. This is a hypothesis generating feasibility study which explores whether early respo

Neuroimmune lipidome dysregulation in major depressive disorder: A meta-analysis of peripheral endocannabinoid and N-acylethanolamine signaling.

Major depressive disorder (MDD) is a prevalent psychiatric condition characterized by substantial biological heterogeneity. Evidence suggests that the endocannabinoid system (ECS), which regulates stress responses, emotional processing, and neuroimmune signaling, may contribute to depression pathophysiology. However, clinical studies examining circulating endocannabinoids (eCBs) and N-acylethanolamines (NAEs) in MDD have produced inconsistent findings. We therefore conducted a systematic review and meta-analysis to quantify peripheral alterations in eCBs and NAEs in individuals with MDD. Follo
