Rat embryos were exposed to aspirin or its metabolite, salicylic acid in culture. In these embryos acute reduction of heart beat was observed during 4 hours of administration compared to that in non-treated one. Protein contents and crown-rump length of cultured embryos were significantly decreased in aspirin-treated group, but were not so decreased in salicylic acid-treated one. The predominant defects of the embryos exposed to aspirin were edematous facial malformations and abnormality of tail. The effects of progesterone 10 mg/kg, estrogen 5 mg/kg and a combination of both at half these doses were studied in rats. Ulcers were induced in rats by means of various experimental models: drug-induced ulcers (aspirin or indomethacin), stress ulcer and Shay rat. Gastric ulcerations were produced in rats by oral administration of aspirin (ASA) suspended in a vehicle consisting of either water or increasing concentrations of HCl (0.005 M to 0.35 M). Studies in guinea pigs showed that some forms of drug-induced bronchospasm are reflexogenic involving afferents in the glossopharyngeal nerve. At least two pathways appear to be involved. One pathway contains H1 receptors and is blocked by mepyramine and sodium cromoglycate (SCG), and its pharmacological characteristics are similar to those of active reflex vasodilation. The other appears to involve peripheral muscarinic receptors. The findings also indicate that SCG may act on efferent as well as afferent pathways.

Seven cases of analgesic nephropathy due to excessive ingestion of paracetamol are reported. None of these patients had been taking any other analgesic. All had radiological features of papillary necrosis. With the increasing use of paracetamol as a mild analgesic it is necessary to be aware of the possibility that paracetamol may induce analgesic nephropathy.

A survey is given on the damaging effect of acetylsalicylic acid, other nonsteroidal anti-inflammatory drugs (NSAID) and corticosteroids on the gastroduodenal mucosa.
