Co-designing a study on perspectives toward pharmacogenomic testing in mental health care among youth and caregivers.

AIMS: Finding suitable psychotropic medications can be a lengthy trial-and-error process, leading to poor symptom control, low adherence, and treatment failure. Pharmacogenomic (PGx) testing can help inform prescribing considerations for some individuals. As PGx becomes more common in Canada, understanding patients' and families' hopes, expectations, and concerns is vital. MATERIALS AND METHODS: This article details our work engaging two advisory councils comprised of five youth with lived/living experience of mental health challenges and five caregivers across four videoconferencing sessions

Designing evaluation infrastructure for psychedelic harm reduction at events: a methodological framework with formative mixed-methods field application.

Psychedelic and other psychoactive substance use is increasing in non-clinical environments, including music festivals, concerts, and other large-scale gatherings. As psychedelic use expands beyond clinical settings, policymakers, healthcare systems, event operators, and researchers require evidence-informed frameworks for understanding and responding to psychoactive-related distress in naturalistic environments. Despite the growing presence of psychedelic harm reduction services, few studies have established reproducible methods for documenting their operation, coordination with event-health

Prochlorperazine inhibits the proliferation of hepatocellular carcinoma cells by promoting phosphorylation of β-catenin.

Hepatocellular carcinoma (HCC) is the most prevalent form of primary liver cancer and ranks as the third leading cause of cancer-related fatalities worldwide. Recently, prochlorperazine (PCZ), a synthetic antipsychotic medication, has gained attention for its potential anticancer properties. However, the specific mechanisms underlying its anticancer effects remain unclear. In this study, we have identified that PCZ effectively inhibits the proliferation of HCC cells and diminishes the number and size of HCC foci in the mouse model. Subsequent studies have revealed that PCZ can directly bind to

Change in HbA1c and weight after initiation of antipsychotic treatment in people with severe mental illness in England: an individual-level interrupted time-series analysis.

BACKGROUND: Second-generation antipsychotics contribute to excess cardiometabolic morbidity in people with severe mental illness, but the timing of early glycaemic change and whether it parallels weight gain remain unclear. We aimed to evaluate glycated haemoglobin A1c (HbA1c) and weight trajectories before and after initiation of antipsychotic treatment. METHODS: We conducted an individual-level interrupted time-series analysis in England using electronic health records (EHR) from the Clinical Practice Research Datalink Aurum database between Jan 1, 2000, and June 7, 2023. Included participa

In Utero Valproic Acid Exposure in Cryptorchidism Reports: Integrating FAERS, Toxicogenomics, Transcriptomics, and Mendelian Randomization.

To screen the FDA Adverse Event Reporting System (FAERS) for valproic acid (VPA)-associated cryptorchidism reports, restrict developmental interpretation to documented transplacental/in utero exposure, and prioritize candidate genes without claiming mechanistic validation, we analyzed 7,232 deduplicated primary-suspect VPA reports in males aged ≤18 years from 2004Q1-2025Q4. Cryptorchidism screening identified 108 reports; exposure-route review defined the prenatal subset. Disproportionality analyses used pediatric male FAERS reports with primary-suspect drugs as the background. Curated human V

Parkinsonism and psychopathology severity in real-world outpatients with schizophrenia: A cross-sectional PRISM study.

Extrapyramidal symptoms (EPS) have been described as intrinsic to schizophrenia spectrum disorders (SSD), beyond medication-induced effects. Among EPS, parkinsonism is the most prevalent subtype and has been linked to poorer clinical and functional outcomes. We aimed to examine the relationships between parkinsonism and psychopathology and social functioning, as well as its demographic and pharmacological correlates, in a real-world sample of outpatients with schizophrenia receiving stable antipsychotic medication. Data were drawn from the PRISM project, an EU-funded, multicentre, cross-sectio

Intramuscular pharmacological treatment and mechanical restraint duration in acute psychiatric settings: Implications for nursing practice - A retrospective observational study.

BACKGROUND: Mechanical restraint is used in psychiatric settings despite international efforts to minimize restrictive practices. Intramuscular medication is routinely administered during the restraint episode as rapid tranquillization with the aim of achieving behavioral control and earlier removal, but evidence regarding its effect on restraint duration in psychiatric inpatient settings is lacking. AIM: To examine whether intramuscular pharmacological interventions are associated with restraint duration in acute psychiatric wards using standardized dosing. METHODS: Retrospective observatio

Short-term lithium treatment modulates excitation/inhibition balance in resting-state electroencephalography (EEG) among survivors of medically severe suicide attempts.

Short-term lithium treatment may reduce impulsivity and suicide risk by stabilizing cortical signaling in high-risk individuals. The present double-blind, placebo-controlled, crossover study enrolled survivors of medically severe suicide attempts and recorded 2-min eyes-open resting-state electroencephalography at baseline (drug-naïve), after 4 weeks of lithium carbonate, and after 4 weeks of placebo (n = 10 completers). Analysis focused on spectral power across traditional frequency bands (delta through gamma) and the aperiodic slope of the power spectrum, a marker of excitation/inhibition ba

Psilocybin's effect on human brain synaptic plasticity.

Psychedelics such as psilocybin have been linked to enhanced neuroplasticity and symptom relief in affective disorders, but the neurobiological mechanisms and impact of environmental context remain unclear. Here, we tested whether a single dose of psilocybin (0.3 mg/kg) alters synaptic density in healthy individuals and whether setting-dependent subjective experience shapes this effect. Fifteen healthy participants had a psilocybin-induced psychedelic experience either inside an MRI scanner or in a therapeutic-like room. We assessed synaptic density changes by measuring the Synaptic Vesicle gl

Enhancement of hippocampal synaptic AMPA receptor function by the antidepressant tianeptine and by opioid peptides.

Management of major depressive disorder (MDD) with conventional therapeutics that enhance monoaminergic neurotransmission requires prolonged treatment prior to clinical benefit and a substantial proportion of patients show little or no improvement. Identifying alternative molecular targets is required to develop efficacious, rapid acting antidepressants. In this regard, tianeptine, an established but atypical clinical antidepressant, is of interest. Tianeptine does not directly influence monoamine levels; in rodents it has a relatively rapid onset of antidepressant-like efficacy and, unusually

Skin and Brain in Bipolar Disorder: A Review of Dermatologic-Psychotropic Drug Interactions.

Importance: Dermatologic and psychotropic therapies intersect frequently in patients with bipolar disorder, in whom systemic dermatologic medications may affect mood stability and mood stabilizers or antipsychotics may produce clinically important cutaneous adverse reactions. Because several medications discussed in this review are also used for psychotic disorders, severe or treatment-resistant major depressive disorder, suicidality, and other psychiatric conditions, this article distinguishes medication-related risks from considerations specific to bipolar disorder. Observations: This narrat

Mdivi-1 augments chlorpromazine's anticancer effect via modulation of mitochondrial dynamics, hallmarks of cancer-related cellular behavior, and oxidative stress.

The multifaceted therapeutic potential of phenothiazines was previously documented. This study investigated the anticancer effect of chlorpromazine (CPZ) in hepatoma and breast cancer cell lines and further explored the implications of mitochondrial dynamics. To establish this aim, the cytotoxic and antimitotic effects of CPZ combined with the mitochondrial fission inhibitor (mdivi-1) were assessed in HepG2 and MDA-MB-231 cells using MTT and clonogenic assays, respectively. Apoptosis and nuclear changes were evaluated with Annexin V/PI, acridine orange/ethidium bromide, and DAPI staining. More

Olanzapine for the Prevention and Treatment of ICU Delirium: Protocol for a Systematic Review With Meta-Analysis.

BACKGROUND: Intensive care unit (ICU) delirium is a multifactorial acute brain dysfunction associated with prolonged hospitalization, increased morbidity and mortality, and higher healthcare costs. Despite these substantial negative impacts of delirium, the optimal prevention and treatment strategies are unclear. Olanzapine, an atypical antipsychotic drug, is commonly used for delirium management. We will perform an updated systematic review and meta-analysis to assess the benefits and harms of olanzapine for the prevention and treatment of ICU delirium. METHODS: This protocol is developed in

LSD reconfigures cortical dynamics through faster brain rhythms and increased fractal dimension.

Lysergic acid diethylamide (LSD) profoundly alters conscious experience, yet the electrophysiological mechanisms by which it reshapes neural dynamics remain incompletely understood. A hallmark of psychedelic states is widespread cortical desynchronization, typically inferred from reductions in spectral power, but whether such effects reflect genuine weakening of neural oscillations or are confounded by shifts in oscillatory peak frequencies remains unresolved. We combine source-resolved magnetoencephalography (MEG), spectral parameterization, temporal complexity metrics, and interpretable mach
