Neurobehavioral and reinforcing effects of pregabalin in mice.

Pregabalin misuse is increasing, but its psychostimulant and rewarding properties remain unclear, especially regarding sex differences and interactions with opioids. The objectives of this study are to investigate pregabalin's behavioral and neurochemical effects in male and female mice, including its impact on cognition, social interaction, reinforcement, and cross-sensitization with morphine. CD-1 mice received pregabalin (30-90 mg/kg, i.p.) acutely or for 5 days. Locomotor activity, Y-maze performance, social behavior, and conditioned place preference (CPP) were assessed. Brain monoamine le

Can the incentive-sensitization theory of addiction incorporate addiction to opioid drugs?

The Incentive Sensitization Theory (IST) of addiction posits that repeated intermittent exposure to potentially addictive drugs can sensitize brain mesolimbic dopamine systems. Those systems normally attribute incentive salience to rewards and their cues, but when sensitized may produce compulsive cue-triggered 'wanting' for drugs that can persist long after the discontinuation of drug use and the cessation of withdrawal symptoms, thus contributing to an enduring propensity to relapse. Much of the original evidence for IST came from studies on psychostimulant drugs, such as amphetamine and coc

[Oxytocin versus dopamine: The delicate balance between social bonding and addiction].

Social behaviour is fundamental to survival and adaptation, relying on complex, interacting neurobiological systems in which the neuropeptide oxytocin (OT) and the neurotransmitter dopamine (DA) play pivotal roles in regulating social bonding, motivation, and reward processing. This review synthesises recent advances in understanding the bidirectional interactions between OT and DA within key nodes of the brain's reward circuitry, including the nucleus accumbens, ventral tegmental area, amygdala, and medial prefrontal cortex. We examine how these interactions support a wide range of social beh

Disposition and effects of alpha-pyrrolidinoisohexanophenone (α-PHiP) in comparison with cocaine: an observational study.

INTRODUCTION: Alpha-pyrrolidinoisohexanophenone (α-PHiP) is a synthetic cathinone structurally related to alpha-pyrrolidinopentiophenone (α-PVP) and other pyrrolidinophenones, acting as a potent monoamine transporter inhibitor. Despite its raising prevalence on the recreational drug market, limited data exist about its human pharmacology. This study aimed to assess its concentrations in different matrices and characterize its acute effects in humans in comparison with cocaine when administered by intranasal route. METHODS: A non-controlled observational study was conducted under naturalistic

4-MEC potentially triggers CAV1 via the BDNF-TrkB signaling pathway.

4-Methylethcathinone (4-MEC), a synthetic cathinone with psychostimulant properties, is increasingly abused as a "designer drug". However, its molecular mechanisms, particularly those related to neuroplasticity regulation, remain poorly understood. Caveolin-1 (CAV1) is a scaffolding protein of membrane lipid rafts and has been confirmed to organize multiple synaptic signaling proteins to regulate synaptic signaling and neuroplasticity. Herein, we investigated whether CAV1 modulates 4-MEC-induced alterations in the BDNF-TrkB signal pathway and neuroplasticity markers in human SH-SY5Y neuroblast

Incretin-Based Therapies: A Novel Pathway in Addiction Treatment.

Addiction poses a major global public health challenge. It is characterized by high prevalence, chronic relapse and limited efficacy of available pharmacotherapies across different substance use disorders. Increasing evidence demonstrates that incretin-based therapies directly modulate metabolic signaling pathways that intercross with central reward and motivational circuits, including hypothalamic-mesolimbic networks and dopaminergic neurotransmission. As a result, agents such as glucagon-like peptide 1 receptor agonists, originally developed for the treatment of type 2 diabetes and obesity,

Sulforaphane Attenuates Methamphetamine-Induced Neurotoxicity via Activation of the PI3K/AKT Pathway.

Methamphetamine (METH) is a widely abused psychostimulant known to induce neurotoxicity through oxidative stress and apoptosis. Sulforaphane (SFN), a natural compound found in cruciferous vegetables, has shown neuroprotective potential, but its role in METH-induced neuronal injury remains unclear. In this study, we integrated network pharmacology together with in vitro and in vivo experiments to investigate the protective effects of SFN and its underlying mechanisms. A total of 235 overlapping targets were identified between SFN and METH-related neurotoxicity, with significant enrichment in ap

Microinjection of CART peptide into the nucleus accumbens medial shell attenuates methamphetamine-induced anxiety-like behaviors via restoration of GABAB receptor membrane expression.

Methamphetamine (METH) abuse often leads to anxiety-like behaviors, and the cocaine- and amphetamine-regulated transcript (CART) peptide, which is abundant in the nucleus accumbens (NAc), has shown promise in mitigating behavioral effects of psychostimulants. However, the role of CART peptide in the NAc medial shell in METH-induced anxiety remains poorly understood. In this study, following METH withdrawal and reinstatement, rats exhibited significant anxiety-like behaviors. Immunofluorescence analysis revealed a substantial increase in the proportion of c-Fos⁺/NeuN⁺ and CART⁺/NeuN⁺ cells with

Prescription psychostimulants, atomoxetine and the risk of psychosis in adults with history of psychosis: a population-based cohort study.

Concerns exist that psychostimulants may increase the risk of psychosis, particularly in individuals with a history of psychosis. This study assessed whether psychosis risk increases after initiating psychostimulant or atomoxetine treatment in individuals with prior psychotic events. In this cohort study, we used Swedish population-based register data that contain data on all Swedish residents. We identified adults aged 18-64, who had a history of psychosis and initiated psychostimulant or atomoxetine treatment between 2008 and 2021. A within-individual design compared the rate of psychotic ev

Sigma receptor ligands in selected affective disorders, psychoses, and addictions - a narrative review.

Sigma receptors (SRs), comprising the sigma 1 (S1R) and sigma 2 (S2R) subtypes, represent a distinct class of intracellular proteins that differ fundamentally from classical neurotransmitter receptors. S1Rs act as ligand-operated molecular chaperones primarily localized at mitochondria-associated membranes (MAM) of the endoplasmic reticulum (ER), whereas S2Rs, identified as transmembrane protein 97 (TMEM97), function as modulators of cellular homeostasis. Both subtypes are densely expressed in brain regions critical for emotional and cognitive processing, including the prefrontal cortex (PFC),

AT1R in Central Nervous System Disorders: Unveiling Novel Mechanisms and Therapeutic Avenues for Addiction.

Although the angiotensin II type 1 receptor (AT1R), a pivotal component of the renin-angiotensin system (RAS), is associated with cardiovascular and renal homeostasis, burgeoning evidence implicates its critical role in neuropsychiatric disorders, particularly addiction. Beyond regulating haemodynamics, AT1R activation in the central nervous system (CNS) modulates neuroinflammatory cascades, dopaminergic signalling plasticity, and stress-responsive neural circuit processes central to addiction pathophysiology. Notably, preclinical studies reveal that AT1R blockade attenuates drug-seeking behav

Optimizing cognitive enhancement through dopamine transporter modulation.

Psychostimulants act on monoamine systems including the dopamine transporter (DAT), a key regulator of dopamine reuptake following synaptic release. While these compounds can enhance energy, cognition, and sociability, their clinical utility is often limited by their abuse potential and peripheral side effects. Research has traditionally focused more on their addictive properties rather than their therapeutic potential. However, certain psychostimulants, such as R-modafinil, enhance cognitive performance without inducing significant euphoria or addiction, making them promising lead candidates

Endothelial adenosine A2A receptor activation-mediated Tau hyperphosphorylation leads to blood-brain barrier breakdown in a chronic methamphetamine mouse model.

INTRODUCTION: Methamphetamine (METH), a psychostimulant, can cause the blood-brain barrier (BBB) breakdown through astrocyte endfeet swelling and endothelial cell impairment. Our previous studies have shown that phosphorylated microtubule-associated protein Tau (p-Tau) levels increase, especially in neurons, after METH exposure. However, whether p-Tau accumulates in the endothelial cells of METH mouse brains remains unclear. The effects of endothelial p-Tau accumulation on BBB integrity in a chronic METH mouse model are elusive. METHODS: The A2AR-GSK3β signaling pathway protein levels were ev

Preclinical and Emerging Clinical Evidence Supporting the Translational Potential of GLP-1 Receptor Agonists for Treating Psychostimulant and Nicotine Use Disorders.

Psychostimulant use disorder (PSUD) and nicotine use disorder (NUD) affect tens of millions of people worldwide, yet no clinically approved medications exist for PSUD, and existing NUD treatments have limited long-term efficacy. Recent preclinical evidence suggests that glucagon-like peptide-1 receptor (GLP-1R) agonists could be repurposed for treating PSUD and NUD. This review summarizes the current literature investigating the efficacy of GLP-1R agonists to reduce psychostimulant and nicotine-mediated behaviors in animal models of PSUD and NUD, respectively. The neural mechanisms underlying

Postnatal social stress impairs cognitive function, while acute methamphetamine enhances performance in adult male rats.

Methamphetamine (MA) is a potent psychostimulant frequently abused during pregnancy, with potential long-term consequences for offspring neurodevelopment. In addition to prenatal exposure, early-life stress is a critical factor influencing cognitive function in adulthood. This study aimed to evaluate the combined effects of prenatal MA or injection stress, early postnatal stressors (social and/or physical), and acute MA administration in adulthood on cognitive performance in male rats. Pregnant female Wistar rats received MA (5 mg/kg, s.c.), saline (1 ml/kg, s.c.), or no treatment throughout g
