Fusobacterium nucleatum enhances amphetamine-induced behavioral responses through a butyrate-driven epigenetic mechanism.

Amphetamines are psychostimulants that are commonly used to treat neuropsychiatric disorders and are prone to misuse. The pathogenesis of amphetamine use disorder (AUD) is associated with dysbiosis (an imbalance in the body's microbiome) and bacterially produced short-chain fatty acids (SCFAs), which are implicated in the gut-brain axis. Amphetamine exposure in both rats and humans increases the amount of intestinal Fusobacterium nucleatum, which releases SFCAs. Here, we found that colonization of gnotobiotic Drosophila melanogaster with F. nucleatum or supplementing the flies' diet with the S

How does methamphetamine affect the brain? A systematic review of magnetic resonance imaging studies.

Methamphetamine (METH) is an addictive psychostimulant that is associated with significant physical and psychological harm. Magnetic resonance imaging (MRI) is a non-invasive imaging modality used to identify structural and functional brain changes. We conducted a systematic review to identify and appraise the existing literature on brain MRI differences associated with METH exposure. A comprehensive literature search was conducted on PubMed, Web of Science, and Scopus. Any study that evaluated the brain in people who used or had been exposed to METH using MRI were included. Data were collecte

Methamphetamine Exposure Induces Neuronal Programmed Necrosis by Permeabilizing Mitochondria via the RIPK1-RIPK3-MLKL Axis.

Methamphetamine (Meth), a psychostimulant drug of the amphetamine type, is widely abused and highly neurotoxic. Meth exposure leads to neuronal necroptosis, and the mitochondrial dysfunction may be involved. However, the underlying mechanisms remain poorly understood. Here, we found that Meth significantly elicited the formation of the RIPK1-RIPK3-MLKL necrosome complex. Intriguingly, the activated MLKL (p-MLKL) translocated to the mitochondrial membrane and displayed pore-forming activity, manifesting as the penetration of MLKL in the cell membranes of the mitochondria, which caused decreased

Unlocking the Potential of High-Quality Dopamine Transporter Pharmacological Data: Advancing Robust Machine Learning-Based QSAR Modeling.

The dopamine transporter (DAT) plays a critical role in the central nervous system and has been implicated in numerous psychiatric disorders. The ligand-based approaches are instrumental to decipher the structure-activity relationship (SAR) of DAT ligands, especially the quantitative SAR (QSAR) modeling. By gathering and analyzing data from literature and databases, we systematically assemble a diverse range of ligands binding to DAT, aiming to discern the general features of DAT ligands and uncover the chemical space for potential novel DAT ligand scaffolds. The aggregation of DAT pharmacolog

Causal Mechanisms of Monoamine Transporter Phosphorylation.

Monoamine transporters are essential proteins located at presynaptic terminals that play a crucial role in regulating neurotransmission of serotonin, dopamine, and norepinephrine by rapid reuptake of released amines from the synapse. Clinically used antidepressants and widely abused psychostimulants exhibit a high affinity for amine transporters. Function and expression of biogenic amine transporter are altered in subjects suffering from psychiatric diseases such as depression and in psychostimulant use disorder. Therefore, proper functional regulation of monoamine transporters is critical in

Novel Ways of Targeting the Dopamine Transporter.

Dopamine (DA) is an important modulatory neurotransmitter that is involved in daily activities such as movement, memory, and reward-oriented learning of essential behaviors and needs. DA signaling is initiated by the release of DA into the synaptic cleft that will bind to dopamine receptors to mediate the physiological response. To terminate the DA response, the DA is taken up by the dopamine transporter (DAT), a surface membrane protein. Psychostimulants, like cocaine and amphetamine, both target DAT and interfere with the DA uptake process, resulting in an increased amount of DA in the synap

Transcriptomic markers of psychoactive substance addiction in peripheral blood.

The regular use of psychoactive substances, resulting in substance use disorders, represents a significant public health concern globally. Due to genetic variability and the substantial impact of environmental factors, identifying specific genes associated with predisposition to addictive behaviors presents a significant challenge. Regular exposure to psychoactive substances is known to change gene expression levels in the brain regions for reward and motivation. These changes affect the dopaminergic, glutamatergic, and cannabinoid systems of the brain. The existing data on cellular gene expre

Role of RET-Regulated GDNF-GFRα1 Endocytosis in Methamphetamine-Induced Neurotoxicity.

Methamphetamine (METH) is a highly addictive synthetic psychostimulant that can induce severe neurotoxicity, leading to neurodegeneration similar to neurodegenerative diseases. The endocytosis of glial cell line-derived neurotrophic factor (GDNF) and its family receptor alpha 1 (GFRα1), regulated by transmembrane receptor tyrosine kinase (RET), has been shown to resist neurodegeneration. Specifically, the endocytosis of GDNF-GFRα1 mediated by RET is crucial in protecting neurons. Although many molecular mechanisms of METH induced neurotoxicity have been explored, the obstacles to the neuroprot

Discovering next-generation dopamine transporter drugs with lower abuse potential.

Many traditional psychostimulants used to treat neuropsychiatric disorders by targeting the dopamine transporter (DAT) exhibit a high potential of abuse. This necessitates the development of next-generation drugs with improved safety. Discovering atypical DAT inhibitors represent a promising strategy that may be facilitated by integration of structural pharmacology and artificial intelligence.

Structure-Function Activity Relationships of Cariprazine Analogues with Distinct G Protein versus β-Arrestin Activities at the Dopamine D3 Receptor.

Psychostimulant use disorder (PSUD) remains an unmet medical need, with no FDA-approved pharmacotherapies currently available. The dopamine D3 receptor (D3R), due to its selective expression in mesolimbic reward circuits, has emerged as a compelling target for PSUD intervention. We used cariprazine (1a), a D3R-preferring antipsychotic that reduces cocaine-related behaviors in preclinical models, as a scaffold for the synthesis of a library of novel derivatives. We employed BRET-based assays to functionally characterize their effects on G protein and β-arrestin2 signaling at D3R. Structure-func

Cocaine and Morphine Converge to Disrupt Chloride Homeostasis in Ventral Tegmental Area GABA Neurons.

Identifying shared neural mechanisms influenced by diverse classes of drugs of abuse is essential for understanding addiction and for developing broad-spectrum treatments for substance use disorders. Previous studies indicate that many drugs of abuse increase dopamine output from the ventral tegmental area (VTA) by altering the balance of excitatory and inhibitory inputs onto dopamine neurons, thereby promoting maladaptive plasticity within reward circuits. Here, we demonstrate in rats that acute injections of morphine and cocaine, but not saline, disrupt chloride homeostasis in VTA GABA neuro

tRF-M2-Regulated Dopamine Receptor D2 Expression Attenuates Methamphetamine Reinstatement Behavior in Rats.

Methamphetamine addiction is a chronic disorder characterized by compulsive drug-seeking and high relapse rates, driven by dopamine-mediated neuroadaptations in reward circuits that induce persistent behavioral and synaptic changes that endure despite abstinence, making treatment challenging. In this study, we demonstrated significant downregulation of the tRNA-derived small RNA (tsRNA) tRF-M2 (tRF-1:32-Gly-GCC-2-M2) in the nucleus accumbens (NAc) of rats exposed to methamphetamine self-administration (METH SA). Functional neuroanatomy of dopaminergic arousal systems: implications for the wake-promoting effect of psychostimulants, with particular reference to modafinil.

Arousal involves activation of the cerebral cortex by inputs from subcortical (hypothalamic, brainstem) wake-promoting nuclei, utilizing monoamine (noradrenaline, dopamine, serotonin, histamine) and neuropeptide (orexin) neurotransmitters. Dopaminergic neurones of the midbrain, originating from distinct nuclei [pars compacta of substantia nigra (SNc), ventral tegmental area (VTA), and other clusters of dopaminergic neurones in the ventral periaqueductal gray (vPAG)] and the pontine dorsal raphe nucleus (DRN), constitute a powerful wake-promoting system. Cortical activation by dopaminergic neur

Functional Variation in RGS12 Should Not Preclude Methylphenidate Use in Bipolar Disorder with Established Mood Stabilization: Preclinical Evidence.

Our goal in this study was to determine whether functional variation in the human RGS12 gene influences behavioral responses to psychostimulants such as methylphenidate, thereby informing whether such genetic findings should affect the clinical use of this central nervous system (CNS)-stimulating agent in bipolar disorder (BD) patients with comorbid attention-deficit/hyperactivity disorder (ADHD). The use of psychostimulants for ADHD in BD remains controversial due to concerns about mood destabilization, although recent systematic reviews indicate that methylphenidates and amphetamines can be

Integrated Assessment of the Cardiotoxic and Neurobehavioral Effects of 3,4-Methylenedioxypyrovalerone (MDPV) in Zebrafish Embryos.

Synthetic cathinones such as 3,4-methylenedioxypyrovalerone (MDPV) are potent psychostimulants with high abuse potential, yet their systemic toxicity and neurobehavioral effects remain poorly characterized during early development. Using Danio rerio (zebrafish) embryos and larvae, we performed an integrated assessment of the cardiotoxic, behavioral, and molecular effects of MDPV. Acute exposure of 3 days post-fertilization (dpf) embryos produced a marked, concentration-dependent bradycardia and atrioventricular (AV) conduction block, leading to reduced ventricular activity and complete AV diss

From Stress to Substance Use Disorders: The Expanding Role of Microglia-Astrocyte Crosstalk in Neuroimmune and Glutamate Alterations in the Nucleus Accumbens.

This review examines convergent neurobiological mechanisms linking stress and drugs that drive stress-induced drug-related behaviors. It first outlines the main theoretical frameworks explaining substance use disorders (SUDs), emphasizing vulnerability factors-particularly stressful life events-that increase addiction risk. The analysis integrates preclinical evidence demonstrating that chronic stress facilitates cross-sensitization to psychostimulants and accelerates drug self-administration, underscoring how stress and drugs converge on glutamatergic and dopaminergic transmission within the
