INTRODUCTION: Expansive processes around the third ventricle have always been a major neurosurgical challenge. Despite all the technological and scientific progress recorded over the last few years, third ventricle tumors are still a very difficult pathology to approach due to their formidable complexity.Treating such a tumor demands a good knowledge of local anatomy, pathophysiology, pathology and a good capacity to integrate all the data gathered from the patient prior to and during surgery. Last but not least, the correct means to approach such a tumor involves using modern neuronavigation

Myasthenia gravis (MG) is an autoimmune disease caused by the production of specific autoantibodies to various components of the neuromuscular synapse, leading to muscle weakness and disabling fatigue. Treatment of MG aims to stop the symptoms and inhibit the triggers of the autoimmune process. For a long time, MG treatment included anticholinesterase agents and nonspecific immunosuppressive and immunomodulatory therapies used alone or in combinations: glucocorticosteroids, cytostatics, plasmapheresis, and intravenous immunoglobulin. Despite the fact that the above drugs are widely used in the

Despite being present in many drugs, guanylhydrazones and semicarbazones are two functional groups that have been little investigated as potential therapeutic strategies for the treatment of Alzheimer's disease (AD). For this reason, we initiated the synthesis and evaluation of these compounds as potential anticholinesterase agents, aiming to offer new alternatives for drug development against AD. In the severe phase of AD butyrylcholinesterase (BChE) becomes the main enzyme responsible for the hydrolysis of acetylcholine (ACh). Therefore, in this project, we present the results of BChE inhibi

INTRODUCTION: In sub-Saharan Africa, autoimmune myasthenia gravis (AMG) is still poorly known and underdiagnosed (delayed diagnosis, poor availability and accessibility of proven effective diagnostic and therapeutic tools), resulting in a poor functional prognosis and high mortality. The aim of the present study was to evaluate the therapeutic and clinical course of AMG in Burkina Faso.
PATIENTS AND METHODS: This was a longitudinal, multicenter study conducted from March 2015 to April 2023. It included patients with clinical signs suggestive of myasthenia associated with the presence of serum

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that affects millions of individuals worldwide. Treatment strategies for AD vary depending on cognitive and behavioral symptoms, with cholinergic replacement therapy using acetylcholinesterase (AChE) inhibitors being one of the primary approaches. Recent studies have also identified human carbonic anhydrases (hCAs) as significant therapeutic targets for AD, offering new opportunities for the development of innovative treatments. Carbonic anhydrase inhibitors have been shown to prevent early mitochondrial damage and inhibit H2

Alzheimer's disease (AD) is the most prevalent form of dementia all around the globe and currently poses a significant challenge to the healthcare system. Currently, available drugs only slow the progression of this disease rather than provide a proper cure for it. Acetylcholinesterase (AChE) enzyme is mainly responsible for the progression of AD, and its management can be done by developing its inhibitors, such as anticholinesterases. We have synthesized a series of novel dihydropyrimidine-oxadiazole hybrids (8a-8ad) via one-pot economic Biginelli reaction using cobalt perchlorate catalyst. T

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by a set of multifactorial conditions that progressively impair memory processing and cognitive function. The study of this pathology is particularly challenging due to its complex etiology, which involves several pathological hallmarks, including amyloid plaque formation, tau protein hyperphosphorylation, neuroinflammation, oxidative stress, and other contributing factors-all leading to neuronal loss. The primary therapeutic approach for AD involves the use of anticholinesterase agents; however, these treatments are associ

Ferrocene-1,2,3-triazole hybrids have emerged as a new class of compounds with significant potential as anticholinesterase agents. These hybrids uniquely combine the biological properties of ferrocene and the 1,2,3-triazole moiety, both of which are known for their stability and versatile interactions with biomolecules. In this study, the synthesis of various ferrocene-1,2,3-triazole derivatives (9-15) was carried out. All synthesized compounds were characterized using standard techniques. The crystal structures of compounds 10, 13, and 14 were elucidated. Their inhibitory effects on acetylcho

The article discusses ambulation disorders in Parkinson's disease, which result from both disease progression and aging. It focuses on multisensory impairments and the critical link between cognitive and motor disorders, particularly the motor-cognitive deficit. Diagnosing ambulation disorders presents several challenges because it's essential to identify the specific cause in each case. Key factors include the ineffectiveness of standard antiparkinsonian therapy, varying degrees of cognitive impairments (including visual and spatial difficulties), and the presence of comorbidities. The potent

Bungarus multicinctus is one of the most venomous and lethal snake species in mainland China, with envenomation resulting in a mortality rate as high as 23%. Currently, antivenin against B. multicinctus remains the gold standard for treating bites from this species. However, in remote mountainous areas or rural regions of China, access to antivenin may be delayed or unavailable following a bite. The lethal components of B. multicinctus venom include α-, β-, γ- and κ-bungarotoxins, which act on either the presynaptic or postsynaptic membrane, thereby altering acetylcholine levels in the synapti

Pyridostigmine (Mestinon®, Bausch Health, Canada Inc.) increases acetylcholine availability at the neuromuscular junction, enhancing transmission. Preclinical studies suggest that neuromuscular junction dysfunction develops early in ALS, and pyridostigmine may temporarily improve neuromuscular transmission. However, altered neuromuscular junction transmission has uncertain benefits in ALS progression. Pyridostigmine does not have other plausible mechanisms that truly modify ALS pathophysiology. People with ALS (PALS) who have positive acetylcholine receptor autoantibodies and no myasthenia sym

Since the development of the "cholinergic hypothesis" as an important therapeutic approach in the treatment of Alzheimer's disease (AD), the scientific community has made a remarkable effort to discover new and effective molecules with the ability to inhibit the enzyme acetylcholinesterase (AChE). The natural function of this enzyme is to catalyze the hydrolysis of the neurotransmitter acetylcholine in the brain. Thus, its inhibition increases the levels of this neurochemical and improves the cholinergic functions in patients with AD alleviating the symptoms of this neurological disorder. In r

BACKGROUND: Alzheimer's disease (AD) is a progressive neurodegenerative disease that compromises the cognitive system and causes dementia. In general, AD affects people over 65 years old, which implies a social impact if we consider future projections due to the increase in life expectancy. The drugs currently marketed only slow the progression of the disease. In this sense, the search for new drugs is a relevant topic in medicinal chemistry. The therapeutic strategy adopted herein is the cholinergic hypothesis, for which acetylcholinesterase enzyme (AChE) inhibitors constitute the main treatm

Synthesis of new substituted pyrrole scaffolds containing substituted thiadiazol-2-amine moiety was successfully developed through one-pot and multi-component tandem condensation reaction utilizing of triethyl ammonium hydrogen sulfate ([Et3NH][HSO4]) ionic liquid as a green media under solvent-free conditions. The chemical structures of all newly synthesized compounds were fully characterized by spectroscopic methods (IR, 1H NMR, 13C NMR) and elemental analyzes. The molecular docking studies were also performed to predict the possible binding sites of the derivatives on the active site gorge

Amphibians represent one of the main natural sources of bioactive molecules of interest to biotechnological research. The Phyllomedusidae family has several species occurring in Brazil and some studies demonstrate the biological potential of poisons of these species, however many still need to be characterized. Phyllomedusa iheringii is endemic in Brazilian and Uruguayan Pampa Biome and has little data in the literature regarding the action of its poison on experimental organisms. Thus, the present work evaluates the biological activity of P. iheringii secretion on the central and peripheral n

Five selected medicinal plants in the family Zingiberaceae, Alpinia nigra, Boesenbergia maxwellii, Etlingera araneosa, Meistera koenigii, and Wurfbainia biflora, were collected from Thailand. HPLC was used to determine the content of phenolic compounds in their ethanolic extracts. The total phenolic contents and the total flavonoid contents of M. koenigii were found to be higher than that of the others. The antioxidant activity of the extracts was assayed by two methods, namely ABTS and DPPH assays using HPTLC and a microplate spectrophotometer. Anticholinesterase activity was screened by the

The use of drugs has undeniable benefits to the elderly, but it is not exempt from undesirable effects. Deprescription is the process of systematic medication review with the target of achieving the best risk-benefit ratio based on the best available evidence. This process is especially important for polymedicated elderly patients as well as those overtreated, frail, terminally ill and at the end of life. The deprescription must be done in stages, establishing a close follow-up in case problems appear after withdrawal. In the decision-making process, it is very important to consider the patien
