A further study of the cardiovascular responses to central administration of acetylcholine in rats.

General characteristics and the nature of cardiovascular responses to acetylcholine (ACh) injected into the lateral cerebral ventricle (LCV), the 4th cerebral ventricle (4th CV) and the cisterna magna (CM) were studied. Acetylcholine, depending on the route of injection, the dose and the peripheral vascular tone, produced both increases and decreases in blood pressure and heart rate. After injection of central anticholinesterase agents (physostigmine, neostigmine, armin), the pressor response to ACh was reversed to a depressor response, with or without a secondary pressor response, accompanied

Influence of intoxication by anticholinesterase agents on core temperature in rats: relationships between hypothermia and acetylcholinesterase inhibition in different brain areas.

The early and late effect of three anticholinesterase agents (physostigmine, paraoxon and soman) on core temperature and brain acetylcholinesterase (AcChE) inhibition are compared. The study was performed in adult male rats using sublethal doses of all drugs. AcChE activity was determined by hypothalamus, striatum, hippocampus, medulla oblongata-pons and rest of the brain. The co-existence of intoxication symptoms, hypothermia and AcChE inhibitions was clearly shown by the early effects. However, no relationship was found between the degree of hypothermia and that of inhibition whatever brain

Clinical characteristics of long-term succinylcholine neuromuscular blockade during balanced anesthesia.

Thumb adductor twitch response to train-of-four (2 Hz for 2 seconds) stimulation of the ulnar nerve was used to assess the clinical characteristics of long-term neuromuscular blockade induced with continuous infusion of succinylcholine during balanced (N2O-O2-narcotic-thiopental) anesthesia. Twitch depression of 80 to 90% was maintained for 86 to 365 minutes by continuous infusion of succinylcholine at 86 +/- 5(SEM) micrograms/kg/min. Of 32 patients, 24 developed phase II block, defined as a train-of-four ratio of less than 50%. There was a large degree of individual variability in sensitivity

Prolonged succinylcholine-induced apnea caused by atypical cholinesterase: report of case.

A case of prolonged apnea after administration of succinylcholine in a patient homozygous for the dibucain variant cholinesterase (genotype E1aE1a) has been presented. Knowledge of the patient's medical history, preoperative laboratory tests, and the length of apnea enabled the surgical team to eliminate liver disease, carcinoma, and malnutrition from the differential diagnosis. This, in addition to the patient's failure to respond to an anticholinesterase agent, led to the belief that the patient had an atypical cholinesterase response to succinylcholine and not one secondary to a decreased p

Cholinergic motor control of sea urchin tube feet: evidence for chemical transmission without synapses.

Isolated tube feet of Strongylocentrotus franciscanus contract briefly when the outer epithelium is touched. Similar twitch-like contractions can be induced by electrical stimulation of the outer surface of the tube foot. These responses appear to be chemically mediated. The following evidence indicates that the transmitter substance may be acetylcholine (ACh): ACh causes muscle contraction. This effect and that of electrical stimuli is potentiated by anticholinesterase agents and is antagonized by cholinergic blocking agents. Anaesthesia with chloralhydrate or chloretone abolishes responsiven

[Anticholinesterase agents and axial teratogenesis in quail embryos].

The results of an investigation made with organo-phosphorous compounds (OP), carbamates (C), and a compound including oxamide groups (GO), indicate that spine anomalies in Japanese quail embryos are linked with cholinesterase inhibition. The blocking effect of these various compounds on cholinesterases has been examined both in vitro and in vivo.In vitro, the embryonic cholinesterases are particularly sensitive to carbamates, whereas high concentrations of organo-phosphorous compounds-and especially those which need to be activated at the level of metabolism-block only part of the relevant act

A pharmacological investigation of the electrically evoked convulsive activity induced by administration of catechol in the anaesthetized rat.

1 The response evoked by electrical stimulation at the wrist has been recorded from muscles of the forelimb of anaesthetized rats induced to convulse by administration of catechol.2 This response can be divided into three temporally distinct components, the characteristics of which have been described.3 The probability of occurrence of the two early components of the response has been measured before and after administration of various drugs. The results show that the first component is not affected by cholinoceptor or adrenoceptor blocking drugs or anticholinesterase agents. The probability o

Effects of neostigmine and pyridostigmine at the neuromuscular junction.

The effect of subcutaneous administration of pyridostigmine or neostigmine for 7 to 15 days on neuromuscular transmission has been studied in the rat phrenic nerve-hemidiaphragm preparation. The quantal contents of end-plate potentials at different stimulus rates and the amplitude and frequency of miniature end-plate potentials were compared with those of untreated controls. The rate of release of acetylcholine quanta at high stimulus rates, and the frequency of miniature end-plate potentials, were reduced by pretreatment with both pyridostigmine and neostigmine. Presynaptic effects differed i

Pheniramine aminosalicylate overdosage. Reversal of delirium and choreiform movements with tacrine treatment.

We describe a case of central anticholinergic syndrome with choreiform movements after an overdose of pheniramine aminosalicylate. The symptoms and signs of the condition were reversed by the intravenous administration of the cholinesterase-inhibitor tetrahydroaminacrine. We also discuss the concept of adrenergic-cholinergic imbalance in relation to this case and recommend the use of centrally acting anticholinesterase agents in the treatment of specific cases of drug overdose.

Uropharmacology: v. choline esters and other parasympathomimetic drugs.

Various parasympathomimetic drugs are discussed, including the choline esters, bethanechol, carbachol, methacholine chloride, and furtrethonium. Other cholinomimetic agents include muscarine, muscarone, arecholine, and pilocarpine. Anticholinesterase agents inhibit or inactivate acetylcholinesterase enzyme and thus result in a prolonged stimulation of cholinergic receptors by endogenous ACh. Bethanechol is the most widely used parasympathomimetic drug in the United States. Its action is mainly muscarinic with activity largely confined to the urinary bladder and to a lesser degree the gastroint

[Effect of anticholinesterase agents on the protein and RNA content in the cells of the supraoptic nucleus during sleep and its disorder].

In rats, administration of anticholinesterase drugs (both the penetrating blood-brain barrier armin and the nonpenetrating preparation Gd-42) prevented the decrease of protein content in the neurons and glia of the supraoptic nucleus, as well as of RNA content in the neurons which usually occurred in intact animals after 24-hr deprivation of the REM sleep by the Jouvet method (also with partial disturbance of the slow wave phase).

Potent reversible anticholinesterase agents. Bis- and mono-N-substituted benzoquinolinium halides.

A number of bis- and mono-N-substituted benzoquinolinium salts and their analogues were prepared and evaluated as inhibitors of acetylcholinesterase (AcChE) and butyrylcholinesterase (BuChE). These compounds were also used to help identify some of the morphologic characteristics of the surface at or near the active sites of the cholinesterases. The shape, size, configuration, and conformation of the onium moieties of the quaternary ammonium compounds were found to be the important factors in their anticholinesterase activity. A high concentration of the positive charge of the quaternary ammoni

An analysis of the mode of action of carbachol on the chick biventer cervicis nerve--muscle preparation.

The mechanism of contracture evoked by carbachol in the isolated chick biventer cervicis nerve--muscle preparation was studied. At concentrations lower than 11 muM, carbachol progressively induced contracture at a rate much slower than did acetylcholine. A spontaneous increase of the response to carbachol, but not to acetylcholine, was observed 2-4 hr after isolation of the muscle. By contrast, no change of the response occurred in the denervated muscle. Anticholinesterase treatment shifted the dose-response curve for carbachol markedly to the left as far as the contracture attained after 4-6

Train-of-four nerve stimulation in the management of prolonged neuromuscular blockade following succinylcholine.

Four patients, all possessing an atypical form of plasma cholinesterase, developed prolonged paralysis following succinylcholine administration. The clinical management of all four cases was facilitated by monitoring the train-of-four stimulus. All patients showed marked "fade" of the train-of-four ratio, the initial ratios of the fourth to the first twitches being 50 per cent or less, indicating variable degrees of nondepolarizing neuromuscular blockade. Reversal of paralysis with anticholinesterase agents was completely successful in three cases, but only partially effective in the fourth be
