Hemodynamic effects of dobutamine in an intact animal model.

BACKGROUND AND METHODS: Actions of dobutamine at the beta 1, beta 2, and alpha 1 adrenoreceptors were studied in anesthetized dogs. Six animals received dobutamine (at infusion rates of 0 to 160 micrograms/kg/min) with and without beta-adrenergic receptor blockade. Five animals received phenylephrine (0 to 16 micrograms/kg/min), with and without concurrent dobutamine (20 micrograms/kg/min); this procedure was repeated in five animals after beta-blockade. RESULTS: Dobutamine (10 to 160 micrograms/kg/min) increased heart rate (HR), cardiac output, and left ventricular change in pressure over ti

[Thrombosis prophylaxis using plasmin and its combination with alpha-adrenoreceptor antagonists].

Using two models of experimental thrombosis (arterio-venous shunt and Wessler's model) the effect of plasmin and its combination with alpha-adrenoreceptor antagonists on the formation of thrombus was studied on white rats. It was established that the efficacy of prophylactic of thrombosis by plasmin only was low: middle ball of thrombosis was 2.5-3.0. The combination of plasmin with alpha-adrenoblockers dihydroergotoxine or prazosin under these conditions is most efficient against the formation of thrombus (middle ball of thrombus in this case was 0.8-1.1). Prazosin under certain conditions ha

Noradrenergic mediation of spinal antinociception by 5-hydroxytryptamine: characterization of receptor subtypes.

The present study examined the involvement of spinal noradrenergic mechanisms in spinal antinociception by the 5-hydroxy-tryptamine (5-HT) receptor-selective agonists CGS 12066B (5-HT1B; 7-trifluoromethyl-4(4-methyl-1-piperazinyl)-pyrrolo[1,2-a]quinoxaline), TFMPP (5-HT1C; M-trifluoromethylphenyl-piperazine) and DOI (5-HT2; 1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane) using the rat hot plate test. Effects of alpha-adrenoreceptor antagonists (phentolamine, yohimbine), the adrenergic neurotoxin 6-hydroxydopamine, and the selective noradrenergic uptake blocker desipramine were determined. CGS 1

Catecholamines stimulate testicular testosterone release of the immature golden hamster via interaction with alpha- and beta-adrenergic receptors.

Several lines of evidence suggest that catecholamines are involved in the regulation of the development of the testis. We have therefore investigated the ability of testicular parenchyma (decapsulated pieces of testes) from 18 to 20-day-old golden hamsters to respond to catecholaminergic stimuli in vitro. Norepinephrine and epinephrine, as well as the beta-receptor agonist isoproterenol and the alpha-adrenoreceptor agonist phenylephrine were able to significantly stimulate testicular testosterone production. Dopamine and serotonin were not effective. The stimulatory action of norepinephrine on

Prolonged administration in vivo of alpha and beta adrenergic agonists decreases insulin binding to rat myocardial membranes in vitro by different mechanisms.

Male Sprague Dawley rats were continuously treated in vivo for 6, 12 and 20 hours with a combination of an alpha- (beta-) adrenoreceptor agonist and a beta- (alpha-) adrenoreceptor antagonist in subcutaneously implanted depot tablets. Crude membranes prepared from myocardial cells exhibited a decreased maximum binding of [125I]-insulin after 20 hours irrespective of the treatment applied. Scatchard and non-linear regression analysis of the displacement curves assuming two non-cooperative binding sites revealed a downregulation of the high affinity receptors for about 85% and a concomitant 2.5-

Hepatic glutathione suppression by the alpha-adrenoreceptor stimulating agents phenylephrine and clonidine.

The effects of alpha-adrenoreceptor stimulation on hepatic glutathione content were examined in ICR male mice using a selective alpha 1-adrenoreceptor stimulating agent, phenylephrine, and a selective alpha 2-adrenoreceptor stimulating drug, clonidine. Phenylephrine produced a dose-dependent depression in hepatic glutathione levels when administered by the intraperitoneal (i.p.) route, with a maximum extent of depression of approximately 30% occurring in both male and female mice. Phenylephrine was ineffective by the intracerebroventricular route, indicating a peripheral site of action which w

Cyclic adenosine monophosphate (cAMP) does not mediate the stimulatory action of norepinephrine on testosterone production by the testis of the golden hamster.

The role of cAMP in mediating the stimulatory effects of norepinephrine (NE) on testosterone (T) production by hamster testes in vitro was examined using tissue from both gonadally active and gonadally regressed hamsters. As expected from our previous studies, the NE-induced increase in T accumulation in this system was prevented by alpha-adrenoreceptor antagonist prazosin, but not by beta-adrenoreceptor antagonist propranolol. In incubations of regressed testes from short photoperiod-exposed hamsters, NE stimulated accumulation of cAMP in media and tissue. These effects were prevented by prop

Examination of the role of catecholamines in hepatic glutathione suppression by cold-restraint in mice.

Cold-restraint stress was found to produce a depression in hepatic glutathione content and to elevate circulating catecholamine levels in four mouse strains--ICR, NIH, B6C3F1, and ND/4. Serum norepinephrine concentrations were significantly elevated after cold-restraint (2--3 h) in all strains, and serum epinephrine levels were increased in the B6C3F1 and ND/4 strains. In time-course studies conducted using ND/4 mice, the decline in hepatic glutathione concentrations was found to slightly precede increases in serum epinephrine and norepinephrine concentrations. Also, pretreatment with phentola

The roles of catecholamines in responses evoked in arterioles and venules of rat skeletal muscle by systemic hypoxia.

1. Studies have been made in the anaesthetized rat of the roles played by alpha- and beta-adrenoreceptor stimulation in determining diameter changes induced in individual arterioles and venules of the spinotrapezius muscle during systemic hypoxia (breathing 6% O2 for 3 min). 2. Topical application to the spinotrapezius of phentolamine, the alpha-adrenoreceptor antagonist, or sotalol, the beta-adrenoreceptor antagonist, had no effect on the fall in systemic arterial pressure and tachycardia induced by hypoxia. 3. All arterioles and venules showed a decrease in diameter in response to topical ap

Spontaneous and noradrenaline-induced transient depolarizations in the smooth muscle of guinea-pig mesenteric vein.

1. Recordings of membrane current were made in the smooth muscle of short segments of mesenteric vein before or during stimulation with noradrenaline (NA). 2. Small veins (diameter less than 150 microns) when cut into short segments (of length less than 250 microns) had the passive electrical characteristics of short cables both before and during activation with NA. 3. Spontaneous transient depolarizations (STDs) or the underlying inward currents (STICs) were recorded in these preparations. STDs were of myogenic origin as they were not blocked by tetrodotoxin or antagonists to the alpha-adreno

Hemodynamic responses to alpha-adrenergic blockade during hypoxemia in the fetal lamb.

The mechanisms responsible for the increase in umbilical venous and hepatic vascular resistance during hypoxemia are poorly understood. To assess the relative importance of alpha-adrenergic receptors, we produced an acute, severe hypoxemia in chronically instrumented fetal sheep. While fetal arterial oxygen saturation was maintained at the same level, we then injected phentolamine, a selective alpha-adrenoreceptor blocker. We found that the hypoxemia-induced vasoconstriction of the umbilical veins and hepatic vasculature was reversed by alpha blockade. Thus, alpha-adrenergic stimulation is nec

[The characteristics of the adrenergic mechanisms regulating the lymph flow in arterial hypertension in dogs].

The influence of i.v. infusion of adrenaline on the lymph flow and arterial pressure was studied in acute experiments on dogs with experimental renovascular hypertension. A reflex increase of the lymph flow by 268% above the initial level was revealed, as well as more obvious alterations in the arterial pressure, decrease of latencies and increase of the response duration in hypertensive dogs as compared with normotensive ones. After infusion of adrenaline together with obsydan we observed the lymph flow to be 2-fold enhanced. Pyrroxan has abolished the effect of adrenaline on alterations in t

Separation of periventricular dopaminergic and alpha-adrenergic systems from the vasopressin-secreting mechanisms activated by prostaglandin D2.

The aim of this study was to evaluate periventricular dopaminergic and alpha-adrenergic receptor functions in vasopressin (AVP) secretion elicited by central applications of prostaglandin D2 (PGD2) in conscious rats. Intracerebroventricular (i.c.v.) injections of PGD2 (70.9 nmol (25.0 micrograms] produced transient rises in plasma AVP 5 min later, without increasing plasma osmolality, sodium and hematocrit. Arterial pressure and heart rate before and after the PGD2 administrations were not significantly different from those of control rats receiving its vehicle. The PGD2-induced AVP response w

Blood pressure maintenance in hypertensive sympathectomized rats. I. Adrenal medullary catecholamines.

To assess the role of the sympathetic nervous system in the development of genetic hypertension, blood pressure (BP) was recorded in conscious adult Lyon hypertensive (LH) and normotensive (LN) rats that had received daily injections of saline or guanethidine at 1-13 wk of age. Guanethidine abolished the pressor response to tyramine, decreased plasma norepinephrine by greater than 70% and plasma 3,4-dihydroxyphenylglycol by approximately 90%, and did not change plasma epinephrine. Bilateral adrenalectomy further reduced plasma norepinephrine to 8 and 5% of control levels in LH and LN rats, res

Phorbol esters and cyclic AMP activate AMP deaminase in adult rat cardiac myocytes.

Using rapid deenergization as a probe for adenylate deaminase activity in intact adult rat cardiac myocytes, we have previously established that IMP formation is enhanced by alpha-adrenergic agonists. In the present study, the effect of adrenergic agents on adenylate deaminase was further characterized. Phenylephrine (PE)3 increased IMP production in a dose-dependent fashion with an EC50 of 8 x 10(-7) M. The response to PE was reversed within 10 min by the alpha 1-antagonist, prazosin. Likewise, adenylate deaminase was also activated in ventricular myocytes challenged with phorbol 12-myristate

Mechanism of the haemotensive action of porcine relaxin in anaesthetized rats.

Abstract Experiments were done to examine the pressor effect of iv porcine relaxin in anaesthetized rats. Acute injections of relaxin caused consistent and sustained rises in systemic blood pressure that were dose-dependent within the physiological range. Pretreatment of rats with a specific vasopressin (V1) receptor antagonist, but not an alpha-adrenoreceptor antagonist, substantially reduced the pressor effect of relaxin. After the vasopressin receptor antagonist, small rises in blood pressure occurred after a longer latent period, compared with the responses in intact animals. The data clea

Thromboxane mediation of the pressor response to infused angiotensin II.

UNLABELLED: The role of thromboxane A2(Tx) in mediating the pressor response to angiotensin II (AII) was studied in anesthetized rats. Intravenous AII (500 ng/kg/min) increased mean arterial pressure (MAP) by 35 +/- 3 mm Hg and increased the excretion of prostaglandin PGE2, the metabolites of prostacyclin (6kPGF1 alpha) and Tx (TxB2) (P less than .05). A similar pressor infusion of the alpha 1-adrenoreceptor agonist phenylephrine (PE) increased the excretion of PGE2 and 6kPGF1 alpha but not TxB2. The increases in MAP and prostaglandin excretion produced by AII were reversed by the AII-receptor
