Acute effects and mechanism of action of ketanserin in patients with primary Raynaud's phenomenon.

This study evaluates the vasoactive effects and mode of action of ketanserin, a selective 5HT2 receptor antagonist, on digital circulation in 11 patients with primary Raynaud's phenomenon. Reflex digital vasoconstriction was induced by moderate body cooling. We found that ketanserin given intravenously in a dose of 10 mg, normalized digital skin temperature and digital blood flow as estimated by venous occlusion plethysmography and laser-Doppler flowmetry. However, effects of ketanserin on transcutaneously measured oxygen pressure were modest. This could imply a preferential effect of ketanser

Halothane anesthesia causes active flow-independent pulmonary vasoconstriction.

We utilized multipoint pulmonary vascular pressure-flow (P/Q) plots to investigate the effects of halothane anesthesia on the pulmonary circulation. Our first objective was to assess the extent to which the P/Q relationship measured in conscious dogs is altered during halothane anesthesia. P/Q plots were constructed by stepwise constriction of the thoracic inferior vena cava to decrease venous return and Q. Compared with conscious dogs, halothane (approximately 1.2% end-tidal) resulted in active, flow-independent pulmonary vasoconstriction (P less than 0.01) at all levels of Q. Halothane also

Synthesis and structure-activity relationship of some new beta-blocking agents with possible alpha-adrenoreceptor activity.

A series of new phenoxypropanolamines with cyclic guanidine and isothiourea moieties has been prepared and investigated for non-specific beta- and alpha-adrenoreceptor activity by receptor binding experiments on crude rat brain membrane. Some of them [9a, 10c] possess affinity for both beta- and alpha-adrenoreceptors.

[A voltametric study of the role of the noradrenaline innervation of the hippocampus in cats in paradoxical sleep].

By the method of cyclic voltammetry with carbon fibrous electrodes the signal of 0.1 V was recorded from the dorsal hippocampus of male cats during natural sleep. Systemic injection of 50 mg of L-DOPA and 2.5 mg of isadrin did not influence the 0.1 V signal but the pyrroxan in a dose of 15 mg caused an increase of the signal amplitude. Parallel record of the EEG, myogram, oculogram and voltammogram showed that the 0.1 V signal, reflecting the level of alpha-adrenoreceptor norepinephrine content in the hippocampus increased in the first and last 1-2 minutes of the paradoxical sleep phase and wa

The effects of yohimbine and four other antagonists on amitraz-induced depression of shuttle avoidance responses in dogs.

The ability of five antagonists to prevent the central nervous system's depressant effects of amitraz in dogs was evaluated using a shuttle avoidance paradigm. All drugs were injected iv into six male dogs trained to avoid a 1-mA shock by jumping over a hurdle within 10 sec of the start of a tone. Dogs were given an antagonist or saline followed in 5 min by 3 mg/kg amitraz dissolved in dimethyl sulfoxide (DMSO) or DMSO alone. After pretreatment with saline, amitraz decreased significantly the means number of avoidance responses and increased significantly the means latencies of avoidance respo

Molecular properties of the histamine H2-receptor. Covalent inhibition by tetraamine disulfides.

A series of tetraamine disulfides related to benextramine (an alpha-adrenoreceptor and H2-receptor antagonist) in which the distance between the inner and the outer nitrogens were changed from five to nine methylenes has been studied. Both effects of the displacement of the disulfide bridge by two methylenes and those of the progressive removal of two of the four nitrogens on the pharmacological profile have been assessed. Peak potency appeared to be associated with eight methylenes between the inner and the outer nitrogens and to four cationic charges as in the most active analogue 4 which wa

Structure-activity relationships in 1,4-benzodioxan-related compounds. 2. Role of the amine function on alpha-adrenoreceptor blocking activity.

Several WB4101 (1)-related compounds were synthesized. Their alpha-adrenoreceptor blocking properties were evaluated in order to understand the kind of interaction occurring between the amine function of (1)-related compounds and the alpha-adrenoreceptor anionic site. The results suggest that a charge reinforced hydrogen bond rather than an ion pairing might play a most important role in receptor binding.

SK&F 86466, a novel alpha-adrenolytic drug: effects on heart rate, blood pressure, and neuroendocrine function in supine resting position and in response to postural and cold stimulation in normal humans.

SK&F 86466 is a novel alpha-adrenoceptor blocking drug shown in preclinical profiling to have relative selectivity for the pre- and postjunctional alpha 2-adrenoreceptor. In the present clinical study, the effects of single oral doses of 10, 25, and 50 mg SK&F 86466 on supine and stimulated circulatory and neuroendocrine function were assessed in eight normal subjects studied in a placebo-controlled balanced cross-over design with the drugs administered in double-blind fashion. SK&F 86466 caused a dose-related increase in stimulated (postural and cold challenge) and supine plasma norepinephrin

Effect of metalaxyl on heart rate in rats: role of alpha 1-adrenoreceptors.

Intraperitoneal injection of metalaxyl (250 mg/kg) produced a decrease in heart rate lasting for more than 60 min in anesthetized rats. Pretreatment of rats with phentolamine (a nonselective alpha-adrenoreceptor antagonist, ip at 20 mg/kg) and prazosin (an alpha 1-adrenoreceptor antagonist, ip at 5 mg/kg) significantly reduced the bradycardia induced by metalaxyl. Yohimbine (an alpha 2-adrenoreceptor antagonist, ip at 10 mg/kg) did not change the effect of metalaxyl on heart rate. The results suggest that alpha 2-adrenoreceptors mediate the bradycardic effect of metalaxyl.

Inhibitory effect of norepinephrine on the single-unit activity of caudally projecting paraventricular neurons.

The role of norepinephrine (NE) in controlling the single-unit activity of paraventricular (PVN) neurons projecting to or passing through the caudal ventrolateral medulla (CVLM) was investigated in adult male rats anesthetized with urethane. Of 72 PVN neurons studied, 19 were antidromically activated by CVLM stimulation (Group I) and 48 were antidromically activated by posterior pituitary (PP) stimulation (Group II). The remaining 5 neurons were antidromically driven by both CVLM and PP stimulation (Group III). In 14 of the 19 Group I neurons and in all the 5 Group III neurons, iontophoretical

Antagonism of bromobenzene-induced hepatotoxicity by the alpha-adrenoreceptor blocking agents phentolamine and idazoxan: role of hypothermia.

A recent study from our laboratory revealed that cotreating mice with the alpha-adrenoreceptor antagonists phentolamine and idazoxan markedly diminished bromobenzene-induced hepatotoxicity. Subsequent studies also revealed that such cotreatment does not alter the pharmacokinetic disposition of bromobenzene in mice nor its bioactivation to reactive metabolites. In the present study, the possible role of hypothermia in the phentolamine antagonism of bromobenzene-induced hepatotoxicity was investigated. Bromobenzene alone caused a significant, dose-related hypothermia. The high dosage regimen (10

Stimulation of postjunctional alpha-1 and alpha-2 adrenoreceptors increases proximal urethral perfusion pressure in the pithed rat.

1. In pithed rats, the effects of postjunctional alpha-1 and alpha-2 adrenoreceptor stimulation on prostatomembranous urethral perfusion pressure (UPP) were characterized by using selective adrenoreceptor agonists and antagonists. 2. Dose-dependent increases in UPP were elicited by the intravenous administration of selective alpha-1 and alpha-2 adrenoreceptor agonists, phenylephrine (1.0-30.0 micrograms kg-1) and BHT-933 (10-3000 micrograms kg-1), respectively. 3. The effects of phenylephrine on UPP were antagonized, in a dose-related manner, by pretreatment with prazosin (0.1 and 0.5 mg kg-1,

Tetrahydrothiadiazoloisoquinolines: synthesis and inhibition of phenylethanolamine-N-methyltransferase.

A series of 7,8-fused heterocyclic tetrahydroisoquinolines were synthesized and tested as inhibitors of rabbit adrenal phenylethanolamine-N-methyltransferase (PNMT). 6,7,8,9-Tetrahydro[1,2,3]thiadiazolo[5,4-h]isoquinoline 5 (SK&F 86607) was found to be a potent inhibitor of PNMT with an IC50 similar to that of 7,8-dichloro-1,2,3,4-tetrahydroisoquinoline (1, SK&F 64139). The isomeric tetrahydro[1,2,3]thiadiazolo[4,5-h]- and tetrahydro[1,2,5]thiadiazolo[3,4-h]isoquinolines, 13 and 20, were also potent PNMT inhibitors. Role of dopaminergic and adrenergic receptors in the pathogenesis of arterial lesions induced by fenoldopam mesylate and dopamine in the rat.

Fenoldopam mesylate (FM), a selective, postjunctional, dopaminergic (DA1) vasodilator, causes a novel lesion of large caliber splanchnic arteries (100 to 800 microns) in the rat characterized by necrosis of medial smooth muscle cells and hemorrhage. FM does not induce lesions in other vascular beds of the rat or in dogs or monkeys. Dopamine, like FM, causes hemorrhagic lesions of large caliber splanchnic arteries in the rat, as well as fibrinoid necrosis of small caliber arteries (less than 100 microns) of the splanchnic, cerebral, coronary, and renal vascular beds. ATP, alpha,beta-methylene ATP and suramin as tools for characterization of vascular P2x receptors in the pithed rat.

1. In pithed rats, the blood pressure effects of ATP, alpha,beta-methylene ATP (mATP), alpha-adrenoreceptor agonists and of electrical stimulation of the thoracolumbar sympathetic outflow were studied in the absence and presence of mATP, suramin and adrenoreceptor antagonists. 2. ATP elicited an initial rise in mean blood pressure followed by a decrease and a second increase. mATP produced a short-lived increase in blood pressure, whereas equieffective doses of noradrenaline, methoxamine and B-HT 920 (6-allyl-2-amino-5,6,7,8-tetrahydro-4H-thiazolo-(5,4-d)-azepine) produced a more prolonged, bi

Influence of the endothelium and alpha-adrenoreceptor antagonists on responses to noradrenaline in the rabbit basilar artery.

1. Noradrenaline (10(-6)-10(-2) M) produced slow, concentration-dependent depolarization of smooth muscle cells in the rabbit basilar artery, which preceded the onset of contraction by around 8 s (n = 18). 2. With concentrations greater than 10(-4) M, noradrenaline produced action potentials and fast rhythmic depolarizations superimposed on the slow depolarization. Each fast event was followed by a clear increase in the rate of smooth muscle contraction. The selective alpha 1-adrenoreceptor agonist phenylephrine produced very similar membrane and contractile responses. 3. Action potentials wer

Prejunctional alpha 2-adrenoreceptors modulate the stimulated release of noradrenaline in isolated follicle strips from bovine ovaries.

1. Strips from the follicle wall of bovine ovaries were incubated in Krebs-Ringer solution containing 3H-noradrenaline for measurement of transmitter liberation during electrical field stimulation (5 Hz frequency, 1 ms pulse duration, 10 V between the electrodes). The effects of noradrenaline as well as selective alpha-adrenoreceptor agonists and antagonists were studied on the electrically induced efflux of radioactivity. 2. Noradrenaline (1 microM) inhibited the stimulated release of radioactivity. The alpha 2-adrenoreceptor agonist, oxymetazoline, significantly reduced the release of radioa

Electrical and mechanical activity of rabbit prostate smooth muscles in response to nerve stimulation.

1. The rabbit prostate contains two smooth muscle bundles: one forms the capsule of the prostate and the other runs longitudinally in the outermost layer of the prostate. The former was contracted by exogenously applied noradrenaline (NA) and the latter by acetylcholine (ACh), and were therefore identified as NA-sensitive and ACh-sensitive muscles, respectively. 2. In the NA-sensitive muscle, exogenously applied NA, but not ACh, depolarized the smooth muscle membrane, and this action of NA was antagonized by prazosin but not by yohimbine. The membrane of the ACh-sensitive muscle was depolarize

No evidence of vascular dopamine receptors in the rat portal vein.

Dopamine (5.3 X 10(-6) to 5.3 X 10(-5) M) decreased the force of spontaneous myogenic contractions of the rat portal vein in a concentration-dependent manner in the presence of the alpha-adrenoreceptor antagonist BE-2254. The DA1-selective agonist fenoldopam and the DA2-preferential agonist (-)-EOE were both inactive. Dopamine-induced relaxation was not inhibited by (+)-butaclamol (10(-8) M), but was reduced by a potent beta 2-selective adrenoreceptor antagonist ICI 118,551 (2 X 10(-8) M). Our results demonstrate that the rat portal vein is devoid of postsynaptic DA1 and DA2 dopamine receptors
