Metalaxyl-induced bradycardia in rats: mediated by alpha-adrenoreceptors.

The effect of the fungicide metalaxyl [methyl N-(2-methoxyacetyl)-N-(2,6-xylyl)-DL-alaniate] on cardiac activity was investigated in albino rats anesthetized with phentobarbitone. Metalaxyl caused dose-dependent bradycardia, and at higher doses (250 and 300 mg/kg body weight) the sustained bradycardia led to cardiac arrest. The acetylcholinesterase activities in brain and heart were not affected in metalaxyl-treated rats. However, the bradycardia induced by the fungicide was blocked by pretreatment of rats with the alpha-adrenoreceptor antagonist phentolamine (20 mg/kg, ip). The data suggest t

Induction of glycogenolysis in cultured Ewing's sarcoma cells by dopamine and beta-adrenergic agonists.

This study describes hormonal regulation of glycogen metabolism in Ewing's sarcoma cells. 3H-Glycogen synthesized in cultured Ewing's sarcoma WE-68 cells from 3H-glucose was hydrolyzed in a concentration-dependent manner by various catecholamines. The order of potency for the glycogenolytic effects of catecholamines was isoproterenol greater than or equal to dopamine greater than norepinephrine greater than epinephrine. The concentrations giving half-maximal effectiveness (EC50) were about 2 x 10(-8) M, 3 x 10(-8) M, 8 x 10(-8) M, and 5 x 10(-7) M for isoproterenol, dopamine, norepinephrine, a

Alpha 1- and alpha 2-adrenoreceptor antagonists differentially influence locomotor and stereotyped behaviour induced by d-amphetamine and apomorphine in the rat.

The importance of dopamine (DA) in mediating locomotor, exploratory and stereotyped behaviour in rodents is well established. Evidence also indicates a modulatory role for noradrenaline (NA) although, due to nonspecificity. of previously available agents, a precise role remains undefined. The effects of the specific and selective alpha-adrenoreceptor antagonists idazoxan (alpha 2) and prazosin (alpha 1) on behaviour induced by amphetamine and apomorphine have been investigated in the rat. d-Amphetamine (2 mg/kg) induced hyperactive locomotion and exploration. Pretreatment with prazosin (1 mg/k

Investigation of mechanism of fade of gastrin-stimulated gastric acid secretion in the cat.

1. In conscious cats prepared with gastric fistulae gastric acid secretion in response to pentagastrin was found to reach a maximum after 45 min of stimulation, and to fade thereafter. Over the period 45-150 min of stimulation the fade was 5.4-7.8% of the maximum response per 15 min. 2. Once the response to pentagastrin had declined, acid secretion could not be restored by doubling the dose of pentagastrin, although an equisecretory dose of histamine could restore it. 3. Low doses of histamine were additive to the pentagastrin acid secretory response; they tended to prolong the peak response,

The effect of temperature on neuromuscular transmission in the main caudal artery of the rat.

1. Excitatory junction potentials (EJPs) recorded in isolated segments of the proximal main ventral artery of the rat tail were reduced in amplitude and prolonged in time course as temperature was lowered from 35 to 15 degrees C. 2. The slow depolarization that followed the EJPs after supramaximal or repetitive perivascular stimulation was markedly slowed in time course, but little affected in amplitude, as temperature was lowered. 3. The time constant (tau EJP) of the exponential decay phase of the EJP recorded from cells deep in the media was similar to the membrane time constant, so that th

Adrenergic modulation of 'non-adrenergic' twitches in the rat vas deferens.

1. Field stimulation of the intramural sympathetic nerves of rat vas deferens produced a biphasic contraction. Post-tetanic twitches were potentiated (PTP) and the relationship of this to the secondary component of tetanic responses was investigated. 2. A tetanus evoked by field stimulation at 4 Hz for 50 s gave a maximum increase in twitch tension of 109 +/- 5% (n = 43). The potentiation was blocked by the alpha-adrenoreceptor antagonists phenoxybenzamine, prazosin and thymoxamine. These drugs also blocked the secondary component of the tetanic response. 3. Increasing the frequency (1-8 Hz) o

Effects of activating spinal alpha-adrenoreceptors on sympathetic nerve activity in the rat.

The effects of several alpha-adrenoreceptor agonists and antagonists administered intrathecally at T10 level on renal sympathetic nerve activity (RSNA) were examined, in chloralose-urethane anaesthetised rats. Intrathecal noradrenaline (NA, 0.5-500 micrograms) produced one of 3 responses depending on dose, an inhibition of RSNA at low doses, an excitation of RSNA at high doses, or a biphasic effect. Intrathecal adrenaline (5-200 micrograms) was inhibitory in the main but some doses elicited poorly repeatable brief excitation followed by prolonged inhibition. Intrathecal methoxamine (ME; 2.5 ng

Dopamine-sensitive receptors that evoke rumination and modify reticulo-ruminal activity in sheep.

Dopamine (20 micrograms/kg) evoked rumination in sheep when injected as a bolus into the coeliac artery or into the left gastric artery but not when injected into the carotid artery. A mixed alpha-adrenoreceptor antagonist (phentolamine) and an alpha 2-adrenoreceptor antagonist (yohimbine) prevented dopamine from evoking rumination, but a dopaminergic antagonist (metoclopramide) did not. These findings suggest that dopamine stimulated rumination by acting upon alpha 2-adrenoreceptors situated in the area supplied by the left gastric artery, whereas dopamine injected intracerebrally may have ev

Antagonism of bromobenzene-induced hepatotoxicity by the alpha-adrenergic blocking agents, phentolamine and idazoxan.

The coadministration of phentolamine, an alpha-adrenoreceptor antagonist, was found to be effective in antagonizing the hepatotoxicity produced by bromobenzene in B6C3F1 mice. Multiple doses of phentolamine, administered in dosages of 10 mg/kg, attenuated almost completely the acute lethality resulting from a 0.5 ml/kg dosage of bromobenzene. Consistent with this decline in lethality, the coadministration of phentolamine significantly altered the magnitude of hepatocellular necrosis, the elevation of serum alanine aminotransferase activity, and the glutathione depression normally produced by t

Structure-activity relationships among benextramine-related tetraamine disulfides. Chain length effect on alpha-adrenoreceptor blocking activity.

Several N'-substituted N,N''-(dithiodi-2,1-ethanediyl)bis(1, omega-alkanediamines) were prepared and evaluated for their blocking activity on alpha-adrenoreceptors in the isolated rat vas deferens and human blood platelets. The results were compared with those obtained for benextramine (N,N''-(dithiodi-2,1-ethanediyl)bis[N'-[(2-methoxyphenyl)-methyl]- 1 ,6- hexanediamine], 10). Bendotramine (N,N''-(dithiodi-2,1-ethanediyl)bis[N'-[(2-methoxyphenyl)- methyl]-1,12-dodecanediamine], 16) proved to be as active as 10 on alpha 1-adrenoreceptors, showing that optimum activity is associated with two ca

Inhibition of rumination in sheep by alpha-adrenoreceptor antagonists.

Rumination activity was recorded in sheep during a period of 3 h. The amount of rumination occurring in 30 min following the intravenous injection of an alpha 1 blocking drug (prazosin, 0.1 mg/kg) and an alpha 2 blocking drug (idazoxan, 0.1 mg/kg) was reduced compared with control days when saline was given. The observation that rumination can be stimulated by a rapid, intravenous dose of adrenalin was confirmed. The induction of rumination by adrenalin was prevented by these alpha-adrenoreceptor antagonists given singly or together at the dose rate mentioned. These findings are discussed in t

No evidence for more than one type of alpha 1-adrenoreceptor in rabbit pulmonary artery.

1. The effects of the alpha-adrenoreceptor antagonists, prazosin and yohimbine, have been examined on the isometric contraction of the rabbit pulmonary artery produced by alpha 1-adrenoreceptor agonists. Cocaine, corticosterone and propranolol were present throughout and either a single or two concentration-response curve method was used. 2. Prazosin and yohimbine competitively antagonized the responses to agonists but did not reveal differences between agonists. 3. Using the single concentration-response curve method, prazosin had pA2 values of 9.06 against clonidine and 8.76 against methoxam

Labetalol protects against the potentiation by propranolol of the bronchospasm to norepinephrine in guinea-pigs.

The increase in airway obstruction observed in asthmatics after treatment with beta-adrenergic blockers may be attributed to an unopposed alpha-adrenergic activity. Labetalol is an antihypertensive agent with beta-adrenoreceptor and alpha-adrenoreceptor blocking properties. Labetalol does not cause a bronchospasm in human asthmatics. Norepinephrine (0.025-0.1 mg/kg, i.v.) caused a bronchospasm in guinea-pigs after the blockade of beta-adrenoreceptors with propranolol (0.1-1 mg/kg, i.v.), but not after beta-blockade with labetalol (0.1-10 mg/kg, i.v.). Labetalol significantly protected against

A comparison of the ability of various alpha-adrenoreceptor agonists to induce EGTA-resistant contractions of rat aorta.

Phenylephrine (PE) and Guanfacine (GU) produced maximum contractions of rat isolated aorta which were 96.8 +/- 1.3% and 96.0 +/- 2.8% respectively of the maximum responses induced by noradrenaline (NA). The responses produced by maximum concentrations of NA following 2 min incubation of the preparations with 3.0 mM EGTA were 38.1 +/- 1.2% of the maxima achieved in normal Krebs. PE produced maximum EGTA-resistant responses which were significantly less than those seen with NA; GU produced EGTA-resistant responses which were significantly less than those to PE. Characterization of alpha-adrenoreceptor mediated reduction of flow in a saline-perfused constant pressure model of rat perfused hindquarters.

An analysis of vasopressor responses elicited by alpha-adrenoreceptor agonists and KCl in a constant pressure saline perfused model of rat perfused hindquarters is presented. Quantitative method for the study of the morphological changes induced by vasoactive agents in single aortic myocytes grown in primary cultures.

Morphological changes of aortic myocytes grown in primary cultures on collagen gels, were induced by two adrenergic agonists, in the absence or presence of specific antagonists. Their effects were quantified by means of the measurement of the surface areas of the images of the cells. The alpha-adrenoreceptor agonist phenylephrine induced a concentration-dependent decrease of the surface areas of the images of the cells, which was competitively antagonized by phentolamine, and was reduced in the presence of CdCl2 (an inhibitor of calcium entry). The beta-adrenoreceptor agonist isoproterenol ind

Antagonism of cocaine-induced hepatotoxicity by the alpha adrenergic antagonists phentolamine and yohimbine.

The ability of the alpha adrenoreceptor antagonists phentolamine and yohimbine to antagonize cocaine-induced hepatotoxicity was determined in phenobarbital-induced B6C3/F1 mice. Hepatotoxicity was assessed by the histologic extent of necrosis, incidence of latent lethality and increases in serum alanine aminotransferase activity. The depression of hepatic glutathione levels also were measured. The administration of a single 5-mg/kg dose of phentolamine antagonized the decrease in glutathione levels and the elevation of aminotransferase activity caused by a 60-mg/kg dose of cocaine. Possible prostacyclin involvement on disparate tonic responses to "in vitro" norepinephrine in circular and in longitudinal preparations from rat thoracic aorta.

In view of existing reports documenting that "in vitro" norepinephrine (NE) contracts ring-shaped rat aortic preparations, whereas it relaxes arterial strips mounted in longitudinal fashion within an organ bath: it was decided to explore possible reasons which may account for such disparate actions of the same agonist on the same tissue. Isolated rings (circular preparations) obtained from rat thoracic aortae responded to increasing concentrations of NE with dose-dependent tonic enhancement, not significantly affected by the presence of indomethacin (10(-6)M); whereas, preincubation with phent

Interactions between neuropeptide Y and adrenergic systems in the stimulation of luteinizing hormone release in steroid-primed ovariectomized rats.

Intraventricular injection of neuropeptide Y (NPY) stimulates LH release in estradiol benzoate- and progesterone-primed (EBP) ovariectomized rats. Because adrenergic neurotransmitters, norepinephrine (NE) and epinephrine (E), show intraneuronal coexistence with NPY in certain brain regions of the rat and there are similarities in the effects of NPY and NE/E on LH release, we investigated the possible interaction of NPY and adrenergic receptor systems in the stimulation of LH release in EBP-treated ovariectomized rats. The experiments were designed to determine whether NPY exerted its effects v
