Circulating Piwi-Interacting RNAs Associate With Childhood Asthma ICS Response With Vitamin D Effect Modification.

BACKGROUND: Piwi-interacting RNAs (piRNAs) have been linked to type 2-high asthma phenotypes. Although vitamin D modulates inhaled corticosteroid (ICS) response via microRNAs, its potential role through piRNAs remains unknown. METHODS: Baseline plasma piRNA expression was profiled by small RNA sequencing in 492 participants from the Childhood Asthma Management Program (CAMP), with 187 children randomised to the ICS arm. Serum vitamin D levels were classified as insufficient (≤ 30 ng/mL) or sufficient (> 30 ng/mL). Linear regression was used to assess associations between piRNA expression and

TrxR Inhibition and Nrf2-FOXO3 Modulation by Repurposed Drugs: A Redox Strategy to Reverse Cancer Multidrug Resistance.

A common cause of multidrug-resistant (MDR) cancer is imbalanced redox signaling, which reduces the effectiveness of chemotherapy and promotes regrowth of cancer cells. Amplification of thioredoxin reductase (TrxR) and activation of the Keap1-Nrf2-FOXO3 pathway may contribute to enhanced drug efflux, strengthens antioxidant defenses, and resistance to oxidative stress-induced apoptosis in certain tumors. Redox-based drug repurposing offers a promising strategy to overcome MDR by targeting these shortcomings. Repurposing drugs including metformin, auranofin, brusatol, and natural polyphenols in

Nutritional Supplements in the Treatment of Endometriosis-Associated Symptoms: A Systematic Review.

Cancer is regarded as one of the leading causes of death worldwide, despite the progress of traditional therapies. Chemotherapy, radiotherapy, and surgery are often accompanied by significant side effects and the development of drug resistance contributes to making the fight against cancer even more challenging, which clearly highlights the urgent need to develop new therapeutic molecular approaches. In this context, natural peptides were introduced into the pharmaceutical market in the last decade and have replenished the ranks of effective anticancer agents due to their structural diversity,

Serum Vitamin A levels in children with sickle cell anemia: relationship with pulmonary function indices.

BACKGROUND: Vitamin A is involved in the proliferation and maintenance of the epithelial cells of the respiratory tract. Since maintenance of the integrity of the respiratory epithelium is important for pulmonary functions, vitamin A may contribute to the reduced pulmonary functions noted in children with sickle cell anemia (SCA). This study aims to relate serum vitamin A levels with pulmonary function indices in children with SCA. METHODS: This cross-sectional analytical study recruited 70 children with SCA aged 6–17 years using systematic random sampling technique. Ethical clearance was obta

Theranostic π Electron-Stabilized Polymeric Micelles for PET Imaging-Guided Drug Delivery to Tumors.

Nanomedicines are increasingly employed in oncology. However, their efficacy is limited by heterogeneous nanoparticle accumulation in different tumors and patients. Nuclear imaging offers non-invasive, patient- and lesion-specific visualization and quantification of nanoparticle uptake, providing a biomarker to predict nanotherapy efficacy. We present a positron emission tomography (PET)-imageable [mPEG-b-p(HPMAm-Bz)]-based polymeric micelle platform for image-guided and tumor-targeted drug delivery, with potential for patient stratification and theranostics. Polymers with 1 or 3 deferoxamine

Designing of sorafenib analogs to target c-Raf for the management of hepatocellular carcinoma: Molecular dynamics and mmPBSA analysis.

Introduction: Sorafenib remains the only approved treatment for advanced hepatocellular carcinoma (HCC), yet its clinical use is hindered by toxicity and the emergence of drug resistance. Sorafenib's anticancer effects are largely attributed to its inhibition of multiple kinases, including c-Raf, a key player in the Ras-Raf-MEK-ERK signaling cascade that promotes cell growth and survival. Given the critical role of c-Raf in tumor progression, targeting this kinase offers a promising strategy for improving therapeutic outcomes. Developing new analogs with stronger c-Raf inhibition, better pharm

FDX1 expression promotes DSF/Cu-induced cuproptosis in gastric cancer cells.

OBJECTIVE: Gastric cancer (GC) is a prevalent malignant tumor that warrants the development of drugs and therapeutic targets. Cuproptosis has emerged as a promising mechanism by which to inhibit tumors because copper homeostasis disorders frequently occur in various malignancies. The combination of disulfiram (DSF) and copper ions (DSF/Cu) has been shown to have significant antitumor effects. This study utilized DSF/Cu to investigate the mechanism underlying cuproptosis in GC cells. METHODS: GC cells were treated with DSF/Cu and protein sequencing was performed to screen for differentially ex

Beyond dabigatran: recognizing direct oral anticoagulants as a cause of exfoliative esophagitis.

The new direct oral anticoagulants, despite their efficacy and safety, may cause adverse gastrointestinal effects, such as exfoliative esophagitis or esophagitis dissecans superficialis. This adverse effect, primarily reported with dabigatran treatment, is uncommon with other anticoagulants. Clinical and endoscopic recognition is essential for the proper management of the patient.

Gut Microbiota Remodeling Mediates the Therapeutic Effects of a Plant-Based Medicine on DSS-Induced Ulcerative Colitis in Mice via the Butyrate-SVCT1-Vitamin C Axis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease with a rising global incidence in recent years. Dengzhan shengmai (DZSM), a plant-based formulation clinically used in the management of cerebrovascular diseases, possesses documented anti-inflammatory and antioxidant properties; however, its effects on UC are unclear. In this study, we investigated the therapeutic potential and underlying mechanism of DZSM in a dextran sulfate sodium (DSS)-induced murine colitis model. Our results showed that DZSM significantly alleviated UC-related parameters. Mechanistically, DZSM remodeled gut

Lack of Differences in the Pharmacokinetics of Therapeutic Monoclonal Antibodies Between Japanese and Non-Japanese Individuals.

Evaluation of the pharmacokinetics (PK) of new drugs in Japanese individuals is regularly requested by the Japanese regulatory agency prior to participation in global clinical trials or for bridging approaches for regulatory approval. However, the need for PK assessments in the Japanese population may be less compelling for monoclonal antibodies (mAbs) compared to other drugs, given that the intrinsic properties of immunoglobulin G molecules are generally considered ethnically insensitive. This prompted an evaluation of the currently available evidence for potential PK differences for mAbs bet

Nanoparticles for antiviral nucleotide analogs: approaches and analytical challenges.

Nucleoside and nucleotide analogs are an impactful class of antivirals, yet their efficacy is often limited by inefficient intracellular accumulation and enzymatic activation. Many analogs are administered as nucleoside prodrugs, dependent on host or viral kinases for sequential phosphorylation, with the first monophosphate-forming step widely considered rate-limiting. Medicinal chemistry strategies such as utilizing masked-phosphate prodrugs may improve uptake and activation, but require alternative metabolic processing to generate the active triphosphate. Nanoparticle delivery provides an al

"SMAC"-down for the apoptosis-restoring drug, Xevinapant: What does its story reveal?

Xevinapant, an antagonist of inhibitor of apoptosis proteins (IAPs) from the SMAC-mimetic drug class, was developed to restore apoptosis in cancer cells. Preclinical studies demonstrated on-target IAP degradation, caspase activation, and pro-inflammatory apoptosis via TNF-α upregulation. Monotherapy activity was modest in both preclinical and phase I studies, pointing to a primarily synergistic role as a (chemo)radiosensitizer, with the strongest antitumor signals observed in head and neck squamous cell carcinoma (HNSCC). In the Debio1143-201 phase II trial in locally advanced HNSCC, adding xe

Multispecific DNA coatings for self-assembly.

DNA-coated particles are promising building blocks for functional and finite-sized assemblies because they can be programmed to exhibit orthogonal interactions through the sequence-specific hybridization of DNA strands. To fully exploit this programmability, it is important to develop particles with coatings that incorporate multiple distinct DNA sequences in tunable ratios and to understand how the coating composition influences self-assembly. Here, we compare two strategies for grafting multiple DNA sequences in tunable and well-defined ratios onto micron-sized colloidal particles. We found

Mapping the effects of specific radiation damage and solvent radiolysis in buffers and crystals with online UV-Vis absorption spectroscopy.

Online UV-Vis absorption spectroscopy is often used during crystallographic data collection to characterize chromophores within proteins. This includes the study of electron-rich or redox-active enzyme intermediates and cofactors that are prone to specific radiation damage, necessitating suitable low-dose data-collection strategies. In crystallo spectroscopy enables the monitoring of X-ray-induced changes in the spectroscopic signatures of proteins and ligands; however, a comprehensive approach that also considers buffer components is required. Here, we present a mapping of X-ray-induced spect
