The Long Splice Variant of Ribosomal Protein S24 Protects Cells from Stress and Is Induced by AMPK-ULK1-Regulated Autophagy Independent of mTORC1.

All living organisms have developed complex mechanisms to detect and react to environmental changes to maintain internal balance and support survival. One strategy that cells employ to respond to stress is to refocus the translation machinery to express select stress response proteins, this includes the emerging field of specialized ribosomes. We have previously shown that a long splice variant of ribosomal protein S24 (RPS24L) is induced by several fold in hypoxic monolayers and spheroids in several human cell lines in an autophagy-dependent manner. Here, we show that an allosteric activator

Directed Evolution of an Escherichia coli Secretor Strain Using the Curli Pathway.

A key goal of many bacterial engineering projects is to produce protein, due to their potent chemical and material functions. However, recombinant proteins are usually produced intracellularly, and secreting arbitrary proteins directly into an extracellular environment is challenging. In this work, we explored recombinant protein secretion using the curli secretion pathway in the probiotic Escherichia coli Nissle 1917 strain (EcN). We developed a Green Fluorescent Protein (GFP) secretion system by fusing GFP of the N-terminus sequence from the curli monomer protein while co-expressing the curl

Rational Design of Hemicyanine Platform for Engineering High-Contrast Activatable NIR-II Probes.

Activatable NIR-II hemicyanine probes are crucial tools for high-contrast deep-tissue biomarker imaging. However, their development is constrained by an inherent dilemma: extending π-conjugation to narrow the energy gap inevitably compromises the fluorescence brightness in the activated state (dim on), while the strong intramolecular charge transfer (ICT) effect makes effective quenching in the locked state difficult to achieve (leaky off), leading to poor response sensitivity and a low on/off ratio. Here, we rationally designed a xanthene-cyanine hybrid NIR-II activatable platform (RC-OH), wh

Molecular drivers of cotranslational protein assembly in bacteria.

Protein complexes are pivotal to all major cellular processes. Translation-coupled complex assembly is ubiquitous in eukaryotes, but its molecular basis and prevalence in bacteria remain poorly understood. Here, we combine ribosome profiling and single-molecule optical tweezers to establish principles governing cotranslational assembly of four structurally diverse complexes in Escherichia coli. We reveal how heterotetramers form cotranslationally via hierarchical assembly pathways, with initial heterodimer formation priming the assembly of higher-order oligomers through a topology-dependent or

Interaction of biochar with multiple amendment combinations affects plant biomass and soil enzyme activities.

Biochar is an important restoration amendment that is often applied together with other amendments, such as microbial inoculum, or organic and inorganic amendments. However, the mechanisms underlying biochar interactions with other restoration amendment combinations on soil functions and plant growth often remain unclear. Here we investigate the interaction of biochar with up to six additional restoration amendments using a novel experimental design. We test 6 distinct restoration amendments (amorphous silica, vermiculite, bentonite, AM fungi, potassium dissolving bacteria, and rhizobium) and

Pre-harvest cold treatment enhances the flavor quality of green tea: Insights from metabolomics, volatilomics, and sensory analysis during processing.

Pre-harvest cold treatment enhances tea flavor, yet the chemical mechanisms during processing remain elusive. This study investigated the impact of cold treatment on green tea quality using metabolomics, volatilomics, and sensory evaluation. Cold treatment significantly reduced astringency and enhanced umami, chemically attributed to a reprogrammed "high-amino acid, low-ester catechin" profile. Crucially, GC × GC-O-Q-TOF-MS revealed a dramatic enrichment of floral and fruity volatiles in the cold-treated group. Odor activity value (OAV) analysis identified trans-β-ionone as the critical impact

Synthesis of 4-(Diethylamino)salicylaldehyde Derivatives and Research on Their Antifungal Mechanism against Rhizoctonia solani.

To enhance the antifungal activity of 4-(diethylamino)salicylaldehyde (DSA), 20 salicylaldehyde Schiff base compounds (L1-L20), including nine novel derivatives, were synthesized and characterized by MS, 1H NMR, and 13C NMR. In vitro assays identified compound L5 as the most active agent, with an EC50 of 10.797 μg/mL (2-fold higher than DSA), and pot experiments confirmed its efficacy against cotton damping-off disease. Mechanistic investigations via electron microscopy, transcriptomics, physiological assays and molecular docking suggest that L5 may target fatty acid synthase (FAS) to interfer

Single-Cell Transcriptomic Profiling of the O-Methyltransferase Characteristics of Flavonoid Biosynthesis in Ziziphus jujuba var. spinosa Fruit.

Sour jujube (Ziziphus jujuba var. spinosa) is a valuable medicinal and edible fruit owing to its health-promoting properties. Methoxylated flavonoids enhance their value by improving stability and bioavailability. However, the cellular basis of their biosynthesis remains unclear. We constructed a single-nucleus transcriptomic atlas of sour jujube fruit. Parenchyma cells were identified as the primary sites for flavonoid biosynthesis and their O-methylation, characterized by high expression of phenylpropanoid pathway genes and O-methyltransferases (OMTs). Among the 48 identified OMTs, four (ZjO

Vacuolar Protein Sorting Genes Contribute to Paraquat Resistance in Eleusine indica.

This study investigates resistance mechanisms in a paraquat (PQ)-resistant Eleusine indica population. The resistant population exhibited 27.5-fold resistance to PQ. Although leaf absorption of 14C-PQ was similar to that in susceptible plants, translocation from treated leaves was significantly lower in resistant plants (3.6% vs 18.8%). Cellular fractionation showed reduced PQ accumulation in chloroplasts but increased levels in the cytosol/vacuole fraction. Following PQ treatment, photosynthetic parameters initially declined but later recovered in the resistant population, which also displaye

A β-Mannanase Gene, MAN2, Coordinates Tomato Fruit and Seed Development and Salt Stress Tolerance.

Tomato (Solanum lycopersicum) β-mannanase is known as playing an important role in mannan degradation; however, its biological functions and transcriptional regulation remain poorly understood. Here, we characterized the roles of mannan-1,4-β-mannanase 2 (SlMAN2) in tomato development and salt stress tolerance. Results showed that SlMAN2 was preferentially expressed in seeds, stems, stamens, and stigmas. SlMAN2 overexpression significantly increased fruit and seed size, accelerated seed germination, and enhanced salt tolerance under 100 mM NaCl, whereas knockout mutants exhibited the opposite

Integrating instrumental analysis and computational simulation to elucidate the effect of dual-enzyme hydrolysis combined with γ-glutamylation on the flavor characteristics of soybean meal.

To improve soybean meal's flavor, this study explored the effect and mechanism of dual-enzyme hydrolysis combined with γ-glutamylation on the flavor characteristics of soybean meal. Sensory evaluation revealed dual-enzyme hydrolysis significantly enhanced umami, saltiness and sweetness, while γ-glutamylation further strengthened umami and kokumi sensations. Specifically, γ-glutamylation increased umami amino acid (18.23 mg/100 mL), reduced bitter amino acids (26.94 mg/100 mL). Meanwhile, it also elevated esters and heterocyclic compounds (P < 0.05). Correlation analysis showed umami/sweet amin

Isolation of polyethylene‑modifying soil bacteria from Egypt and Genome-based functional characterization of Sphingobacterium spiritivorum ASb3.

Polyethylene (PE) has been widely utilized, resulting in its accumulation in the environment due to its recalcitrant nature. In this study, seven bacterial isolates from garden soil and garbage dumps in Cairo and Giza, Egypt, grew on PE films as the sole carbon source, reduced PE weight, and formed biofilms. Scanning electron microscopy revealed surface perforations, while Fourier transform infrared spectroscopy detected new carbonyl and hydroxyl groups. Based on 16 S rRNA gene sequencing and phylogeny, the isolates exhibited high similarity to Sphingobacterium spiritivorum, Rhodococcus pyridi

Deciphering the Enzyme-Mediated Oxidation of Dehydroabietic Acid and Other Diterpenoid Resin Acids.

Cytochrome P450 enzymes are versatile monooxygenase biocatalysts. Members of the bacterial CYP226 family are hypothesized to catalyze the oxidation of diterpenoid resin acids as the first step in their catabolism. Two CYP226 enzymes (CYP226A3 and CYP226A31) from the thermophilic bacterium Zestomonas thermotolerans were characterized. Both could bind the diterpenoids in gum rosin. Oxidation metabolites were formed using peroxide-driven reactions with the wild-type enzymes and variants with modifications at the dioxygen-binding groove. The CYP226A31 enzyme was used to enantioselectively hydroxyl

Enhanced biodegradation of bisphenol S by laccase encapsulated in copper-nucleotide metal-organic frameworks.

Bisphenol S (BPS) is widely used as a substitute for bisphenol A (BPA) but has also been identified as an endocrine-disrupting compound with considerable biotoxicity. Compared with BPA, BPS exhibits higher chemical stability, and studies on its biodegradation remain limited. In this work, an efficient strategy for BPS biodegradation was developed using a bacterial laccase immobilized in copper-nucleotide metal-organic frameworks (MOFs). The laccase gene from Bacillus licheniformis was heterologously expressed in Escherichia coli, achieving an extracellular activity of 373.8 U/L. The recombinan

Crowder Environments Control Interfacial Dynamics and Topological Stability in a Knotted Protein.

Knotted proteins are unique biomolecules where backbone entanglement imposes intrinsic constraints on structure and dynamics. The interaction of such a topologically restrained protein with molecular crowders remains unexplored. Using molecular dynamics simulations, we investigate the effects of polyethylene glycol (PEG) and dextran on the structure, interactions, and topological dynamics of the trefoil-knotted enzyme AOTCase. Persistent, heterogeneous accumulation of dextran near the protein surface is observed, whereas PEG exhibits a transient interaction pattern. The distinct interfacial be
