Imine reductase-catalyzed enantiodivergent synthesis of axially chiral alkylidene cyclohexane amines.

Axially chiral alkylidene cycloalkanes (ACACs) are valuable yet underexplored scaffolds, and biocatalytic access to their enantioenriched amine derivatives remains limited. Herein, we report an imine reductase (IRED)-catalyzed asymmetric reductive amination of 4-alkylidene cyclohexanones with primary amines for the enantiodivergent synthesis of axially chiral alkylidene cyclohexane amines. Upon enzyme screening and reaction condition optimization, it was found that the use of AtIRED and MeIRED afforded a variety of (S)- and (R)-configured amine products in up to 99% conversion and 99 : 1 enant

Antidepressant desvenlafaxine identified in wastewater promotes transformation and antibiotic resistance risk in Acinetobacter baylyi ADP1 via metabolic adaptations.

Wastewater treatment plants (WWTPs) are known reservoirs of antibiotic resistance genes (ARGs). Non-antibiotic compounds, such as antidepressants, may further promote ARG acquisition through horizontal gene transfer (HGT). Desvenlafaxine, a serotonin-norepinephrine reuptake inhibitor (SNRI) listed on the EU Surface Water Watch List, is among the most frequently detected antidepressants in WWTP effluents, yet its role in HGT has not been examined. Here, we detected desvenlafaxine at the highest concentrations among four antidepressants monitored across three municipal WWTPs in western New York.

Dynamic Ionic Double-Network Hydrogels of Agricultural Lignin/Zwitterionic Starch via 3D Printing for a Rapid-Rebound and Real-Time Feedback Flexible Sensor.

Agricultural biomass hydrogels are environmentally friendly candidates for flexible sensors, yet their application in advanced AI devices is restricted by persistent trade-offs among mechanical strength, tensile performance, and sensitivity. Lignin can strengthen hydrogels benefiting flexible sensing, but its steric hindrance lowers photocuring cross-linking density, triggering mechanical hysteresis and slow rebound under dynamic deformation, which deteriorates sensing signal reliability. Herein, we develop a vat photopolymerization (VPP) 3D-printed double-network elastomer. Modified lignosulf

Structure-Guided Engineering of Xylitol 4-dehydrogenase Unlocks Efficient L-Xylulose Biosynthesis: Mechanistic Insights into Catalytic Enhancement.

L-Xylulose, a potent α-glucosidase inhibitor and indispensable chiral precursor for L-nucleoside drugs, is highly valuable. However, chemical synthesis often suffers from poor atom economy and tedious protection and deprotection steps of functional groups. Here, a green biocatalytic route was explored by retrieving five XDH4 genes and selecting AYXDH4 as the best candidate. Through homology modeling and site-directed mutagenesis targeting potential synergy sites in the α-helix, the double mutant T171A/S172A was generated, which exhibited 43.97% higher catalytic efficiency (kcat/Km) than the wi

Metabolite-Peptidome Flavor Synergy and Predicted Receptor-Level Interactions in Black Tea across Diverse Cultivars.

This study elucidated flavor-synergistic mechanisms of black teas from 11 diverse cultivars using sensomics, targeted metabolomics, de novo peptidomics, and AlphaFold 3 structural modeling. A total of 566 water-soluble oligopeptides and 158 modified peptides were identified, with dominant modifications including glutamine-ethylation and pyroglutamate-cyclization. Newly identified theanine-substituted peptides (e.g., EAKDEGYRQ+28.03 m/z) and glycosylated peptides may serve as potential thermal precursors during black tea processing. Their contributions to aroma formation warrant further experim

Sequential Molecular Events in the Solid-State Acid-Base Incompatibility of Amlodipine Besylate and Citric Acid: 2D-COS-PXRD and 2D-COS-FTIR as Orthogonal Probes of Reaction Sequence in Drug-Excipient Solid-State Incompatibility.

Drug-excipient incompatibility in solid oral dosage forms can generate toxic degradants and compromise potency without producing a visible macroscopic signal in early development. We reported, for the first time, a mechanistically resolved, temperature-sequenced characterization of the solid-state acid-base incompatibility between amlodipine besylate (AMLO; pKa ≈ 9.4) and citric acid (CA; pKa1 3.13; ΔpKa = 6.27) using six orthogonal techniques: DSC, TGA, isothermal titration calorimetry (ITC), powder X-ray diffractometry with two-dimensional correlation spectroscopy (2D-COS-PXR), FTIR with 2D-

HumanToxPredict: An interpretable data-driven framework for human oral TDLo prediction and toxicity assessment of pharmaceuticals and bioactive compounds.

Predicting the human toxic dose low (TDLo) is critically important for pharmaceutical safety assessment and regulatory toxicology. A global computational toxicology framework that integrates toxicity data for children, infants, men, and women was developed using indicator-variable-based demographic encoding. Initially, Partial Least Squares (PLS)-based quantitative structure-activity relationship (QSAR) and quantitative read-across structure-activity relationship (q-RASAR) models were developed using interpretable molecular descriptors and similarity-based descriptors. A structure-level group-

Co-production of gelatin, hydroxyapatite and calcium-binding peptides from Tilapia scales: Structural characteristics and physicochemical properties.

A novel co-production strategy was developed to valorize tilapia scale waste into gelatin (TSG), hydroxyapatite (TSHA), and calcium-binding peptides. TSG was obtained by hot-water extraction, and the residues were calcined at 700 °C for 3 h to produce TSHA, which served as a self-prepared affinity medium. Alkaline protease hydrolysis yielded the highest calcium-binding capacity (CBC, 4.86 μg/mg). After TSHA-based affinity purification, the recovered fraction (THAC) showed improved CBC (5.71 μg/mg). Isothermal titration calorimetry suggested spontaneous and exothermic calcium-binding (Ka = 2.4

MASTL Kinase Mediates the DNA Replication Stress Response Through PP2A/B55 Phosphatase Modulation.

MASTL kinase promotes mitosis by inhibiting PP2A-B55 through ENSA/ARPP19 phosphorylation. Our analyses of cancer datasets support the involvement and diagnostic/prognostic potential of the MASTL-ENSA-PP2A/B55 module. In head and neck and other cancers, increased MASTL/ENSA or reduced B55 is associated with resistance to multiple chemotherapeutic agents, including anti-replication drugs. Experimentally, MASTL enhances cellular tolerance by promoting replication fork progression under stress, supporting fork restart, and promoting new origin firing. These effects require PP2A/B55 suppression, wh

Screening and identification of halofuginone direct binding target proteins based on thermal proteomics profiling.

Anticoccidial drugs remain the primary strategy for controlling avian coccidiosis caused by Eimeria parasites. However, widespread drug resistance and limited understanding of the molecular mechanisms underlying the action of existing anticoccidials severely impede the development of novel agents. In this study, simplified thermal proteome profiling coupled with precipitate-supported thermal proteome profiling was employed to globally identify halofuginone-binding proteins in Eimeria tenella. Sixteen candidate interacting proteins were prioritized through integrated bioinformatics analyses and

Cellulose-based dual-network hydrogels combining reversible Schiff base interactions and photopolymerized covalent networks.

Polysaccharide-based hydrogels have attracted considerable interest for biomedical applications due to their inherent biocompatibility, structural flexibility, and renewable origin. Among them, cellulose and its derivatives represent particularly promising building blocks; however, the limited mechanical stability and rapid degradation of conventional polysaccharide hydrogels under physiological conditions remain major challenges for their practical implementation. In this work, we report the development of cellulose-based dual-network hydrogels combining reversible Schiff base interactions an

Advances in plant polysaccharide-based self-healing hydrogels.

Traditional hydrogels suffer from irreversible structural damage under mechanical stress, which shortens service lifetimes and restricts their practical deployment in dynamic scenarios. Self-healing hydrogels constructed from plant-derived polysaccharides stand out against synthetic polymers and other biopolymers as promising biomaterials, owing to their abundant terrestrial resources, favorable biocompatibility, and inherent biodegradability. This review systematically summarizes recent advances in self-healing hydrogels based on major plant polysaccharides, including cellulose, hemicellulose

CAZyme-guided modification of cactus and regional polysaccharides for food hydrocolloid applications.

Food-hydrocolloid performance depends on polymer architecture and processing history rather than ingredient name alone. This critical narrative review evaluates CAZyme-guided modification of cactus-derived pectic materials and related regional polysaccharides for food hydrocolloid applications. Opuntia mucilage and pectic fractions are the focal substrates; non-cactus pectins provide the closest mechanistic evidence, and arabinoxylan is retained only as a bounded comparison for substitution and oxidative crosslinking. Starch is considered only as a limited process benchmark and is not used to

Therapeutic Drug Monitoring Considerations in Pregnancy.

Physiological changes during pregnancy can impact the pharmacokinetics of medications. Due to a variety of practical and ethical factors, clinical pharmacokinetic studies are challenging to perform during pregnancy. In addition, information on fetal toxicity of drugs requires years of study. As a result, evidence-based guidance for optimizing drug dosing in pregnancy, including therapeutic drug monitoring (TDM), is currently sparse. The most complete practice guidelines are related to antiseizure medications, with data also accumulating for antipsychotic and antidepressant medications. There a

The Drug's the Same but the Range Has Changed: Revisiting Therapeutic Ranges for Older Medications.

Therapeutic drug monitoring is an essential tool for optimizing pharmacotherapy and avoiding toxicity, yet for many older drugs, therapeutic ranges are derived from historical studies that may not reflect contemporary clinical practice. Changing clinical indications, greater recognition of patient-specific factors, and new pharmacokinetic data may support the need for updated ranges. This review examines methodological, clinical, and analytical challenges in defining and updating therapeutic ranges, including heterogeneous study designs, variable clinical endpoints, drug interactions, and inte

Eco-sustainable organic solvent-free spectrophotometric method for the concurrent determination of a ternary pharmaceutical mixture.

In recent years, green analytical chemistry has become essential in pharmaceutical research, aiming to mitigate environmental hazards associated with conventional analytical practices. The current focus lies in optimizing analytical methods to achieve both environmental and economic sustainability. In this framework, three environmentally sustainable spectrophotometric methods were developed for simultaneous analysis of three co-formulated drugs: acetaminophen, dextromethorphan hydrobromide, and pseudoephedrine hydrochloride, which are often used to alleviate symptoms associated with COVID-19.

What Have We Learned About Incentive Mechanisms?

This comment assesses the evidence from four papers on the development process for four breakthrough drugs or treatments. The comment notes that the papers show all the key breakthroughs in the development process were financed either through the National Institutes of Health or various foundations or charities, not drug companies looking to be compensated through patent monopolies. It argues that this evidence suggests patent monopolies are an unnecessary distortion that results in high drug prices and creates perverse incentives. Expanded public funding could replace patent monopolies, allow

Impact of Clinical Pharmacist-Led Interventions on the Interception of Medication Errors and Error-Related Toxicity in Pediatric Oncology Patients.

BACKGROUND: Pediatric oncology patients are highly susceptible to medication errors, because chemotherapeutic agents are high-alert drugs with a narrow therapeutic index requiring individualized, weight-based dosing. Prospective intervention-based evidence from resource-limited settings is scarce. This study evaluated the effect of clinical pharmacist interventions on medication errors in pediatric oncology inpatients. METHODS: In a prospective pre-post study in the central teaching hospital of pediatrics in Baghdad, Iraq (September 2025-April 2026), 125 patients receiving high-alert medicati

Investigator-Initiated Trials as a Clinical Development Approach for Orphan Drugs Compared with Industry-Sponsored Trials in Japan.

Orphan drug development faces substantial economic and operational challenges, particularly in small patient populations where conventional industry-sponsored trials (ISTs) may be difficult to conduct. In Japan, investigator-initiated trials (IITs) have been used to support the development of treatments for rare diseases and other areas of high unmet medical need; however, the characteristics of orphan drug approvals supported by IITs compared with those supported by ISTs have not been fully evaluated. This study analyzed pivotal trials supporting orphan drug approvals in Japan between 2015 an
