The Acetylcholine Esterase Inhibitor Donepezil Increases Dopamine Levels in the Nucleus Accumbens and Blocks the Alcohol Deprivation Effect in Rats.

Alcohol use disorder is associated with substantial morbidity and mortality arising from both medical consequences and accidents. Currently available pharmacotherapies are limited, and novel treatment options are needed. One putative target for pharmacotherapy is the mesolimbic dopamine system and its major target, the nucleus accumbens. Mesolimbic dopamine signaling is regulated by multiple neurotransmitters, among which acetylcholine is a key modulator. This study investigated whether the acetylcholine esterase inhibitor donepezil affects basal and/or alcohol-induced increases in extracellul

Efficacy and Safety of Baxdrostat in Resistant Hypertension: A Systematic Review and Meta-Analysis of Randomized Controlled Trials.

Resistant hypertension (RH) is characterized by persistently elevated blood pressure despite treatment with three or more antihypertensive agents, including a diuretic. Excess production of aldosterone is a key contributor to this condition. Baxdrostat is a novel, highly selective aldosterone synthase inhibitor that reduces aldosterone synthesis without affecting cortisol production. This systematic review and meta-analysis evaluated the efficacy and safety of baxdrostat in patients with RH. Following a preregistered PROSPERO protocol (CRD420251038564) and the Preferred Reporting Items for Sys

Trends of new diagnosis and treatment initiation of hypertension during the COVID-19 pandemic in Korea.

The COVID-19 pandemic has broadly disrupted healthcare utilization, potentially affecting the diagnosis and treatment of hypertension. While most reports focus on countries with strict lockdowns, the impact on care in countries like South Korea that did not impose lockdowns remains unclear. This study aimed to investigate the changes in trends of new diagnosis and treatment initiation of hypertension in Korea during the pandemic. Using Korean nationwide claims data from 2015 to 2020, we identified new diagnosis and treatment initiation of hypertension. New diagnosis was defined as a claim for

The NPR1 agonist antibody XXB750 in heart failure: a phase 2 randomized trial.

Therapies targeting the natriuretic peptide system have the potential to reduce death or heart failure events in heart failure with reduced ejection fraction. Here we assess XXB750, a human monoclonal antibody activating natriuretic peptide receptor 1, in patients with heart failure. Patients with heart failure and a left ventricular ejection fraction <50% were enrolled. Those patients who were on background angiotensin-converting enzyme inhibitor or angiotensin receptor blocker treatment were randomized to receive 60 mg XXB750, 120 mg XXB750 or placebo in a blinded fashion or sacubitril/valsa

Remission of severe familial proteinuria in a Bracco Italiano dog.

The objective of this report was to document clinical remission of suspected familial proteinuria in a Bracco Italiano dog. A 4-year-old intact female Bracco Italiano with a familial history of amyloidosis was presented because of unilateral chemosis, polyuria, and polydipsia. Hypoalbuminemia and severe proteinuria were detected via initial diagnostics. Although no renal histopathologic assessment was done, there was a strong clinical suspicion for renal amyloidosis. The dog was treated with telmisartan and colchicine (in addition to other therapies) for proteinuria and achieved clinical remis

Prescribing patterns for hypertensive disorders of pregnancy in low- and middle-income countries: a systematic review.

BACKGROUND: Hypertensive Disorders of Pregnancy (HDPs), encompassing gestational hypertension, pre-eclampsia, and eclampsia, affect approximately 18 million pregnancies globally. While international guidelines specify management for these disorders, their implementation in low-and middle-income countries (LMICs) is unassessed. Therefore, this systematic review aims to assess the prescribing patterns for HDPs to gauge suitable recommendations for practitioners in LMICs. METHODS: A systematic review was conducted using PubMed, CINAHL, Scopus and Google Scholar using relevant terms for HDPs, pres

Environmental sustainability assessment of reverse-phase liquid chromatography for retention and ionization profiling of angiotensin-converting enzyme inhibitors.

This paper presents a sustainable chromatographic framework using reverse-phase liquid chromatography (RPLC) method with an ultraviolet detector (UV) to determine the ionization behavior of three angiotensin-converting enzyme (ACE) inhibitors, benazepril, cilazapril, and quinapril, in a water-organic solvent binary mixture. In this study, retention-mobile phase pH data under various hydroorganic conditions were analyzed using nonlinear regression (NLREG) and linear solvation energy relationship (LSER) approaches to calculate the dissociation constant (pKass) values of the compounds. Accurate a

Discovery of ferulic acid carbamate derivatives as dual-targeting agents of BuChE and Nrf2 for Alzheimer's disease.

Given the multifactorial aetiology of Alzheimer's disease, multi-target strategies have emerged as a promising therapeutic approach. In this study, we designed and synthesised a series of ferulic acid carbamate derivatives to selectively inhibit BuChE and stimulate Nrf2 pathway. The biological evaluation revealed that compound 5c and 5e were the most potent, exhibiting over 150-fold selectivity for BuChE. Also, 5c, 5g and 5h significantly reversed both H2O2- and Aβ-induced toxicity in HT22 cells. These compounds were further shown to eliminate ROS accumulation induced by Aβ and upregulated HO-

Antihypertensive Drug Target Genes and Epithelial Ovarian Cancer Risk: A Two-Sample Mendelian Randomization Study.

PURPOSE: Findings on the relationship between the use of antihypertensive drugs and the risk of epithelial ovarian cancer (EOC) have been inconsistent. We performed a two-sample Mendelian randomization (MR) using gene expression of target genes as genetic proxies for antihypertensive drug action to interrogate this. METHODS: Genetic instruments for expression of antihypertensive medication target genes were identified with expression quantitative trait loci (eQTLs) in blood. Summary statistics for overall EOC and high-grade serous ovarian cancer (HGSOC) were obtained from genome wide associat

Effects of Genetically-Proxied Antihypertensive Drug Targets on Preeclampsia and Birth Weight.

BACKGROUND: Preeclampsia is a leading cause of maternal and perinatal morbidity and mortality, and a major contributor to low birth weight. Beta blockers (BB) and calcium channel blockers (CCB) are the most commonly recommended agents to treat hypertension in pregnancy. Yet it remains unknown whether these agents alter the risk of preeclampsia (PE), and if so, whether effects arise through maternal physiology or through direct fetal mechanisms. OBJECTIVES: To use drug-target Mendelian randomization (MR) to estimate the effects of genetically-proxied inhibition of beta-adrenergic and L-type ca

Regulation of the decision threshold by the locus coeruleus.

A fundamental challenge for decision-making under uncertainty lies in balancing speed and accuracy. Humans and animals solve this problem by adjusting decision thresholds-the criterion that determines how much information is required before committing to a choice. While brain regions associated with this process have been identified, the neural circuits that directly alter decision thresholds remain unknown. Here, we investigate the role of the locus coeruleus (LC) norepinephrine (NE) system in controlling this balance. Through cell-type-specific chemogenetic manipulations, we discovered that

Oleuropein-based olive leaf extract enhances muscle mitochondrial bioenergetics response to moderate - but not maximal - intensity exercise in humans.

Sprint interval exercise (SIE) induces skeletal muscle mitochondrial adaptations that are comparable to, or greater than, those observed with moderate-intensity continuous exercise (MICE), despite requiring a lower training volume. Previous work has shown that these adaptations are at least partly mediated by enhanced mitochondrial bioenergetics, including increased mitochondrial Ca2+ uptake and resulting pyruvate dehydrogenase (PDH) activation. In parallel, the natural compound oleuropein from olive leaf extract (OLE) promotes mitochondrial Ca2+ uptake and activates PDH in mouse skeletal musc
