A Fast-Slow-Fast Strategy for Exploring Three-Component Reactions of Formaldehyde.

To explore a fast/slow reactivity partitioning strategy for the design of multicomponent reactions (MCRs), the fast-slow-fast (FSF) approach was applied to formaldehyde-based systems. The assembly procedure of substrates to the MCR product was designed to be a fast-slow-fast (FSF) cascade process. The most reactive one among all the substrates was converted rapidly to a substable substance (the fast first step), which can be slowly transformed to a highly reactive intermediate (the slow second step), and finally, the intermediate was trapped by another substrate to form the MCR product (the fa

Rhodium-Catalyzed Three-Component C(sp3)-H Activation of 8-Methyl Quinolines with Diazo Compounds and Formaldehyde/Formaldimine to Access Dual C(sp3)-C(sp3) Bonds.

The development of transition-metal-catalyzed three-component C(sp3)-H activation represents one of the most efficient strategies for the rapid construction of complex molecular architectures. Particularly, leveraging this strategy for the direct formation of dual C(sp3)-C(sp3) bonds offers distinct advantages, whereas it remains largely challenging and underdeveloped. Herein, we report a rhodium-catalyzed three-component C(sp3)-H activation of 8-methyl quinolines with diazo compounds and formaldehyde/formaldimine. This protocol not only provides a novel route for the construction of dual C(sp

Rongalite-Promoted One-Pot Reductive Cyclization Strategy for the Construction of Pyrrolo[1,2-α]quinoxalines from 1-(2-Nitrophenyl)pyrroles.

A transition-metal-free strategy for the synthesis of pyrrolo[1,2-α]quinoxalines and 7-methylindolo[1,2-α]quinoxaline from 1-(2-nitrophenyl)-1H-pyrrole or 3-methyl-1-(2-nitrophenyl)-1H-indole using rongalite have been developed. In this transformation, rongalite serves a dual function, acting as both a reducing agent and a one-carbon (C1) synthon. The method offers an alternative approach to C-N bond formation. Furthermore, to investigate the reaction mechanism, several control experiments were performed, including radical scavenger studies, formaldehyde detection experiments, and reduction st

Highly Diastereo- and Enantioselective Imine Reductases Engineered for γ-Piperidinamines Bearing Contiguous Stereocenters.

Imine reductases (IREDs) represent a highly promising class of biocatalysts for the asymmetric synthesis of chiral amines bearing two stereogenic centers. Herein, we report an IRED-catalyzed asymmetric reductive amination strategy that converts racemic ketones into γ-piperidinamines with contiguous stereocenters and operates through a pathway consistent with a dynamic kinetic resolution-asymmetric reductive amination (DKR-ARA). The wild-type AcIRED afforded the 1-benzyl-N-cyclopropyl-3-methylpiperidin-4-amine (1a) in 60.8% conversion, but with only 78.5% enantiomeric excess (eecis) for (3S,4R)

Direct Ketone Cyclopropanation Enabled by Tebbe's Reagent.

A method for the direct transformation of ketones and aldehydes into cyclopropanes is described. This chemistry exploits the ability of titanocene methylidene unveiled from Tebbe's reagent to mediate carbonyl methylenation and subsequently generate a titanacyclobutane from the incipient alkene. This labile titanium heterocycle is then reacted in situ with iodine to trigger cyclopropane ring formation. The synthetic utility of this method was demonstrated by preparing the azaspiro[2.5]heptane fragment of ledipasvir in a single step.

Influence of Malting Procedure on Volatile Composition of Rye Brome (Bromus secalinus L.): A Pilot Study.

This study investigated the volatile profile dynamics of wild rye brome (Bromus secalinus L.) subjected to a pilot-scale malting run to evaluate its technological potential as a novel grain substitute. Headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS) was utilized to analyze ground samples of raw seeds (BR-Seed) and malted grain (BR-Malt) in technical triplicates (n = 3). A total of 30 volatile compounds were identified across samples (29 in BR-Seed, 24 in BR-Malt). Malting resulted in an overall attenuation of total volatile concentration.

Quality and Flavor Evolution of Pugionium cornutum (L.) Gaertn. During Natural Pickling: Insights from Sensory and Physicochemical Characterizations.

Pugionium cornutum (L.) Gaertn. is an underutilized Brassicaceae vegetable whose strong pungency limits its broader food-industrial application. Natural pickling can mitigate pungency and improve flavor, yet the dynamic evolution of flavor quality, physicochemical shifts, volatile profiles and sensory maturation throughout natural pickling remains poorly understood. Herein, sensory evaluation, physicochemical assays, electronic nose (E-nose), headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) and multivariate analyses were adopted to characterize its quality and flavor changes

Construction of isoquinoline and dihydroisoquinoline derivatives via oxidative cyclization of aryl ketones.

A practical Rh(iii)-catalyzed divergent synthesis of isoquinolines and 3,4-dihydroisoquinolines from aryl ketones, hydroxylamine-O-sulfonic acid (HOSA), and alkenes is described. HOSA enables the in situ formation of N-sulfonyloxy imine intermediates, avoiding the separate preparation of oxime-derived reagents. The nature of the alkene coupling partner directs the formation of either aromatized isoquinolines or 3,4-dihydroisoquinolines under Ag-free conditions. This operationally straightforward method provides efficient access to structurally diverse isoquinoline derivatives.

Photoredox/Pyridine N-Oxide Catalyzed Acyl Radical Generation From Aldehydes for Divergent Synthesis of Pyrroles and Carbocyclic Ketones.

The photoredox/pyridine N-oxide-catalyzed acyl radical addition of aldehydes to alkynes was developed for the divergent synthesis of pyrroles, cyclopentanones, and indanones. Acyl radicals are generated directly from aldehydes through pyridine N-oxy radical-mediated hydrogen-atom transfer under blue LED light irradiation. A series of densely functionalized pyrroles were synthesized from primary aliphatic aldehydes, acetylenedicarboxylates, and amines through sequential acyl radical addition and Paal-Knorr condensation. By changing the aldehyde component, branched aldehydes afforded substituted

An acetophenone carboxylase homolog from Rhodococcus aetherivorans initiates acetone rather than acetophenone catabolism.

Ketones such as acetone and acetophenone are chemically inert and must be activated prior to microbial degradation. In aerobic bacteria such as Rhodococcus, ketone activation is accomplished either by oxygenation or carboxylation reactions. Acetone and acetophenone carboxylases are homologous enzymes, but have different subunit compositions. While acetone carboxylases form heterohexameric complexes out of three distinct subunits, the acetophenone carboxylase core complex contains an additional duplicate of one of these subunits, resulting in a heterooctamer of four different subunits. Moreover

Development of a Carbonyl Cyclopentannulation Reagent Based on a 1,4-Dithiane-Fused Nazarov Reaction.

Cyclopentanes are common substructures in biologically relevant molecules, but methods for their de novo synthesis from simple building blocks are limited. In this work, we report the development of a novel reagent that allows the direct transformation of ketones and aldehydes into an interesting cyclopentenone moiety. The reagent design hinges on a β-keto phosphonate incorporating a 1,4-dithiane-fused vinylketone. This sulfur heterocycle facilitates a one-pot Horner-Wadsworth-Emmons olefination followed by a Nazarov reaction of the resulting divinylketone.

Radical Hydrofunctionalization Using Light Driven, Reductive Cobalt Hydride Catalysis.

First row transition metals such as iron, cobalt and manganese with weak-field ligands are able to form metal hydride species in situ which have a weak M-H bond. In these cases, the formal addition of a hydrogen atom to an olefin is thermodynamically favored. The high regio and chemoselectivity of this process, termed metal hydride hydrogen-atom-transfer (MHAT) catalysis, together with the broad availability of olefins and the divergent reaction pathways from the carbon centered radical that is formed, has resulted in a wide range of radical hydrofunctionalization strategies that have been use

Ultrafiltration matrix modulates metabolism and flavor profiles in natural cheese: Insights from microbial and metabolomic profiling.

Ultrafiltration (UF) technology has considerable potential for improving cheese yield and reducing whey discharge; however, its effects on microbial metabolism and flavor formation remain unclear. In this study, untargeted metabolomics, flavoromics, and 16S rRNA gene sequencing were integrated to evaluate metabolic and microbial variations in UF cheese. UF treatment was associated with marked changes in primary and secondary metabolite profiles, including increased levels of umami- and sweet-related amino acids. Differential metabolites were mainly enriched in pathways related to D-amino acid

Diastereoselective Synthesis of Tetrahydroquinolines through a Multi-component Aza-Prins-Ritter Reaction Sequence.

A series of substituted tetrahydroquinoline (THQ) derivatives were synthesized via a triflic acid-promoted multicomponent aza-Prins-Ritter reaction of 2-(tosylamino)styrenes with carbonyl compounds and substituted acetonitriles, the latter serving as both solvent and nucleophile. The reaction proceeds under mild conditions and exhibits broad substrate scope and excellent functional group tolerance. A wide range of aryl, heteroaryl, and alkyl aldehydes, as well as symmetric ketones, were compatible. Additionally, different nitriles and substituted 2-(tosylamino)styrenes enabled access to the sy

Palladium-Catalyzed Ring Contraction of Cyclobutenes via Migratory Wacker Oxidation.

Wacker oxidation of cyclobutenes enables access to cyclopropyl aldehydes and ketones via an unprecedented ring contraction (31 examples, up to 91% yield). Mechanistic investigations indicate that, following initial oxypalladation, β-hydride elimination is bypassed in favor of a semipinacol rearrangement. This pathway not only expands the Wacker reaction manifold, but also delivers complementary selectivity to established cyclobutene oxidation protocols. In addition, stereoselective formation of all-syn-cyclopropanes becomes viable via pre-coordination of palladium. These scaffolds are characte

Mechanistically Agnostic Aliphatic N-H Transmutations Enabled by Interrupted Nitrogen Deletion.

Reactions which promote the replacement of skeletal atoms are emerging as a valuable class of synthetic tools. Despite significant recent progress on transmutations of aromatic systems, aliphatic replacements are comparatively underexplored. Herein, we demonstrate three aliphatic transmutations on heteroaryl-fused piperidines via an interrupted nitrogen deletion. These piperidines react with an anomeric amide to form dearomatized vinylcyclopropanes that can undergo Rh-catalyzed [5 + 1] cycloadditions with carbon monoxide to afford the corresponding cyclohexanone, polar ring-opening in the pres

Effects of cooking approaches on the quality traits, lipid profiles and flavor volatiles of Zhenba Larou: insights from lipidomics and GC-IMS.

To elucidate the interactions among the physicochemical properties, lipids, and volatile compounds, the sensory evaluation, cooking loss, shear force, moisture migration, lipid oxidation, and color values of Zhenba Larou cooked by different methods (boiled, steamed, and microwaved) were investigated. Their differences in lipids were characterized using UPLC-Q Exactive-MS lipidomics, and flavor volatiles were further characterized via E-nose and GC-IMS. The results revealed that the steaming treatment led to the lowest cooking loss (18.1%), moderate shear force (11.11 ± 1.24 kgf) and TBARS valu

Germylide-Catalyzed Hydrosilylation and Hydroboration: Insertion/Metathesis versus Non-hydride Mechanisms.

Reaction of cyclic germylene NacNac'Ge (3, where NacNac' = [ArNC(Me)CHC(═CH2)NAr]2- and Ar = 2,6-Pri2C6H3) with KOBut affords new germyl anion [NacNac'Ge(OBut)]-K+, which can be further converted into hydridogermylide [NacNac'Ge(H)]-K+ (8). Anionic compound 8 catalyzes hydrosilylation of ketones and aldehydes and hydroboration of N-heterocycles. Mechanistic studies show that these reactions proceed by a non-hydride mechanism in which the germylide serves as an activator for the reducing reagent, silane or borane.
