An enzymic assay for the specific determination of methanol in serum.

This method for the specific determination of methanol in serum is based on the following two reactions: (formula; see text) Alcohol oxidase is not specific: it converts all lower alcohols to their corresponding aldehydes; however, formaldehyde dehydrogenase is specific and thus the transformation of NAD+ to NADH (which is used to monitor the reaction) proceeds only if methanol is originally present in the sample. The method was automated with a Roche COBAS FARA centrifugal analyzer. The calibration curve is linear between 0.6 and 12 mmol/L. The detection limit is about 0.6 mmol/L. The CV is 4

Characterization of rat testicular alcohol dehydrogenase.

A protein from rat testes that catalyzes the oxidation of ethanol in the presence of NAD+, but not NADP+, has been characterized enzymatically and compared to that of hepatic alcohol dehydrogenase obtained from the same animals. The testicular enzyme, like the hepatic enzyme, has a Km value for ethanol in the 0.5-1.0-mM range and can utilize other alcohols such as n-propanol, n-butanol, and isobutanol, although the Km values for these other alcohols are considerably lower (0.03-0.08 mM) that that for ethanol. The testicular enzyme is more heat-labile than is the hepatic enzyme. Finally, the te

Effect of ethanol and other aliphatic alcohols on the intestinal transport of folates.

The effect of ethanol and other aliphatic alcohols on the intestinal transport of 5-methyltetrahydrofolate and folic acid was examined using everted sacs from rat jejunum. Ethanol added to the mucosal medium inhibited the transport of both folate compounds in a parallel manner, and the inhibition increased with increasing ethanol concentration (0.5-10% v/v). Ethanol at 3% v/v in the mucosal medium caused: depression in the pH dependency of the active transport of 5-methyltetrahydrofolate; higher inhibition in the transport of low concentration (0.1 microM) than high concentration (10 microM) o

Acute effects of ethanol and other short-chain alcohols on the guanylate cyclase system of murine neuroblastoma cells (clone N1E-115).

The acute effects of ethanol were studied on the guanylate cyclase system of cultured murine neuroblastoma clone N1E-115. Using intact cells, we found that although ethanol had no effect on basal levels of cyclic GMP synthesis, it rapidly inhibited in a concentration-dependent manner cyclic GMP synthesis mediated by the agonists histamine (histamine H1 receptor) and carbachol (low-affinity muscarinic receptor) and by ionophore X537A and melittin, agents which bypass these receptors. At 200 mM ethanol, inhibition was about 40 to 50% with the agonists, X537A and melittin. Ethanol had no effect o

Membrane solubility of ethanol in chronic alcoholism. The effect of ethanol feeding and its withdrawal on the protection by alcohol of rat red blood cells from hypotonic hemolysis.

Membranes from ethanol-fed rats are resistant to the in vitro effects of ethanol on membrane structure and function. We have proposed that the resistance arises from adaptive changes in membrane composition which lower the solubility (partition coefficient) of ethanol in these membranes. The partition of ethanol (and other alcohols and anesthetics) into red blood cells protects the cells from hypotonic hemolysis. Here, we show that the protection by alcohols and anesthetics of red blood cells from ethanol-fed rats is greatly attenuated. This finding indicates that the membrane solubility of th

Intestinal hydrolysis of pyridoxal 5'-phosphate in vitro and in vivo in the rat: effect of ethanol.

The effect of ethanol on intestinal hydrolysis of pyridoxal 5'-phosphate (PLP), the first step in PLP absorption, was evaluated in the rat. Models included PLP decay during in vitro incubation and PLP luminal disappearance during in vivo perfusion of jejunal segments. Ethanol inhibited PLP in vitro decay (pH = 7.4 and 3.0) and in vivo disappearance (pH = 7.4) in a concentration-dependent manner (1-4% w/v). At pH = 7.4, 4% w/v ethanol inhibited in vitro PLP decay by 48% (p less than 0.001) and in vivo PLP disappearance by 55% (p less than 0.001). This inhibition was reproduced in vitro with oth

Growth of methanogenic bacteria in pure culture with 2-propanol and other alcohols as hydrogen donors.

Two types of mesophilic, methanogenic bacteria were isolated in pure culture from anaerobic freshwater and marine mud with 2-propanol as the hydrogen donor. The freshwater strain (SK) was a Methanospirillum species, the marine, salt-requiring strain (CV), which had irregular coccoid cells, resembled Methanogenium sp. Stoichiometric measurements revealed formation of 1 mol of CH(4) by CO(2) reduction, with 4 mol of 2-propanol being converted to acetone. In addition to 2-propanol, the isolates used 2-butanol, H(2), or formate but not methanol or polyols. Acetate did not serve as an energy substr

Breakthrough behavior of 17.5 mol% water in methanol, ethanol, isopropanol, and t-butanol vapors passed over corn grits.

Ground corn is now used in industry as an adsorbent to remove water from ethanol vapors. It is stable and inexpensive at 10 cents/lb (22 cents/kg). For regeneration it requires less than 2000 Btu/gal of 190 proof ethanol processed. If necessary, it could be readily saccharified and fermented into ethanol after use. This renewable resource has further exciting potential as an inexpensive adsorbent for water removal from other alcohols, including methanol, isopropanol, and t-butanol. Water sorption capacity in a fixed bed, nonisothermal adsorption column appears to be a function of the heat capa

New instrument using gas sensors for the quantitative analysis of ethanol in biological liquids.

A gas sensor was built into an instrument to measure ethanol in biological liquids by determining head space ethanol concentrations without chromatography. The analysis of plasma, urine, and whole blood containing ethanol over the range 20-640 mg/dl determined by this novel instrument is fast (30 sec), accurate (r = 0.99), and precise (coefficients of variation 0.6-1.8%) when compared with gas chromatography. The addition of alcohol dehydrogenase, beta-nicotinamide adenine dinucleotide, and semicarbazide to samples allows ethanol to be distinguished from other alcohols. The instrument can be b

Differing effects of short-chain alcohols on body temperature and coordinated muscular activity in mice.

Administration of single doses of ethanol, 1-propanol, 1-butanol or 1-pentanol to mice caused hypothermia and impairment of rotarod performance. Repetitive doses, at 24-72 hr intervals led to development of tolerance to the hypothermic effects of ethanol but not of the other alcohols. No tolerance was seen in the impairment of rotarod performance with repeated doses of any of the alcohols. Ethanol did show an intersession tolerance on rotarod performance; at 20 and 80 min after injection, blood levels were similar, while performance was impaired at 20 but not at 80 min.

Immunochemical evidence for induction of the alcohol-oxidizing cytochrome P-450 of rabbit liver microsomes by diverse agents: ethanol, imidazole, trichloroethylene, acetone, pyrazole, and isoniazid.

Isozyme 3a of rabbit liver microsomal cytochrome P-450, also termed P-450ALC, was previously isolated in this laboratory from animals administered ethanol or imidazole, and the purified cytochrome was shown to function in the reconstituted system as an oxygenase in catalyzing the oxidation of ethanol and other alcohols. Although liver microsomes from animals treated in various ways exhibit increased alcohol-oxidizing activity, evidence was not available as to whether this was due to enzyme induction or to other factors influencing the activity. Immunochemical quantitation of P-450 isozyme 3a h

Teratological assessment of methanol and ethanol at high inhalation levels in rats.

Alcohols are widely used as industrial solvents. In spite of the fact that ethanol is a human teratogen, there has not been systematic investigation of the potential teratogenic effects of other alcohols, particularly using the inhalation route of exposure, as would be appropriate in assessing occupational and environmental types of experience. As part of a large teratological examination of industrial alcohols, methanol and ethanol were administered by inhalation to groups of approximately 15 pregnant Sprague-Dawley rats. Methanol was administered at 20,000 ppm (20ME), 10,000 ppm (10ME), 5000

1,4 Butanediol, gamma-hydroxybutyric acid and ethanol: relationships and interactions.

We have investigated the interaction of 1,4 butanediol (1,4 BD) with ethanol and the involvement of the major metabolite of 1,4 BD, gamma-hydroxybutyric acid (GHB) in the ability of 1,4 BD to produce behavioral and EEG changes in rat as well as the toxic side effects of 1,4 BD. Behavioral, electrical, and biochemical studies in rats suggest that the effects of 1,4 BD are indeed mediated by GHB. Further, ethanol appears to block conversion of 1,4 BD to GHB. 1,4 BD however potentiates some of the behavioral effects of ethanol perhaps by a mechanism of action similar to that of other alcohols. Metabolism of alcohol and ketone by cytochrome P-450 oxygenase: fluoren-9-ol in equilibrium with fluoren-9-one.

Fluoren-9-ol and fluoren-9-one were used as model substrates to study microsomal metabolism of alcohols and carbonyl compounds. It was found that there was an oxidoreductase(s) present in the microsomal preparation that catalyzed interconversion of this alcohol and ketone using pyridine nucleotides as cofactors. The alcohol could also be oxygenated to the ketone by microsomes and NADPH. It required molecular oxygen. This oxygenation was inhibited by SKF 525-A and CO, suggesting the involvement of P-450 oxygenase. Using highly purified P-450 oxygenase components, it was shown that fluoren-9-ol

Alcohol oxidation by isozyme 3a of liver microsomal cytochrome P-450.

Liver microsomes from rabbits treated chronically with ethanol were solubilized and fractionated to yield a new isozyme of cytochrome P-450 in a homogeneous state. This cytochrome, designated as isozyme 3a on the basis of its relative electrophoretic mobility, is distinct from the known terminal amino acid sequences. In addition, peptide mapping by high performance liquid chromatography following trypsinolysis indicates that form 3a is a unique gene product. This cytochrome has unusually high activity in the oxidation of ethanol and other alcohols to aldehydes and in the rho-hydroxylation of a

Alcohol interactions with brain opiate receptors.

Ethanol, added in vitro to mouse caudate membranes, inhibited high-affinity binding of 0.2 nM 3H-dihydromorphine (3H-DHM) over an ethanol concentration range of 250-1,000 mM. At lower, physiologically-attainable ethanol concentrations (e.g.: 50 mM), 3H-DHM binding was significantly increased. Over the concentration range of 50-1,000 mM, ethanol inhibited 0.5 nM 3H-[D-Ala2,D-Leu5]enkephalin (3H-ENK) binding to mouse caudate tissue, and no stimulation of 3H-ENK binding was noted at any of these concentrations of ethanol. Uptake of monohydric alcohols by liver: demonstration of a shared enzymic space.

Multiple-indicator dilution studies of the hepatic uptake of straight-chain C1-C5 monohydric alcohols were carried out in anesthetized dogs, with either no preceding or saturating infusions of ethanol, and at different steady-state levels for the C2 experiments. Labeled red cells were utilized as a vascular reference, and labeled water was used as a second reference entering liver cells. Kinetic analysis of the data provided estimates of both an uptake rate constant and the space of distribution available to label. From the decrease in the uptake rate constant for labeled ethanol with bulk con

Phenylethanolamine-N-methyltransferase:alcohol and the mechanism of action.

Mixed-solvent systems of methanol and other alcohols and water were used to study the properties of bovine phenylethanolamine-N-methyltransferase. The presence of methanol decreased the binding affinity of the enzyme for its amine substrate but did not alter the maximum velocity. The change in binding was accompanied by an alkaline shift in the pK of an ionizable group in the active site. The well-known property of enzyme inhibition by substrate was also alleviated. Increasing the pH of the medium, in the presence or absence of methanol, increased the maximum velocity but did not alter substra
