We examined effects of ethanol and dimethyl sulfoxide on the regulation and apparent thermodynamic properties of moderate affinity Na+ and K+ binding that regulates the K+-dependent phosphatase activity of (Na+,K+)-ATPase. Ethanol and other alcohols reduced the apparent affinity for Na+ and K+ at their moderate affinity sites and increased the negative delta H and delta S of cation binding. Dimethyl sulfoxide had the opposite effects. Inhibition by ethanol was favored by high temperature or low K+. Ethanol potentiated inhibition of K+ binding by ATP or Mg2+. Ethanol also shifted the equilibriu

Agents that are known to be scavengers of hydroxyl radicals inhibit lymphocyte mitogenesis induced by phorbol myristate acetate (PMA) to a greater extent than they inhibit mitogenesis induced by concanavalin A or phytohemagglutinin. These agents include dimethyl sulfoxide, benzoate, thiourea, dimethylurea, tetramethylurea, L-tryptophan, mannitol, and several other alcohols. Their inhibitory effect is not associated with cytotoxicity. The hydroxyl radical scavengers do not inhibit PMA-dependent amino acid transport in T cells or PMA-induced superoxide production by monocytes. Thus, they do not

The effect of 1,2,4-triaminotriazole on the activity of catalase was studied in the resting cells of Candida mycoderma when they oxidized glucose, ethanol and other alcohols. The compound was shown to inhibit the activity of catalase in the cells when the endogenous substrate and glucose were oxidized, but it had no effect on its activity during the oxidation of alcohols (C1-C4). Catalase is supposed to be involved in the oxidation of lower alcohols forming a complex with them and thus preventing the inhibiting action of 1,2,4-triaminotriazole.

The effect of alcohols on the spectral properties of riboflavin derivatives in non-polar solvent was studied by various spectroscopic methods in order to support the view point that alcohol may directly interact with the isoalloxazine moiety of FAD and enhance the catalytic activity of D-amino acid oxidase (DAAO). The most likely association complex between alcohol and riboflavin is 1 : 1 stoichiometric complex through the 3-N imino and the 2-C carbonyl groups of the isoalloxazine ring and the hydroxyl group of alcohols. It appears that methanol has a larger association constant than any other

The substrate specificity of a bacterial analogue of the plasma enzyme lecithin:cholesterol acyltransferase (LCAT) has been examined with small unilamellar liposomes and Triton mixed micelles. In contrast to LCAT, the microbial enzyme is capable of using all of the naturally occurring phospholipids as acyl donors. In general reaction rate depends more on the length or degree of unsaturation of the acyl chains than on the nature of the phospholipid head group. Among a series of disaturated phosphatidylcholines in liposomes, dilauroylphosphatidylcholine is the preferred acyl donor. Like LCAT, th

Evaluation has been made of ultrastructural variations which obtain following dehydration and infiltration of glutaraldehyde plus osmium fixed CHO cells with the weakly protic alcohols (methanol, ethanol, ethylene glycol, 1-propanol, 2-propanol, propylene glycol and glycerine), acetone alone, and with use of acetone and propylene oxide as transition solvents subsequent to dehydration with ethanol, methanol and ethylene glycol. It appears that the weakly protic alcohols, applied without transition solvent, in addition to some extraction by themselves, variously condition the components of membr

The rat paw edema due to carrageenan, yeast, and dextran was dose-dependently antagonized by oral administration of ethanol. Carrageenan-induced paw edema was also inhibited by methanol, 1-propanol, and 1-butanol. The rat carrageenan pleurisy was reduced by oral pretreatment with methanol, ethanol, 1-propanol, 1-butanol, and dimethyl sulfoxide. Central depression, altered adrenal functions, increased osmolality, and hypothermia were no important factors for these actions of solvents. It is concluded that the inhibitory effect of ethanol on inflammatory responses increases the susceptibility of

The effects of alcohols on human glycophorin were monitored by circular dichroism, solvent perturbation of absorption spectra, fluorescence of 8-anilino-1-naphthalene sulfonate, and sedimentation equilibrium in the ultracentrifuge. Both ethanol and 2-chloroethanol gradually increase the alpha helix in glycophorin and its sialic acid free counterpart. The same alcohols do not cause a cooperative transition in the structure of the polypeptide chain of glycophorin. Other alcohols also increase the alpha-helix content of glycophorin. Binding of ANS to glycophorin is abolished at relatively low alc

Alcohol is mutagenic, cancerogenic and teratogenic in man. Ethanol is mutagenic via its first metabolite, acetaldehyde. This is substantiated by the findings that acetaldehyde induces chromosomal aberrations, sister-chromatid exchanges and cross-links between DNA strands. Methanol, a contaminant of many alcoholic beverages, is also mutagenic via its metabolite, formaldehyde. In addition, different indirect pathways may lead to mutations by alcohol. The cancerogenic activity of alcohol remains unverified by modern standard carcinogenicity tests. Ethanol and other alcohols, as well as aldehydes,

We describe a case of ethanol and isopropanol ingestion that resulted in coma. The concentration of ethanol and isopropanol was 0.90 and 1.65 g/liter in serum and 3.12 and 5.34 g/liter in gastric contents. With an enzymatic (alcohol dehydrogenase) method for ethanol determination we obtained erroneous analytical results. Because of partial cross reactivity with isopropanol, ethanol concentration was overestimated and total alcohol (i.e., the contribution of isopropanol) was underestimated. This was recognized by measuring serum osmolality. Differences between measured and calculated serum osmo

The allozyme polymorphism at the alcohol dehydrogenase locus in Drosophila melanogaster was studied in order to obtain experimental evidence about the maintenance of this polymorphism. Populations started with different initial allele frequencies from homozygous F and S lines showed a convergence of frequencies on regular food at 25 degrees, leading to values equal to those in the base populations. These results were interpreted as due to some kind of balancing selection. In populations kept at 29.8 degrees, a lower equilibrium F frequency was attained. Addition of ethanol and some other alcoh

In this study ethanol and certain other short-chain aryl (benzyl and phenethyl) and aliphatic (methyl, propyl, butyl, and amyl) alcohols produced up to 10-fold increases in cyclic AMP (cAMP) concentrations in purified human peripheral blood lymphocytes. Ethanol concentrations as low as 80 mg/dl produced significant elevations in lymphocyte cAMP. Significant but less marked augmentation of cAMP in response to alcohols was observed in human platelets, human granulocytes, and rabbit alveolar macrophages. The mechanism of the alcohol-induced cAMP accumulation is probably secondary to membrane pert

The role of alcohols in potentiating the toxicity of halogenated hydrocarbon solvents has been reviewed. The toxicity of carbon tetrachloride and chloroform can be markedly potentiated by prior treatment with ethanol or phenobarbital. Trichloroethylene toxicity may also be potentiated by ethanol ingestion. Prior ethanol ingestion acts by altering biochemical parameters that result in an increased response to subsequent solvent exposure. Simultaneous exposure to both ethanol and trichloroethylene allows for competitive substrate inhibition of metabolism since these compounds share several commo

Stability of C1- and C2-cellulases, CX-exo- and CX-endoglucanases and beta-glucosidase of Aspergillus awamori was studied as affected by monoatomic aliphatic alcohols --methanol, ethanol, propanol and isopropanol; bi- and triatomic alcohols - ethylene glycol and glycerol, urea as well as detergents of dodecyl sulphate and sodium nonilate. The mentioned enzymes are established to manifest the highest activity in 40-60% glycerol. It is also shown that their stability is changed differently under the effect of other alcohols, urea and detergents. The latter testifies to the fact that the studied

Ethanol and other alcohols stimulate adenylate cyclase activity in various tissues and potentiate its stimulation by some hormones. This effect, however, usually requires a high alcohol concentration. In some cases, an unknown substance, different from cyclic AMP, was formed from ATP in the presence of an alcohol and mimicked stimulation of adenylate cyclase. Ethanol inhibits phosphodiesterase activity in some tissues. In the brain, only the low affinity enzyme of pons-medulla region is inhibited. ATP levels and ATPase activities are affected by ethanol treatment and this can lead to secondary

Ethanol through its primary catabolite, acetaldehyde, competitively inhibits oxidation of aldehyde dehydrogenase substrates. As a consequence biogenic amines form increased quantities of alcohols rather than the corresponding acids. During this biotransformation, condensation reactions between deaminated and intact amines may occur which can yield tetrahydropapaverolines. These compounds are closely related to precursors of opioids which is cause to link ethanol abuse to morphine addiction. There is, however, no pharmacological or clinical evidence suggesting similarities between ethanol depen

The rate of penetration of the simpler alcohols into brine shrimp cysts and cucumber seeds was studied. In solutions below 70% the rate of penetration is related to lipid solvent capacity of the alcohol. In concentrations above 70%, particularly in absolute alcohols, methanol penetrates brine shrimp rapidly and ethanol penetrates slowly. All the other alcohols tested did not pepetrate the dormant structures. Ethionine and deuteroxy-methanol did not affect the rate of penetration of methanol. It is suggested that in dehydrated membranes the lipid moiety is protected by a continuous sheet of pro

The rate constants for alcoholic solvolysis of the hydrochloride salts of diethylaminodibenzo(a,d)cycloheptene and related amino congeners were determined. The objective was a study of the comparative ease of cleavage of the C-N amino linkage by various aliphatic alcohols. The interaction of protonated amines of this series with alcoholic hydroxyls presumably leads to formation of the corresponding ethers in a manner somewhat analogous to alkoxide reaction with alkyl bromides. The methyl ether produced from solvolysis of diethylaminodibenzocycloheptene hydrochloride was isolated and identified

The ability to utilize methanol was found in the yeast strains isolated from forest substrates (Zygowillia pini IBFM y-655, Torulopsis molicshiana IBFM y-657) and in the collection yeast cultures (Candida boidinii IBFM y-588 and IBFM )-587). These yeasts assimilated not only methanol but other alcohols (ethanol, glycerol, erythrol, mannitol, and sorbitol), nitrates or nitrites. Their fermentation ability was low.
