Silybum marianum peptide-rich fraction alleviates acute liver injury by inhibiting TRAF3/NIK/NF-κB and is associated with changes in gut microbiota composition.

Acute liver injury (ALI) is a severe and prevalent condition that can progress to irreversible liver damage and failure. This study aimed to investigate the prophylactic hepatoprotective potential of Silybum marianum peptides (SMP) in ALI and elucidate its underlying mechanisms. Using a carbon tetrachloride (CCl4)-induced ALI mouse model, we evaluated the dose-dependent effects of SMP. Additionally, we explored the modulatory effects of SMP on the NF-κB pathway, gut microbiota composition, and metabolite profiles. The role of SMP in the TRAF3/NIK/NF-κB signaling axis was further validated in a

Distinct collagen resolution dynamics in early versus advanced liver fibrosis: a quantitative SHG/TPEF study in mice.

BACKGROUND & AIMS: Liver fibrosis regression occurs after the source of injury is removed, but its dynamics and histologic features remain poorly characterized. Second-harmonic generation/two-photon excited fluorescence (SHG/TPEF) microscopy enables quantitative, high-resolution collagen assessment. We employed SHG/TPEF to characterize spatiotemporal collagen remodeling during spontaneous regression in carbon tetrachloride (CCl4)-induced mouse models. METHODS: Early- and advanced-stage liver fibrosis were established by intraperitoneal CCl4 injection for 8 or 12 weeks. After peak injury, mice

Dietary Supplementation of Auraptene, a Major Component in Citrus reticulata Blanco, Prevents Acute Liver Injury Associated with Apoptosis and Inflammation.

Auraptene (AP), a coumarin derivative abundant in citrus peels, exhibits diverse bioactivities; however, its role in acute liver injury and the underlying mechanisms remain elusive. This study investigates the hepatoprotective effects of AP against carbon tetrachloride (CCl4)-induced acute liver injury in mice and explored its mechanistic basis. In vitro, transforming growth factor-beta (TGF-β) was employed to stimulate AML-12 hepatocytes to model the injury microenvironment. The impact of AP on the toll-like receptor 2 (TLR2) signaling pathway was assessed through a combination of siRNA gene

Unveiling the Role of the Soil Matrix in Trichloroethylene Degradation by Ball-Milled Sulfidated Microscale Zero-Valent Iron.

Ball-milled sulfidated microscale zerovalent iron (S-mZVIbm) is highly effective in degrading trichloroethylene (TCE) in anoxic water. This study investigated its performance and soil-mediated regulation in a soil-water mixed system (SWMS) using two natural soils. The results show that soil addition markedly enhanced the reactivity of S-mZVIbm. Compared with the aqueous system, the degradation rate constant of TCE (kTCE) exhibited a 6.0-8.4-fold increase at a soil-to-water ratio (S/W) of 0.5 to 2.0, while H2 production concurrently increased 210%-450%. This activation was accompanied by a decr

Antifibrotic activity of Schistosoma japonicum-derived sja-miR-2a-3p in carbon tetrachloride-induced liver fibrosis.

BACKGROUND: Liver fibrosis is a common consequence of chronic liver injury and is driven largely by persistent activation of hepatic stellate cells and excessive extracellular matrix deposition. We previously identified Schistosoma japonicum-derived sja-miR-2a-3p as a parasite-derived microRNA with antifibrotic activity in schistosomiasis-associated liver fibrosis. Whether this cross-species regulatory molecule also modulates fibrogenesis beyond the context of schistosome infection remains unclear. METHODS AND RESULTS: We investigated the effects of sja-miR-2a-3p in a carbon tetrachloride-ind

High-Fat Diet-Induced Metabolic Stress Durably Reprograms the Liver Regenerative Compartment Toward a Senescence-Like State.

Chronic metabolic stress is increasingly recognized as a driver of aging-like tissue dysfunction, but whether it durably reprograms liver regenerative cells, potentially compromising tissue maintenance and repair, remains unknown. Here, we show that high-fat diet (HFD)-induced metabolic stress promotes a persistent senescence-like state in the liver regenerative compartment. In male rats, chronic HFD feeding induced hepatic steatosis together with oxidative stress, inflammatory responses, and senescence-associated changes. Following carbon tetrachloride-induced liver injury, HFD-fed animals ex

TGF-β/p38 signaling axis regulates hepatocellular senescence and promotes Mallory-Denk body formation.

Mallory-Denk bodies (MDBs), a hallmark pathological feature of metabolic dysfunction-associated steatotic liver disease (MASLD), have been associated with hepatocellular senescence; however, the underlying regulatory mechanisms remain incompletely understood. We investigated whether the transforming growth factor-β (TGF-β)/p38 mitogen-activated protein kinase (p38 MAPK) signaling axis serves as a shared regulatory pathway driving both MDB formation and hepatocellular senescence. To address this, we established two MDB-positive mouse models: a metabolic dysfunction-associated steatohepatitis (M

Spatial transcriptomics reveals renal ER stress in alcohol-associated kidney injury.

Alcohol misuse damages numerous organs, including the liver, heart, brain, and pancreas. The kidney remains a severely understudied target of alcohol-induced injury despite renal injury being commonly observed in end-stage alcohol-associated liver disease (ALD) patients. A 6-week murine ethanol (1-6% v/v) model for early-stage ALD combined with carbon tetrachloride (CCl4) treatment was utilized to reveal mechanisms of renal injury. This paradigm yields elevated kidney damage markers when compared to ethanol consumption alone, including elevated urinary KIM-1 in 3 weeks. Hypothesis generating t

Nuciferine Suppresses Inflammatory Hepatocarcinogenesis by Targeting cPLA2-Dependent Arachidonic Acid Metabolism.

Chronic inflammation is a hallmark of hepatocellular carcinoma (HCC) and is closely linked to aberrant arachidonic acid (AA) metabolism; however, whether AA mobilization is driven by defined upstream regulatory mechanisms and can be therapeutically targeted remains largely unresolved. Here, we identify cytosolic phospholipase A2 (cPLA2) as a central regulator of AA-dependent inflammatory signaling in hepatocarcinogenesis and a direct pharmacological target of nuciferine (NF). Using diethylnitrosamine (DEN)/carbon tetrachloride (CCl4)-induced HCC mouse models with transcriptomic and lipidomic p

Baicalein Attenuates Early Molecular and Proliferative Changes in MASH-Driven Hepatocarcinogenesis by Inhibiting the NLRP3 Inflammasome and Pyroptosis Pathway.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) is a major risk factor for hepatocellular carcinoma (HCC). The nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome and pyroptosis are key drivers of inflammation and tumorigenesis. Baicalein (BAI), a flavonoid derived from Scutellaria baicalensis, has anti-inflammatory and antitumor properties. OBJECTIVES: This study aimed to evaluate the effects of BAI on early molecular and proliferative changes in a MASH-driven hepatocarcinogenesis model, using validated surrogate m

Synthesis and anti-liver fibrotic activity study of a chalcone derivative through anti-inflammatory effects and inhibition of JNK/NF-κB signaling pathways.

Liver fibrosis is a progressive disease caused by chronic inflammation and the activation of hepatic stellate cells (HSCs). This disease manifests as the abnormal proliferation and migration of HSCs, as well as the excessive deposition of the extracellular matrix. Chalcone analogues exhibit various biological activities, including anti-inflammatory, anti-proliferative, and apoptotic modulation properties, making them promising candidates for anti-fibrotic drug development. To enhance anti-fibrotic activities and decrease their side-effects, 31 novel chalcone derivatives were synthesized and ev

Integrative metabolomics and malonyl-proteomics reveal the crosstalk between metabolic reprogramming and lysine malonylation in liver fibrosis.

Lysine malonylation (Kmal), a recently identified post-translational modification (PTM) of proteins, has been linked to energy metabolism and implicated in various pathological disorders, including liver diseases. However, the specific roles of Kmal in hepatic fibrosis remain enigmatic. This study aims to investigate novel post-translational modifications and the underlying mechanisms involved in liver fibrosis. Utilizing untargeted metabolomics on carbon tetrachloride (CCl4)-induced mouse fibrotic liver tissues, we observed significant metabolic dysregulation during the liver fibrotic process

Targeting activated hepatic stellate cells through ferroptosis: phenethyl isothiocyanate attenuates CCl4-induced liver fibrosis by regulating iron metabolism, redox homeostasis, and autophagy.

Phenethyl isothiocyanate (PEITC), a dietary compound from cruciferous vegetables, is known for its chemopreventive activity, but its effects on liver fibrosis remain largely unexplored. In this study, we show that PEITC administration attenuated carbon tetrachloride (CCl4)-induced hepatic injury and fibrosis in mice, as evidenced by reduced collagen deposition, hydroxyproline content and fibrotic marker expression, and improved liver function. PEITC inhibited cell survival and fibrogenic phenotypes in transforming growth factor β (TGF-β)-stimulated LX-2 human hepatic stellate cells (HSCs) whil

Gypensapogenin I ameliorates liver fibrosis by targeting MST1, restoring Hippo pathway homeostasis, and inhibiting HSCs activation.

Gypensapogenin I (GI) is a new dammarane type triterpenoid saponin extracted from Gynostemma pentaphyllum. This study explored the therapeutic potential and mechanism of GI on liver fibrosis (LF). LF is a key pathological stage of chronic liver disease, and the current treatment is limited. In the mouse LF model induced by carbon tetrachloride (CCl4) and the LX-2 hepatic stellate cell model activated by transforming growth factor-β1 (TGF-β1), GI showed significant antifibrotic effects in vitro and in vivo, and could reduce liver injury, inflammation, collagen deposition and cell activation. Xiaoyao San restores NK cell anti-fibrotic activity through regulation of EZH2 in liver fibrosis.

BACKGROUND: Liver fibrosis poses a major global health issue, with few effective treatments currently available. Natural killer (NK) cells possess the capability to target activated hepatic stellate cells (HSCs) via their activating receptors, such as natural killer group 2 member D (NKG2D) and natural killer p46. Nonetheless, the fibrotic environment often limits their effectiveness in tackling fibrosis. Although the functions of Enhancer of zeste homolog 2 (EZH2) in the activation of HSCs and the regulation of NK cells are established, its influence on NK cell-driven anti-fibrotic activities

Zfp469 Deletion Attenuates Liver Fibrosis by Suppressing Extracellular Matrix Production in Hepatic Stellate Cells.

BACKGROUND: Liver fibrosis is driven by excessive extracellular matrix (ECM) deposition following chronic injury. Mutations in the human zinc finger protein 469 (ZNF469) cause Brittle Cornea Syndrome, a systemic connective tissue disorder, yet its functional role in hepatic fibrogenesis remains unexplored in vivo. This study investigated the impact of the murine ortholog, Zfp469, on liver fibrosis using CRISPR-generated knockout (KO) models and integrated transcriptomic and cellular analyses. Chlorogenic Acid Attenuates Liver Fibrosis in Mice via HIF-1α Inhibition and M2 Macrophage Polarization.

INTRODUCTION: Liver fibrosis, a key pathological step preceding cirrhosis, is characterized by the abnormal accumulation of extracellular matrix (ECM) components, a process largely driven by hepatic stellate cell activation and persistent chronic inflammation. Chlorogenic acid (CGA), a natural polyphenolic compound, has exhibited promising anti-fibrotic activity. However, the precise mechanisms underlying its effects are still not fully elucidated. This study investigates CGA's therapeutic effects and underlying pathways in carbon tetrachloride (CCl4)-induced liver fibrosis in mice. METHODS:

Molecular structure dependence of thermo-diffusion in binary mixture using thermal Lens spectroscopy.

Thermo-diffusion (Soret effect) in binary mixtures arises from the coupling of heat and mass fluxes in an inhomogeneous temperature field. In this work, we investigated how molecular structural isomerism influences thermo-diffusion in binary mixtures of butanol isomers (n-, s-, i-, and t-BuOH) with carbon tetrachloride (CCl4) using femtosecond laser-induced thermal lens spectroscopy (FTLS). The thermal lens (TL) response of the mixtures reflected the interplay between heat conduction and mass transport, which evolved systematically with composition. The magnitude and sign of the Soret coeffici
