Inflammatory bowel disease (IBD) is frequently accompanied by intestinal fibrosis, with nonresponse to long-term anti-TNF-α therapy occurring in approximately 23%-46% of patients. Integrated analysis of single-cell and bulk RNA-seq datasets revealed an expansion of IL11+ fibroblasts in inflamed intestine and their significant enrichment in nonresponders. We further identified IL11+ fibroblasts as a central communication hub that engaged in extensive crosstalk with monocytes and may contribute to inflammatory amplification and fibrotic remodeling. Additionally, we employed machine learning appr

Chrysin, a flavone nutraceutical, possesses several beneficial pharmacological properties, which has gained much emphasis in recent years. The biological effects of chrysin are exerted due to impeding or activating multifarious cellular and molecular pathways. Our findings indicated that chrysin inhibited tumor progression in various cancer cell lines by repressing the formation of a sphere and upregulated protein expression of Src homology region 2 domain-containing phosphatase-1 (SHP-1), alleviating phosphorylated-signal transducer and activator of transcription 3 (p-STAT3) and transaction w

The activation of hepatic stellate cells (HSCs), characterized by transdifferentiation from a quiescent state to a fibrogenic phenotype, is a core process of liver fibrosis. The metabolic reprogramming of HSCs plays a major role in this process to meet the high energy demands of myofibroblastic HSCs with multiple functions, such as extracellular matrix synthesis, migration, and proliferation. AMP-activated protein kinase (AMPK) is a gatekeeper of intracellular energy homeostasis, but its role in the activation of HSCs and the progression of liver fibrosis remains unclear. Here, we found that t

INTRODUCTION: Salvianolic acid B (SAB), as one of the major water-soluble compounds of Salvia miltiorrhiza, has proved to effectively reduce elevated portal pressure in cirrhotic rats. However, the short halflife and in vivo retention time of SAB affect its pharmacodynamics. Therefore, in this study, we prepared albumin nanoparticles loaded with SAB (SAB-ALB-NPs) to improve the in vivo retention time of the drug and enhance bioavailability.
METHODS: We prepared and characterized SAB-ALB-NPs, including particle size, polydispersity index (PDI), zeta potential, stability, EE, in vitro release,

INTRODUCTION: Hepatocellular carcinoma (HCC), the predominant form of primary liver malignancy, remains a major global health concern owing to its aggressive progression, limited therapeutic efficacy, and high fatality rate. A significant proportion of HCC arises from chronic exposure to chemical carcinogens, which trigger hepatocarcinogenesis through oxidative stress, DNA damage, and dysregulation of signalling networks. Natural compounds, particularly beta-carboline alkaloids, are emerging as safer, multi-targeted candidates with promising hepatoprotective and anticancer potential. This revi

It has been a consensus that hepatic microenvironment composed by the non-parenchymal cells networks play a critical role during liver fibrogenesis, with the crosstalk between hepatic stellate cells (HSCs) and macrophages being of paramount importance. Interleukin 11(IL-11) has been implicated as a pro-fibrogenic cytokine, where its function in/between hepatocytes and HSCs has been revealed. But whether IL-11 participates in the interaction of HSCs and macrophages and related mechanism remains obscure. Our research demonstrates that HSC-derived IL-11 operates through a dual mechanism of autocr

BACKGROUND & AIMS: Liver fibrosis significantly burdens global health, and increased protein synthesis during hepatic stellate cell (HSC) activation plays a crucial role in its progression. Ribosome is the site of protein synthesis. RNA polymerase I (Pol I) is a protein that regulates the transcription of ribosomal DNA (rDNA) genes into ribosomal RNA (rRNA) in ribosomal biogenesis. Therefore, we investigated the role and mechanism of Pol I-regulated ribosome biogenesis in HSCs activation and liver fibrosis progression.
METHODS: Initially, we assessed the expression levels of Pol I in the seru

Rationale: Nuclear factor erythroid 2-like 1 (Nrf1/NFE2L1) is a crucial redox-sensitive factor essential for mitochondrial homeostasis. However, its function in controlling macrophage-associated liver inflammation and fibrosis remains to be fully understood. Herein, this study was conducted to elucidate the roles of macrophage Nrf1 in regulating liver fibrosis. Methods: Expression levels were analyzed in human liver tissues collected from individuals diagnosed with or without liver fibrosis. High-fat diet feeding, carbon tetrachloride injection or bile duct ligation was performed respectively

Chronic liver injury represents a core pathological substrate in a spectrum of hepatic disorders, wherein gut-liver axis homeostasis critically drives progression. Although bovine milk extracellular vesicles (mEVs) positively regulate intestinal homeostasis, the mechanisms underlying their gut microbiota-linked hepatoprotection remain unclear. Herein, we demonstrated that mEVs (0.6 mg/kg/d) effectively alleviated carbon tetrachloride (CCl4, 1 mg/kg)-induced liver injury, as evidenced by reduced hepatic inflammation and fibrosis. Concurrently, mEVs also markedly attenuated colonic barrier disru

We report a catalyst-free, regioselective [3 + 2] cycloaddition reaction between benzofuran-derived azadienes (BDAs) and N,N'-cyclic azomethine imines, where regioselectivity is controlled by modulating solvent polarity. Spiro[benzofuran-2,2'-pyrazolo[1,2-a]pyrazole] is obtained in high yields (up to 93%) using acetonitrile as the solvent, while the novel spiro[benzofuran-2,1'-pyrazolo[1,2-a]pyrazole] forms in moderate to good yields (42%-73%) with carbon tetrachloride. This solvent-controlled protocol exhibits excellent diastereoselectivity (dr >20:1) for both products, along with a broad sub

Correlated vibrational spectroscopy (CVS) is a hyper-Raman-based vibrational spectroscopy that retrieves separate spectra of individual (self-correlated, SC) and interacting (cross-correlated, CC) molecules. The spectra are recorded in the >40 cm-1 THz/mid-IR frequency range and contain modes that are IR and/or Raman active. Here, we further develop CVS and apply it to investigate intra- and intermolecular interactions using room temperature liquid carbon tetrachloride (CCl4), a nonpolar liquid, and acetonitrile (CH3CN), a polar liquid, as case studies. CVS spectra of CCl4 display no intermole

BACKGROUND: Liver fibrosis poses a significant global health challenge with limited treatments. Baoganning (BGN) decoction, a traditional Chinese medicine formula administered for liver fibrosis, has shown promise in preliminary studies by targeting indoleamine 2,3-dioxygenase 1 (IDO1), a key immunoregulator in the liver microenvironment. However, the precise underlying mechanisms remain unclear.
PURPOSE: This study aimed to elucidate the mechanism by which BGN alleviates liver fibrosis via IDO1-mediated regulation of macrophage function.
METHODS: The anti-fibrotic effects of BGN were evalua
