Punicalagin Alleviates Acute Liver Injury via Dual STAT1/NF-κB Inhibition and STAT3 Activation to Orchestrate M1-to-M2 Macrophage Polarization.

Acute liver injury (ALI) is characterized by excessive inflammation and macrophage polarization, with the M1-to-M2 phenotypic shift emerging as a critical regulatory node. Punicalagin (PUN), a polyphenolic compound derived from pomegranate, has demonstrated hepatoprotective effects in preclinical models; however, its molecular mechanisms underlying macrophage polarization and inflammatory signaling remain unclear. This study aims to investigate the role of PUN in modulating macrophage polarization and inflammatory pathways in chemical- and drug-induced ALI. CCL4- and APAP-induced ALI mouse mod

Silk fibroin scaffolds loaded with growth factors can reverse liver fibrosis.

Currently, there are no effective pharmacological interventions in clinical practice to reverse liver fibrosis. This study explores the therapeutic potential of regenerated silk fibroin (RSF) scaffolds loaded with dual growth factors for reversing hepatic fibrosis. A chronic liver fibrosis mouse model was induced using carbon tetrachloride (CCl4) combined with a high-fat diet. RSF scaffolds, with or without hepatocyte growth factor (HGF) and fibroblast growth factor-4 (FGF-4), were implanted onto the liver surface to assess antifibrotic efficacy. Liver function was evaluated using biochemical

Goosecoid promotes hepatic stellate cell epithelial-mesenchymal transition via transcriptional activation of serum- and glucocorticoid-induced protein kinase 1 in liver fibrosis.

BACKGROUND: Goosecoid (GSC) is a transcription factor implicated in epithelial-mesenchymal transition (EMT) during embryogenesis and cancer, but its function in organ fibrosis remains poorly understood. This study investigates the role and mechanistic targets of GSC in hepatic stellate cell (HSC) EMT during liver fibrogenesis. METHODS: We analyzed GSC and EMT marker expression in primary HSCs isolated from murine liver fibrosis models induced by carbon tetrachloride (CCl₄) or bile duct ligation (BDL), and in TGF-β-stimulated LX-2 cells. The functional role of GSC was validated via in vitro an

ATP6V0d2 alleviates acute liver injury via promoting hepatocyte autophagy.

Liver injury is the pathological basis of various liver diseases, and severe acute liver injury can progress to acute liver failure with a high mortality. Although continuous progress has been made over the past few decades, the molecular mechanism underlying the progress of acute liver injury is still unclear. ATP6V0d2, a subunit of the vacuolar ATPase (V-ATPase) complex, is implicated in organelle acidification and intracellular degradation processes. In this study, three liver injury datasets were analyzed to screen the differentially expressed genes (DEGs) and identify Atp6v0d2, which was

Curcumol Induces Necroptosis of Hepatic Stellate Cells by Targeting KAT8 to Suppress HK2 Lactylation and Promote HUWE1-Dependent Ubiquitination.

Liver fibrosis is a pathological outcome of chronic liver injury and is primarily driven by the continuous activation of hepatic stellate cells (HSCs). During activation, HSCs depend on aerobic glycolysis to maintain their fibrogenic characteristics, suggesting that metabolic reprogramming could serve as an effective therapeutic approach. In this study, we demonstrate that Curcumol, a natural compound derived from plants in the Zingiberaceae family, selectively removes activated HSCs by interfering with the lactate-KAT8-HK2 regulatory pathway. In HSCs, lactate produced through glycolysis activ

Klotho-derived peptide 1 ameliorates hepatic fibrosis induced by αKlotho deficiency and liver injury.

Hepatic fibrosis, driven primarily by hepatic stellate cells (HSCs) activation induced by TGF-β, currently lacks effective therapies. In this study, we demonstrated that deficiency of αKlotho, an extrahepatic antiaging protein, due to genetic ablation in kl/kl model or aging caused spontaneous hepatic fibrosis, as evidenced by an increased collagens deposition and TGF-β signaling hyperactivation. KP1, a small peptide derived from human αKlotho protein, recapitulated its anti-fibrotic potential and blocked HSCs activation induced by TGF-β1. Mechanistically, KP1 acted as a competitive TGF-β rece

[Elucidation of a Novel Protective Function of AADAC Against Drug-induced Liver Injury through Ferroptosis Suppression].

Drug-induced liver injury (DILI) remains a major cause of drug withdrawal from the market, and safety concerns in clinical settings. Reactive metabolites, primarily produced by cytochrome P450 enzymes, are known to induce oxidative stress and hepatotoxicity through covalent binding to macromolecules and the generation of reactive oxygen species. Recent evidence suggests that ferroptosis, an iron-dependent form of lipid peroxidation-mediated cell death, is involved in DILI triggered by compounds such as carbon tetrachloride (CCl4) and acetaminophen (APAP). Arylacetamide deacetylase (AADAC), a h

TGFBI promotes liver fibrosis through remodeling the profibrotic microenvironment by a positive feedback regulatory loop.

Liver fibrosis is a major global health burden with limited treatment options. Transforming growth factor-beta-induced protein (TGFBI) is crucial in fibrotic diseases and tumors, however, its precise mechanism in liver fibrosis remains unclear. Here we show that TGFBI promotes liver fibrosis in male C57BL/6 mice. TGFBI is upregulated in fibrotic livers and derived from non-parenchymal cells. Genetic TGFBI deficiency alleviates liver fibrosis in both CCl4 (carbon tetrachloride) injection and bile duct ligation (BDL) models. Mechanistically, PDGFRβ is identified via RNA sequencing as a key downs

Development of hybrid nanoparticles based on Zr(iv) and perylene-3,4,9,10-tetracarboxylic acid with visible-light photoredox activity.

Herein, we investigate perylene-3,4,9,10-tetracarboxylic dianhydride (PTCDA) as a linker in Zr-clusters. The photostable, 3D metal-organic nanomaterial obtained by a solvothermal synthesis procedure in the presence of formic acid as modulator, named ZIPER, shows strong absorption in the visible (400-560 nm) and an intense photoluminescence (PL) in the 600-700 nm range. PL quenching experiments strongly indicate that the ZIPER excited state (ZIPER*) behaves primarily as a strong oxidant and a mild reductant with redox couples E(ZIPER*/ZIPER˙-) = 1.8-1.2 V and E(ZIPER˙+/ZIPER*) = -0.44--0.48 V (

Compressed MgCl2 Reveals Multiple Pathways to Cotunnite Structures.

The phase transition sequences of MeX2 compounds (Me = metal or, more generally, an electropositive element), whose constituent atoms contribute 16 valence electrons per formula unit under high pressure, are of fundamental importance in materials science, high-pressure chemistry, and mineral physics. Here, we report the first observation of trigonal prismatic coordination in this class of materials, realized in magnesium dichloride MgCl2. We synthesized anhydrous MgCl2 by the direct reaction of elemental magnesium with carbon tetrachloride (CCl4) in laser-heated diamond anvil cells from 7(2) t

Hepatic stellate cell derived lipid droplets drive protumoral M2 macrophage polarization in hepatocellular carcinoma.

BACKGROUND: During chronic liver injury, hepatic stellate cells (HSCs) lose their vitamin-A-rich lipid droplets (LDs), yet whether these organelles are merely degraded or released via vesicles and functionally relevant remains unclear. We investigated the fate of HSCs LDs and their impact on hepatic macrophage phenotype and hepatocellular carcinoma (HCC) development. METHODS: Chronic liver injury was induced in C57BL/6 mice using carbon tetrachloride (CCl4) for up to 12 weeks. HSCs activation and lipid droplet dynamics were assessed by immunofluorescence, transmission electron microscopy, and

miR-10a Delivered via MSC-derived Extracellular Vesicles Modulates Inflammation in a CCl4-induced Liver Fibrosis Model.

Liver fibrosis is a significant global health issue characterized by an abnormal accumulation of extracellular matrix proteins, that disrupts normal liver architecture and function. Mesenchymal stem cells (MSCs) show therapeutic potential by releasing extracellular vesicles (EVs) containing regulatory microRNAs like miR-10a. This study evaluates miR-10a-enriched human umbilical cord MSC (hUCMSC)-EVs in a CCl4-induced model of liver fibrosis, focusing on inflammatory marker modulation. Liver fibrosis was induced in experimental animals using CCl4 administration. MSCs were isolated from the huma
