Integrated stress response promotes acute liver failure by activating SETD7 and enhancing NLRP3 methylation.

While suppression of the integrated stress response (ISR) has been shown to restore proteostasis and mitigate organ injury in various diseases, its role in acute liver failure (ALF) remains poorly defined. Here, we discovered that, during drug-induced ALF, hepatocytes exhibited early and transient activation of the eIF2α-ATF4 signaling pathway, whereas macrophages displayed delayed but sustained activation. Hepatocyte-specific deletion of ATF4 (ATF4ΔHep) significantly protected mice from acetaminophen (APAP)-induced liver injury, whereas myeloid-specific ATF4 deletion (ATF4ΔMye) increased susc

Nanoceria drive stem cell hepatic differentiation and microenvironment remodeling for acute liver failure therapy.

Stem cell-derived hepatocyte therapy holds great potential for treating acute liver failure (ALF) by rapidly restoring liver function and stimulating regeneration. However, significant challenges remain in efficiently driving stem cells toward a hepatic lineage and ensuring transplanted cell survival and functionality in the hostile, injured liver microenvironment. To overcome these hurdles, we developed a novel ceria nanoparticle@microRNA complex that facilitates hepatic differentiation of mesenchymal stem cells. We show that ceria nanozymes synergize with microRNA-122 to enhance hepatic diff

Co-administered internalizing RGD peptide boosts anti-PD-L1 therapy in hepatocellular carcinoma.

BACKGROUND & AIMS: Immune checkpoint inhibitor (ICI) therapy has significantly improved the treatment of solid tumors such as hepatocellular carcinoma (HCC); however, most patients fail to respond. Here, we examined whether co-administration of the tumor-penetrating internalizing (i)RGD peptide, which selectively increases tumor vascular permeability in a neuropilin-1-dependent manner, enhances intratumoral delivery and therapeutic efficacy of αPD-L1 in mouse models of HCC. METHODS: αPD-L1, with or without iRGD, was administered intravenously to mice bearing endogenous HCCs (TGFα/c-myc and di

Subacute and chronic CCl4 exposure induces time-dependent neuropsychiatric disorders via MAOA-mediated serotonin dysregulation: A liver-brain axis mechanism.

Carbon tetrachloride (CCl4), a potent hepatotoxin, is known to induce liver injury, but its effects on neuropsychiatric behavior through the liver-brain axis remain poorly understood. This study investigates how subacute versus chronic CCl4 exposure drives distinct behavioral phenotypes via monoamine oxidase A (MAOA)-mediated serotonin dysregulation. Male C57BL/6 mice were subjected to subacute (23 days) or chronic (49 days) CCl4 exposure (n = 10/group). Liver injury was assessed through serum ALT/AST, histopathology, and fibrotic markers. Neurobehavioral changes were evaluated using open fiel

Preclinical approaches to studying liver and kidney fibrosis: models and methodologies.

Organ fibrosis, notably affecting the liver and kidneys, remains a major contributor to global morbidity. This review examines the pathophysiology, molecular mechanisms, and preclinical models used to study hepatic and renal fibrosis. In liver fibrosis, hepatic stellate cell activation, chronic inflammation, and extracellular matrix accumulation are central features, while renal fibrosis involves myofibroblast activation and redox-mediated signaling pathways. The present review highlights both in vitro and in vivo models such as the carbon tetrachloride, bile duct ligation, and dimethylnitrosa

Ultrafast carbon tetrachloride dechlorination by assembling electron and hydrogen bridges over sulfidated zero-valent iron interfaces.

Sulfidated zero-valent iron (S-ZVI) offers an attractive and sustainable approach for dechlorinating persistent chlorinated organics. However, it remains challenging to efficiently dechlorinate chlorinated organics with highly-symmetrical structure and extreme hydrophobicity, due to the limited contact interface and the insufficient electron/proton transfer efficiency of S-ZVI. In this study, we report a surfactant-mediated interfacial engineering strategy that achieves unprecedented carbon tetrachloride (CT) dechlorination kinetics (8.36 h-1) over S-ZVI, representing the highest reported rate

Physcion-8-O-β-D-monoglucoside protects hepatocytes from TNF-α-mediated apoptosis by suppressing the PI3K/AKT/NF-κB signaling pathway.

Physcion-8-O-β-D-monoglucoside (PMG) is one of the active ingredients of Radix et Rhizoma Rhei, which has been used for treating liver diseases for hundreds of years in China. However, the hepatoprotective effects of PMG remain poorly understood. This study aimed to investigate the mechanism of the protection effects of PMG on tumor necrosis factor-α (TNF-α)-induced hepatotoxicity. We developed both in vitro and in vivo models of liver injury to assess the protective effects of PMG against TNF-α-induced hepatotoxicity. The in vitro model employed TNF-α/actinomycin D in AML-12 cells, while the

Acetylshikonin mitigates diet-induced MASLD by targeting PPARγ-mediated metabolic dysfunction.

INTRODUCTION: The liver, as the central metabolic hub of the body, is highly susceptible to diet-induced injury. The increasing prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD) highlights the urgent need for effective clinical interventions. Currently, there are no specific therapeutics for MASLD, and dietary patterns are closely associated with its pathogenesis, making the exploration of natural bioactive compounds a promising strategy. METHODS: In this study, we identified acetylshikonin (AS), a component derived from traditional Chinese medicine (TCM), as a co

ITPR3 promotes liver fibrosis by damaging hepatocytes via the Ca2+/NF-B/LECT2 pathway.

Hepatocyte damage is the initial factor in liver injury. De novo expression of inositol 1,4,5-triphosphate receptor type 3 (ITPR3) typically occurs in hepatocellular carcinoma and may contribute to its pathogenesis. However, the role of de novo expression of ITPR3 by hepatocytes in the pathological processes of liver fibrosis remains unclear. Expression analyses of ITPR3 in hepatocytes were combined with an evaluation Ca2+/nuclear factor-κB (NF-κB)/leukocyte cell-derived chemotaxin 2 (LECT2) pathway and characterization of the effect of ITPR3 expression on hepatocytes damage. The effect of ITP

Branched-Chain Amino Acids Alleviate Carbon Tetrachloride-Induced Kidney Fibrosis and Splenic Hemorrhage in Rat Models.

Branched-chain amino acids (BCAAs) are recognized for their roles in protein synthesis, mitigation of oxidative stress, and maintenance of cellular health in models of carbon tetrachloride (CCl4) toxicity, which is associated with industrial chlorofluorocarbon emissions. Male Wistar rats were randomly assigned to four groups: control, CCl4, CCl4 plus 10 g BCAAs, and CCl4 plus 20-g BCAAs. Each rat received 1 mL/kg of CCl4. After 10 weeks, kidney function biomarkers, including glutathione peroxidase (GPX) and malondialdehyde (MDA), were assessed in kidney homogenates. Histomorphological changes

MicroRNA-200a as a therapeutic agent for acute and chronic liver pathologies and regeneration.

Effective therapies targeting the molecular pathogenesis of liver diseases remain an unmet clinical need. Here, using acute and chronic liver pathology models, we demonstrate a dual therapeutic role for miR-200a encapsulated in poly(lactic acid) (PLA) nanoparticles (NPs) in both treating liver pathology and stimulating regeneration. This rationale is based on miR-200a's central role in regulating oxidative and endoplasmic reticulum (ER) stress, key drivers of liver diseases, enhancing liver regeneration and preventing disease development. Herein, we report the design and therapeutic validation

Identification and clustering analysis of drug-responsive temporally varying genes through high-frequency longitudinal RNA sequencing.

High-frequency longitudinal RNA sequencing has emerged as a powerful approach for capturing dynamic transcriptional responses to therapeutic interventions, yet traditional differential expression analysis fails to identify genes with temporal variability that lack static expression differences. We used our previously developed computational framework for identifying Temporally Varying Genes (TVGs) from daily blood samples collected over 10-21 days in Sprague-Dawley rats treated with hepatotoxic compounds including tetracycline, isoniazid, carbon tetrachloride, and valproate. Our methodology em

Acute exercise activates hepatic Nrf2 signaling by ROS, AMPK and epinephrine to protect against acute liver injuries.

While exercise is well-established as a protective strategy against non-alcoholic fatty liver disease, its role in acute liver injury (ALI) remains poorly understood. Nuclear factor erythroid 2-related factor 2 (Nrf2), a master regulator of antioxidant response element (ARE)-dependent gene expression, plays an important role in the pathogenesis of liver diseases, where oxidative stress contributes significantly to both chronic progression and acute injury. This study investigated whether exercise confers protection against ALI via the Nrf2 signaling pathway and elucidated the underlying molecu
