Tandem RNA and Protein Extraction: A Platform for Maximizing the Use of Limited Ex Vivo Tissue Samples.

Human tissue samples represent the gold standard for obtaining clinically relevant and translatable insight into disease processes that in vitro systems cannot fully reproduce. However, patient-derived samples are often limited in size and availability, limiting the number of downstream assays that can be performed. To maximize the use of invaluable human samples, we present a protocol for the tandem extraction of high-quality RNA and protein from the same tissue section. This method has been optimized for 15-30 mg tissue sections, enabling more experimental conditions and technical replicates

Symmetry-breaking charge separation in perylene diimide trimers: effects of aggregation and solvent polarity.

Symmetry-breaking charge separation (SB-CS) is a fundamental process in natural photosynthetic systems, serving as the primary trigger for electron transfer and ultimately leading to the formation of a charge separated state. This mechanism has also been leveraged in optoelectronic devices to minimize energy loss and improve solar energy conversion efficiency. However, in organic solar cell (OSC) studies, SB-CS has predominantly been observed in polar environments, and the roles of molecular aggregation in modulating this process remain unclear. Herein, we investigated the influence of aggrega

Developing an E. coli heterologous expression system for characterizing a marine debrominase from Roseobacter sp.

Reductive dehalogenases (RDases) are a family of enzymes that employ a cobamide cofactor and two iron-sulfur clusters to selectively cleave carbon-halogen bonds. The identification of yet-uncharacterized RDases is crucial for advancing our understanding of dehalogenation processes in both natural and engineered environments, particularly for bioremediation. In this study, we developed a heterologous expression system in Escherichia coli DH5α to produce a chloroform-reducing RDase (TmrA) from Dehalobacter sp. UNSWDHB, using a pTrcHisA vector without requiring an additional vector for vitamin B1

Solvent-Substrate Interaction-Controlled Single-Step Inkjet Printing of Micro-Inlaid IOLEDs.

Inkjet printing offers a scalable, material-efficient route for the patterning of optoelectronic devices, but the conventional pixel definition depends on the bank structures and multistep alignment, increasing the process complexity. Here, a single-step, interphase-controlled inkjet printing strategy is reported for the direct fabrication of micro-inlaid inverted organic light-emitting diodes (μ-inlaid IOLED) pixels on zinc oxide (ZnO) electron-transport layers. Chloroform-based inks containing a blend of small-molecule semiconductor solutes are deposited onto poly(4-vinylpyridine) (P4VP) ins

Complex-Valued Chemometrics for Analyzing Absorbance or Raman Spectra.

Complex-valued chemometrics offers a promising extension of classical regression methods by exploiting both real and imaginary spectral components. Here, we show that conventional absorbance (χ(1)) and Raman (χ(3)) spectra can be transformed into complex-valued forms by combining the measured intensities as imaginary parts with their Kramers-Kronig-derived real parts. We benchmark four regression methods─classical least squares (CLS), inverse least squares (ILS), principal component regression (PCR), and partial least-squares regression (PLSR)─across four representative systems: the quasi-idea
