Perturbation of Metabolic Pathways Associated with Exposure of Human Renal Proximal Tubular Cells to Nephrotoxic Pollutants and Drugs.

Mammalian kidneys, although exposed to a diverse array of environmental pollutants and commonly used drugs that are dose-limited by nephrotoxicity, have a significant but limited capacity for repair and regeneration. Primary cultures of human proximal tubular (hPT) cells were used to investigate the cellular response to exposures to one of two environmental toxicants (mercuric chloride [HgCl2] or S-(1,2-dichlorovinyl)-L-cysteine [DCVC; key metabolite of trichloroethylene] or one of three nephrotoxic drugs (cisplatin [CDDP], polymyxin B [PmxB], tenofovir disoproxil fumarate [TDF]) at concentrat

Trichloroethylene and perchloroethylene metabolites have fetal sex-specific RNA expression impacts in a full term placental villous explant model.

Exposure to volatile organic chemicals (VOCs) like trichloroethylene (TCE) and perchloroethylene (PCE) has been associated with adverse pregnancy outcomes. We conducted an analysis of gene expression responses to the TCE metabolite S-(1,2-dichlorovinyl)-L-cysteine (DCVC) and the PCE metabolite S-(1,2,2-trichlorovinyl)-L-cysteine (TCVC) in placental explants. Explants from placentas with male and female fetuses were treated with either metabolite at 10μM or 20μM (n=4). We measured gene expression using RNA sequencing, conducted differential gene expression analysis, cell type deconvolution and

Trichloroethylene-Associated Hypersensitivity: Clinical Patterns Supporting an Idiosyncratic Solvent-Induced Liver Injury Framework.

Occupational exposure to trichloroethylene (TCE) has been associated with rare but severe systemic hypersensitivity reactions frequently accompanied by hepatic dysfunction, yet these reactions are often discussed within exposure-centered toxicological paradigms. This review synthesizes clinical and epidemiologic evidence on occupational trichloroethylene-associated hypersensitivity to clarify its characteristic phenotype and interpret the condition within a framework of idiosyncratic solvent-induced liver injury (ISILI). Across published reports, a highly consistent phenotype was observed, cha

An Optimized Fibrous Network by Collaborative Slowing Aggregation Kinetics and Enhancing Intermolecular Interactions via a Co-Solvent Combined With Solid Additive Strategy Enables 20% Efficient Organic Solar Cells.

A well-defined fibrous network is important for efficient exciton dissociation and charge transport in organic solar cells (OSCs). However, forming such fibrous morphology is difficult to control in polymer:non-fullerene acceptor (NFA) systems, mainly because of the relatively weak intermolecular interactions between components and fast film formation dynamics. Herein, we propose a co-solvent combined with solid additive strategy to synchronously regulate film formation kinetics and tune the intermolecular interaction in D18:L8-BO systems. The high-boiling-point co-solvent, trichloroethylene (

A simple approach to identifying community and healthcare locations for hand hygiene interventions during public health emergencies - Uganda, 2020-2022.

BACKGROUND: As part of Uganda's COVID-19 emergency response, two districts along high-volume trucker routes linked to early clusters, Amuru and Tororo, were prioritized for hand hygiene (HH) interventions. While HH is a critical non-pharmaceutical intervention, tools to efficiently identify high-risk community locations during outbreaks are limited. METHODS: We adapted the Population Connectivity Across Borders (PopCAB) tool, developed by the U.S. Centers for Disease Control and Prevention, to identify and prioritize high-risk community locations and healthcare facilities (HCF) and inform tar

Dual role of phenol Co-contamination in trichloroethylene detoxification in EK-BIO systems: Microbial inhibition versus stimulated aerobic cometabolism.

Phenol often co-occurs with trichloroethylene (TCE) and can inhibit the activity of organohalide-respiring bacteria, posing challenges for in-situ bioremediation. This study evaluated one-dimensional columns operated under electrokinetic-enhanced bioremediation (EK-BIO) for simultaneous TCE detoxification with phenol co-amendment at 1 mM (replenished every 14 days). EK-BIO achieved effective TCE removal despite 6 injection cycles (2100 μmol total), with pore water concentrations decreasing from 500 μM to 79.03 μM at day 28, and 5.86 μM by day 84. Phenol-free batch experiments exhibited earlier

Use of upcycled biosolids for bioremediation of groundwater contaminated with chlorinated solvents.

Trichloroethene (TCE) is a widespread toxic groundwater contaminant; thus, the development of inexpensive and accessible remediation materials is needed. Bioremediation of TCE under field conditions often requires the addition of carbon amendments. This study examines the use of biosolids at a field site as a sustainable carbon-rich and microbial amendment. A trench with amendments was installed upstream of a TCE-contaminated site, which was filled with biosolids, limestone, and biochar. Field monitoring of water quality parameters was compared with microbial community changes in soil samples

Application of the solvent effect on bioluminescent reporter bacteria as a real-time membrane toxicity assay.

Bioluminescent bioreporters are widely used across various scientific disciplines due to the well-characterized bacterial bioluminescence mechanism. However, solvent-induced membrane perturbations may confound the use of bioreporters in assessing cellular toxicity from environmental contaminants. This study investigated the solvent effect, wherein membrane damage increases intracellular availability of bioluminescent reaction precursors, increasing the light produced. A new online in situ monitoring system was also tested with multiple bioluminescent reporters, including a newly constructed Ps

Sulfidation Doses of Nanoscale Zerovalent Iron Particles Need to Be Tuned to Achieve High Reactivity to Different Chlorinated Solvent Compounds.

The sulfur content in sulfidated nanoscale iron (S-nZVI) alters the degradation efficiency of trichloroethene (TCE), a common groundwater contaminant, but its impact on other chlorinated hydrocarbon contaminants (CHCs) has not been characterized. In this study, the anaerobic degradation of carbon tetrachloride (CT), chloroform (CF), trichloroethane (1,1,1-TCA) and TCE was assessed using S-nZVI at seven S loadings ([S/Fe] = 0.01-0.303). S-nZVI0.01 yielded the highest degradation rate constants for CT, 1,1,1-TCA, and CF, which were 1.77-10.9 times higher than those of nZVI and other S-nZVI. In c

Integrated behavioral, transcriptomic and metabolomic analysis reveals the neurotoxicity of trichloroethylene in adult zebrafish (Danio rerio).

Trichloroethylene (TCE), a persistent environmental pollutant, poses potential health risks through multiple exposure routes, such as inhalation of vapor-phase TCE or ingestion of contaminated water and food. While recognized as a carcinogen, its neurobehavioral effects remain poorly characterized. This study investigated the mechanisms of TCE neurotoxicity in adult zebrafish following chronic exposure to environmentally relevant concentrations (20, 60, and 180 μg/L). Behavioral assessments revealed concentration-dependent anxiety-like behavior, reduced exploration, and memory impairment. Tran

Immune effects of trichloroethylene: key findings of occupational hypersensitivity syndrome and its pathogenesis.

Trichloroethylene (TCE) remains a versatile organic solvent used globally. In recent years, immunological occupational diseases such as hypersensitivity syndrome (HS) and systemic sclerosis (SSc) have attracted attention in terms of the number of patients and the life-threatening consequences. This review summarizes mechanisms underlying HS pathogenesis based on the available literature. TCE-HS is a disease attributable to anti-CYP2E1 autoantibodies and HLA-B13:01 as susceptibility gene and is characterized by systemic skin lesions, severe-to-moderate liver damage, fever above 38 °C, leukocyto

EBV-driven rewiring of Drp1-Mediated mitophagy exacerbates trichloroethylene-induced hypersensitivity syndrome.

The environmental pollutant trichloroethylene (TCE) can induce T-cell-mediated hypersensitivity syndrome. Clinical evidence closely links Epstein-Barr virus (EBV) reactivation to aggravated disease severity, yet the mechanism underlying this chemical-viral synergy remains unknown. Based on the view that hypersensitivity essentially involves sustained activation of effector T-cells and disruption of immune homeostasis, this study aims to investigate how EBV reactivation cooperates with TCE metabolite trichloroethanol (TCOH) to regulate the T-cell apoptosis process by affecting mitochondrial dyn

Pore-scale optimization of H2O2-based TCE remediation using stabilizer-enhanced oxidant delivery and follow-up water injection.

Hydrogen peroxide (H2O2)-based in situ chemical oxidation (ISCO) is a commonly applied technique for remediating groundwater contaminated by dense non-aqueous phase liquids (DNAPL) such as trichloroethylene (TCE). However, the overall remediation efficiency is often constrained by oxidant self-decomposition and gas-induced pore blockage, which hinder oxidant accessibility to contaminants. To improve the remediation performance, this study experimentally investigated three optimization strategies: follow-up water injection, chemical stabilization, and staged oxidant injection. These three strat

Analyte Recovery of Volatile Organic Compounds: A Passive Sampling Analysis via Photothermal Desorption Compatible Diffusive Samplers.

Volatile organic compounds (VOCs) are common sources of occupational exposure throughout a variety of industries. To protect personnel from overexposure, field industrial hygienists must conduct compliance sampling. In efforts to improve upon analytical sensitivity and time-to-knowledge of existing VOC exposure assessment methods, the industrial hygiene research group at UAB has developed a preanalytical technique known as photothermal desorption (PTD), which uses pulses of high-energy light to desorb analytes from thermally conductive, carbonaceous sorbents. To-date, the theoretical and conce

Trichloroethylene induces cardiomyocyte senescence through an AhR-ROS-IL-1 axis and amplified by Wnt/β-catenin suppression.

Trichloroethylene (TCE), a pervasive groundwater contaminant, has been epidemiologically linked to cardiovascular diseases, yet its potential to initial cardiac aging remains uncharacterized. Using H9c2 cardiomyocytes and zebrafish larvae, we dissected the molecular mechanisms underlying TCE-induced cardiac senescence. Exposure to an environmentally relevant dose of TCE (60 μg/L) triggered a robust premature aging program, characterized by flattened morphology, elevated senescence-associated SA-β-gal activity, up-regulation of p16/p21 expression, and reduced Lamin B1 levels, without compromisi

Role of Histone Modifications and miRNA Regulations in the Toxicity of Trichloroethylene in Terms of Cell Cycle Regulation in Human Kidney Cells.

Trichloroethylene (TCE) is an organic solvent with various industrial applications. Therefore, the study aimed to determine the effects of TCE on cytotoxicity, apoptosis, reactive oxygen species (ROS) production, global histone modifications, and regulation in microRNAs (miRNAs) related to cancer pathways in human embryonic kidney cells (HEK-293) after exposure to TCE at concentrations of 0.03, 0.1, and 0.3 mM for 24 h. Results indicated that TCE caused dose- dependent cytotoxicity without induction of significant apoptosis or ROS production at concentrations of ≤ 0.3 mM. Upregulation in myc,

Synergistic reductive dechlorination and oxidative mineralization of chlorinated organic compounds in a UV-anaerobic Fenton system: UV-driven Fe(II)/Fe(III) cycling and continuous radical generation.

The effective remediation of chlorinated organic compounds (COCs) in industrial wastewater is hindered by the high C-Cl bond dissociation energy and toxic chlorinated intermediates formation in traditional advanced oxidation processes (AOPs). This study developed a UV-anaerobic Fenton system that synergizes oxidative and reductive pathways for efficient degradation and dechlorination of p-chloroaniline (p-CA). The system achieved excellent performance with 93.5 % TOC removal and 89.8 % dechlorination within 90 min. Mechanistic analysis revealed that UV light drove Fe(III) reduction through bot

Orientation Enhancement in Donor-Acceptor π-Conjugated Polymer Thin Films by a Cooled High-Speed Bar-Coating Method.

π-Conjugated polymers are promising materials for next-generation flexible and lightweight electronics owing to their solution processability and excellent charge transport properties, where molecular orientation is critical for efficient charge transport. However, maintaining high molecular orientation at high coating speeds required for industrial production remains challenging, particularly when using volatile, low-boiling-point solvents. In this study, we employed a cooled, high-speed bar-coating method at 50 mm s-1 to fabricate highly oriented films of poly{2,2'-[(2,5-bis(2-octyldodecyl)-

Solar-Driven Photocatalytic Trichloroethylene Mineralization with High CO2 Selectivity.

Sunlight-driven photocatalytic oxidation, using ambient air as the oxidant, offers a sustainable route for removing chlorinated volatile organic compounds (VOCs) such as trichloroethylene (TCE). Yet, practical implementation is hindered by low reaction rates and undesirable product selectivity. Here, we present a material design strategy that overcomes both challenges. Size-controlled anatase TiO2 nanocrystals with truncated bipyramid (nanoTBP) morphology achieve exceptional photocatalytic activity for TCE degradation, comparable to rates reported for energy-intensive thermal catalysis, with u
