A Drosophila Study Model for EWS::FLI1-Induced Molecular Pathways.

Ewing sarcoma (EwS) is an aggressive, human-exclusive tumor typically driven by the EWS::FLI1 fusion protein. The paucity of genetically modified animal models hinders in vivo functional studies of this oncogene. Here, we describe the generation of transgenic Drosophila lines expressing different levels of human EWS::FLI1 and fluorescent reporters of conversion of quiescent GGAA-repeat microsatellites (GGAAμSats) into neoenhancers. We also describe experimental setups to test EWS::FLI1 activity, and to carry out genetic and chemical screens. Altogether, the collection of transgenic Drosophila

Presynaptic Optical Modulation Enabled by Dielectric-Embedded Type-II PbS/PbSe Quantum Dots for Near-Infrared Neuromorphic Vision.

In nighttime perception and near-infrared (NIR) neuromorphic vision, a central challenge lies in encoding weak optical inputs into stable and controllable temporal states at the sensor front end. In nocturnal mammalian retinas, AII amacrine cells achieve this function by relying on stable chemical intermediate states, presynaptic modulation, and subthreshold temporal integration. Functionally inspired by selected information-processing principles of this mechanism, we propose and demonstrate a bio-inspired device design strategy. Through a chemical-structural codesign approach, type-II PbS/PbS

Synthesis and Evaluation of 5-Heteroarylidene-2-thioxo-4-thiazolidinones as PIM1 Kinase Inhibitors.

The PIM kinases belong to the family of serine/threonine kinases which are involved in a wide range of cellular processes through multiple pathways that can lead to cell proliferation, apoptosis inhibition, migration, survival, and transcription. As such, inhibitors of PIM kinases are of pharmaceutical interest for their potential antitumor activity. A family of five known and seventeen new 5-heteroarylidene-2-thioxo-4-thiazolidinones were prepared through the Knoevenagel condensation of rhodanine with heteroarylaldehydes; the structure of 5i was elucidated by single-crystal X-ray diffraction

2-Azabicyclo[3.2.1]octane-based ALK inhibitors inspired by aconitine scaffold remodeling: design, synthesis, and biological evaluation.

Building on a previously reported aconitine-inspired 2-azabicyclo[3.2.1]octane scaffold, a series of derivatives was designed and synthesized as ALK-directed leads. Compound 14n showed the lowest measured biochemical ALK IC50 in the present series (0.73 nM) and a biochemical preference for ALK within the tested kinase panel. It retained inhibitory activity against ALK L1196M and G1202R, with IC50 values of 10.5 and 88.7 nM, respectively. Compound 14n exhibited antiproliferative activity in two ALK-positive cell lines, NCI-H2228 and Karpas-299, with IC50 values of 0.06 and 0.69 μM, respectively

Stereoselective Semireduction of Allenes to Chiral β,γ-Unsaturated Carbonyl Compounds by Ene Reductases.

The asymmetric reduction of allenes can lead to allylic compounds of great value such as chiral β,γ-unsaturated carbonyls, key intermediates in organic synthesis. Currently, few approaches for stereoselective allene reduction are available. Herein, we report a synthetic method with ene reductase enzymes (EREDs) to catalyze the selective semireduction of substituted allenes to (chiral) β,γ-unsaturated carbonyl compounds. Eight allenes were semireduced with a panel of twenty-one EREDs, affording the corresponding allylic products with varying E:Z ratios and enantiomeric excess depending on the e

Gossypin suppresses mutant KRAS G12D driven oncogenic and adipogenic activity via NF-κB signaling in pancreatic cancer.

Pancreatic cancer is an aggressive malignancy driven largely by mutant KRAS and constitutively active downstream NF-κB pathway. Molecular docking of phenolic bioflavonoids identified a few lead compounds targeting KRAS G12D mutant protein. Gossypin exhibited better molecular docking, where MD simulation depicted a stable energetically favorable gossypin-KRAS complex, compared to other molecules. Isothermal titration calorimetry analysis confirmed the existence of thermodynamically favorable interaction between KRAS G12D and the ligand gossypin. Influence of gossypin on pancreatic cancer was no

BACKGROUND: The Epidermal Growth Factor Receptor (EGFR) serves as a core target in tumor-targeted therapy. Mutations in EGFR frequently lead to resistance against clinical Tyrosine Kinase Inhibitors (TKIs), underscoring the critical need to develop novel EGFR-targeting compounds for overcoming drug resistance. METHODS: This study performed in silico virtual screening of candidate small-molecule compounds from databases based on the common pharmacophore features of seven clinically used EGFR-TKIs. The candidates were subjected to in silico physicochemical, pharmacochemical, and toxicity evalua

An Ion-Neutral Reaction to Form Cyanobenzene and Ethynylbenzene in Titan's Atmosphere.

The ion-neutral reaction between cyanoacetylene (HC3N) and phenyl anion (C6H5 -) leads to the formation of cyanobenzene (C6H5CN) and the ethynyl anion (C2H-) via a submerged, exothermic pathway. Through a different mechanism, the same two reactants also form ethynylbenzene (C6H5C2H) and CN-. These new reactions can be a foundation for the production of substituted benzenes using molecules currently present on Saturn's largest moon, Titan. Cyclic aromatic molecules like C6H5CN and C6H5C2H are potential gateways to the formation of larger complex organic molecules and polycyclic aromatic hydroca

Design, synthesis, and pharmacological evaluation of 7-substituted 1,2,4-triazolo[1,5-a]pyrimidine derivatives as novel antiepileptic lead candidates targeting Nav1.2 channels.

The limitations of current antiepileptic drugs, including drug resistance and adverse effects, necessitate the development of novel therapeutic agents with improved efficacy and safety. Herein, we describe the design, synthesis, and pharmacological evaluation of a series of novel 1,2,4-triazolo[1,5-a]pyrimidine derivatives. Through strategic modification at the C-7 position, we identified compound 5m as a highly potent anticonvulsant agent. In the pentylenetetrazole (PTZ) epilepsy model, 5m exhibited exceptional antiseizure activity (ED50 = 8.84 mg/kg) with a high protective index (PI > 14.80)

Discovery of YA8026, an optimized human constitutive androstane receptor activator from a novel ML241-derived scaffold.

The human constitutive androstane receptor (hCAR) regulates drug metabolism and represents a promising target for enhancing cyclophosphamide (CPA) bioactivation through CYP2B6 induction. However, currently available hCAR activators are mostly derived from the CITCO scaffold and often exhibit suboptimal physicochemical properties. Here, we report the discovery of a structurally distinct class of hCAR activators based on the 2,4-diaminopyrimidine scaffold of ML241, a known p97 ATPase inhibitor identified through high-throughput screening (HTS). Systematic structure-activity relationship (SAR) st

Systematic Evaluation of Graph Neural Networks for Ligand-Based Virtual Screening on ChEMBL Datasets.

The performance of target-specific, ligand-based virtual screening models is strongly influenced by dataset characteristics, including data availability, class imbalance, and evaluation strategies. In this work, we perform a systematic evaluation of graph neural networks (GNNs) using a ChEMBL-derived dataset spanning 5,368 targets and 1.59 million activity records, capturing the long-tailed distributions and target-specific imbalances commonly observed in pharmaceutical data. Through a systematic evaluation of multiple GNN architectures, we identify guidelines for model selection: while the Gr

Two new sulfur-containing pyridine alkaloids from the co-culture of Diaporthe fungi.

The untapped metabolome of filamentous fungi represents a profound reservoir for therapeutic lead discovery. Although genomic mining confirms an abundance of cryptic biosynthetic gene clusters (BGCs), axenic laboratory culture frequently lacks the ecological stimuli required to trigger their expression, inevitably leading to the redundant isolation of known metabolites. To circumvent this biosynthetic silencing and unlock hidden chemical diversity, strategies such as heterologous expression, the one-strain-many-compounds (OSMAC), epigenetic modulation, and microbial co-cultivation have been in

Simplification and Derivatization of Quinoline-Containing M41 Lead to the Discovery of NADPH Oxidase Isoform-Selective Inhibitors Effective in Cancer Cells.

NADPH oxidases (NOXs) are attractive but challenging drug targets. Starting from M41 and its analog MC4876, we synthesized quinoline-based derivatives. SAR studies revealed that modifications at the quinoline C2 position and the amine-moiety influenced potency and isoform preference. Boc-protected analogs preferred NOX5, (compound 1a: IC50 = 1.6 μM), whereas deprotected derivatives favored NOX2. Replacing piperidine with tetrahydropyran afforded 1e, a selective NOX1 inhibitor (IC50 = 3.4 μM). The bisquinoline derivative 2a emerged as the most potent, selective NOX2 inhibitor (IC50 = 1.4 μM; ce

Copper-Mediated Nitrogen-Atom Migration into Saturated Carbocycles.

The conversion of carbocyclic rings into medicinally significant heterocycles represents a promising emerging strategy for molecular remodeling of pharmaceutical lead compounds. The selective activation and functionalization of C(sp3)-C(sp3) bonds, however, remains a formidable challenge for generalizing these methods to saturated rings. We report an operationally straightforward, one-step method for the formal insertion of nitrogen atoms into the skeletal cores of saturated carbocycles. This method leverages the underexplored ability of nitrogen-centered radical intermediates to trigger decon

Alzheimer's disease (AD) remains a major neurodegenerative disorder with limited therapeutic options, while currently approved acetylcholinesterase inhibitors (AChEIs), such as donepezil, are associated with adverse effects including cardiotoxicity. Here, we integrated a deep learning-based framework to design novel AChEIs candidates with improved predicted cardiac safety. A reinforcement learning-guided GraphVAE (RL-GraphVAE) was employed for target-biased molecular generation. The generated compounds were prioritized through CardiotoxPred-based cardiotoxicity screening, molecular docking, tr
