Targeting Oxidative Stress and Neuroinflammation: Analgesic Effects of 4-(phenylsulfanyl)butan-2-one on Chronic Constriction Injury-Induced Peripheral Neuropathic Pain in Rats.

Neuropathic pain arises from complex pathological processes involving oxidative stress and neuroinflammation. Excessive production of reactive oxygen species (ROS) contributes critically to the initiation and persistence of neuropathic pain following nerve injury and inflammatory insults, and antioxidant-based interventions have shown consistent efficacy across multiple preclinical models. This study investigated the analgesic effects of 4-(phenylsulfanyl)butan-2-one (4-PSB-2), a compound with known anti-inflammatory properties, using the chronic constriction injury (CCI) rat model. Intratheca

A solid magnesium zincate for hydrogen-methane evolution.

Hydrogen-methane blends are gaining attention as transitional fuels, but their safe storage and controlled release remain a challenge. Advancing this approach necessitates the development of compounds and materials capable of solid-state storage of mixed fuel gases. Molecular precursors capable of delivering both gases from a single source are not known. In this context, we report a low-molecular weight magnesium hydridomethylzincate, Mg[(CH3)ZnH3], a crystalline, hydride-rich organometallic compound that does not rely on bulky supporting ligands. Under solvent-free, anaerobic conditions, it r

Site-specific protein functionalization by late-stage reversible cysteine masking.

Site-selective functionalization strategies are essential for generating complex proteins for basic research and biomedical applications. However, achieving precise transformations across protein sequences remains a significant challenge and often requires protein engineering with orthogonal handles or sequence interventions to achieve selectivity. Here, we report a modular strategy for site-selective protein functionalization that enables the selective targeting of desired Cys residues through late-stage reversible thiol caging. This strategy involves splitting the protein sequence into two s

Crown Ether-Engineered Porous Liquid-Based Electrolytes Decouple Redox Kinetics and Stability in Zinc-Sulfur Batteries.

Aqueous zinc-sulfur batteries are promising for a new generation of safe and abundant energy storage devices, yet their realization is hindered by sluggish solid-state sulfur redox kinetics at the cathode and parasitic shuttling of iodine mediators in the electrolyte. Here, we demonstrate a path toward resolving these dilemmas by engineering a novel porous liquid-based electrolyte that decouples the essential catalytic activity from deleterious side reactions via microenvironment modulation. The macrocyclic molecular host leverages its intrinsic structural duality-dynamically coordinating a su

Structural and mechanistic insights into zinc-binding inhibitors of metallo-β-lactamases: From boronic acids to peptide mimics for combating carbapenem resistance.

The global dissemination of multidrug-resistant Gram-negative pathogens, primarily driven by the hydrolytic activity of metallo-β-lactamases (MBLs) such as the New Delhi metallo-β-lactamase-1 (NDM-1), necessitates the urgent development of effective inhibitors to resensitize these bacteria to β-lactam antibiotics. This review comprehensively summarizes recent crystallographic and enzymological insights into the rational design of MBL inhibitors. We critically evaluate key chemotypes equipped with diverse zinc-binding groups (ZBGs), including boronic acids, thiosemicarbazones, phosphonates, sul

In silico discovery and cellular validation of PBM-342: a novel PPAR-β/δ modulator for skeletal muscle differentiation.

Modulation of the peroxisome proliferator-activated receptor beta/delta (PPAR-β/δ) holds therapeutic potential for skeletal muscle regeneration and metabolism. However, the clinical development of classical agonists has been hampered by severe issues of non-specificity and metabolic toxicity. In this study, we implemented an in-silico strategy coupled with in-vitro functional validation to discover selective and safe modulators. Through a ligand-based virtual screening of the ZINC database, using the crystallographic agonist 'Compound 9' (PBM-0) as a structural template to favor isotype select

Tropolone and hinokitiol act against Staphylococcus aureus by concentration-dependent iron restriction or toxic iron overload.

UNLABELLED: Transition metal homeostasis is a critical component of bacterial physiology and thus represents a promising opportunity for antimicrobial intervention. Here, we demonstrate that the naturally occurring chelators tropolone (TRO) and hinokitiol (HNK) inhibit Staphylococcus aureus growth through concentration-dependent shifts in metal stress. Integrated metallomic and transcriptomic profiling revealed that low doses of both compounds promote toxic iron loading and a PerR-regulated oxidative stress response, consistent with iron-induced redox toxicity. However, at higher concentration

DeepFusion-ACSM: an interpretable hybrid ML-DL ensemble model for anticancer small-molecule activity prediction.

Cancer remains one of the leading causes of mortality worldwide, and the discovery of novel anticancer drugs continues to be a costly and time-consuming process. Although machine learning (ML) and deep learning (DL) have significantly advanced computational drug discovery, many existing predictive models rely on a single learning algorithm and often fail to generalize across diverse molecular descriptors, limiting their predictive reliability. In this study, we propose an interpretable hybrid ML-DL ensemble framework for the prediction of anticancer small molecules by integrating complementary

Bis(pyridinium) tetra-chlorido-zincate(II), [Hpy]2[ZnCl4].

Single crystals of the title compound, (C5H6N)2[ZnCl4] or [Hpy]2[ZnCl4], were grown from an aqueous solution of pyridinium chloride and zinc dichloride. The compound is isostructural with the corresponding cobalt compound and exhibits an almost tetra-hedral [ZnCl4]2- anion and two different pyridinium cations. The structure is described with respect to the bond lengths and angles of the anion and the π-π inter-actions between the cations, whilst mention is made of the N-H⋯Cl and C-H⋯Cl inter-actions already known from the cobalt compound.

Synergistic effect of zinc-chitosan co-modified rice husk biochar on nutrient retention and controlled release of fertilizer.

The key challenges in the application of biochar in slow-release fertilizers due to the defects of poor pores and few functional groups. In this study, a chitosan-modified biochar-based infiltration nitrogen fertilizer (ZBC@CS-BUF) was synthesized using zinc chloride-chitosan co-modified biochar (ZBC@CS) as a fertilizer carrier with molten urea. The results showed that ZnCl2 promoted the formation of micropores in biochar (SSA is 306.04 m2/g), and the introduction of chitosan increased the abundance of functional groups containing nitrogen (-NH2) and oxygen (-CO, -OH). The fitting results of t

Effect of inorganic retarders on setting, mechanical properties, and microstructure of fly ash-slag geopolymer mortars under hot-weather conditions.

Under hot-weather construction conditions, the rapid setting problem of geopolymer mortar as a sustainable material severely restricts its practical application. This study systematically investigated the retarding behavior of representative inorganic salts (ZnCl2, Na2SO4, BaCl2) in fly ash and slag-based geopolymer mortar at 30-50°C using multi-scale characterization. The results revealed three distinct retardation pathways: ion competition depletion (ZnCl2), precipitation equilibrium regulation (Na2SO4), and interfacial barrier-phase separation (BaCl2). Among the three, BaCl2 exhibited the s

Multifunctional epoxy/ZnO/GO superhydrophobic coatings for integrated marine protection and hydrodynamic drag reduction.

Marine biofouling limits the efficacy of epoxy (EP) based marine coatings due to their inherent hydrophilic properties and easy bio-adaptability, leading to loss of barrier performance and increased hydrodynamic drag. To overcome these constraints, the present work demonstrates a multifunctional EP-based antibiofouling coating modified with acetic acid (AA) and stearic acid (SA), containing functionally active zinc oxide (ZnO) and graphene oxide (GO) additives. Three coatings were fabricated, namely EP, EP/ZnO (EPZ), and EPZ/GO (EPZG). Surface chemical modification showed a shift in water cont

Novel photo-electro-Fenton process with natural air diffusion electrode coating recycled Zn-Mn oxides/C from spent Zn-Mn dry battery: fast generation and activation of H2O2.

The development of photo-electro-Fenton (PEF) technology could significantly enhance the efficiency of pollutants degradation. However, the high costs of efficient electrodes and catalysts for generating and activating hydrogen peroxide (H2O2) constrained its industrial application. To address these challenges, a natural air diffusion electrode (NADE) was introduced and coated with recycled Zn-Mn oxides/C catalysts from spent Zn-Mn dry battery to fabricate the recycled Zn-Mn/NADE (rZM/NADE) cathode, for fast generation and activation of H2O2. NADE could generate H2O2 by atmospheric oxygen with

Optimizing the Production of a Condensed Tannin-Based Bioflocculant for Toxic Metal Remediation.

Conventional water treatment typically employs chemical coagulants and flocculants, such as aluminum sulfate and iron salts. However, the intensive use of these products can lead to negative environmental impacts and pose significant health risks. In this context, bioflocculants derived from renewable sources, such as tannins extracted from açaí seeds (Euterpe oleracea), represent a promising alternative for sustainable production. This study investigated the efficiency of a cationic bioflocculant (BioTAC), developed from açaí seed tannins and chemically modified via the Mannich reaction, for

[Zinc homeostasis in plants: the role of ZIP, MTP and HMA transporters and chelating agents].

Zinc (Zn) is an essential micronutrient involved in numerous physiological processes in plants, serving catalytic, structural, and regulatory functions. Maintaining its appropriate concentration requires precise control of uptake, transport, distribution, and storage, as both Zn deficiency and excess lead to disruptions in plant growth and development. This paper presents the mechanisms regulating zinc homeostasis in plants, with a particular focus on the role of membrane transporters and chelating compounds. The ZIP, MTP, and HMA protein families play a key role, being responsible for cellula
