Functional validation and characterization of Slc39a8 genotype effects on ethanol consumption and related phenotypes.

Alcohol use disorders are heritable, and large-scale genome-wide association studies have recently identified novel genetic loci that are correlated with alcohol consumption and problematic alcohol use. One such gene is Slc39a8 , which encodes the ion transporter ZIP8, responsible for cellular import of zinc and other metals. Slc39a8 is highly pleiotropic, and a common missense variant has been associated with decreased problematic alcohol use. Here, we used Slc39a8 heterozygous (HET) knockout mice and their wild type (WT) littermates to characterize the effects of reduced gene function on two

Lysosomal Expansion Compartments Mediate Zinc and Copper Homeostasis in Caenorhabditis elegans.

Zinc is an essential transition metal that participates in many biological processes. In C. elegans, excess zinc is stored in lysosomes in intestinal cells; this process involves increasing the expression of the zinc transporter CDF-2 and remodeling of lysosomes characterized by an increase in the volume of the expansion compartment. To determine if this is a more general property, we investigated other metals. Here we report that lysosomes are remodeled in response to excess copper, manganese, and cadmium, with each metal causing an increase in the volume of the expansion compartment. Mutants

Fe(III) oxide respiration by a Desulfovibrio species lacking canonical outer-membrane electron transfer systems.

Desulfovibrio species play an important role in the extracellular reduction of Fe(III) oxides and other metals in diverse environments, but their extracellular electron transfer (EET) mechanisms are poorly understood. We found that Desulfovibrio strain JY grew with H2 or lactate as the electron donor and poorly crystalline Fe(III) oxide as the electron acceptor, demonstrating the capacity to conserve energy from Fe(III) respiration despite the absence of genes for porin-cytochrome conduits, outer-surface cytochromes, or electrically conductive pili. Washed cell suspensions reduced Fe(III) citr

Gold Contact Sensitization, Clinical Characteristics and Associations With Autoimmune Disease: A Retrospective Cohort Study With Registry Linkage.

BACKGROUND: Gold sensitization is common, but its clinical and immunologic significance remains unclear. European data from consecutively patch tested patients are scarce. Animal models suggest that gold can induce systemic autoimmunity, but studies in humans are limited. OBJECTIVES: To characterize gold sodium thiosulphate (GST) patch test positive patients, including co-sensitization patterns to other metals and determine whether gold sensitization is linked to autoimmune diseases. METHODS: A retrospective register-based study including 3436 dermatitis patients consecutively patch tested b

From Population to Individual: Bulk and Single-Cell Analysis Reveal Fe Bioavailability, Limitation Thresholds, and Biological Responses for Marine Diatom Chaetoceros debilis.

In marine ecosystems, Fe fundamentally regulates algal productivity and drives biogeochemical cycling in the hydrosphere, yet its environmental controls and cellular responses remain insufficiently resolved, particularly at the single-cell level. Herein, we combined bulk and single-cell analysis to systematically investigate Fe bioaccumulation process in the marine diatom Chaetoceros debilis under varying Fe species, initial pH, and concentration conditions. Fe bioavailability followed the order Fe-ethylenediaminetetraacetate > Fe-deferoxamine B > Ferrihydrite and decreased with declining init

Mechanistic insights for the preparation of improved metal-fluoride based radioimaging agents.

Metal-fluoride complexes incorporating fluorine-18 are advancing PET imaging, and creating new opportunities for Total Body PET and theranostics, but the factors governing metal-chelator selection, radiofluorination mechanisms, and hydroxide competition remain unclear. Although several Al-18F tracers based on NOTA and NODA chelators have reached clinical use, mechanistic understanding is limited and has hindered both extension to other metals and chelators and understanding of the relative merits of "one-pot" (radio)fluorination protocols vs. late-stage (radio)fluorination of pre-formed comple

Optimized colorimetric detection of cobalt ions (Co2+) using alliin-Ag-Au nanoparticles.

Cobalt is a vital trace element that is necessary for biological functions but poses toxicity risks at higher levels, necessitating its accurate detection in environmental and occupational contexts. Traditional cobalt detection methods, including spectroscopy and chromatography, often lack affordability and environmental friendliness. This study explores the development of a highly sensitive, selective, and eco-friendly nanosensor for detecting cobalt ions (Co2+) using bimetallic silver-gold nanoparticles (Ag-Au NPs) synthesized with alliin extracted from garlic. The properties of the synthesi

Evaluation of the new role of the invasive species (Procambarus clarkii) as an environmental indicator in polluted watersheds.

Procambarus clarkii represents an invasive species of crayfish common in the rivers of the Iberian Peninsula, which replaced a previous invasive species of crayfish (Austropotamobius pallipes) and is now being replaced by a new invasive species (Pacifastacus leniusculus) in northern rivers. Leaving aside the ecological problems that these invasive species can cause, these organisms can be excellent candidates to act as environmental indicators in pollution processes that involves the interface water-sediment. In this work, the potential role of Procambarus clarkii as a bioindicator of pollutio

Developmental toxicity of metals: Three decades of insights into mechanisms, epigenetics, and transgenerational inheritance.

Since the early 1990s, considerable progress has been made in understanding the teratogenic and embryotoxic effects of metals in mammalian systems. The present updated review synthesizes over three decades of research findings, examining the developmental toxicity of metals across four categories: (a) metals of greatest toxicological significance (arsenic, cadmium, lead, mercury, and uranium), (b) essential trace metals (chromium, cobalt, manganese, selenium, and zinc), (c) other metals with evident biological interest (nickel and vanadium), and (d) metals of pharmacological interest (aluminum

[Pollution Characteristics and Potential Risks of Heavy Metals in Reservoir Basin Sediments of the Hilly Areas in the Upper Yangtze River].

The upper Yangtze River is an important water conservation area, and its hilly terrain is prone to soil erosion, which causes pollutants such as heavy metals to migrate into the water and then accumulate in the reservoir sediments. It may pose a potential threat to drinking water safety and water ecological health. In this study, the Quanmin Reservoir Basin of Sichuan Province located in the upper reaches of the Yangtze River was selected, and the basic sediment physicochemical properties as well as the spatial distributions and occurrence characteristics of heavy metals (including Cr, Mn, Ni,

Waste-to-waste valorization: Sustainable palladium recovery from real spent catalytic converter leachates using chicken feathers.

The increasing demand for platinum group metals (PGM), mainly driven by their extensive use in automotive catalytic converters, has heightened the need for efficient and green recovery methods. Simultaneously, the poultry industry generates significant amounts of keratin-rich chicken feathers (CF), which are commonly disposed of by incineration or landfilling, raising environmental concerns. This study explores the use of CF as a sustainable and cost-effective biosorbent for the selective recovery of Pd from synthetic multimetallic solutions and HCl-based spent autocatalytic converters leachat

Heavy metal blood concentrations in smelters and chimney sweepers in Norway.

BACKGROUND: Smelter workers and chimney sweepers may be exposed to heavy metals via particulate matter through airways and skin uptake. We hypothesized that these workers have higher blood concentrations of arsenic, cadmium, chromium, lead, and mercury compared to an office-worker group. Furthermore, we hypothesized that their concentrations increased cross-week despite the use of properly fitted respiratory protective equipment (RPE). Our aim was to uncover workers' general heavy metal concentrations and cross-week changes to provide further insight into industrial occupational exposure. MET

Histidines promote zinc over cadmium binding to the single type 4 metallothionein from Great Millet (Sorghum bicolor).

Zinc and cadmium share similar chemical properties; however, while zinc is an indispensable microelement involved in several physiological processes, cadmium is highly toxic. Cadmium toxicity results at least to some extent from replacing zinc (and other metals) from their active sites in enzymes and other proteins. This highlights why the correct population of metalloproteins with metals is crucial for proper cellular metabolism. In the face of growing demand for food, both in terms of quantity and quality, a rapid development of crop cultivars containing a higher amount of bioavailable zinc

Envelope destabilization by AcrAB2NodT overexpression links antibiotic resistance to metal sensitivity in Caulobacter vibrioides.

Antibiotic (AB) resistance often arises through mutations that alter AB targets, reduce membrane permeability, or increase efflux activity, yet its physiological costs remain poorly understood. In Caulobacter vibrioides, a ΔtipR mutant that constitutively upregulates the RND efflux pump AcrAB2NodT exhibits heightened sensitivity to copper (Cu), revealing a trade-off linked to excessive pump expression. Deleting acrAB2nodT or impairing its transport activity restored cell envelope integrity and, ultimately, Cu resistance, uncoupling the effects of pump abundance from active efflux. The ΔtipR mu

Molecular dynamics study of catalytic H2/O2 recombination on Pd, Pt, Cu, Ag, and Au nanoclusters using the universal neural network potential.

The H2/O2 catalytic recombination reaction, essential for safely utilizing hydrogen energy, was simulated rapidly and accurately using the molecular dynamics (MD) simulation with a machine-learning potential known as the 'universal neural network potential' (UNNP). The catalytic activities of Pd, Pt, Cu, Ag, and Au nanoclusters were systematically investigated under a 100 bar H2/O2 atmosphere at 500 K. This approach enables the investigation of the entirely dynamic catalytic reaction, rather than assembling a static configuration at 0 K. Throughout the simulation, the ability to catalyze H2O f

An Effective Method for Isolating Ergothioneine Analogues for Use in Peptide Synthesis: 2-Thiohistidine and Fmoc-2-Thiohistidine.

Here, we report on an improved synthesis of the novel amino acid 2-thiohistidine and its Fmoc-derivative through the addition of sodium chloride to the isolation steps of each synthetic procedure. These compounds must be synthesized in either water or a mixture of water/organic solvent and have proven difficult to isolate by traditional methods. We serendipitously discovered that the addition of sodium chloride caused each compound to precipitate from the solution during the workup. Since 2-thiohistidine is the amino acid analogue of the natural product ergothioneine, we hypothesize that sodiu

Effect of hydrotreating on the pyrolysis oil composition: GC-MS studies.

The processing of waste tires is a significant challenge. The pyrolysis of waste tires offers a sustainable pathway to produce fuels, addressing both waste management and energy recovery challenges. However, due to the presence of a large amount of unsaturated, aromatic hydrocarbons and sulfur compounds, their direct use as fuel is not possible. Catalysts based on Pd, Pt, and other metals are commonly used for the hydrotreating of pyrolysis oil for producing alternative transportation fuels. The effect of hydrotreating with the commercial Pt/Al2O3 and NiMo/Al2O3 catalysts on the chemical compo

Spatial Variability of Metal Concentrations in Seaweeds, Mussels and Surface Sediments in Gemlik Bay: an Extensive Assessment of Contamination Sources and Associated Health Risks.

Surface sediments, mussels, and macroalgae were collected to investigate metal accumulation, distribution, pollution levels, and sources from Gemlik Bay in the southeastern Sea of Marmara. The elements Cu, Zn, V, Co, Cr, Pb, Ni, and Mn were determined in macroalgae (Ulva lactuca Linnaeus,1753), (Ulva intestinalis, Linnaeus, 1753) and mussel (Mytilus galloprovincialis, Lamarck, 1819) samples, while Al and Fe were also determined in sediment samples in addition to these metals. Element accumulation and metal contamination levels were determined, and health risks from mussel and macroalgae consum

Acid Resistant and Highly Silver Selective Membranes Based on Charged Nanoporous 2D-Materials.

The amount of waste electrical and electronic equipment (WEEE) is growing rapidly, yet less than a quarter of it is recycled. These "urban mines" contain valuable and critical metals that must be recovered. Therefore, developing more sustainable and energy-efficient separation techniques is essential. Membrane technology has demonstrated this capability, particularly those based on 2D materials, due to their atomic thickness and the ability to precisely tune pore size. This report introduces a novel nanofiltration membrane fabricated from charged, fully exfoliated, nanoporous 2D nanosheets. De

Facet-dependent Antimony Isotope Fractionation during Sb(V) Adsorption on Hematite.

The widely distributed hematite in soil and water environments can affect the environmental fate of antimony (Sb) through adsorption, yet little is known about the role played by various hematite facets in Sb isotopic fractionation. This study investigated the Sb(V) adsorption mechanism on the (001) and (012) facets of hematite and the associated isotope fractionation mechanism, combining batch adsorption experiments, extended X-ray absorption fine structure (EXAFS), and density functional theory (DFT) calculations. The results indicate that the adsorption capacity of the (012) facet is strong
