Intravesical mycobacteria reshape bladder and gut microbiota in a murine bladder cancer model.

The bladder microbiota may influence tumor progression and responses to intravesical therapies, yet the effects of Mycobacterium bovis BCG instillations on bladder and gut microbiota remain unexplored. This study evaluates, for the first time, the impact of M. bovis BCG and Mycobacterium brumae, a non-pathogenic immunomodulator, on microbiota composition in bladder tissue and cecal stool of healthy and tumor-bearing mice. Mycobacterial instillations increased gut microbial richness but reduced Actinobacteria. In tumor-bearing mice, Muribaculaceae and Bacteroidaceae were enriched, while M. bovi

Rv2656c: A Potential Candidate Antigen Associated with Latent Tuberculosis Infection.

Background/Objectives: Several subunit vaccines for tuberculosis (TB), such as MVA85A and H4:IC31, have not demonstrated ideal protective efficacy in clinical trials, which may be attributed to their limited antigenic profile and lack of effective latency-associated antigens. In this study, we combined bioinformatics with experimental validation to screen for latency-associated antigens that have immune-protective effects. Methods: Highly expressed antigens were identified from models related to latent infections, such as hypoxia and nutritional starvation. Their physicochemical properties and

Systemic Granulomatous Mycobacteriosis in Orbiculate Batfish (Platax orbicularis) Associated with Mycobacterium marinum-like Organism in an Aquarium in South Korea.

In this report, we describe systemic granulomatous mycobacteriosis in an orbiculate batfish from an aquarium in South Korea. Gross examination of the deceased fish showed multifocal nodular lesions in multiple internal organs including the gills, spleen, and kidney. Histopathological analysis demonstrated severe chronic systemic granulomatous inflammation, and Ziehl-Neelsen staining highlighted abundant intralesional acid-fast bacilli. Molecular analysis based on partial sequencing of the 16S ribosomal RNA (rRNA) and heat shock protein 65 (hsp65) genes showed that the detected organism was mos

Detection of nontuberculous mycobacteria in milk and nasal swab of cattle tested positive by tuberculin skin test, Interferon-γ release assay and MycoPac dual lateral flow assay.

The Single Intradermal Comparative Cervical Tuberculin (SICCT) and Interferon-γ release assay (IGRA) test represent a major antemortem diagnostic test for preliminary detection of bovine tuberculosis in a herd. Very few studies have focused on identifying the mycobacterial species in the milk and nasal swab of SICCT- and IGRA-positive animals. This study was carried out to identify the presence of Mycobacterium in the milk and nasal swab from SICCT- and IGRA-positive cows in a herd in West Bengal, India. Out of the study population of 250 lactating animals, 12 (4.8%) cows were positive for the

Mycobacterium tuberculosis urine ESAT-6 antigen homogeneous immunoassay through spatial confinement and cascade material amplification.

Tuberculosis (TB) remains one of the leading causes of death from infectious diseases. Early secreted antigenic target-6 (ESAT-6) is a specific biomarker for active Mycobacterium TB infection; however, its ultrasensitive detection in urine specimens remains challenging. Herein, a homogeneous immunoassay for ESAT-6 detection integrating spatial confinement-mediated DNA conformational switching with cascade signal amplification is reported. The system employs antibody-bound triplet DNA molecules stabilized by C-Ag+-C coordination; specific antigen binding induces conformational changes that disr

Delineating the potential of bioenergetics in drug discovery against Mycobacteriumabscessus.

Pulmonary and skin infections caused by Mycobacterium abscessus, a rapidly growing non-tuberculous mycobacterium (NTM), are becoming increasingly prevalent worldwide. Managing these infections is particularly challenging due to the bacterium's inherent resistance to many antibiotics, often resulting in extended treatment periods or therapeutic failure. With few effective drugs available, identifying new therapeutic targets is critical. A promising approach involves targeting the pathogen's energy metabolism, specifically the electron transport chain (ETC), which plays a vital role in its survi

Advancing national preparedness for the introduction of novel tuberculosis vaccines in South Africa: policy, evidence, and system readiness insights from a national workshop.

Tuberculosis (TB) remains a leading cause of morbidity and mortality in South Africa despite substantial progress in diagnosis and treatment. Several candidate vaccines targeting adolescents and adults are now in advanced clinical development, raising the prospect that a new TB vaccine could become available as early as 2029. However, translating clinical trial success into population-level impact will require early policy planning, health system preparedness, and coordinated national action. To support these efforts, the South African National Department of Health and the World Health Organiz

Guidance for chest-CT in children and adults with cystic fibrosis: A European perspective (part 2: special situations/respiratory complications).

This manuscript covers guidance proposed by The European Cystic Fibrosis Society Clinical Trials Network (ECFS-CTN) for the use of chest computed tomography (CT) scans in special situations such as haemoptysis, aspergillus-related lung disease, non-tuberculous mycobacterial (NTM) infection and pneumothorax in people with CF. Guidance development followed a three-step process. The most important questions were first identified via anonymous online survey, then a comprehensive literature search was made to support a first set of statements. They were finally validated by a Delphi process to obta

Bone tuberculosis in the distal metaphysis of a child's radius: case report.

INTRODUCTION: tuberculosis is an important public health problem associated with patient's morbidity and increased public expenses in Brazil. The disease typically affects the lungs, but can affect other sites as well. Extrapulmonary manifestations are estimated to occur in approximately 20% of patients with tuberculosis. The diagnosis is often difficult and neglected, which can generate important consequences for the patients. Radiological images, bacteriological exams and histopathology of the tissue allowing the isolation of pathogens and identification of the bacillus are part of the diagn

Genome-wide association study reveals a novel tuberculosis susceptibility locus in multiple East Asian and European populations.

BACKGROUND: Tuberculosis (TB) continues to be a leading cause of morbidity and mortality worldwide. Although numerous genome-wide association studies (GWAS) have explored TB susceptibility across various ethnic groups, multi-population replication of findings has been very limited, particularly outside the HLA region, and a significant portion of TB heritability remains unexplained. METHODS: We conducted GWAS in the Singapore Chinese and Vietnamese, followed by a comprehensive meta-analysis incorporating 4 independent East Asian datasets (N = 11,841 cases; N = 197,373 controls). The transfera

Comparison of active and passive case finding on diagnostic delay and treatment outcomes in older patients with pulmonary tuberculosis: a cohort study using real-world data.

BACKGROUND: Tuberculosis (TB) case detection in China predominantly relies on passive case finding (PCF). To evaluate the effectiveness of active case finding (ACF), a retrospective analysis of routine surveillance data from ACF programs implemented among individuals aged 65 years and older in Quzhou from 2020 to 2022 was conducted. However, Comparative studies evaluating these two strategies remain limited. This study analyzes the basic characteristics of ACF and PCF, evaluates the impact of ACF on diagnostic delay and treatment outcomes, and provides evidence-based support for optimizing TB

Drug-Resistant Tuberculous Spondylitis Treated with Bedaquiline-Containing Regimens in South Korea: Two Case Reports.

Background: South Korea continues to report a considerable burden of drug-resistant tuberculosis (TB). Bedaquiline-containing regimens are recommended for multidrug-resistant pulmonary TB, but evidence regarding the optimal treatment for extrapulmonary manifestations such as spinal TB remains limited. Case presentation: Herein, we report two cases of drug-resistant tuberculous spondylitis that were successfully managed using bedaquiline-containing regimens. Case 1 involved a 67-year-old man who was receiving chemotherapy for lymphoma and had a history of spinal TB treated 20 years earlier. The

Bactericidal Mechanism of Chlorous Acid Water in the Inactivation of Non-Tuberculous Mycobacteria.

The global prevalence of pulmonary infections caused by non-tuberculous Mycobacteria (NTM), particularly the Mycobacterium avium complex (MAC), is increasing. Since NTM are ubiquitous in moist environments and resistant to standard disinfectants, this study evaluated the efficacy of chlorous acid water (CAW) against them. CAW demonstrated superior sanitizing effects compared to sodium hypochlorite (NaClO), efficiently inactivating NTM at 100 mg/L free available chlorine even in the presence of organic matter, where 1000 mg/L NaClO failed. Instead, subcellular fractionation and protein analysis

Multilevel Determinants of Tuberculosis Treatment Interruption in Rural South Africa: Insights from Primary Healthcare Nurses.

BACKGROUND: Tuberculosis (TB) remains a major public health challenge globally, particularly in high-burden countries such as South Africa. Treatment interruption is a critical barrier to effective TB control, contributing to poor treatment outcomes, increased risk of multidrug-resistant tuberculosis (MDR-TB), and continued community transmission. Understanding the determinants of treatment interruption in rural healthcare settings is essential for strengthening TB programme implementation. METHODS: This qualitative study explored the factors influencing TB treatment interruption from the per

Infectious Spondylodiscitis of Bacterial Causes in Adults: Epidemiology, Pathophysiology, Diagnostic and Treatment Challenges.

Spinal infections in general, and infectious spondylodiscitis in particular, are increasingly diagnosed in the Western world, in recent decades. This rise in incidence is associated with an ageing population and with an increased availability of accurate diagnostic modalities. Even so, due to the non-specific nature of clinical manifestations, and of the implicated blood and serum markers, there is a risk of underdiagnosis or misdiagnosis of the disease in its initial stages. Ionizing radiation methods, such as plain radiography (X-ray) and computed tomography (CT), are also not reliable in th

Genetic Characterization of First-Line Drug-Resistance Mutations in Multidrug-Resistant Mycobacterium tuberculosis.

BACKGROUND: Resistance to first-line anti-tuberculosis drugs in Mycobacterium tuberculosis represents a significant public health challenge, particularly in high-burden tuberculosis (TB) settings such as Pakistan, where multidrug-resistant (MDR) forms further complicate disease control efforts. Drug resistance is primarily associated with mutations in rpoB, inhA, katG, embA, embB, embC, and pncA. The emergence of novel, region-specific variants underscores the urgent need for integrating genomic surveillance into routine TB diagnostics and regional control programs. This study aimed to identif
