α-Mangostin attenuates Pseudomonas aeruginosa virulence by targeting PqsR and inhibiting PQS-mediated quorum sensing.

Due to the increasing global antibiotic resistance spread, novel antibacterial strategies targeting virulence factors instead of bacterial growth are clinically crucial. One promising approach involves interfering with bacterial quorum-sensing systems, which regulate virulence and enhance adaptability. To this end, exploring quorum sensing inhibitors from Chinese medicine monomers have gained attention. The key quorum-sensing systems of Pseudomonas aeruginosa (las, rhl, and pqs) control multiple virulence factors and contribute to pathogenicity. This study screened a library of Chinese medicin

Heart Transplantation and Donation After Circulatory Death in Children. A Review of the Technological, Logistical and Ethical Framework.

Metal solubilization from discarded electrical material and electronic devices (e-waste) using the bioleaching capabilities of bacterial cells is highly effective. However, gaps in understanding about the microbiological processes involved in the bioleaching reaction leads to less efficient metal solubilization in large-scale e-waste processing. In this study, bacterial species belonging to the genera Acidithiobacillus and Pseudomonas were used to leach copper and gold from discarded printed circuit boards (PCB). Through modulation of the cell-to-cell communication system in these bacteria, ph

Allelopathic Effects of Sorghum/Sorghum-Sudangrass Hybrids Against Rotylenchulus reniformis.

Reniform nematode (Rotylenchulus reniformis) is a damaging and difficult-to-manage pest on many agricultural crops. Sorghum and sorghum-sudangrass hybrids (SSgHs) have shown potential in managing plant-parasitic nematodes by releasing toxic hydrogen cyanide gas through the hydrolysis of dhurrin, a cyanogenic glucoside found in leaf epidermal cells. The objectives of this study were to evaluate the effect of different SSgH varieties and their age on the suppression of R. reniformis and to quantify their dhurrin contents. Shoot biomass of seven SSgH varieties was harvested at 1, 2, and 3 months

Constitutive chemical defense mechanism of inner bark of Sorbus commixta against Trametes versicolor.

Tree bark is a crucial tissue that defends tree stems from invasions by microorganisms. However, our understanding of the constitutive chemical defense mechanisms of the tree barks remains limited. Our group recently discovered that the inner bark of Sorbus commixta Hedl. exhibited potent inhibitory effects on the growth of the white-rot fungus, Trametes versicolor (Linnaeus) Lloyd. It was hypothesized that this growth suppression was due to hydrogen cyanide (HCN) originating from cyanogenic glycosides such as amygdalin and prunasin, which are secondary metabolites in the inner bark of S. comm

Computational exploration of the electrochemical oxidation mechanism of thiocyanate catalyzed by cobalt-phthalocyanines.

In this study, we focused on the mechanism of the electrocatalytic oxidation of thiocyanate, which in traditional electrodes typically requires high overpotentials. As models for reducing these overpotentials and catalyzing the reaction, we used a set of modified cobalt phthalocyanines (CoPc), known as electrocatalysts. Using DFT calculations, we explored how modifications to CoPc by adding electron-donating and withdrawing groups and the coordination of 4-amino thiophenol impact the oxidation process. The reaction mechanism for the electrooxidation of thiocyanate has remained elusive, where o

The GacS/GacA two-component system strongly regulates antimicrobial competition mechanisms of Pseudomonas fluorescens MFE01 strain.

UNLABELLED: Pseudomonas fluorescens MFE01 is an environmental bacterium characterized by an hyperactive type 6 secretion system (T6SS) and a strong emission of volatile organic compounds (VOCs). In a previous study, a transposition mutant, 3H5, exhibited an inactive T6SS and altered VOC emission. In 3H5, the interruption of trpE gene by the transposon was insufficient to explain these phenotypes. To determine the actual impact of this insertion, a comparative transcriptomic analysis was performed on the two-component system GacS/GacA, known to influence numerous phenotypes in Pseudomonas. The

Anticancer Attributes and Multifaceted Pharmacological Implications of Laetrile and Amygdalin.

Laetrile, known as vitamin B17, is often used interchangeably with amygdalin. Laetrile is a semi-synthesis product of amygdalin, whereas amygdalin is a naturally occurring substance in many plants. Both compounds have a nitrile functional group that, when activated by the intestinal enzyme β-glucosidases, releases hydrogen cyanide. The two compounds have been considered for a long time as alternative therapy for cancer treatment however, findings available in the literature are discordant on the real efficacy of laetrile/amygdalin for the treatment of cancer, often highlighting a negative bene

Interplay of Tetrel, Hydrogen, and Halogen Bonds in F3GeOCl and HCN Complexes: A Comprehensive Theoretical Study of Dimers, Trimers, and Tetramers.

This study investigates the nature and interplay of noncovalent interactions (NCIs)─tetrel bonds (TB), hydrogen bonds (HB), and halogen bonds (XB)─in molecular assemblies formed between trifluorogermyl hypochlorite (F3GeOCl) and hydrogen cyanide (HCN). Using a combination of high-level computational methods, we explored the geometric, energetic, and electronic properties of dimers, trimers, and tetramers formed in different molar ratios of interacting reagents. Various analyses reveal a significant cooperativity between TB and HB, which mutually reinforce each other, while XB interactions are

Numerical investigation of the effects of hydrogen/ammonia addition to n-heptane combustion and emissions for a dual injection compression-ignition engine.

This research employs a detailed chemical mechanism to investigate the spraying of hydrogen and ammonia through dual-channel injector nozzles in a compression ignition engine. This method allows n-heptane, as the diesel primary reference fuel, and hydrogen/ammonia, as carbon-neutral fumigants, to be directly injected into the combustion chamber. The main objective is to predict how substituting a portion of the primary carbon-based fuel with these synthetic fuels affects engine performance and exhaust emissions. Numerical analysis is conducted to evaluate the impact of combustion parameters var

Improved Selectivity of CeMnOx/Pt@SnO2 Laminated MOS Sensor for Hydrogen Cyanide Under Temperature Dynamic Modulation.

Poor selectivity is one of the main bottlenecks restricting the development of metal oxide semiconductor (MOS) sensors. In this paper, using hydrogen cyanide (HCN) as the target gas, CeMnOx as the catalytic layer material and Pt@SnO2 as the gas-sensitive layer material, we have proposed a scheme to improve the selectivity of a catalytic layer/gas-sensitive layer-laminated MOS sensor under dynamic temperature modulation. We tested HCN and 12 kinds of battlefield environment simulation gases, and the results showed that the CeMnOx/Pt@SnO2 sensor, under the condition of temperature dynamic modula

Thermal Characterisation and Toxicity Profile of Potential Drugs from a Class of Disubstituted Heterofused Triazinones.

The thermal characterisation and toxicity profile of a class of disubstituted heterofused triazinones were revealed in this article for the first time. The thermal behaviour of molecules 1-12 was investigated by means of TG and DSC analyses performed in an air atmosphere and by the coupled TG/FTIR technique in a nitrogen atmosphere. The heating atmosphere affects both the stability of compounds and the degradation mechanism. A two-step degradation occurs in air, while a one-step degradation takes place in nitrogen, both preceded by a melting process. Compound 3 shows the highest thermal stabil

Quorum sensing regulation by the nitrogen phosphotransferase system in Pseudomonas aeruginosa.

In the opportunistic pathogen Pseudomonas aeruginosa, the nitrogen-related phosphotransferase system (PTSNtr) influences multiple virulence behaviors. The PTSNtr is comprised of three enzymes: first PtsP, then the PtsO phosphocarrier, and the final PtsN phosphoacceptor. We previously showed that ptsP inactivation increases LasI-LasR quorum sensing, a system by which P. aeruginosa regulates genes in response to population density. LasI synthesizes a diffusible autoinducer that binds and activates the LasR receptor, which activates a feedback loop by increasing lasI expression. In this study, we
