We have studied receptor-mediated generation of an activator of soluble guanylate cyclase in cultured mouse neuroblastoma cells (clone N1E-115) by ESR/spin trapping spectroscopy. A spin adduct was detected during the activation of muscarinic receptors by carbamylcholine in the presence of the spin trap 3,5-dibromo 4-nitrosobenzene sulphonate (DBNBS). The spin adduct does not correspond to that originating from the free radical nitric oxide or hydroxylamine. A pivotal biochemical event in the thyroid physiology is identified unravelling a superoxide anion radical-mediated activation of iodine into an active I.- form, which could be the intermediate that is incorporated onto tyrosine. This active iodine species gives fairly stable spin-adducts with PBN that could be characterized using EPR spectroscopy. Thus, a long-lasting puzzle regarding the iodine intermediate formed before iodination of tyrosine seems to be solved.

N-Tert-butyl-alpha-phenylnitrone (PBN), a spin trap agent, reduced ischemic hippocampal damage and the associated locomotor hyperactivity in Mongolian gerbils. Cerebral ischemia was induced in unanesthetized gerbils by a bilateral 5 min occlusion of the carotid arteries. An epidemiological study is presented to demonstrate, in addition to the viral aetiology, the influence of airpollution and various weather conditions to the incidence of croup. By means of a special statistic tool (baseline data curves) we could demonstrate that rapid changes in airpollutants (especially NO and NO2) are followed by increased occurrence of croup. In addition, the quotient NO2/NO seems to correlate directly to the O3 concentration. The noticeable influence of airpollution as well as changing climatic conditions demonstrate the multiform aetiology of croup.

Human platelet soluble guanylate cyclase activity was studied with respect to the function of its heme-containing regulatory subunit. The effects of oxygen on the metabolism of lipophilic nitroxide and the membrane structure of microsomes were investigated with an ESR spectrometer equipped with a hand-made gas-permeable sampling tube. The half life of gas change in the membrane was confirmed to be 48 s by the line-width of the ESR signal. The signal intensity of lipophilic nitroxide in microsomal membranes decreased with NADPH under N2 and increased under O2. The change of signal intensity was reversible, suggesting that lipophilic nitroxide can be used as an indicator of the redox state in membranes for in vivo ESR imaging.

Local fire in an enclosed environment was simulated, and the composition and toxicity of compounds outgassed by the polymers were investigated. Highly toxic compounds, e.g. prussic acid, carbon monoxide, benzene, nitrogen oxides, formaldehyde, were identified. The toxic hazard of the products depended on the mass and chemical composition of the burnt materials, time of exposure and enclosure volume. In the case of a local fire the enclosed atmosphere should be monitored by rapid diagnostic methods using indicator tubes.

The in vivo measurement of nitroxide radicals in whole mouse was carried out by L-band ESR spectroscopy. Spectra were successively observed in hepatic and bladder domains of female mice after intravenous administration of spin-labeled compounds (CPROXYL or TEMPOL).
