Tissue Preservation and FFPE Samples: Optimized Nucleic Acids Isolation in Ewing Sarcoma.

Different methods have been described for the preservation of biopsy or resection samples. In routine pathology, the cheapest and most used method is fixation of samples in formalin and embedding in paraffin (FFPE samples). This method preserves tissue samples for a very long time and is suitable for several specialized techniques, such as fluorescence in situ hybridization (FISH) and immunohistochemistry, the latter being the most frequent and often the only additional method used for the establishment of the final diagnosis. However, in light of the growing need for next-generation sequencin

Preparation of Postmortem Human Formalin-Fixed Paraffin-Embedded Frontal Cortex Tissue for Profiling Pyramidal Neurons Via Digital Spatial Profiling.

Digital spatial profiling (DSP) facilitates RNA sequencing (RNA-seq) of small populations of cells, specifically ~20 pyramidal neurons in postmortem formalin-fixed paraffin-embedded (FFPE) human brain tissue. This approach enables precise expression profiling via RNA-seq data linked to spatially characterized samples utilizing tissue-bound probes. We present a comprehensive protocol for manual slide preparation using the GeoMx DSP system as preliminary steps to spatial characterization of lamina-specific pyramidal neurons in human postmortem Brodmann area 9 (BA9) frontal cortex brain tissue.

A Formaldehyde-Free, Soy Protein-Based Adhesive with P-N-Si Synergistic Flame Retardancy for Plywood Application.

To overcome the flammability and formaldehyde-emission problems associated with plywood, a novel soy protein-based intumescent flame-retardant adhesive with phosphorus-nitrogen-silicon (P-N-Si) synergism (C@Si@APP) was developed in this study. C@Si@APP was initially prepared by coating silane-modified ammonium polyphosphate with collagen. Subsequently, C@Si@APP was combined with lignin and adenine and incorporated into a soy protein, collagen, and isocyanate (MDI)-based adhesive matrix to fabricate a flame-retardant adhesive for eucalyptus plywood. Cone calorimeter (CONE) analysis demonstrated

Evolutionary Inferences Based on Thoracic Limb Muscles of South American Coati (Nasua nasua): Brachium and Craniolateral Antebrachium.

The South American coati (Nasua nasua) is a mammal of the order Carnivora and family Procyonidae, characterized by scansorial locomotion. Accordingly, several anatomical and functional studies examined its thoracic limbs. Nevertheless, the anatomical descriptions and terminology differ among those studies, and none considered the evolutionary derivations of the brachium and antebrachium muscles based on topology, innervation and anatomical variants. Therefore, the present study aimed to infer the muscle derivations and reconstruct muscle maps of the brachial and craniolateral antebrachial musc

Simultaneous valorization of nitrate and formaldehyde waste streams to hexamethylenetetramine via photo-electro-thermo-catalysis.

To remediate nitrate (NO3-) pollution and recover resources from formaldehyde wastewater, this work presents a waste valorization strategy integrating photoelectrocatalytic nitrate reduction (PEC NO3RR) to ammonia (NH3) with downstream chemical upgrading. Herein, a hollow MoO2@CuFe2O4@MnFe2O4 Z-scheme-Schottky dual heterojunction nanoreactor array is developed to spatially separate proton and nitrate adsorption sites. The MoO2@CuFe2O4@MnFe2O4 improves photogenerated charge separation and retaining robust redox capability, while the oxygen vacancy-rich MoO2 layer exhibits favorable photothermal

In Vivo Chemical Cross-Linking Coupled with Reversed-Phase Co-Fractionation Mass Spectrometry Enables Protein Complex Analysis.

Understanding protein-protein interactions (PPIs) and the architecture of protein complexes is essential for elucidating cellular functions and drug action mechanisms. Co-fractionation mass spectrometry (CF-MS) has emerged as an effective tool for analyzing protein complexes under near-native conditions, however, its resolution and structural fidelity are often compromised by complex dissociation during biochemical processing. Here, we present an analytical strategy that integrates in vivo formaldehyde cross-linking with reversed-phase cofractionation mass spectrometry (XL-RP-CF-MS) to identif

A Fast-Slow-Fast Strategy for Exploring Three-Component Reactions of Formaldehyde.

To explore a fast/slow reactivity partitioning strategy for the design of multicomponent reactions (MCRs), the fast-slow-fast (FSF) approach was applied to formaldehyde-based systems. The assembly procedure of substrates to the MCR product was designed to be a fast-slow-fast (FSF) cascade process. The most reactive one among all the substrates was converted rapidly to a substable substance (the fast first step), which can be slowly transformed to a highly reactive intermediate (the slow second step), and finally, the intermediate was trapped by another substrate to form the MCR product (the fa

Rhodium-Catalyzed Three-Component C(sp3)-H Activation of 8-Methyl Quinolines with Diazo Compounds and Formaldehyde/Formaldimine to Access Dual C(sp3)-C(sp3) Bonds.

The development of transition-metal-catalyzed three-component C(sp3)-H activation represents one of the most efficient strategies for the rapid construction of complex molecular architectures. Particularly, leveraging this strategy for the direct formation of dual C(sp3)-C(sp3) bonds offers distinct advantages, whereas it remains largely challenging and underdeveloped. Herein, we report a rhodium-catalyzed three-component C(sp3)-H activation of 8-methyl quinolines with diazo compounds and formaldehyde/formaldimine. This protocol not only provides a novel route for the construction of dual C(sp

Rongalite-Promoted One-Pot Reductive Cyclization Strategy for the Construction of Pyrrolo[1,2-α]quinoxalines from 1-(2-Nitrophenyl)pyrroles.

A transition-metal-free strategy for the synthesis of pyrrolo[1,2-α]quinoxalines and 7-methylindolo[1,2-α]quinoxaline from 1-(2-nitrophenyl)-1H-pyrrole or 3-methyl-1-(2-nitrophenyl)-1H-indole using rongalite have been developed. In this transformation, rongalite serves a dual function, acting as both a reducing agent and a one-carbon (C1) synthon. The method offers an alternative approach to C-N bond formation. Furthermore, to investigate the reaction mechanism, several control experiments were performed, including radical scavenger studies, formaldehyde detection experiments, and reduction st

Compartment-specific oxylipin-derived carbonyl profiles in spinach chloroplasts.

The first compartment-resolved map of chloroplast carbonyls was established, revealing distinct carbonyl compositions and patterns of accumulation in thylakoids, envelopes, and stroma. Oxylipin-derived carbonyls, diverse aldehydes and ketones generated from oxygenated lipids, function as key cytotoxic agents and signaling molecules in oxidative stress responses of plants. However, the intracellular and sub-organellar sites at which individual carbonyl species occur remain poorly defined. Here, we analyzed carbonyl compositions in spinach (Spinacia oleracea L.) leaves, isolated chloroplasts, an

Toward Cadaveric Proteomics: A Theoretical Framework for Mass Spectrometry Analysis of Embalmed Human Tissues.

Mass spectrometry-based proteomics has been applied with considerable success to the analysis of formalin-fixed, paraffin-embedded (FFPE) tissues, and this success raises the question of whether the same methods can be applied to the embalmed human cadaveric specimens that are used in anatomical education and research. In this review, a theoretical framework is presented for the application of proteomic methods to embalmed cadaveric tissues, in which the feasibility of the approach is evaluated on the basis of the methods that have been established for FFPE tissue, and the critical gaps in the

Reactive molecular dynamics simulations reveal hydrogen fate steering and the catalytic promotion mechanism of MgH2 nanoparticles in methanol combustion.

Magnesium hydride nanoparticles are promising combustion promoters with poorly understood atomistic mechanisms. Using ReaxFF molecular dynamics simulations, we elucidate the mechanism by which MgH2 nanoparticles promote methanol combustion. MgH2 reduces the apparent activation energy of methanol combustion by 58% and accelerates methanol decomposition. H-element tracking reveals that MgH2 draws methyl hydrogen from the formaldehyde intermediate and the abstracted hydrogen atoms are oxidized to form H2O, with 73.1% of MgH2-derived hydrogen forming H2O instead of H2, raising the CO2/CO ratio fro

Selective Methylation of Phenol with CO2/H2 to o-Cresol and 2,6-Xylenol.

Employing CO2/H2 as a sustainable methylating agent offers a transformative strategy for advancing conventional methylation processes, yet achieving controllable C-C coupling with high efficiency remains a formidable challenge. Here, we report a novel tandem process that integrates CO2 hydrogenation with phenol methylation over a Zr-doped In2O3 solid-solution catalyst. The optimized In7Zr3Ox catalyst exhibits >95% selectivity toward ortho-methylation with tunable product distribution, ranging from 92% o-cresol at 16% conversion to 86% 2,6-xylenol at 97% conversion, while maintaining stable ope

A single archaeon couples long-chain fatty acid oxidation to methanogenesis.

Anaerobic degradation of complex organic compounds to methane and carbon dioxide is common in the deep subsurface biosphere and has long been considered to require syntrophic cooperation, with methanogens confined to the terminal use of simple C1 and C2 substrates. Thermodynamic considerations, multi-omics analyses and microcosm experiments suggest that archaeal lineages are capable of methanogenic degradation of complex substrates1-6. However, experimental validation in pure culture was lacking. Here we report the isolation of a thermophilic archaeon, Methanoglobus dolicholipophagus AA-75, af

Enhanced alkene productivity in methanol-to-hydrocarbons conversion using a secondary catalyst component and hydrogen co-feeds.

The methanol-to-hydrocarbons conversion over unidimensional 10-membered ring zeolites offers an attractive route to produce C3-C5 alkenes, central building blocks for the production of sustainable aviation fuels. However, the application of such zeolites is limited by a rapid deactivation due to coke formation. We present a novel catalyst design concept to significantly enhance coke resistance, achieved by physically mixing the zeolite with a secondary catalyst component exhibiting hydrogenation activity and operating under near ambient-pressure hydrogen co-feeds. Among the tested materials, P

Evaluation of alternative pretreatment solutions for the Masson's trichrome stain: replacements for Bouin's solution.

Masson's trichrome stain is a standard stain used to differentiate extracellular matrix components, primarily collagen, in histologic tissue samples. The classic pretreatment solution, Bouin's solution, contains formaldehyde, picric acid, and acetic acid. The chemical components introduce safety concerns encompassing toxicity, respiratory, explosive hazards, and environmental hazards. Three pretreatment solutions (citric acid solutions pH 4 and pH 6, and Gram's iodine) as well as a no pretreatment condition, were assayed alongside Bouin's solution on various human and animal tissue samples by

Tobacco Composition, Nicotine, and Carbonyl Emissions Across IQOS Product Generations: ILUMA/TEREA Compared with IQOS 2.4/HEETS.

INTRODUCTION: Heated tobacco products (HTPs) are marketed as reduced-exposure alternatives to combustible cigarettes. In its initial iteration, Philip Morris International's IQOS used an electrically powered metal blade to heat a reconstituted tobacco plug (HEETS). The current iteration, the IQOS ILUMA, uses electromagnetic induction to heat a metal element embedded within the tobacco plug (TEREA). This change in heating architecture may involve altered emissions and tobacco filler constituents. METHODS: Mainstream aerosol emissions from Sienna- and Turquoise-flavored TEREA sticks used with t

Dual-phase formaldehyde detection via catalyst-triggered AIE in metal-coordinated supramolecular polymers.

It is essential to develop a simple method for detecting formaldehyde (FA) because of its harmful effects on human health. Herein, we present a novel metal-coordinated supramolecular polymer chemosensor utilizing a complex between the tetraphenylethylene (TPE) derivative (T4J) and Cu2+ for FA detection in both gaseous and aqueous phases. In this system, Cu(NO3)2 acts as both a linker and a promoter, enabling selective FA detection by T4J-Cu through an aggregation-induced emission (AIE) mechanism. This configuration affords a clear fluorescence turn-on response that can be recognized by the nak

Effects of Chemical Fixation on Stimulated Raman Scattering Microscopy.

Stimulated Raman scattering (SRS) microscopy enables label-free imaging of cells and tissues in their native state with chemical specificity. However, there are often experimental advantages of chemical fixation of samples prior to imaging, which can introduce perturbations that may alter the native state of the samples and possibly impact the SRS signal. In this study, we systematically characterize the effects of common fixatives (paraformaldehyde, formalin, glutaraldehyde, methanol, ethanol) on the preservation of cellular integrity and molecular composition in a HeLa cell model as observed

Laboratory Intercomparison of Multiple Ethylene Oxide Measurement Methods Under Simulated Near-Source Monitoring Conditions.

Ethylene oxide (EtO) is a widely used industrial chemical that poses potential health risks due to its carcinogenic and mutagenic properties. Accurate and reliable measurement of EtO concentrations is crucial for assessing public health risks and regulatory compliance. However, the lack of a standard reference method for EtO measurement poses significant challenges for researchers and analysts. In this study, we compared various EtO measurement methods under simulated, atmospherically relevant, laboratory conditions by investigating the impact of water vapor, potentially interfering gases (inc
