Decorating SrTiO3 With Cu@Pt Core-Shell Cocatalysts to Promote the Selective Thermally Assisted Photocatalytic Methanol Dehydrogenation to Formaldehyde.

Photocatalytic methanol conversion, a process of particular interest in the context of hydrogen generation, also holds promise as a sustainable approach to chemical synthesis of value-added products, including formaldehyde. In this study, Cu-core/Pt-shell modified SrTiO3 photocatalysts are demonstrated to enhance the thermally assisted photocatalytic methanol dehydrogenation in the gas phase by 50% compared to Pt/SrTiO3 photocatalysts prepared by conventional photodeposition, while maintaining a high formaldehyde selectivity. Cu-core/Pt-shell structures were prepared by means of galvanic displ

Protein engineering of formolase for the synthesis of 1,3-dihydroxyacetone from formaldehyde.

Formaldehyde is an attractive one-carbon building block for sustainable biomanufacturing, but efficient conversion of formaldehyde into higher-value chemicals remains challenging. Formolases, a class of engineered thiamine pyrophosphate-dependent enzymes, catalyze the condensation of formaldehyde to produce glycolaldehyde or 1,3-dihydroxyacetone (DHA), yet their catalytic performance is often insufficient for practical application. This chapter presents a workflow for engineering a benzoylformate decarboxylase (BFD)-derived formolase through a channel-modulating helix strategy. The protocol co

Use of dihydroxyacetone synthase for the assimilation of methanol in E. coli.

Dihydroxyacetone synthase (Das) is a key enzyme of the xylulose monophosphate (XuMP) pathway, catalysing the assimilation of formaldehyde in methylotrophic yeasts. In this study, we first performed a systematic screening of twenty-five Das variants in combination with a methanol dehydrogenase (Mdh) from Acinetobacter gerneri to identify the Das variants supporting the most efficient methanol assimilation.13C-methanol labelling experiments revealed marked differences in 13C incorporation into central carbon metabolism among the various Das variants and enabled the identification of the Pichia a

Indoor Air Pollutants of PM2.5, Carbon Monoxide, Nicotine, and Carcinogens in Waterpipe Bars.

INTRODUCTION: Waterpipe smoking has become popular in East Asia and other regions. However, few studies have investigated the indoor air quality, including the presence of carcinogens, in waterpipe bars (WBs). This study aimed to compare indoor air quality between WBs and non-WBs in Hong Kong, focusing on differences observed during weekdays and weekends. METHODS: Air samples from 8 WBs and 6 non-WBs were collected to investigate levels of fine particulate matter (PM2.5), carbon monoxide (CO), nicotine, volatile organic compounds (VOCs), and polycyclic aromatic hydrocarbons (PAHs) in the ambi

Mapping of RBP-Mediated RNA-RNA Interactome with RIC-Seq.

RNA-RNA interactions tend to be mediated by RNA-binding proteins (RBPs) and are involved in every step of gene regulation. Faithfully mapping RBP-mediated intra- and intermolecular RNA-RNA interactions is a prerequisite for simultaneously inferring noncoding RNA structures and targets. We present a recently developed RIC-seq (RNA in situ conformation sequencing) technology for global profiling of RBP-mediated in situ RNA-RNA interactions. Our approach fixes RBP-mediated RNA-RNA interactions in living cells with formaldehyde for subsequent permeabilization, micrococcal nuclease fragmentation, p

MCS-Stain: Boosting FFPE-to-HE Virtual Staining With Multiple Cell Semantics.

The diagnosis of cancer primarily relies on pathological slides stained with hematoxylin and eosin (HE). These slides are typically prepared from tissue samples that have been fixed in formalin and embedded in paraffin (FFPE). However, the traditional process of staining FFPE samples with HE is time-consuming and resource-intensive. Recent advances in virtual staining technologies, driven by digital pathology and generative models, offer a promising alternative. However, the blurred structures in FFPE images pose unique challenges to achieving high-quality FFPE-to-HE virtual staining. In this

Genetic polymorphisms of GSTM1 and GSTT1 genes: effects on susceptibility to formaldehyde-induced hematotoxicity.

Formaldehyde (FA) is a known human carcinogen for the upper respiratory tract. However, its hematotoxicity remains unclear. This study was performed to assess probable effects of FA on blood parameters and to determine the common polymorphisms in the detoxification enzymes GSTM1 and GSTT1 as biomarkers of susceptibility. Sixty-four workers with high occupational FA exposure and 57 non-exposed controls were studied. Complete blood count was performed. Also, GSTM1/GSTT1 genotypes were determined. After adjusting for potential confounding variables, FA exposure was only associated with the levels

A streamlined technique for in-depth ubiquitomics analysis of formalin-fixed paraffin-embedded tissues.

Formalin-fixed paraffin-embedded (FFPE) tissues offer advantages over fresh frozen (FrFr) tissues in terms of preserving tissue integrity, reducing long-term storage costs, and increasing sample availability, thus providing an invaluable resource for biomarker discovery and exploration of disease mechanisms. As a reversible and highly versatile post-translational modification (PTM), ubiquitination is involved in protein homeostasis and the regulation of almost all physiological and pathological processes, but ubiquitomics studies of FFPE tissues are lacking due to technical challenges. In this

Efficacy of glyoxal acid-free (GAF®) as a non-toxic fixative for fish gonads, gametes and larvae: widespread implications for laboratory studies, field sampling and applied aquaculture.

CONTEXT: Traditional fixatives such as buffered formaldehyde (BF) and paraformaldehyde (PFA) are widely used but raise health concerns because of their toxicity. Their use is increasingly subject to regulatory restrictions, calling for the development of non-toxic alternatives. Glyoxal acid-free (GAF®) has emerged as a potential substitute. AIMS: This study evaluated GAF® for preserving gonadal tissues, oocyte, sperm, and larvae of the European seabass (Dicentrarchus labrax). METHODS: GAF formulations - standard and an optimized, marine-adapted formulation - were compared to conventional fix

Construction of hierarchical 2D/2D few-layered MoS2/S-doped g-C3N4 heterojunctions for enhanced photocatalytic formaldehyde removal.

Modification of graphitic carbon nitride (g-C3N4) with semiconductor nanomaterials has been widely investigated as an effective method to enhance its photocatalytic activity. However, the construction of efficient g-C3N4-based heterojunction photocatalysts via an environmentally benign method remains critical. Herein, a two-dimensional (2D) few-layered MoS2/S-doped g-C3N4 hierarchical heterojunction was successfully synthesized by a simple strategy via combining small molecule-assisted liquid exfoliation, calcination and hydrothermal strategy, which was subsequently utilized for photocatalytic

Engineering oxygen vacancies in Au/MnO2 catalysts for complete formaldehyde removal at near-freezing temperatures.

Formaldehyde (HCHO) is a major indoor air pollutant that poses serious risks to human health, making its efficient removal a critical environmental concern. Catalytic oxidation at room and sub-ambient temperatures has attracted significant attention due to its potential to completely decompose HCHO into harmless CO2 and H2O. However, practical implementation remains challenging because of low reaction activation energy and limited catalyst performance at reduced temperatures. In this study, Au-loaded manganese oxide nanowire catalysts (x% Au/MnO2-NWs) were synthesized using a colloidal deposit

Is formalin-fixed paraffin embedded tissue good enough for studies of transmission electron microscopy of tumors? a comparative study with wet tissue.

Transmission electron microscopy (TEM) has historically been pivotal in diagnosing neoplasms with challenging histopathology. However, its use has declined due to cost and the advent of ancillary techniques like immunohistochemistry. This study compares the diagnostic utility of formalin-fixed paraffin-embedded (FFPE) tissue versus wet glutaraldehyde-paraformaldehyde fixed tissue (WT) for TEM in neoplasms. A retrospective analysis of 746 cases (2014-2023) from the National Cancer Institute of Mexico - Instituto Nacional de Cancerología de Mexico (INCan) was performed. Cases were divided into G

Optimization of Brain Tissue Preservation for Nucleic Acid Stability.

Postmortem human brain tissue is an important resource for brain research. Spatial transcriptomics is a novel technology that utilizes formalin-fixed, paraffin-embedded (FFPE) or cryosections, with the former having good cytoarchitecture but poor RNA quality and vice versa for frozen tissue. Sheep brains (n=16) were used to test various protocols that simulate the conditions around the preparation and dissemination of human postmortem FFPE and frozen brain tissue to optimize for future spatial transcriptomic work. FFPE and frozen tissues were investigated for RNA quality, while hematoxylin and

Genome sequence analysis of a highly formaldehyde-resistant Methylomicrobium alcaliphilum strain.

Formaldehyde is a highly reactive and cytotoxic compound, and its efficient detoxification is essential for cellular survival. This requirement is particularly critical in methylotrophic microorganisms, where formaldehyde serves as a central intermediate in methanol assimilation. Despite the toxicity of this compound, methylotrophic and methanotrophic bacteria have evolved mechanisms that allow limited tolerance, yet the isolation of highly resistant variants remains extremely challenging due to the intrinsic difficulties of cultivating and purifying these organisms. Here, we report the isolat

Formaldehyde detoxification by the moss Racomitrium japonicum.

As a pervasive indoor air pollutant, formaldehyde (FA) poses significant environmental and health risks. This study investigates the molecular and physiological mechanisms underlying the FA remediation potential of the moss Racomitrium japonicum. The moss employs a dual defense strategy: activation of antioxidant systems (SOD/POD/CAT enzymes and AsA-GSH cycle) under moderate FA stress, and metabolic detoxification via upregulation of key genes (FDH, formate dehydrogenase, and MS, malate synthase) that channel FA into the Calvin cycle and glyoxylate pathway. High FA concentrations induced oxida

Low-energy glycine formation and spectral masking in star-forming regions.

The presence of amino acids in comets and meteorites has long suggested that prebiotic molecules may have formed in space and contributed to the origins of life on Earth. Glycine, the simplest amino acid, has been identified in several extraterrestrial environments, although its detection in the interstellar medium, including prestellar cores and protostellar regions, remains elusive. Here, we investigate a novel catalytic pathway for glycine formation on silicate grains during relatively warm (> 150 K) stages of star formation. Using atomistic simulations, the feasibility of a Strecker-type s
