Gasotransmitters in cancers: multiple mechanisms, crosstalk, and therapeutic implications.

Nitric oxide, carbon monoxide, and hydrogen sulfide, once recognized primarily for their toxicity or vasoactive properties, have emerged as multifaceted signaling molecules in cancer biology. This review provides an update on current findings on how gasotransmitters support cancer processes. Gasotransmitters act through activation of soluble guanylate cyclase, redox-dependent post-translational modifications such as S-nitrosylation and persulfidation, mitochondrial metabolic regulation, calcium signaling, and crosstalk with pathways, and telomerase-related networks. In addition, the same gasot

NO- and H2S-derived post-translational modifications and stress resilience.

Nitric oxide (NO) and hydrogen sulfide (H2S) have emerged as key gaseous signaling molecules in plants, playing central roles in regulating responses to a myriad of abiotic stresses. A major mechanism through which these molecules exert their effects is via post-translational modifications (PTMs) of proteins, including tyrosine nitration, S-nitrosation, and persulfidation. The last two correspond to redox-based modifications (oxiPTMs) that can rapidly and reversibly alter protein activity, stability, localization, and interactions, thereby enabling dynamic cellular reprogramming under stress c

Evaluating operational efficiency and hydrogen sulfide mitigation in fluidized bed co-gasification of sewage sludge.

This study investigated the operational efficiency and in situ H2S mitigation of municipal sludge (MS) co-gasification with drinking water treatment sludge (DWTS) in a lab-scale fluidized-bed reactor. Gasification experiments were performed at 700-900 °C, ER 0.2-0.4, and 10-20 wt% DWTS addition. Increasing temperature and DWTS ratio enhanced syngas production and reduced tar yield. The highest syngas yield for MS alone was 79.10% at 900 °C and ER = 0.2, while co-gasification with 20 wt% DWTS increased syngas yield to 87.12% and reduced both heavy and light tars substantially. Under the same co

Dimethyl Phthalate Disrupts Sulfur Cycling and Triggers Toxic H2S Pollution in Sludge Anaerobic System.

Dimethyl phthalate (DMP), a crucial plastic additive widely employed, has been detected in sewers, sediments, and sewage sludge at elevated levels, particularly in regions where active sulfur cycling occurs. However, the effect of DMP on anaerobic sulfur conversion, especially toxic H2S production, has not been documented. This study aimed to explore the fate of DMP in anaerobic system and its influence on sulfur conversion through both batch and long-term tests using sludge as substrate. Experimental results showed that sludge can effectively adsorb DMP, which conformed to the Freundlich mode

Microporous Materials for Therapeutic Gas Delivery: A Focus on Metal-Organic Frameworks.

Gas therapy offers powerful clinical potential but remains limited by poor control of gas concentration, localization, and stability. Microporous materials, particularly metal-organic frameworks and zeolites, present a path toward precision gas therapy by allowing selective adsorption, protection, and controlled release of therapeutic gases such as oxygen, nitric oxide, carbon monoxide, and hydrogen sulfide. Their permanent porosity, tunable pore size, and active metal centers enable the design of materials that respond to biological or external stimuli with high selectivity. This review highl

Exogenous H2S suppresses traumatic stress-induced changes of HMGB1 and ERS in rat liver and lungs.

OBJECTIVES: Traumatic stress triggers systemic inflammation and organ injury via HMGB1 and endoplasmic reticulum stress (ERS), but whether H2S modulates this pathway remains unclear. This study aimed to investigate the effects of exogenous H2S (NaHS, an H2S donor) on traumatic stress-induced HMGB1 and ERS changes in rat liver and lungs. METHODS: Male SD rats underwent bilateral hindlimb crush. Sixty rats were assigned to sham, crush, NaHS pretreatment (14, 28, 56 μmol/kg), delayed NaHS (120 μmol/kg), and NaHS-only groups (n=6). Serum ALT/AST, histopathology, HMGB1 (ELISA/IHC), and ERS markers

HYPOXIC INTENSITY REVEALS DISTINCT CAROTID BODY O2 SENSING MECHANISMS.

Carotid bodies (CBs) sense arterial blood O2 levels. Hypoxemia activates the carotid sinus nerve (CSN) afferent activity to elicit cardiorespiratory reflexes to maintain tissue oxygenation. This brief review presents emerging evidence suggesting that CBs employ distinct O2-sensing mechanisms depending on the severity of hypoxia. During physiologically relevant moderate hypoxia (pO2 ~40 mmHg), glomus cells use heme oxygenase-2 (HO-2)/carbon monoxide (CO)/cystathionine-γ-lyase (CSE)/hydrogen sulfide (H2S) signaling pathway to stimulate CSN activity and breathing response to hypoxia. H2S enhances drought resistance by orchestrating alternative splicing and persulfidation of ETFQO.

Identifies ETFQO as a central target, showing H2S induces its alternative splicing to generate functionally distinct isoforms that are differentially persulfidated, coordinating stomatal and respiratory responses for drought adaptation. Hydrogen sulfide (H2S) is an important gasotransmitter known to enhance plant stress tolerance, but its specific role in reprogramming energy metabolism under drought remains poorly understood. Here, we demonstrate that electron transfer flavoprotein: ubiquinone oxidoreductase (ETFQO) functions as a significant target through which H2S enhances drought toleranc

Gas Sensing and Photocatalytic Applications of Graphene Oxide Annealed at Various Temperature.

Researchers employed the improved Hummers' method to synthesize graphene oxide (GO) from graphite, chosen for its faster kinetics, enhanced safety, and reduced toxicity. Thermal annealing was conducted at 0°C, 200°C, 300°C, and 400°C. The acidity of GO was minimized through repeated centrifugation, rinsing, and decanting. The resulting films were characterized for structural, morphological, elemental, optical, gas-sensing, and photocatalytic properties. FESEM micrographs revealed nanoflake distributions across the substrate, while EDS confirmed elemental composition with precise mass and atomi

CD28+ CD8+ Tem cells with a STAT1-dependent glucocorticoid receptor deficit contribute to steroid-refractory acute GVHD.

Steroid-refractory (SR) disease develops in a substantial fraction of patients with grade II to IV acute graft-versus-host disease (aGVHD) and is associated with poor long-term survival. Improved mechanistic insight is needed to identify reliable predictors of steroid resistance. We retrospectively profiled peripheral blood collected before glucocorticoid treatment from allogeneic hematopoietic cell transplantation recipients without aGVHD, with steroid-sensitive aGVHD, and with SR-aGVHD using an integrated multiomics approach, and validated our findings in an independent multicenter cohort. Sulfur and Selenium Modifications at Phosphorus Atom in Nucleoside Monophosphates, Activity and Potential Applications.

Nucleotides and nucleosides play an essential role in many cellular processes but have low physiological stability, which limits their usefulness. Nucleosides modified with chalcogen at the phosphorus atom are more stable in body fluids and tissues. They can act as activators or inhibitors in many processes, including signal transduction through receptors and intracellular signaling. Some of them are also used as drugs or prodrugs that can serve as potential therapeutics in cancer and other diseases. This review focuses primarily on the activity and potential application of the nucleoside mono

Naturally Occurring H2S-Donors in Vascular Diseases.

Hydrogen sulfide (H2S) is an endogenous gasotransmitter able to exert a pivotal role in different organs and systems, strongly influencing cardiovascular health. It is an endowed antioxidant, a vasorelaxant, and has cardioprotective properties, thanks to the activation of different classes of potassium channels and the interaction with several pathways including those involving sirtuins, nuclear factor erythroid 2-related factor 2 (Nrf2), nuclear factor kappa B (NF-κB), and nitric oxide (NO) production. Alterations in the homeostasis of H2S represent an etiopathogenetic factor in the onset and

The Role of Hydrogen Sulfide-Derived Perivascular Adipose Tissue in Vascular Diseases.

Perivascular adipose tissue (PVAT) is a metabolically active, endocrine organ that plays a crucial role in regulating blood vessel tone, endothelial function, vascular smooth muscle cell growth, and proliferation and contributes significantly to the onset and progression of cardiovascular diseases. In a healthy state, PVAT displays anticontractile, anti-inflammatory, and antioxidative properties, which are critical for maintaining vascular homeostasis. However, under certain pathophysiological conditions, PVAT exerts pro-contractile effects by decreasing the production of anticontractile and/o

Protein persulfidation: a ubiquitous modification regulating a broad spectrum of biological processes.

Hydrogen sulfide signaling occurs mainly through protein persulfidation, an important post-translational modification and a highly dynamic process in plants. Beyond enzyme activity, persulfidation affects protein localization, structure, and interactions. This review provides an overview of the mechanisms governing this modification and the pivotal role of the thioredoxin system mediating depersulfidation, which is essential for maintaining redox homeostasis. Proteomic studies in Arabidopsis thaliana and crop species reveal that protein persulfidation is a widespread post-translational modific

A FRET-based NCA fluorescent probe for highly sensitive and selective detection of hydrogen sulfide.

A Förster resonance energy transfer (FRET)-based fluorescent probe (NCA) was developed for highly sensitive and selective detection of hydrogen sulfide (H2S). In this design, coumarin (CA) serves as the fluorescent donor and nitrobenzoxadiazole (NB) acts as an energy acceptor and reactive site. Upon exposure to H2S, cleavage of the NB unit interrupts the FRET process and restores CA fluorescence, producing a 16-fold signal enhancement at 463 nm with a detection limit of 0.11 µM. Under optimized conditions (Britton-Robinson (BR) buffer, pH 8, 15 min), NCA enables rapid and selective detection o

Crosstalk Between H2S and Hormones: The Bilateral Relationship and Molecular Mechanisms.

Hydrogen sulfide (H2S) is increasingly recognized as a key gasotransmitter with diverse physiological roles across a range of organisms. Accumulating evidence has clearly shown that disruptions in H2S homeostasis are linked to various endocrine disorders. Hormones are crucial in regulating H2S metabolism and signaling, while H2S influences hormones by modulating their biosynthesis, secretion, transport, and signaling pathways. H2S and hormones function cooperatively to regulate cell survival, metabolism, reproduction, development, and stress responses, etc. The interaction between H2S and horm

Hydrogen Sulfide Consumption and Cell Bioenergetics in Human (Patho)physiology.

Hydrogen sulfide (H₂S), once considered merely a toxic gas, is now recognized as a key endogenous signaling molecule with profound effects on vascular tone, inflammation, and cytoprotection. Central to its physiological roles is a tightly regulated balance between synthesis and degradation. While much attention has been given to H₂S biosynthesis and its signaling functions, its catabolism - particularly through the mitochondrial sulfide oxidizing pathway - has received comparatively less pharmacological exploration. This pathway not only serves as a critical detoxification mechanism but also l
